Effects of ruminally degradable starch levels on performance, nitrogen balance, and nutrient digestibility in dairy cows fed low corn-based starch diets

  • Objective: This trial was performed to examine the effects of ruminally degradable starch (RDS) levels in total mixed ration (TMR) with low corn-based starch on the milk production, whole-tract nutrient digestibility and nitrogen balance in dairy cows. Methods: Eight multiparous Holstein cows (body weight [BW]: $717{\pm}63kg$; days in milk [DIM]: $169{\pm}29$) were assigned to a crossover design with two dietary treatments: a diet containing 62.3% ruminally degradable starch (% of total starch, low RDS) or 72.1% ruminally degradable starch (% of total starch, high RDS). Changes to the ruminally degradable levels were conducted by using either finely ground corn or steam-flaked corn as the starch component. Results: The results showed that dry matter intake, milk yield and composition in dairy cows were not affected by dietary treatments. The concentration of milk urea nitrogen was lower for cows fed high RDS TMR than low RDS TMR. The whole-tract apparent digestibility of neutral detergent fiber, acid detergent fiber and crude protein decreased, and that of starch increased for cows fed high RDS TMR over those fed low RDS TMR, with no dietary effect on the whole-tract apparent digestibility of dry matter and organic matter. The proportion of urinary N excretion in N intake was lower and that of fecal N excretion in N intake was higher for cows fed high RDS TMR than those fed low RDS TMR. The N secretion in milk and the retention of N were not influenced by the dietary treatments. Total purine derivative was similar in cows fed high RDS TMR and low RDS TMR. Consequently, estimated microbial N flow to the duodenum was similar in cows fed high RDS TMR and low RDS TMR. Conclusion: Results of this study show that ruminally degradable starch levels can influence whole-tract nutrient digestibility and nitrogen balance in dairy cows fed low corn-based starch diets, with no influence on performance.

Effect of Simulated Heat Stress on Digestibility, Methane Emission and Metabolic Adaptability in Crossbred Cattle

  • The present experiment was conducted to evaluate the effect of simulated heat stress on digestibility and methane ($CH_4$) emission. Four non-lactating crossbred cattle were exposed to $25^{\circ}C$, $30^{\circ}C$, $35^{\circ}C$, and $40^{\circ}C$ temperature with a relative humidity of 40% to 50% in a climatic chamber from 10:00 hours to 15:00 hours every day for 27 days. The physiological responses were recorded at 15:00 hours every day. The blood samples were collected at 15:00 hours on 1st, 6th, 11th, 16th, and 21st days and serum was collected for biochemical analysis. After 21 days, fecal and feed samples were collected continuously for six days for the estimation of digestibility. In the last 48 hours gas samples were collected continuously to estimate $CH_4$ emission. Heat stress in experimental animals at $35^{\circ}C$ and $40^{\circ}C$ was evident from an alteration (p<0.05) in rectal temperature, respiratory rate, pulse rate, water intake and serum thyroxin levels. The serum lactate dehydrogenase, aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase activity and protein, urea, creatinine and triglyceride concentration changed (p<0.05), and body weight of the animals decreased (p<0.05) after temperature exposure at $40^{\circ}C$. The dry matter intake (DMI) was lower (p<0.05) at $40^{\circ}C$ exposure. The dry matter and neutral detergent fibre digestibilities were higher (p<0.05) at $35^{\circ}C$ compared to $25^{\circ}C$ and $30^{\circ}C$ exposure whereas, organic matter (OM) and acid detergent fibre digestibilities were higher (p<0.05) at $35^{\circ}C$ than $40^{\circ}C$ thermal exposure. The $CH_4$ emission/kg DMI and organic matter intake (OMI) declined (p<0.05) with increase in exposure temperature and reached its lowest levels at $40^{\circ}C$. It can be concluded from the present study that the digestibility and $CH_4$ emission were affected by intensity of heat stress. Further studies are necessary with respect to ruminal microbial changes to justify the variation in the digestibility and $CH_4$ emission during differential heat stress.

