Anti-Mullerian Hormone Serum Concentrations in Prenatal and Postnatal Period in Murine

  • Mullerian inhibiting substance (MIS) is a member of the TGF-${\beta}$ (transforming growth factor-${\beta}$) family whose members play key roles in development, suppression of tumour growth, and feedback control of the pituitary-gonadal hormone axis. MIS is expressed in a highly tissue-specific manner in which it is restricted to male Sertoli cells and female granulose cells. The serum levels of MIS in prenatal and postnatal ICR mice were measured using the enzyme-linked immuno-solvent assay (ELISA) using the MIS/AMH antibody. Mice were grouped by age: the significant periods were at the onset of development. During sex organ differentiation, no remarkable difference between female and male foetus MIS serum levels (both<0.1 ng/ml) was observed. However, MIS serum levels in pregnant mice markedly changed (4.5~12.2 ng/ml). After birth, postnatal female and male mice serum MIS levels changed considerably (male: <0.1~138.5 ng/ml, female: 5.3~103.4 ng/ml), and the changing phase were diametrically opposed (male: decreasing, female: fluctuating). These findings suggest that MIS may have strong associations with not only develop-ment but also puberty. For further studies, establishing the standard MIS serum levels is of importance. Our study provides the basic information for the study of MIS interactions with reproductive organ disability, cancer, and the effect of other hormone or menopause. We hypothesise that if MIS is regularly injected into middle-age women, meno-pause will be delayed. We detected that serum MIS concentration curves change with age. The changing phase is different between males and females, and this difference is significant after birth. Moreover, MIS mRNA is expressed during the developmental period (prenatal) and also in the postnatal period. This finding indicates that MIS may play a significant role in the developmental stage and in growth after birth.

  • To evaluate fetal sex-hormonal status before delivery, testosterone and follicle stimulating normone(FSH) levels were measured in 64 amniotic fluid samples at midgestation by radioimmunoassay method. The mean concentration of testosterone in amniotic fluid of 37 cases carrying male fetus was 90.7 pg/ml and 27 cases carrying female fetus was 62.3 pg/ml. The mean :amniotic fluid FSH concentration of male fetus was 1.15 mIU/ml and of female fetus was 11.98 mIU/ml. The amniotic fluid testoserone and FSH concentrations had statistical difference between male and female fetuses. The ratio of testosterone over FSH in the amniotic fluid was 231.2 in male, 9.8 in female respectively and very significant difference was noticed. The levels of testosterone/FSH greater than 25 were found over 92% of male fetus and lesser than 25 were found over 92% female fetus. Measurement of testosterone and FSH especially testosterone/FSH ratio in amniotic fluid in midgestation may be an adjunct to other method of fetal sex determination.

  • the aim of this study was to investigate whether osteoporosis can be advanced after cerebral infarction Of not and role of female sex hormone. all animals were classified 4 groups; group of ovariectomy (OVXgroup) group of cerebral infarction( INF group), group of combination ovariectomy and cerebral infarction(OVX + INF group), naturally intact group for control data (NOR group ). cerebral infarction was made by Chen,s method with some modification and ovariectomy was performed by Wayforth,s method. experimental data was collected at 15 days, 1 month, 3 months and 6months after starting observation. serum 17 ${{\beta}-estradiol\;(E_2)}$ was determined by radioimmunoassay, for comparision of osteoporosis formation, bone density, serum osteocalcin, Serum total calcium and phosphorus, Serum AST Concentration, Serum ALT Concentration, Creatinine Concentration were also calculated. we have found that cerebral ischemia decreases not only the serum concentration of 17 ${{\beta}-estradiol\;(E_2)}$ and inhibits but also the physiologically compensatory function of the ovariectomized rats and that the decreased estrogen concentration followed by cerebral infarction have not produced osteoporosis, regretfully.

Protective effects of saffron against zearalenone-induced alterations in reproductive hormones in female mice (Mus musculus)

  • Objective: Zearalenone (ZEA) is a mycotoxin with potent estrogenic effects. Saffron is an herbal product that has antioxidant activities. The objective of this study was to investigate the protective role of saffron against reproductive toxicity induced by ZEA in female mice. Methods: Ninety 8-week-old female mice were randomly allocated into three treatment groups. The first group received an intraperitoneal injection of ZEA (2.5 mg/kg) on alternate days. The second group received ZEA (2.5 mg/kg) on alternate days plus oral saffron daily (50 mg/kg). The third group was treated with a vehicle of 1% dimethyl sulfoxide (DMSO) on alternate days, as a control. Ten mice were euthanized from each group at 30, 60, and 90 days of treatment. Serum levels of luteinizing hormone (LH), follicle-stimulating hormone (FSH), estradiol ($E_2$), and progesterone (P) were assessed. The uterus and ovaries were examined for changes in size or morphology. Results: Serum levels of LH, FSH, $E_2$, and P in the female mice treated with ZEA plus saffron were significantly higher than in those treated with ZEA alone, and were not significantly different from those treated with 1% DMSO. The female mice treated with ZEA alone showed a reduction in size of the uterus and abnormal architecture of the ovaries. Conclusion: The administration of saffron to female mice resulted in a significant reduction in ZEA-induced alterations in reproductive hormone levels, the size of the uterus, and the morphology of the ovaries.

  • Recently, As the prevalence of precocious is increased in Korea This study was evaluated the physical growth and female sex hormone characteristics of girls with Idiopathic precocious puberty. Retrospective analysis about the medical record of 113 girls for evaluation of signs of precocious puberty except for organic causes was done. Physical growth features and Female sexual hormone were analyzed. Height SDS and Weight SDS were significantly increased in true sexual precocity compared to pseudo sexual precocity. Hormone studies showed that the level of basal LH, FSH, E2 was significantly higher in true precocious puberty. The finding of this study suggest that we need to develop intervention about physical and psychological problems for precocious puberty girls.

