The effect of water extract of antler on serum level of female hormone in ovariectomized rats (녹용의 물 추출액 투여가 난소를 절제한 흰쥐의 여성호르몬 변화에 미치는 영향)

  • The effect of water extract of antler on serum level of female hormones was investigated in ovariectemized rats. Sprague-Dawley rats were ovariectomized(ox). After a further 40 days the animals were administrated with water extract of antler (625mg/kg daily) for 10, 20 and 30 days, respectively. Serum level of estradiol, progesterone, leutenizing hormone and follicle stimulating hormone were measured. Significant increase of serum estradiol level was elicited at 20 and 30 days after treatment with water extract of antler, respectively, Associated with the increase of serum estradiol level, there was a concomitant decrease in serum follicle stimulating hormone level. Serum progesterone level was also significantly increased at 30 days after treatment with water extract of antler. Although serum leutenizing hormone level of ox rats treated with water extract of anther was slightly lower than that of untreated ox rats, the decrease was not significant.

A Study on Diagnosis of the Fertility of one Aged Female Gorilla by using the Fecal Sex Hormone Metabolites

  • The aim of this study was to diagnose the fertility of a female western lowland gorilla kept in Seoul Zoo, in accordance with age by analyzing the fecal sex hormone metabolites. The study was conducted in two period of times, when the animal was from 35 to 37 years old and when the animal was from 40 to 42 years old. Non-invasive method by using fecal samples was used for safe and efficient fertility diagnosis. We collected the feces from the enclosure at least three times a week. Then $17{\beta}$-estradiol and progesterone, which are fecal sex hormone metabolites, were measured by time-resolved fluoro-immunoassay to compare the menstruation cycle and the annual reproductive cycle. For the duration of the primary study (when the animal was 35~37 years old), irregular menstruation and high concentrations of estradiol and progesterone were observed. However, menstruation was hardly observed and the concentrations of both hormones were statistically very low in the period of secondary study (when the animal was 40~42 years old). This observed phenomenon in our study was very comparable to menopause in adult women; therefore, it was confirmed that our female gorilla has reached menopause because of the natural aging, as they become older.

  • [Purpose] Sex hormones deficiency leads to dramatically bone loss in particular postmenopausal women. Royal jelly has anti-osteoporosis effect due to maintain bone volume in that condition. We hypothesized that royal jelly protein (RJP, a latent residue after extracting royal jelly) also prevents bone deficient in ovariectomized (OVX) female rats, the animal model of postmenopausal women. [Methods] Female Sprague-Dawley rats (n = 30, 6 weeks age old) were sham operated (Sham; sham operated group, n = 7), OVX control group (OC, n = 7), OVX with low RJP intake group (ORL, n = 8), and OVX with high RJP intake group (ORH, n = 8) during 8 weeks experimental periods. In the end point of this experiment, the bone samples (lumbar spine, tibia, and femur) were surgically removed under anesthesia. These bone samples were evaluated bone mineral density (BMD) and bone strength. [Results] BMD of lumbar spine in RJP intake groups (ORL, ORH) were higher than that in OC group (p < 0.05 and p < 0.01) in RJP intake volume dependent manner. BMD of tibial proximal metaphysis and diaphysis in RJP intake groups were also higher than these in OC group (p < 0.01 and p < 0.01 / p < 0.05 and p < 0.001). In addition, breaking force of femur in RJP intake groups were significantly increase compared with that in OC group (p < 0.001 respectively). [Conclusion] These findings indicate that RJP contribute to prevent sex hormone related bone abnormality.

  • The locus coeruleus(LG) is known to be observed a sexual dimorphism in rat CNS. LC is the largest collections of norepinephrine(NE)-containing neurons in the mammalian brain. Especially in rat, all LC neurons contained NE unlike other mammalians, so that specific reactions were found in the tyrosine-hydroxylase(TH) immunoreactive neurons. Sexual dimorphism of rat LC has affected by genes before sex hormone appeared, thereafter affected by sex hormones. In these day, many scientists founded morphological differences between male and female LC morphology, but differences of entire structure was not founded. Thus we investigated sex differences of the LC neuron's morphology in rat by three-dimensional(3-D) reconstruction using Confocal laser scanning microscopy(CLSM). We reported that neuron's shape was relatively-large multipolar neurons and neuron's processes in dorsal LC proceeded to ventral direction in the male and female rat. Male had a longer anterior-posterior length than female had in dorsal LC. In addition to middle-LC, male rat's LC had a more thicker posterior region but had not viewed in a previous study. In reverse, female rat's LC had a thicker anterior region like a previous study. This results using 3-D reconstruction by CLSM showed that the male's LC was more wide-ranging than female's relatively.

