  • These studies were conducted to evaluate the Properties of lactic acid Producing bacteria(LAB), isolated from broiler and laying hens cecum and select the optimum strains to improve the performance, environment of poultry house, immunity, and intestinal microflora of broiler and laying hens. In experiment I , 23 LAB strains were isolated from broiler and laying hens cecum as a colony form. Six strains were selected by acid tolerance, bile salt tolerance, viability, enzyme release, antagonism, and antibiotics susceptibility. In Experiment II, selected LABs from Ex. 1 were conducted to investigate the effects of feeding various Lactobacillus on performance, nutrients digestibility, intestinal microflora, villi development and observation of epithelium surface, blood chemicals and fecal noxious gas of broiler chicks. One thousand eighty one day old broiler chicks were fed into Lactobacillus crispatus avibrol(LCB), Lactobacillus reuteri avibro2(LRB), Lactobacillus crispatus avihen1(LCH), and Lactobacillus vaginalis avihen2(LVH) at the level of 10$^4$ and 10$\^$7/cfu/g diet. Weight gam of chicks fed Lactobacillus tended to increase from the first week and was higher from 50 to 100g in Lactobacillus treatments than control. Feed intake and feed conversion were not statistically different of all treatments. Dry Matter digestibility of Lactobacillus treatments was prone to improve compared to that of control, but was not significantly different. Protein and Ca digestibility were also tended to improve in Lactobacillus treatments relative that of control. Lactobacillus treatments showed improved tendency in crude ash and fat compared to those of control, whereas phosphorus digestibility was not consistency. Nutrients digestibilities of bird fed LCH were superior to those of other treatments, It showed significantly higher in Ca and P digestibility than control(P〈0.05). Total Lactobacillus spp. of birds fed various Lactobacillus was significantly higher in illeum for five weeks(P〈0.05), but was not different at cecum. Yeast was thought to be not completely attached to intestinal lumen for one week. However, total number of yeast was significantly increased in cecum and illeum of three weeks old chicks (P〈0.05). The number of anaerobes exhibited to tendency the increase in Lactobacillus treatments from one week old of age at both ileum and cecum.

Effect of Dietary Supplementation of Cu-methionine Chelate and Cu-soy Proteinate on the Performance, Small Intestinal Microflora and Immune Response in Laying Hens (사료내 Cu-methionine Chelate와 Cu-soy Proteinate가 산란계의 생산성, 소장내 미생물 균총 및 면역체계에 미치는 영향)

  • This study was conducted to investigate the effect of dietary supplementation of Cu-methionine chelate(Cu-Met) and Cu-soy proteinate(Cu-SP) on the performance, small intestinal microflora and immune response in laying hens. A total of 960 Hy-line $Brown^{(R)}$ laying hens of 39 wks old were assigned to one of the following 6 dietary treatment: control(C), antibiotic(Avilamycine 6 ppm), Cu-Met 50 and Cu-Met 100(50 and 100 ppm Cu as Cu-methionine chelate), Cu-SP 50 and Cu-SP 100(50 and 100ppm Cu as Cu-soy proteinate). Each treatment was replicated 4 times with forty birds per replication, housed in 2 birds per cages. Forty birds units were arranged according to randomized block design. Feeding trial lasted 6 wks under 16 hours lighting regimen. Hen-day and hen-house egg production of groups treated with Antibiotic and Cu supplements tended to be higher than the control with significant difference (P<0.05) shown between Cu-Me 100 and control. Egg weight was significantly (P<0.05) heavier in antibiotic and Cu-SP treatments than Cu-Met treatments but they were not significantly different from the control. Eggshell strength, egg shell thickness, egg yolk color and Haugh unit were not significantly different among treatments. There were no significant differences in leukocytes and erythrocytes in the chicken blood. But mean corpuscular hemoglobin value(MCH) was significantly (P<0.05) higher in Cu-SP 100 than antibiotic treatment. The concentrations of serum IgG and IgA were not significantly different among treatments. Copper concentration in the liver tended to increase as the level of copper supplementation increased, that of Cu-SP 100 being significantly (P<0.05) higher than those of the control and antibiotic treatment. Concentrations of iron and zinc of the liver were not significantly influenced by treatments. Populations of Cl. perfringens and Lactobacilli in the small intestinal content were significantly (P<0.05) influenced by treatments. Population of Cl. perfringens decreased and that of Lactobacilli increased in the copper supplemented groups. The result of this experiment showed that Cu-Met and Cu-SP are comparable to antibiotic in improving egg production in laying hens. Birds fed diets supplemented with Cu-SP produced heavier eggs than those fed diets with Cu-Met. There were no significant differences in the performances between 50 ppm and 100 ppm copper supplementation as organic forms.