  • The purpose of this study finds out the effect of red ginseng extract (1.0g/kg) on TBTO (10, 20, and 40mg/kg) which poisons against some organs like thyroids gland, liver, kidney, testis, ovary, sex hormone activity of rats are examined by gastric tubing for 3 weeks. The weight of each organ in treated group were increased, especially liver in female and those of testis in males were significantly increased at 10, 20 and 40mg/kg (P<0.05, P<0.01). In case of sex hormone activity of each sex, the estradiol activity of female and testosterone activity of males were significantly decreased rather than the control group (P<0.05, P<0.01) According to between the TBTO treated group and 10+ rGe group of the testosterone activity each sex were significantly increased (P<0.01).

  • The effect of calcium gluconate on estrogen (estradiol) serum level as well as follicle stimulating hormone level was studied. Our results revealed that oral administration of calcium gluconate (100 mg/kg body wight) to adult non-pregnant female rabbits caused a significant increase of serum levels of estradiol and follicle stimulating hormone. On the other hand, oral contraceptive (Norminest tablets) decreased significantly follicle stimulating hormone serum level, while combined administration of calcium gluconate and oral contraceptive caused significant increase of serum level of follicle stimulating hormone compared with control values. Also, concurrent administration of calcium gluconate and Norminest tablet increased significantly the rate of conception compared with group recieved Norminest tablets only. These results indicated that combined administration of calcium and oral contraceptives must be cautiously.

Changes in Sex Steroid Hormones and Ovarian Development during Artificial Maturation of Female Eel, Anguilla japonica

  • The present study demonstrates the changes in body weight (BW) and plasma sex steroid hormone profiles during artificial maturation induced by human chorionic gonadotropin (HCG) or salmon pituitary extract (SPE) injections in cultured eel, Anguilla japonica, kept in seawater for 3 months. In the weekly SPE-injected female group, BW was relatively stable during vitellogenesis. Following induction of vitellogenesis, females exhibited a rapid increase of BW, and the oocytes were observed to be in the migratory nucleus stage at the end of the experiment. Plasma testosterone (T) and $estradiol-17{\beta}$ ($E_2$) levels increased slightly during vitellogenesis and peaked at an average of 5.82 ng/mL and 4.76 ng/mL, respectively, at the end of the experiment. In the weekly control and HCG-injected female groups, BW slowly decreased during the experimental period, and the oocytes of the two groups were observed to be at the primary yolk globule stage. In the weekly HCG-injected female group, plasma T and $E_2$ levels increased slightly during vitellogenesis and decreased afterward. In the control female group, however, plasma T and $E_2$ levels were not altered during the experimental period. Furthermore, plasma $17{\alpha},20{\beta}-dihydroxy-4-pregnen-3-one$ (DHP) was not detected in all experimental groups. Fertility and hatching rates of SPE-injected females were significantly higher in those that ovulated 15 h after DHP injection than 18 h. These results indicate that long rearing in seawater increases responsiveness to SPE in ovarian maturation of the Japanese eel, resulting in shortened period from completion of vitellogenesis by sex steroid hormone production.

A Study on Diagnosis of the Fertility of one Aged Female Gorilla by using the Fecal Sex Hormone Metabolites

  • The aim of this study was to diagnose the fertility of a female western lowland gorilla kept in Seoul Zoo, in accordance with age by analyzing the fecal sex hormone metabolites. The study was conducted in two period of times, when the animal was from 35 to 37 years old and when the animal was from 40 to 42 years old. Non-invasive method by using fecal samples was used for safe and efficient fertility diagnosis. We collected the feces from the enclosure at least three times a week. Then $17{\beta}$-estradiol and progesterone, which are fecal sex hormone metabolites, were measured by time-resolved fluoro-immunoassay to compare the menstruation cycle and the annual reproductive cycle. For the duration of the primary study (when the animal was 35~37 years old), irregular menstruation and high concentrations of estradiol and progesterone were observed. However, menstruation was hardly observed and the concentrations of both hormones were statistically very low in the period of secondary study (when the animal was 40~42 years old). This observed phenomenon in our study was very comparable to menopause in adult women; therefore, it was confirmed that our female gorilla has reached menopause because of the natural aging, as they become older.

Thyroid Hormone-like Activity of Alachlor as R Endocrine Disruptor in Rats and HeLaTRE Cell Culture (랫드와 HeLaTRE Cell에서의 Alachlor에 의한 갑상선 호르몬성 영향 연구)

  • This study was designed to investigate the suitability of the pubertal assay and the enhanced TG 407 as methods for detection of endocrine-mediated effects, especially thyroid function. Male and female Sprague-Dawley rats were gavaged daily with 0, 12.5, 25, 50 mg/kg alachlor in corn oil during 30 days. The effects of alachlor on thyroid gland, the genital organs and thyroid hormone were measured in male and female rats. Dose of alachlor 25, 50 mg/kg/day increased relative weight of testis and thyroid gland in exposed male rats and decreased relative weight of vagina in exposed female rats. Relative weight of thyroid gland was decreased in alachlor 25 mg/kg/day exposed female rats. Dose of alachlor 25, 50 mg/kg/day decreased plasma T4 and testosterone in female rats. Another purpose of this study was to investigate the effects of endocrine disruptors as like thyroid hormone in vitro. Luciferase activity was measured to dectect reaction of test chemicals and thyroid hormone response elements in HeLaTRE cell. Dose of alachlor 1 nM-1000 nM increased 100-134% luciferase activity compared with control.
