Objectives: The present study aimed at investigating the effects of Schisandrae Fructus (SF) on menopause symptoms in ovariectomized mice. Methods: 6 weeks old female mice were ovariectomized (OVX), and the OVX mice were fed with a three doses of SF (25, 100, and 400 mg/kg, respectively) or $17{\beta}$-estradiol (E2, 0.5 mg/kg) 3 times a week for 6 weeks. Body weight was measured at every week and uterus weight was also measured at the end of experiment. Lipids and osteocalcin levels in serum were analyzed. Results: The uterine weight was not changed in SF treated group, while E2 intake increased the reduced uterine weight by OVX. Although Body weight was increased since two weeks after ovariectomy compared with sham operated group, there was no change in SF or E2 treated group compared with OVX. But LDL was significantly reduced in SF (400 mg/kg) or E2 fed mice and SF or E2 treatment decreased enhanced osteocalcin level by OVX. Conclusion: These results indicated that SF would be effective in the improvement of LDL level and osteoporosis on menopause.
We have investigated the efficacy of liposome encapsulated N-(phosphonacetyl)-L-aspartic acid (PALA) for the treatment of the C-26 murine colon tumor in Balb/c mice, and have compared it in this regard to free PALA. Healthy female Balb/c mice and C-26 tumor inoculated mice were randomized for the maximum tolerated dose (MTD) study and the in vivo therapy study, and the survival was measured after a single intraperitoneal injection of the drug. The maximum tolerated dose for intraperitoneally administered drug was found to be 750 mg/Kg for free PALA, and was greater than the maximum dose possible (150 mg/Kg) for PALA encapsulated in both DSPC and DSPG liposomes. When drug was administered one day after tumor implantation, 150 mg/Kg of PALA in DSPG liposomes increased the percentage of tumor bearing mice surviving at day 36 from 8% (buffer control) to 88%. In contrast, 150 mg/Kg free PALA increased the day 36 surviving percentage to only 25%. A 150 mg/Kg dose of PALA in DSPC liposomes increased the surviving percentage to 50%, while a 75 mg/Kg dose of PALA in sterically stabilized liposomes increased the surviving percentage to 78%. These results show that PALA in negatively charged or sterically stabilized liposomes can exhibit considerably greater potency than free PALA in C-26 tumor bearing mice.
Acute toxic effects of the Woo Whang Chung Shim Won suspension and pills were studied with the doses of 60, 74, 90, and 110 ml/kg in mice. In this study, we daily examined number of deaths, clinical signs, body weights, and pathological findings for 7 days after administration of Woo Whang Chung Shim Won. All mice given the highest dose (110ml/kg) died at 24 hour after administration of Sam-Sung Woo Whang Chung Shim Won suspension (Byunbang, SS-BS and Wonbang, SS-WS), one male and two female mice given 90 ml/kg dose of SS-BS died, and a few mice given middle dose (74 ml/kg) of SS-WS died. In control group (110 ml/kg) treated with vehicle (SS-BS and SS-WS), three out of 5 males and two out of 5 females mice died during the study. However, in animals treated with Kwang-Dong Woo Whang Chung Shim Won suspension (KD-S) and pill (KD-P), deaths were not observed. In the clinical signs, increase of drooling and decrease of spontaneous motor activities were observed in the highest dose group (110 ml/kg). No significant dose-related change in body weight was observed. The results suggest that the toxic effect of SS-BS and SS-WS may be atttributed to the solution for the Woo Whang Chung Shim Won suspension.

Purpose : We hope to evaluate the effectiveness of Citri Reticulatae Viride Pericarpium for the bronchial asthma using assesment on the vascular endothelial growth factor after Citri Reticulatae Viride Pericarpium was intravenously administered OVA-sensitized and -challenged mice. Material and methods: Eleven female mice, 8-10 weeks of age and free of murine specific pathogens, were used. Of the eleven mice, one mouse was not sensitized and ten mice were sensitized by intraperitoneal injection of OVA. Of the sensitized mice, three mice didn't administrate Citri Reticulatae Viride Pericarpium and seven mice administrated Citri Reticulatae Viride Pericarpium. Mice were sensitized on days 1 and 14 by intraperitoneal injection of 20 ㎍ OVA. On days 21, 22, and 23 after the initial sensitization, the mice were challenged for 30 minutes with an aerosol of 1 % OVA in saline. Citri Reticulatae Viride Pericarpium administered 200mg/kg in the tail of the mouse, one time per day, for 7 days, beginning 14 days after first sensitization. Bronchoalveolar lavage was performed 72 hours after the last challenge, and level of VEGF in the BAL fluid were measured by Western blot analysis. Results: Western blot analysis revealed that VEGF protein levels were increased in the all three mice which were challenge with OVA without administered Chung-pi compared the normal mouse. However, in the groups of the administered Chung-pi, the VEGF protein level markedly decreased in six of seven mice. Conclusion : Citri Reticulatae Viride Pericarpium might be effect the treatment of the bronchial asthma as a inhibition of the VEGF.

