Fed-batch fermentations were carried out in order to improve the efficiency of hirudin production by recombinant Saccharomyces cerevisiae. A fed-batch fermentation done with the optimized semi-synthetic medium resulted in a maximum hirudin concentration of 342mg/$\ell$ by keeping a galactose concentrations between 10 and 30g/$\ell$ which corresponded to a 11.4-fold increase in hirudin concentration compared with the simple bach fermentation done with the same medium. Comparison of the chromatographic pattern of proteins in the growth medium clearly showed that the use of the semi-synthetic medium is more advantageous for separation of hirudin than the case o fusing the complex medium. Continuous cell recycle fermentation done at dilution rate of 0.1h-1 and an inlet galactose concentration of 100g/$\ell$ yielded a maximum hirudin productivity of 19.1mg hirudin/$\ell$$.$h.
Fed-batch cultures of Enterococcus faecalis RKY1 were performed to maximize the L(+)-Iactic acid concentration in the bioreactor. The highest lactic acid concentration was obtained at around 225 g/L by intermittent feeding the concentrated glucose media containing 500 g/L of glucose and 15 g/L (or 75 g/L) of yeast extract. However, in all fed-batch cultures, volumetric productivities of lactic acid gradually decreased due to the inhibitory effect of lactic acid produced during the fermentation. The highest value of lactic acid concentration obtained in this work corresponded to around 1.5-fold increase compared with conventional batch fermentation.
A thermostable $\alpha$-L-arabinofuranosidases ($\alpha$-L-AFase) is an industrially important enzyme for recovery of L-arabinose from hemicellulose. The recombinant $\alpha$-L-AFase from Thermotoga maritima was expressed in Escherichia coli by using a constitutive pHCE or an inducible pRSET vectors. In batch fermentation, the constitutive expression system resulted in slightly faster growth rate (0.78 vs. 0.74/hr) but lower enzyme activity (2,553 vs. 3,723 units/L) than those of the induction system. When fed-batch fermentation was performed, biomass and enzyme activity reached the highest levels of 36 g/L and 9,152 units/L, respectively. The fed batch cultures performed superior results than batch culture in terms of biomass yield (4.62-5.42 folds) and enzyme synthesis (3.39-4.00 folds). In addition, the fed-batch induction strategy at high cell density resulted in the best productivity in cell growth as well as enzyme activity rather than the induction method at low cell density or the constitutive expression.
Production of poly-$\beta$-hydroxybutyrate (PHB) in fed-batch fermentation was studied. Utilization of carbon for PHB biosynthesis was investigated by using feeding solutions with different ratios of carbon to nitrogen (C/N). It was observed that at a high C/N ratio carbon source was more preferably utilized for PHB accumulation while its consumption for cellular metabolism appeared to be more favored at a low C/N value. A high cell concentration (184 g/l) was achieved when ammonium hydroxide solution was fed to control the pH, which was also utilized as the sole nitrogen source. For the mass production of PHB, two-stage fed-batch operations were carried out where PHB accumulation was observed to be stimulated by switching the ammonium feeding mode to the nitrogen limiting condition. A large amount of PHB (108 g/l) was obtained with cellular content of 80% within 50 hrs of operation.
A process for the production of mannitol from fructose (5% to 25%) using Leuconosyoc mesenteroides NRRL B-1149 was investigated. Fermentations were carried out in bat도 of fed-batch fermentations without aeration at 28℃, pH 5.0. When 5% fructose was used in batch culture fermentation, the yield of mannitol was 78% of that expected theoretically. When the fructose concentration was increased to 10%, the yield dropped to 59.6% of the theoretical value. However, in the fed-batch culture, using 10% fructose, the yield was 81.9% of the theoretical value. In a 15% fructose fed-bat도 culture, with 5% fructose being added initially and the other 10% fructose being added as a continuous supply, the final yield was 83.7% of the theoretical yield. When 20% fructose was used in the same manner, the yield was 89.5% of theoretical yield.
The green alga, Dunaliella salina under fed-batch cultivation produced 51.12 mg of hydrocarbon per liter with maintaining 0.313 (g dry wt/l). About 20% of hydrocarbon production yield based on dry biomass was obtained from both batch and fed-batch processes. Optimum culture conditions of light intensity, pH and salt concentration were obtained as 0.0080 (kJ/$cm^2$/h), 8.0 and 1.4 (g of NaCl/l), respectively by response surface analysis. The production of hydrocarbons in D. salina was closely correlated to cell growth. Fed-batch cultivation produced more hydrocarbons and maintained better cell growth than a batch process.
The artificial gene coding for anticoagulant hirudin was placed under the control of the GAL 10 promoter and expressed in the galactokinase-deficient strain (Δgal1) of Saccharomyces cerevisiae, which uses galactose only as a gratuitous inducer in order to avoid its consumption. For efficient production of recombinant hirudin, a carbon source other than galactose should be provided in the medium to support growth of the Δgal1 strain. Here we demonstrate the successful use of glucose in the fed-batch fermentation of the Δgal1 strain to achieve efficient production of recombinant hirudin, with a yield of up to 400 mg hirudin/L.
An extractive fermentation process using pervaporation was studied in a 7 liter fermentor. Pervaporation was performed using a silicone membrane module and a low-acid-producing strain Clostridium acetobutylicu, B18 was used to produce butanol. In batch culture without pervaporation pH 5.5 and initial glucose concentration of 60 g/L resulted in the highest butanol productivity (0.216 g/L$.$h) with butanol yield of 0.261 Butanol flux through the membrane was best at 2.0 L/min-tubing of air flow rate In batch and fed-batch fermentation glucose consumption rate increased by 1.3 times with pervaporation.