말 분변을 이용한 주스박의 in vitro 발효 특성 (In vitro Fermentation Characteristics of Juice Pomaces Using Equine Fecal Inoculum)

  • 본 연구는 바실러스, 효모 그리고 유산균으로 발효한 다양한 주스박들과 말 분변을 첨가한 in vitro 발효 시험을 통해 pH, 건물 분해율, 암모니아성 질소, 가스 발생량, 휘발성 지방산의 변화를 조사하였다. 주스박 첨가에 따른 발효액의 pH의 범위는 6.4~7.1로 미생물 발효에 의한 소화가 정상적으로 나타났다. 48시간 후 발효액에서 $10^9CFU/m{\ell}$ 정도 미생물이 증식함에 따라 사용한 주스박들이 장내 미생물에 의한 소화작용에 유용하게 이용될 것으로 판단되었다. 건물 분해율은 배양 12시간 이후부터 증가하여 48시간째에 사과박 39.19%, 포도박 38.22%, 당근박 37.02%, 감귤박 36.2% 그리고 혼합박 34.35%로 나타났다. 암모니아성 질소의 농도는 배양 12시간까지는 전 처리구에서 $1.5mg/100m{\ell}$의 수준이었으며 배양 24시간 이후부터 급격히 증가하였다. 가스 발생량은 혼합박이 가장 낮았으며, 배양 12시간 이후부터 급격히 증가하였다. 총 VFA는 배양 24시간부터 증가하여 48시간에 가장 높았다. 본 연구결과로부터 발효 주스박은 말 in vitro 소화과정에서 적정 pH를 유지하면서 건물 분해가 진행되었고 말의 맹장과 대장에서 섬유소 분해 미생물의 작용이 활발히 이루어질 수 있음을 제시하였다.

Molecular Analysis of Colonized Bacteria in a Human Newborn Infant Gut

  • The complex ecosystem of intestinal micro flora is estimated to harbor approximately 400 different microbial species, mostly bacteria. However, studies on bacterial colonization have mostly been based on culturing methods, which only detect a small fraction of the whole microbiotic ecosystem of the gut. To clarify the initial acquisition and subsequent colonization of bacteria in an infant within the few days after birth, phylogenetic analysis was performed using 16S rDNA sequences from the DNA iso-lated from feces on the 1st, 3rd, and 6th day. 16S rDNA libraries were constructed with the amplicons of PCR conditions at 30 cycles and $50^{\circ}C$ annealing temperature. Nine independent libraries were produced by the application of three sets of primers (set A, set B, and set C) combined with three fecal samples for day 1, day 3, and day 6 of life. Approximately 220 clones ($76.7\%$) of all 325 isolated clones were characterized as known species, while other 105 clones ($32.3\%$) were characterized as unknown species. The library clone with set A universal primers amplifying 350 bp displayed increased diversity by days. Thus, set A primers were better suited for this type of molecular ecological analysis. On the first day of the life of the infant, Enterobacter, Lactococcus lactis, Leuconostoc citreum, and Streptococcus mitis were present. The largest taxonomic group was L. lactis. On the third day of the life of the infant, Enterobacter, Enterococcus faecalis, Escherichia coli, S. mitis, and Streptococcus salivarius were present. On the sixth day of the life of the infant, Citrobacter, Clostridium difficile, Enterobacter sp., Enterobacter cloacae, and E. coli were present. The largest taxonomic group was E. coli. These results showed that microbiotic diversity changes very rapidly in the few days after birth, and the acquisition of unculturable bacteria expanded rapidly after the third day.

  • This study aimed to determine the effects of dietary microalgae (Tetradesmus sp. (TO)) on intestinal immunity and microbiota of pre-starter broilers. One hundred and twenty 1-day-old birds (Ross 308) were allocated to two dietary treatment groups with six blocks in a randomized complete block design. The two experimental diets consisted of a corn-soybean meal-based basal diet and a diet with 0.5% TO powder instead of cornstarch in the basal diet. After feeding the experimental diets for ten days, all birds' body weight and feed intake were measured, and representative eight birds were selected from each treatment group. Small intestinal lamina propria cells were isolated using flow cytometry to examine the frequency of immune cells. Cecal feces were harvested for 16s rRNA gut microbiota analysis and fecal IgA levels. Here, we found that 0.5% TO supplementation increased CD3+CD4+ T cells in the small intestine, but decreased CD3+CD8+ T cells in the small intestine. Gut microbial analysis showed that TO supplementation significantly increased the alpha diversity of the gut microbiome. Taxonomic analysis showed that TO treatment increased the abundance of Firmicutes and decreased that of Bacteroidetes at the phylum level. The distribution of Enterobacteriaceae containing many harmful bacteria at the family level, was lower in the TO group. In the LEfSe analysis, the TO group had a significantly enriched abundance of Agathobaculum at the genus level. Overall, results show that Tetradesmus sp. supplementation influences intestinal T-cell immunity and induces the expansion of beneficial gut microbes in pre-starter broiler chickens.