Effects of Dietary Addition of Sucrose, Propylene Glycol and Tween 80 on the Performance of Transitional Holstein Cows (Sucrose, Propylene Glycol, Tween 80의 첨가가 전환기 젖소의 생산성에 미치는 영향)

  • This study was conducted to evaluate the effects of dietary addition of sucrose, propylene glycol and Tween 80 (Polysorbate 80 : Non-ionic Surfactants) on pre-partum (21 d) and post-partum (21 d) nutrients intake, blood metabolites, occurrence of metabolic disorders, milk yield and its composition in Holstein cows. Two basal diets were formulated each for pre- and post-partum period. The diets were mixed daily and fed at ad libitum to transitional cows. Forty cows of similar parity and milk yield were randomly divided into four groups (ten animals in each). The cows in three groups were supplemented either with 280g of sucrose/day (SU), SU+64g propylene glycol/day (SUP) or SUP+50g Tween80/day (SUPT). The feed for the fourth group was not supplemented and this group served as control (C). Pre-partum DM, total digestible nutrients (TDN), and crude protein (CP) intake was similar in cows fed C, SU, SUP, and SUPT diets. Post-partum DM, TDN, and CP intakes were the highest with SUPT diet followed by SU, SUP and control diets. Pre-partum blood non-esterified fatty acids (NEFA) concentration was noticed significantly higher in cows fed control diet compared to those fed SU, SUP and SUPT diets. The concentration of NEFA was similar at calving and during post-partum period across cows fed different experimental diets. Blood glucose and Ca concentration during pre- and post-partum periods were not significantly different in cows fed C, SU, SUP and SUPT diets. Milk yield (kg/day) was similar in cows fed different experimental diets. However, milk fat percent and 4% fat corrected milk yield were higher in cows fed SU diet (p<0.05) followed by SUP, SUPT and C diets. One case of ketosis was recorded in cows fed control diet however its occurrence was not observed in cows fed other diets. Occurrence of retained placenta and mastitis was numerically higher in cows fed control diet compared with those fed SU, SUP and SUPT diets. In conclusion, the NIS and propylene glycol feeding along with sucrose could improve the 4% fat corrected milk and fat yield in early lactating cows with significant reduction in NEFA and metabolic disorders during transitional period.

Anti-diabetic Effect of Crude Polysaccharides from Grifola frondosa in $KK-A^{y}$ Diabetic Mouse and 3T3-L1 Adipocyte (3T3-L1지방세포 및 제2형 당뇨모델($KK-A^{y}$)에서 잎새버섯(Grifola frondosa) 조다당체 추출물의 항당뇨 효과)

  • This study was performed to investigate the anti-diabetic mechanism of crude polysaccharides isolated from the fruiting bodies of Grifola frondosa. We treated 3T3-L1 adipocyte cells to observe whether the crude polysaccharides isolated from Grifola frondosa would stimulate insulin sensitivity. Significant insulin sensitizing activity was observed in the 3T3-L1 adipocytes, and giving the crude polysaccharide of Grifola frondosa with 1 nM of insulin caused glucose uptake to increase to a similar level as giving 50 nM of insulin alone. To confirm the mechanism for the anti-diabetic effect of the crude polysaccharides, we performed further examinations within $KK-A^{y}$ mice, an animal model of type 2 diabetes. The crude polysaccharides reduced blood glucose levels in the $KK-A^{y}$ mice for 2 weeks after feeding, and also significantly lowered plasma insulin levels. These results suggest that the anti-diabetic mechanism of the crude polysaccharide of Grifola frondosa is related to the enhancement of insulin sensitivity.

