Primordial follicles are formed prenatally in mammalian ovaries, and at birth they are fated to be activated to primary follicles, to be dormant, or to die. During the early stage of folliclulogenesis, the oocyte undergoes dynamic alterations in expression of numerous genes, which are regulated by transcription factors. Several germ-cell specific transcriptional regulators are critical for formation and maintenance of follicles. These transcriptional regulators include: Figla, Lhx8, Nobox, Sohlh1, and Sohlh2. A subset of these transcriptional regulators is mutated in women with ovarian insufficiency and infertility. Establishment of this oocyte pool is essential for fertility. This review focuses on these transcriptional regulators of female primordial follicles.
Transcription factors govern diverse aspects of cell growth and differentiation as major switches of gene expression. Etv5, a member of the E26 transformation-specific family of transcription factors, has many stories to share when it comes to reproduction. Etv5 deficient mice show complex infertility phenotypes both in males and females. In males, the infertility phenotype exhibited by Etv5 deficiency is sexually dimorphic, and it involves both somatic cells and germ cells. In $Etv5^{-/-}$ female mice, the problem is more complicated by hormonal involvement. This review synthesizes old and new information on this versatile transcription factor-from the inadvertent discovery of its role in the testes to its newly discovered role in maintaining spermatogonial stem cells.
The culture phenomenon, which the desire of self expression is noticeable and the diversity of gender identity is widely accepted, simply makes a difference in the lifestyle of one human being rather than the dichotomous classification of male and female. The mansumer power, which does not care too much about the money for the emotionally attached items, has enabled various fashion styles. After analyzing the diversified clothing behavior conducted by these people in connection with the social phenomenon, First, this shows the phenomenon of emotional value pursuit that finds pleasure over the clothing as the item of augmented reality is added to the concept of play, in which the real space referred to as garment and virtual space of playing the rock, paper and scissors game meet together within the augmented reality. Second, the convergence concept has enabled the coordination of new style by obscuring the area of design concept and this refers to the changes in design from the development of new items and transformation into double-style details. Third, the divergence that intensively provides specific use/convenience and specialized value shows a change in the fashion market from the phenomenon that admits various gene rations of culture and specifically, takes differently about the recognition of middle-aged males. Fourth, the variety seeking tendency receives attention as the value of future design together with the phenomenon of discriminative value pursuit. In the male fashion, it is linked to the collaboration with the design area and this tells that the fashion with the narcissistic cross-dressing and motto of neutral gender without being sided to male/female is rising.

Autism spectrum disorder (ASD) remains unexplained and untreated despite the high attention of research in recent years. Aside from its various characteristics is the baffling male preponderance over the female population. Using a validated animal model of ASD which is the telomerase reverse transcriptase overexpressing mice (TERT-tg), we conducted ASD-related behavioral assessments and protein expression experiments to mark the difference between male and females of this animal model. After statistically analyzing the results, we found significant effects of TERT overexpression in sociability, social novelty preference, anxiety, nest building, and electroseizure threshold in the males but not their female littermates. Along these differences are the male-specific increased expressions of postsynaptic proteins which are the NMDA and AMPA receptors in the prefrontal cortex. The vGluT1 presynaptic proteins, but not GAD, were upregulated in both sexes of TERT-tg mice, although it is more significantly pronounced in the male group. Here, we confirmed that the behavioral effect of TERT overexpression in mice was male-specific, suggesting that the aberration of this gene and its downstream pathways preferentially affect the functional development of the male brain, consistent with the male preponderance in ASD.
The milk of transgenic animals may provide an attractive vehicle for large-scale production of hEPO. Since glycosylation is cell type specific, recombinant human EPO (rhEPO) produced in different host cells contain different patterns of oligosaccharides, which could affect the biological functions. However, there have been no reports on the characteristics of rhEPO derived from milk of transgenic animals. To address this objective, several transgenic mice by using pWAPhEPO and/or pBC1hEPO expression vector were produced. However, 2 lines of pWAPhEPO founder female mouse died during late gestational day (day 18) before offspring could be obtained. They showed a severe splenomegaly, Unlike those of pWAPhEPO, mammary gland epithelial cells from biopsies of lactating pBC1hEPO transgenic mice had marked immunoreactivity to EPO and any activity was not detected in other tissues. The expression level of rhEPO is about 0.7% of mammary gland cellular total soluble proteins and an amount of 300~500 mg/L rhEPO is secreted into milk. Furthermore, the pBC1hEPO transgenic mice transmitted this character to their progeny in mendelian manner. In order to determine the extent of glycosylation variation, N-linked oligosaccharide structures present in the milk-derived rhEPO were characterized. Most of milk-derived rhEPO is fully glycosylated. the biological activity of milk-derived rhEPO was comparable to that of purified CHO-derived rhEPO, and milk-derived rhEPO showed relatively stable after freezing and thawing. Taken together, the results illustrate the potential of transgenic animals in the large-scale production of biopharmaceuticals.