Changes in Sex Steroid Hormones and Ovarian Development during Artificial Maturation of Female Eel, Anguilla japonica

  • The present study demonstrates the changes in body weight (BW) and plasma sex steroid hormone profiles during artificial maturation induced by human chorionic gonadotropin (HCG) or salmon pituitary extract (SPE) injections in cultured eel, Anguilla japonica, kept in seawater for 3 months. In the weekly SPE-injected female group, BW was relatively stable during vitellogenesis. Following induction of vitellogenesis, females exhibited a rapid increase of BW, and the oocytes were observed to be in the migratory nucleus stage at the end of the experiment. Plasma testosterone (T) and $estradiol-17{\beta}$ ($E_2$) levels increased slightly during vitellogenesis and peaked at an average of 5.82 ng/mL and 4.76 ng/mL, respectively, at the end of the experiment. In the weekly control and HCG-injected female groups, BW slowly decreased during the experimental period, and the oocytes of the two groups were observed to be at the primary yolk globule stage. In the weekly HCG-injected female group, plasma T and $E_2$ levels increased slightly during vitellogenesis and decreased afterward. In the control female group, however, plasma T and $E_2$ levels were not altered during the experimental period. Furthermore, plasma $17{\alpha},20{\beta}-dihydroxy-4-pregnen-3-one$ (DHP) was not detected in all experimental groups. Fertility and hatching rates of SPE-injected females were significantly higher in those that ovulated 15 h after DHP injection than 18 h. These results indicate that long rearing in seawater increases responsiveness to SPE in ovarian maturation of the Japanese eel, resulting in shortened period from completion of vitellogenesis by sex steroid hormone production.

  • These experiments were carried out to investigate the effects of cumulus cells for in vitro fertilization and development of bovine follicular oocytes matured in vitro. The bovine ovaries were obtained at a slaughter house and the follicular oocytes surrounded by cumulus cells were collected by puncturing follicles with 2~6 mm of diameter. Bovine oocytes were matured in vitro for 24~26 hours in a CO2 incubator with 5% CO2 in air at 39$^{\circ}C$. The medium used for maturation was TCM-199 supplemented with hormones, pyruvate, FCS and antibiotics. Epididymal spermatozoa were capacitated by in vitro culture for 2~3 hours in BO solution containing BSA(5mg/ml) and caffeine(2.5mM). Insemination was made by introducing about 10~15 matured oocytes into the suspension of capacitated spermatozoa. Six hour after insemination the eggs were transferred to TCM-199 supplemented with FCS(10%) and HEPES(25mM), cultured for 7~8 days with 10~15 eggs/well in 4-well multidishes(Nunc Co.) forming cumulus cell monolayer. The results obtained in these experiments were summarized as follows ; 1. The majority of the follicular oocytes with compacted cumulus cells existed in GV stage while those with dispersed or denuded cumulus cells existed GVBD and M II stage. 2. After 24~26 hours maturation, the maturation rates of the follicular oocytes cultured in TCM-199 containing hormones were slightly higher than those of oocytes cultured in medium without hormones, and the frequency of cumulus compacted or denuded oocytes reaching M II stage cultured in medium containing hormones was 75.7% or 51.7%, respectively(P<0.05). 3. After 20 hours in vitro insemination, percentages of ova fertilized were 61.4% or 51.4%, respectively, for cumulus oophorus intacted or removed, and increased frequency of ova with both male and female pronuclei was found when cumuli were present(P<0.05). 4. The rates of embryos developed to 2-, 4-, 8-, 16-cell and morula or blastocyst stage after cocultured with cumulus cells were 65.0%, 45.3%, 34.7%, 28.0% and 22.7%, respectively. The results for momla or blastocyst stage were significantly higher than those of the embryos cultured in the basic medium(P<0.05).

Changes in Plasma Steroid Hormone Level in Rockfish (Sebastes inermis) by the Controlled Water Temperature and Photoperiod (수온과 광주기 조절에 의한 볼락 (Sebastes inermis)의 혈장내 성 스테로이드호르몬 농도의 변화)

  • Plasma levels of sex steroid hormones in rockfish, Sebastes inermis were examined monthly in relation to gonadosomatic index (GSI) under a controlled water temperature and photoperiod, The GSI of a control group (C) in female began to increase from November and reached a maximum in January, Sample fish under a controlled water temperature and photoperiod (Tr) were divided into a responded group (Tr-r) and a un-responded group (Tr-n) by the gonadal maturation condition and GSI. The GSI of females in Tr-r reached a maximum in March. But the female GSI in Tr-n kept lower than 1.2 during the experimental period. No differences in male GSI were noticed between C and Tr. The $estradiol-17\beta$ and testosterone levels of female plasma in Tr reached a maximum in October, later than those in C. In males, these was no difference in 11-ketotestosterone and testosterone between C and Tr. When rockfish was reared in September under the controlled water temperature and photoperiod which were equivalent to those in July, that is two months earlier, the maturation of females was delayed in comparison with C. This finding suggested that delayed maturation in ovary was caused by the secretion of sex steroid hormones in relation to the water temperature and photoperiod of the hypothalamus-pituitary-gonad axis.