Effects of floor type and hanging type environmental enrichment on the behavior of growing pigs (바닥형과 현수형 환경 보조물이 육성돈의 행동에 미치는 영향)

  • Animal Welfare is spreading throughout the world, but remains weak in Korean swine farms. Therefore, the aim of this study was to identify the effect of floor type and hanging type environmental enrichment on the behavior of growing pigs under the traditional feeding environment. A total of 45 crossbred pigs (Yorkshire${\times}$Landrace${\times}$Duroc; average weight, $33.35{\pm}5.5kg$) were assigned into three treatments consisting of control, hanging type(T1) and floor type(T2) with three replicates in semi-slurry pen. The hanging enrichment was suspended at shoulder height of the pigs, and the floor enrichment was fixed in the center of the pens. Growth and cortisol were estimated at the end of the experimental period. Behaviour patterns were analyzed on the first and eighth days after starting the experiment. Growth was not differed among control and treatments. However, cortisol was decreased in T2 compared to control(25.28 vs. 46.75 ng/mL; p<0.05). On the first day, movement and aggression were lower in both treatments than in control(p<0.01). On the eighth day, time and frequency of playing action were increased in T2 compared to T1(p<0.01) and both treatment groups showed more active behaviour than control(p<0.01). These results suggested that the enrichment may meet the natural action requirement of pigs. Therefore, our data can be utilized as basic information for welfare with environmental enrichments in farm animals.

Effects of supplementation of dietary betaine on apparent nutrient digestibility and physiological responses in finishing pigs (사료 내 비테인 첨가 급여가 비육돈의 영양소 소화율 및 생리학적 변화에 미치는 영향)

  • This study was conducted to investigate the effects of supplementation of dietary betaine on nutrient digestibility and physiological responses in finishing pigs. A total of twelve pigs with a body weight of $80.1{\pm}3.7kg$ were individually caged, and randomly assigned to one of the two experimental diets containing 0 (control) or 5 g/kg (treatment) of the betaine in a $2{\times}2$ Latin square design. The experimental period was 14 days-7 days adaptation and 7 days trial period-per phase. All data for the difference between control and treatment groups were statistically analyzed by student's t-test. Dry matter and crude protein digestibility in the treatment group were significantly improved by 1% and 1.3%, respectively, as compared with those in the control (p<0.05). The apparent absorption of dietary energy was increased from 82.3% to 83.7% by dietary betaine supplementation. Thus, the retention of energy was also significantly increased to above 6% in the treatment group compared with the control group (control 4,057 vs treatment 4,314 kcal; p<0.01). The physiological parameters indicating serum biochemical contents and stress-, immune-, and inflammatory- responses were not changed by the supplementation of dietary betaine. In conclusion, dietary betaine improves the nutrient digestibility without any negative effects in terms of physiology in finishing pigs. It suggests that the supplementation of dietary betaine may increase the productivity through the improvement of weight gain and feeding efficiency.

