Changes in Esterase Activity and Acetylcholinesterase Sensitivity of Insecticide-selected Strains of the Brown Planthopper(Nilaparvata lugens Stal) (저항성 벼멸구의 효소활성 변화에 관한 연구)

  • Acetylcholinesterase(AChE) and esterase activities as mechanisms of resistance to fenobucarb, carbofuran and diazinon in the insecticide-selected brown planthopper strains were investigated. Although there was no significant difference in AChE activity from suscept tible and resistant strains, AChE insensitivity was highly increased in the carbam없e insecticide-selected strains. On the other hand, esterase activity was moderately increa잃d in all the s selected strains. It is concluded that the cross-resistance and the level of resistance in the b brown planthopper can be explained by the combination of altered AChE and high esterase a activity, although a possible involvement of other factor(s) can not be excluded.

The Absorption and Metabolism of Fenobucarb and Carbofuran by Susceptible and Carbamate Insecticide-selected Strains of the Brown Planthopper (Nilaparvata lugens Stal) (Fenobucarb 및 Carbofuran의 저항성 벼멸구 체벽 투과량과 체내대사에 관한 연구)

  • Cuticular penetration and detoxication as mechanisms of resistance to the carbamate insecticides in fenobucarb-selected($R_{f}$) and carbofuran-selected($R_{c}$) strains of the brown planthopper (N. lugens Stal) were investigated. Rates of penetration were not significantly different in the susceptible and resistant strains. However, total amount of excretion of the $R_{f}$ and $R_{c}$ strains were much larger than that of the susceptible strain. Fenovucarb and carbofuran were in vivo metabolizd much faster in the $R_{f}$ strain than in the susceptible strain. OSBP(o-sec-butyl phenol) and 3-ketocarbofuran phenol were invitro the major metabolites of fenobucarb and carbofuran in the brown planthopper, respectively. Total amount of the two major metabolites were produced abotu 2 times larger in the $R_{f}$ and $R_{c}$ strains compared to the susceptible strain. OSBP and 3-ketocarbofuran phenol were not so toxic to the brown planthopper ($LD_{50}$ >100 $\mu\textrm{g}$/g hopper). Based on our data, detoxication plays a large role in resistance to fenobucarb and carbofuran in the resistant strain of BPH, although several resistance factors maybe involved.

Adsorption Characteristics of Organophosphorus and Carbamate Pesticides in Four Soils and the Evaluation for Their Leaching Potential Using Two Screening Models (유기인계 및 카바메이트계 농약의 토양흡착성과 간이선발모형을 이용한 용탈 잠재성 평가)

  • Soil adsorption study was carried out to define the mobility of pesticides and to evaluate leaching potential in soils. Nine pesticides including metolcarb, molinate, fenobucarb, isazofos, diazinon, fenitrothion, dimepiperate, parathion and chlorpyrifos-methyl were subjected to adsorption experiment for four types of soils, such as upland, paddy, forest and volcanic ash soil. Based on Koc values, metolcarb and molinate were classified as mobile, fenobucarb as mobile or moderately mobile isazofos as moderately mobile, diazinon, fenitrothin, dimepiperate, and parathion as slightly or moderately mobile and chlorpyrifos-methyl as slightly mobile. Two evaluation methods, Groundwater Ubiquity Score (GUS) index and standard indices of soil-chemical adsorption and biodegradation (half-life), were used for the estimation of pesticide leaching potential. Leachabilities of metolcarb, molinate and fenobucarb were evaluated as high, and isazofos, dimepiperate and diazinon as a little potential, while fenitrothion, parthion and chlorpyrifos-methyl showed very low leaching potential. The leaching potential of pesticides was determined on the basis of intrinsic properties of the pesticides and the soil properties. Among the soil properties, organic matter gave a great influence on the leachability in soil. Therefore, leachabilities of the pesticides used were expected less in Sineom soil with relatively higher organic matter than Gangseo, Jungdong and Yesan soil with lower organic matter.

Cross-resistance and Inheritance of Resistance in Laboratory-selected Strains of the Brown Planthopper(Nilaparvata lugens Stal) (벼멸구의 저항성 유발, 교차저항성 및 저항성 유전에 관한 연구)

  • Cross-resistance and inheritance of resistance in laboratory-selected strains of the brown planthopper to various types of the insecticides were investigated. The fenobucarb-selected ($R_{f}$), carbofuran-selected($R_{c}$), and diazinon-selected($R_{d}$) strains were 50.3, 49.2 and 5.8 times less sensitive to the corresponding insecticides than th susceptible strain. both $R_{f}$ and $R_{c}$ strains were highly resistant to the other carbamate insecticides, and moderately resistant to cypermethrin and deltamethrin, but nearly not resistant to fenvalerate and the organophosphorus insecticides except malathion and phenthate. Moderate resistance to malathion and phenthoate in the $R_{f}$ and $R_{c}$ strains was obtained at the rate of 13.0-12.0 and 8.5-7.5 times, respectively. The $R_{d}$ strain showed low levels of resistance to the carbamate, organophosphorus and pyrethroid insecticides, but negatively correlated cross-resistance to fenvalerate. Resistance of the brown planthopper to all the test insecticides was inherited by partially dominant autosomal factor(s).

Pesticide Multiresidues Analysis of Environmental-friendly Agricultural Soils by the Complex Cleanup Method of Accelerated Solvent Extraction (ASE) and Solid Phase Extraction (SPE) (ASE 및 SPE 복합정제법을 이용한 친환경농업토양의 다성분잔류농약 분석)

  • Fifty substances of pesticide were selected for analysis through the historical investigation of pesticides detected from environmental-friendly agricultural soil, and the environmental-friendly agricultural soils in Gyeongnam area were collected and then were accepted Anve (accelerated solvent extraction) and SPve (solid-phase extraction) as multiresidue extraction and clean up methods suitable to the soils. The pesticide residues were analyzed by using GC/vCD/NPD, HPLC/UV/FL, GC/MSD, or HPLC/MSD. 50 kinds of pesticides for the soils were an average of 95.5% from retrieval ratio of the 72 to 118% range, and the average of 3.0% for CV (%). Among 40 samples of soil, 20 components were detected from pesticide residues of 21 samples, and average amounts detected for these components were 0.035 for endosulfan, 0.043 for ethoprophos, 0.020 for chlorpyrifos, 0.023 for chlorfenapyr, 0.047 for flufenoxuron, 0.070 for fenvalerate, 0.266 for cypermethrin, 0.016 for lufenuron, 0.022 for bifenthrin, 0.025 for fenobucarb/BPMC, 0.043 for difenoconazole, 0.059 for fenarimol, 0.020 for kresoxim-methyl, 0.026 for tetraconazole, 0.039 for isoprothiolane, 0.017 for iprobenfos, 0.014 for nolrimol, 0.156 for fluquinconazole, 0.047 for tebuconazole, and 0.045 mg/kg for oxadiazon. Therefore it is infered that the establishment of pesticide residues limit for environmental-friendly agricultural soil is needed as soon as possible.