Diphlorethohydroxycarmalol (DPHC) is a known to modulate the expression of extracellular matrix (ECM) components in 3T3-L1. However, the possible role of DPHC in integument stability during obesity induction is not clear yet. We evaluated the effects of DPHC on collagen or elastic fiber quantity in integument during obesity induction with high-fat diet. The dorsal back integument sections were stained with hematoxylin-eosin, Masson trichrome, and Verhoff-Van Gieson. The intensities of collagen fibers and elastin fibers were analyzed with ImageJ. The number of fibroblasts was counted at ${\times}1,000$ fields. The number of fibroblast was increased by obesity induction, but DPHC suppressed it in a concentration-dependent manner both in lean and obese mice. On the other hand, the intensities of collagen fibers were increased by DPHC treatment in obese mice groups but not in lean mice groups. The intensities of collagen fibers of obese mice were lower than that of the lean mice in 0% group. However, the number became similar between lean and obese mice by the treatment of DPHC. The intensity of elastic fibers was increased in the lean mice with the concentration of DPHC. In the obese mice group, there were increasing patterns but only significant at 10% DPHC group. The intensity of elastic fibers of obese mice was higher than lean mice in 0%, 1%, and 10% groups. Histologically epithelial cells and follicle cells which were diffused nuclear staining forms were increased by DPHC treatment. The results suggest that the activity of integument cells during obesity induction can be modulated by DPHC.

Mitochondrial NADP+-dependent isocitrate dehydrogenase 2 (IDH2) produces NADPH, which is known to inhibit mitochondrial oxidative stress. Ureteral obstruction induces kidney inflammation and fibrosis via oxidative stress. Here, we investigated the role and underlying mechanism of IDH2 in unilateral ureteral obstruction (UUO)-induced kidney inflammation using IDH2 gene deleted mice (IDH2-/-). Eight- to 10-week-old female IDH2-/- mice and wild type (IDH2+/+) littermates were subjected to UUO and kidneys were harvested 5 days after UUO. IDH2 was not detected in the kidneys of IDH2-/- mice, while UUO decreased IDH2 in IDH2+/+ mice. UUO increased the expressions of markers of oxidative stress in both IDH2+/+ and IDH2-/- mice, and these changes were greater in IDH2-/- mice compared to IDH2+/+ mice. Bone marrow-derived macrophages of IDH2-/- mice showed a more migrating phenotype with greater ruffle formation and Rac1 distribution than that of IDH2+/+ mice. Correspondently, UUO-induced infiltration of monocytes/macrophages was greater in IDH2-/- mice compared to IDH2+/+ mice. Taken together, these data demonstrate that IDH2 plays a protective role against UUO-induced inflammation through inhibition of oxidative stress and macrophage infiltration.
Brazilin, the main constitutent of Caesalpinia sappan, was examined for its immunomodulating activities in normal CBA mice. Mitogen induced proliferation and production of ConA induced T-cell growth factors (TCGF) of splenocytes were significantly reduced in brazilin treated group, cmpared to control group. It was also found that suppressor activities of splenocytes in brazilin treated group was significantly increased compared to those in normal control group.
SCi-B-Vac, the 3rd hepatitis B vaccine , was selected for clinical evaluation on the basis of toxicologic profiles in preclinical studies. These studies were performed to obtain information on its toxic signs, organs which are mainly affected, and to estimate its lethality in mice and rats given Sci-B-Vac through two routes of administratin. In male and female rats given a single intragastrical dose of Sci-B-Vac, we estimated that $LD_{50}$ values were over 2.00 ml/100g B.W. (10ng/ml), respectively.

This study investigated whether swimming exercises improves obesity through regulation of angiogenesis in white adipose tissue. Female mice with high-fat diets were divided into sham-operated group (Sham), ovariectomized group (OVX), and swim-trained ovariectomized group (OVX + Swim). Compared to the Sham, OVX increased body weight, adipose tissue mass and size of adipocyte. However, these factors (: such as body weight, adipose tissue mass and size of adipocyte) of OVX + Swim decreased compared with OVX. Compared with the Sham, OVX increased the mRNA expression of angiogenic activator and MMPs and decreased the mRNA expression of angiogenic inhibitors in white adipose tissue. But OVX + Swim decreased the mRNA expression of angiogenic activator and MMPs and increased the mRNA expression of angiogenic inhibitors in white adipose tissue, compared with the OVX. Theses results suggested that swimming exercises the angiogenesis in white adipose tissue, resulting to improve obesity in high-fat diet-fed female OVX mice.
KO mice provide an excellent tool to determine roles of specific genes in biomedical filed. Traditionally, knockout mice were generated by homologous recombination in embryonic stem cells. Recently, engineered nucleases, such as zinc finger nuclease, transcription activator-like effector nuclease and clustered regularly interspaced short palindromic repeats (CRISPR), were used to produce knockout mice. This new technology is useful because of high efficiency and ability to generate biallelic mutation in founder mice. Until now, most of knockout mice produced using engineered nucleases were C57BL/6 strain. In the present study we used CRISPR-Cas9 system to generate knockout mice in FVB strain. We designed and synthesized single guide RNA (sgRNA) of CRISPR system for targeting gene, Abtb2. Mouse zygote were obtained from superovulated FVB female mice at 8-10 weeks of age. The sgRNA was injected into pronuclear of the mouse zygote with recombinant Cas9 protein. The microinjected zygotes were cultured for an additional day and only cleaved embryos were selected. The selected embryos were surgically transferred to oviduct of surrogate mother and offsprings were obtained. Genomic DNA were isolated from the offsprings and the target sequence was amplified using PCR. In T7E1 assay, 46.7% among the offsprings were founded as mutants. The PCR products were purified and sequences were analyzed. Most of the mutations were founded as deletion of few sequences at the target site, however, not identical among the each offspring. In conclusion, we found that CRISPR system is very efficient to generate knockout mice in FVB strain.