Effect of $Bio-Silverlite^{(R)}$ on Performance, Weight of Organ, Intestinal Villus and Intestinal Microbial in Broiler Chicks (브로일러 사료에 $Bio-Silverlite^{(R)}$의 첨가가 사양성적, 장기중량, 소장 Villus 및 장내 미생물 성상에 미치는 영향)

  • This study was conducted to investigate the feeding effect of $bio-silverlite^{(R)}$ on growth performance, organ phenomenon and cecum microflora in broiler chicks. The $bio-silverlite^{(R)}$ was made by an ion exchange between illite and $silver(Ag^+)$. There were four treatment groups: negative control group(non-treatment), antibiotic supplement group (positive control), 0.5% $bio-silverlite^{(R)}$ supplment group and 1.5% $bio-silverlite^{(R)}$ supplement group. Total 200 birds was assigned for this five replication tests, allocating 10 birds into each treatment. Experimental diets were formulated on isocalories and isonitrogen for the whole experimental period. Body weight gain was higher in antibiotic supplementation (+C) and $bio-silverlite^{(R)}$ supplement groups(S 0.5% and 51.5%) than the negative control group(-C), and feed efficiency was significantly enhanced with increase of the level of $bio-silverlite^{(R)}$ supplement. The length of small intestine was longer in +C than in -C and $bio-silverlite^{(R)}$ supplement groups (P<0.05), and the weight of small intestine was proportional to the level of $bio-silverlite^{(R)}$ supplement. Crop weight was lower in $bio-silverlite^{(R)}$ supplement group than in -C and +C groups (P<0.05), and the cecum weight was heavier in $bio-silverlite^{(R)}$ supplementation group. Intestinal villi height was longer in 51.5% group at 3 weeks and 6 weeks of age than in -C and +C groups. With the respect of the formation of intestinal microflora, TBC and CBC was not affected by age and feed additive. However, the number of LAB was slightly higher in $bio-silverlite^{(R)}$ supplement group than in -C and +C groups.

Effects of Dietary Effective Microorganism (EM) on Growth Performance, Microflora Population and Noxious Gas Emission in Broiler (유용 미생물 첨가가 육계 생산성, 맹장 내 균총 및 유해가스 발생량에 미치는 영향)

  • This study investigated the effects of dietary supplementation with the effective microorganism (EM) on the growth performance, blood parameter, small intestinal microflora, and noxious gas emission of broilers. A total 720 1-d old ROSS 308 was randomly assigned to 6 dietary treatment groups: control, virginiamycin (6 mg/kg), 0.1% PB 0.1% EM, 0.5% EM, and 1.0% EM. Each treatment was fed to 4 replicates of 30 birds per diet for d 35. Two-phase feeding program with a starter diet from 0 to 3 wk, and a finisher diet from 4 to 5 wk was used in the experiment. Within each phase, a diet was formulated to meet or exceed NRC requirements of broilers for macro- and micronutrients. The diet and water were available ad libitum. Result indicated that during overall periods of the experiment, final weight, body weight gain, and feed intake were not different among dietary treatments. Feed conversion ratio was less (P<0.05) for EM treatments than control, antibiotics, and PB. Total cholesterol (TC), triglyceride (TRG), glucose (GLU), total protein (TP), calcium (CA), aspartate aminotransferase (AST) and alanine aminotransferase (ALT) were not different among dietary treatments. White blood cell (WBC), hemoglobin (Hb), heterophils (HE), lymphocyte (LY), monocytes (MO), and eosinophils (EO) were not different among dietary treatments. HE:LY was less (P<0.05) for EM0.5 treatments than control, antibiotics, and PB. Lactobacillus was greater (P<0.05) for EM treatments than control and antibiotics. E. coli and Salmonella were not different among dietary treatments. $NH_3$ and $CO_2$ wereless (P<0.05) for EM treatments than control. These results indicated that EM treatments were effective feed conversion ratio, noxious gas emission and micro flora population on the cecum in broilers.