The effects of the estrogenic compound 4-nonylphenol (4-NP) on vitellogenesis in juvenile olive flounder ($Paralichthys$$olivaceus$) and rockfish ($Sebastes$$schlegeli$) exposed continuously at 10, 50 and $100{\mu}g{\ell}^{-1}$ levels for 7 days were compared. The expression of VTG mRNA level and protein using specific probes were examined. The levels of plasma estradiol-17 ${\beta}(E_2)$ and testosterone (T) were assessed by radioimmunoassay (RIA). Plasma $E_2$ concentrations increased significantly in two female fish species exposed to $100{\mu}g{\ell}^{-1}$ of 4-NP over concentrations in control fish. Plasma T concentrations increased in $P.$$olivaceus$. Four days after exposure, the level of VTG mRNA expression increased in $P.$$olivaceus$ and $S.$$schlegeli$ exposed to $20{\mu}g{\ell}^{-1}$ of 4-NP. In addition, plasma VTG protein expression was seen in $P.$$olivaceus$ and $S.$$schlegeli$. In $S.$$schlegeli$ and $P.$$olivaceus$ exposed to 4-NP, the changes were noticed mainly in hepatocytic vaculation after 7 days of exposure. Thus, 4-NP may disrupt vitellogenesis in immature fish both directly and indirectly via disrupted steroidogenesis and liver pathology. Immature $S.$$schlegeli$ were the most sensitive to 4-NP exposure in vitellogenesis.
방사선 치료는 가장 일반적으로 사용되고 있는 항암치료로, 암 환자의 수 증가와 동반하여 방사선 활용도는 더욱 증가하고 있다. 방사선치료에 대한 환자들의 심리적 거부감과 산화적 스트레스, 저산소증, DNA 손상을 포함하여, 치료 후 나타나는 간 섬유화는 방사선 치료의 가장 큰 문제점으로 대두되고 있다. 간암의 경우 대부분의 환자들이 섬유화를 동반하고 있어서, 방사선에 의해 암세포가 제거된다 하더라도, 기존에 남아있는 손상 부위에서의 섬유화와 방사선 조사에 의한 정상조직의 섬유화는 간경변 유발 확률을 높이고 있다. 간 섬유화는 여러 간질환에서 흔히 관찰되는 증상으로, 간 섬유화 진행 기작의 규명은 만성질환으로의 진행을 억제할 수 있는 치료책 개발로 연계될 수 있지만, 아직까지 명확히 규명되지 않음에 따라, 섬유화를 억제할 수 있는 효과적인 방법이 없는 실정이다. 최근 간 섬유화 생성 및 진행에 대한 헤지호그의 역할이 밝혀지면서, 간암 및 만성질환으로의 진행을 이해하기 위한 연구대상으로 대두되고 있다. 헤지호그는 손상된 간에서 발현되어 보수과정에 기여하게 된다. 헤지호그의 발현은 간 손상 정도에 비례하여 발현되어, 간 줄기세포 및 간 성상세포의 증식을 조절한다. 또한, TGF-${\beta}1$와, EMT를 유도하고 근섬유화세포의 활성을 조절하여, 간 섬유화 진행에 중심적 역할을 한다. 방사선이 조사된 생쥐의 간에서도 헤지호그의 발현증가가 관찰되었고, 이에 따른 간 줄기세포의 증식 및 EMT 유도, 콜라겐의 축적이 관찰되었다. 또한, 방사선이 조사된 암컷 생쥐의 간에서 헤지호그의 차별적 증가는 줄기세포 및 섬유화 증대로 연결되었고, 이는 방사선 민감성에 대한 성별차이를 설명할 수 있는 생물학적 근거가 될 수 있다. 그러나, 방사선에 의해 유도된 간 섬유화에 대한 연구결과들이 헤지호그를 비롯하여, 표면적인 결과보고만 있을 뿐, 섬유화 진행에 대한 구체적 기작을 밝히지 못하고 있다. 여러 간 질환진행에서 규명된 헤지호그 작용에 대한 지식을 방사선에 의해 유도된 간 섬유화를 이해하기 위한 기초자료로 활용하고, 사람에서의 방사선 부작용과 유사한 동물모델의 정립 및 헤지호그 역할을 직접적으로 규명할 수 있는 추가적 연구을 통해서, 간 섬유화 형성 기작을 이해할 수 있는 연구결과가 기대된다.