Induction of Sexual Maturation in Female Eels Anguilla japonica by Recombinant Eel Gonadotropin Produced in Silkworm (누에에서 생산된 뱀장어 재조합 생식소자극호르몬이 암컷 뱀장어(Anguilla japonica)의 성성숙 유도에 미치는 영향)

  • This study investigated the effects of recombinant eel gonadotropin hormone (rJeGTH) produced in silkworms, with and without a carboxyl-terminal peptide from equine chorionic gonadotropin (eCG), on the induction of sexual maturation in female eels Anguilla japonica. Experiments were conducted both in vivo and in vitro. In in vitro trials, germinal vesicle breakdown (GVBD) induction did not significantly differ between rJeFSH and $rJeFSH{\cdot}eCG$ treatments and the control group. However, previous studies did find that rJeLH and $rJeLH{\cdot}eCG$ treatments induced GVBD in female eels. Our in vitro exploration of $estradiol-17{\beta}$ ($E_2$) levels in immature ovarian tissues revealed significantly higher $E_2$ levels in the group treated with $rJeFSH{\cdot}eCG$ $1{\mu}g/mL$ than in the control group. In contrast, the in vivo experiments showed no effect of recombinant hormones on the sexual maturation of feminized eels. Previous studies and our own in vitro results have clearly shown that rJeGTH and $rJeGTH{\cdot}eCG$ have a positive effect on the sexual maturation of feminized eels. To develop the activity of rJeGTH in vivo, further studies should confirm circulation time and activity of these hormones in eels' bloodstream, modify the structure of the recombinant gene, and implement additional glycosylation.

Sex Reverse in Teleost Culture II. Sex Reversal of Guppy and Tilapia by $17\beta$-Estradiol and $17\alpha$-Methyltestoserone (경골어류의 성전환에 관한 연구 II. $17\beta$-Estradiol 및 $17\alpha$-Methyltestosterone을 이용한 Guppy 및 Tilapia의 성전환에 관한 연구)

  • Guppy fry were treated for the first 40 days of life with 0, 20, 40, 60 & 100$\mu\textrm{g}$ of estradiol per gram of food in order to change the sex of normal males to functional females(genetic male). The present investigation deals with the effects of steroid hormones, such as $\beta$-estradiol and testosterone, on the sex differentiation in guppy and tilapia. The results obtained were summarized as follows. 1. In B (20$\mu\textrm{g}$/g diet) group 17$\beta$-estradiol-treated, 67.8% of male offsprings were produced. 2. In D (60$\mu\textrm{g}$/g diet) group 17$\beta$-treated, 67% of female offsprings were produced. 3. B, D groups of genetic male brooders had significantly different effects (P<0.01) upon sex ratios of their progeny. 4. This strongly indicates that sex direction has been achieved and that the male is the heterogametic sex. 5. The group that produced the highest percentage of male offspring(male percentage of observed number to expected number was 91%) contained only full-sibling male brooders to the sex-reversed female brooders. 6. After 7 months following treatment, the sex-reversed males had ovarian portion in the anterior region and a testicular portion in the posterior region of the same intersexual gonad, respectively. 7. At 7 months after treatment, the ovareis revealed a complete arrest of the ovarian formation, and appearances of spermatogenetic cell cysts among surviving auxocytes. 8. In most of sex-reversed fish, anterior portion of test is was devoid of sperm ducts including the seminal vesicle and vas deferens. 9. The male transferrin showed two strong bands, while the female transferrin showed a single weak band. 10. One of the two bands of male transferrin showed the same mobility with band of female transferrin.

Effect of Testosterone on Free Radical Generating Enzyme and Lipid Peroxidation (지질과산화 반응과 Free Radical 생성계 효소활성에 미치는 Testosterone의 영향)

  • Sex hormones not only regulate external sexual characteristics but several internal biochemical processes. It is well accepted that life-span of female is longer than that of male. Life-span is closely related with aging process in which free radicals are known to be involved. We investigated the effect of testosterone on free radical generating systems and lipid peroxidation based on the sexual difference. Lipid peroxide levels of male and female mouse were increased proportionately with age, especially in male mouse. Increase in enzyme activity of aldehyde oxidase with age was observed in male mouse, while no siginificant change in enzyme activity was found in female mouse. Enzyme activity of xanthine oxidase also showed similar results. It, however, was not significant statistically. Lipid peroxide level and xanthine oxidase type conversion ratio of male and female mouse liver homogenate incubated at $37^{\circ}C$, increased remarkably in proportion to incubation time, especially in male mouse. Lipid peroxide level and aldehyde oxidase activity were measured in normal male mouse, castrated mouse and testosterone treated-castrated mouse. Castrated mouse group showed decrease in lipid peroxide level and aldehyde oxidase activity compared with normal group. Treatment of castrated mouse with testosterone, however turned the level of lipid peroxide and aldehyde oxidase activity back to normal. From the above results, it might be concluded that testosteron could increase the activities of free radical generating enzymes which would result in the formation of lipid peroxide, consequently leading to aging.