Effects of Supplementing Aqueous Direct-Fed Microbials on In Vitro Fermentation and Fibrolytic Enzyme Activity in the Ruminant Nutrition (반추가축영양에 있어서 액상미생물제제의 첨가가 In Vitro 발효성상과 섬유소분해효소활성에 미치는 영향)

  • This study was conducted to determine effects of supplementation levels of aqueous direct-fed microbials (DFM; Bacillus spp.) to TMR(exp. 1.) and aqueous DFM addition under the various ratios of starch and cellulose(exp. 2.) on ruminal fermentation and fibrolytic enzyme activity. In experiment 1, ruminal fluids taken from rumen-cannulated Holstein cows were incubated during 24 hr by using TMR as substrates. Aqueous DFM was applied at a rate of 0, 0.025 and 0.05%, respectively. The pH of 0.025% treatment was not significantly different from that of control at 6 and 9 hr, but it was significantly lower (P<0.05) than 0.05% treatment. Concentrations of ammonia-N and VFAs were not affected by supplementing aqueous DFM. The A:P ratio of 0.05% treatment was significantly increased(P<0.05) by supplementation of aqueous DFM as compared with that of control at 24 hr. Although overall fibrolytic enzyme activities were not significantly affected by supplementing aqueous DFM, CMCase(carboxymethylcellulase) activity showed significant increase(P<0.05) compared to control at 6hr. However, the xylanase activity of 0.05% treatment significantly decreased(P<0.05) at 12 hr due to the application of aqueous DFM. There was no significant difference for in vitro dry matter disappearance among treatments. In experiment 2, ruminal fluids were incubated under the condition of various ratios of starch to cellulose(90:10, 70:30, 50:50, 30:70 and 10:90) with or without aqueous DFM(0.025%). Ruminal pH was unaffected by the addition of aqueous DFM, however, as increased level of starch, ruminal pH partially showed significant decrease(P<0.05). Ammonia-N concentration was not affected by aqueous DFM and ratio of starch and cellulose. On 9 hr incubation, DFM addition at a ratio of 70:30 showed significantly (P<0.05) lower value of ammonia-N(35.65 mg/dL) than that(65.05 mg/dL) of control. Concentrations of VFAs were significantly increased(P<0.05) by aqueous DFM addition compared with control at the same ratio on 6 hr incubation. The overall CMCase activity was not affected by aqueous DFM addition. However, the xylanase activity by aqueous DFM partially showed significant differences at the ratios of 90:10, 30:70 and 10:90. Our results indicated that supplementation of aqueous DFM did not significantly improve in vitro fermentation and fibrolytic enzyme activity. In addition, the DFM utilized in this study did not show consistent results by having various effects on ruminal fermentation under different feeding regimens.

Effect of Dietary Silkworm Droppings on Chemical Composition of Pork Loin (잠분(蠶糞) 급여가 돈육 등심의 화학적 조성에 미치는 영향)

  • This study investigated the effects of silkworm droppings(SWD) added diet feeding on chemical composition of pork loin meat. Pigs were divided into 7 treatment groups(10 pigs/group) and subjected to one of seven treatment diets(O, 1 0/0, 2 0/0, 3% SWD for 4 weeks, 1 0/0, 2 0/0, 3% SWD for 8 weeks, SWD diets; total fed diets) before slaughter. Pork loin were collected from the animals(1l0 kg body weight) slaughtered at a commercial slaughter house. Pork loin meat were aerobic packaged and then stored at$4^{\circ}C$ for 2, 5, 8, or 12 days. Samples were analyzed for TBARS, sensory evaluation, myoglobin content, fatty acid composition, amino acid and mineral content. TBARS value was not significantly different between the control and SWD treatment groups. Myoglobin content of SWD fed group pork was significantly increased than that of control group(P<0.05). Marbling score and acceptability were increased in SWD fed pork compared to those in control pork. Na, Mg, Ca, Mn and Fe content of SWD fed group pork was significantly decreased than those of control group(P<0.05). P content was significantly increased than that of control group(P< 0.05). In the change of amino acid composition, the contents of aspartic acid, serine and tyrosine were decreased by dietary SWD-supplementation, whereas the SWD-supplementation resulted in the higher glutamic acid, alanine and isoleucine. As dietary SWD was increased in feed, the content of oleic acid was significantly increased, but stearic acid was significantly decreased(P<0.05).