레티놀 결합 단백질(retinol-binding protein, RBP)은 고등 척추동물에서 혈류를 통해 특이적으로 레티놀을 표적세포에 운반해 주는 중요한 역할을 한다. 우리나라의 연안에 서식하고 있으며 산업적으로 중요한 조피볼락(Sebastes schlegeli)을 대상으로 4-nonylphenol(NP)가 RBP mRNA 발현에 미치는 영향을 구명하기 위해 간으로부터 cDNA library를 제작하고 RBP 단편의 염기서열을 분석하였다. 분석된 RBP mRNA 서열로부터 아미노산 서열을 추정하여 다른 종의 RBP 아미노산 서열과 비교한 결과 Sparus aurata와는 80%, 무지개 송어(Oncorhynchus mykiss)와는 72%, 유럽 뱀장어(Anguilla anguilla)와는 78%의 높은 상동성을 보였다. 조피볼락에서 4-NP에 대한 RBP와 VTG mRNAs 발현에 미치는 영향을 northern blot 분석 방법에 의해 조사하였다. 4-NP를 10 mg/kg BW로 주사한 실험구에서는 암수 모두 VTG mRNA 발현에 영향을 주지 않았으나, RBP mRNA 발현은 수컷에서 48시간 후에 감소하였다. 4-NP를 25 mg/kg BW으로 주사한 실험구에서는 24시간 후에 암수 모두 VTG mRNA 발현이 증가하였으며, RBP mRNA 발현은 48시간 후에 암수 모두 감소하였다. 이들 결과로부터 조피볼락에서는 4-NP 등의 에스트로겐 유사물질이 RBP와 VTG 유전자 발현에 상반되는 효과를 유도함을 알 수 있었다.
콩이나 적포도주의 식물성 에스트로겐(phytoestrogen)은 건강에 부정적이기보다는 긍정적인 효과를 갖는 것으로 알려져 있는데, 특히 콩류 섭취는 유방암이나 골다공증, 그리고 심혈관계 질환 예방과 높은 상관관계가 있는 것으로 보인다. 그러나 콩류, 특히 그 주성분인 genistein(GS)이 상기한 긍정적인 효과 외에도 여성의 생식계에 잠재적으로 부정적인 영향을 미칠 가능성에 대한 의문이 계속되어왔다. 선행 연구에서 본 연구자들은 사춘기 전에 genistein(GS)을 경구 투여했을 때 암컷 흰쥐의 생식계가 활성화되어 사춘기 개시가 조기에 유도되고, 암컷 성체에 GS를 뇌실내로 미세주입했을 때 kisspeptin-GnRH 뉴런회로 활성화가 일어남을 관찰하였다. 본 연구에서는 사춘기 전 암컷 흰쥐에서의 시상하부 특이적인 GS 투여 효과와 이에 관여하는 에스트로겐 수용체 아형($ER{\alpha}$과 $ER{\beta}$)을 조사하였다. 사춘기 전암컷 흰쥐(SD strain, PND 30)를 마취시킨 후 GS(3.4 ${\mu}g$/animal)를 1회 뇌실내로 미세 주입하고, 2시간 후 희생시켰다. 시상하부내 생식조절 유전자 발현을 조사하기 위해, RNA를 추출한 후 semi-quantitative reverse transcription polymerasechain reaction(RT-PCR)을 시행하였다. GS 투여는 KiSS-1 유전자 발현의 상위조절자인 mTOR(1:$0.361{\pm}0.058$ AU, p<0.001)발현을 유의하게 감소시켰고, GnRH 분비의 상위조절자인 GAD67(1:$1.285{\pm}0.099$ AU, p<0.05) 발현을 유의하게 증가시켰다. GS 투여는 KiSS-1(1:$1.458{\pm}0.078$ AU, p<0.001) mRNA 수준을 유의하게 증가시켰지만, kisspeptin 수용체인 GPR-54(1:$1.29{\pm}0.08$ AU) mRNA 수준은 변화가 없었고, GnRH(1:$0.379{\pm}0.196$ AU, p<0.05)의 경우는 유의하게 감소시켰다. GS투여군에서 $ER{\alpha}$(1:$1.180{\pm}0.390$ AU) 발현은 대조군 대비 차이가 없었지만, $ER{\beta}$(1:$4.209{\pm}0.796$ AU, p<0.01) 발현은 유의하게 증가했다. 본 연구결과는 사춘기 전 암컷 흰쥐에서 GS의 단기 노출이 시상하부의 GnRH 조절시스템을 직접 변화시킴을 보여준 것으로, 이러한 GS의 시상하부 특이적 효과에 $ER{\beta}$ 경로가 관여함을 강력히 시사한다. 이는 잘 알려진 $ER{\beta}$ 경로를 매개로 하는 GS의 유방암 억제 효과와 일치한다.
어류 vitellogenin은 난황전구체로 난형성과정동안에 estradiol의 체내 순환에 의해 암컷의 간에서 생산되며 alkylphenol류와 같은 비이온성 계면활성제 등의 내분비계 장애물질에 의해 수컷에서도 생산된다. 물 환경의주요 생물종인 어류는 내분비계장애물질에 의해 암컷에서는 번식률 저하와 함께 수컷에서는 정소의 축소 및 이에 따른 암컷화가 관찰된다. 특히 수컷에서 내분비계 장애물질에 의해 유도된 vitellogenin의 생성을 이용하여 환경오염에 의해 유도된 생물체의 유전자 발현 변화 뿐만 아니라 이를 토대로 특정지역의 환경오염을 모니터링할 수 있다. 본 논문에서는 어류의 vitetlogenin를 이용하여 수환경의 내분비계장애물질의 검출과 환경오염모니터링을 위한 biomarker로서의 유용성을 검토하였다.