학교급식에 Cook/Chill System 적용을 위한 품질보증연구(II) - 돼지불고기 - (A Quality Assurance Study for the Application of Cook/Chill System in School Foodservice Operation (II) - Pork Bulgogi (Broiled Sliced Pork with Sauces) -)

  • 본 연구는 학교급식에서 Cook/chill system으로 생산 가능한 음식으로 돼지 불고기를 선정하고 모의실험을 통해 급식생산체계를 반복 실시함으로서 식품 위해 분석 중요관리점(HACCP)을 규명하고, 저장기간중의 음식품질 평가를 통해 합리적인 저장기한을 설정하고자 수행되었으며, 그 연구결과는 다음과 같다. 돼지불고기는 냉동상태로 운반되지 않아 온도상승이 일어난 검수 단계를 제외하면 각 생산 단계별 기준이 준수된 양호한 상태 하에서 생산된 것으로 나타난다. 생산단계별 미생물 분석결과, 원재료($4.26{\pm}0.11\;Log\;CFU/g$)에서 중온균수가 조리하지 않은 식품 기준 이내이나 다소 높게 나타났고, 양념장($5.97{\pm}0.04\;Log\;CFU/g$)에서는 조리하지 않은 식품 기준에 근접한 수준이었으며, 양념으로 재우는 과정($5.56{\pm}0.21\;Log\;CFU/g$)도 위험 수준이었다. 가열 조리 후 최종 음식온도가 $8.25{\pm}3.54^{\circ}C$에 도달하였으나 중온균수($5.17{\pm}0.04\;Log\;CFU/g$)가 급식 단계 음식기준을 초과한 위험한 수준이었고, 기타 미생물은 검출되지 않았다. 급속 냉각과 저장 1일, 3일, 5일 동안도 중온균수가 급식단계 음식 기준에 근접한 위험한 상태였고 기타 미생물은 검출되지 않았다. 재가열 처리에 의해 저장 1일($4.62{\pm}0.22\;Log\;CFU/g$), 3일($4.55{\pm}0.20\;Log\;CFU/g$), 5일($4.25{\pm}0.16\;Log\;CFU/g$) 모두 중온균수는 감소하여 급식단계 음식기준에 충족한 상태가 되었다. 배분 3조건에서도 급식단계 음식 기준 이내에 들었다. 저장 5일간 이화학 분석 결과 pH, 산가, 휘발성 염기 질소 모두 저장 5일에 유의적으로 증가하였고, 관능평가에서는 모든 항목들이 유의적인 차이를 보이지 않았다. 티아민 정량 분속 결과 ,가열 전의 티아민 함량을 100%으로 했을 때, 가열 후에는 78.6%로 손실이 일어났으며, 냉각과 저장 1일, 3일은 티아민 손실을 거의 일으키지 않는 것으로 나타났다. 그러나 저장 5일에는 티아민이 현저히 저하되어 62.5% 보유에 그쳤다. 저장기간에 따른 미생물적, 이화학적, 관능적 품질을 분석한 결과와 티아민 함량의 변화를 고려하여 돼지불고기의 저장기한을 3일로 제안하며, 생산 단계별 온도-소요시간 측정 및 미생물 분석을 통해 규명된 중요관리점은 돼지고기와 양념장재료인 파, 마늘, 생강의 구입 및 검수, 가열조리, 냉각, 저장, 재가열과 배식단계였다.

청보리 사일리지 첨가 발효사료가 임신돈의 분 성상 및 악취물질 농도에 미치는 효과 (The Effect of Fermented Diet with Whole Crop Barley Silage on Fecal Shape and Odorous Compound Concentration from Feces in Pregnant Sows)

  • 본 연구는 청보리 사일리지가 첨가된 발효사료를 임신돈에 급여하였을 때 분에서의 악취물질 농도와 미생물 변화를 조사하고, 변비예방 효과를 확인하기 위하여 수행되었다. 1. in vitro 시험결과 페놀은 타피오카, 비트펄프, 밀기울, 루핀종실에서 검출되지 않았으며, p-크레졸은 9.62~52.11 mg/L 범위로 검출되었는데, 루핀종실이 가장 높았고, 타피오카가 가장 낮았다. 또한 타피오카와 비트펄프에서는 인돌과 스카톨이 검출되지 않아 악취저감용 원료사료로 타피오카와 비트펄프가 가장 유용한 것으로 평가되었다. 2. 비트펄프를 기초로 배합한 악취저감사료를 28일간 임신돈에 급여한 후 분을 채취하여 성상을 조사한 결과 대조구에 비하여 발효사료 급여구에서 수분함량이 높고 경도가 낮았으며, 대장균 (E. coli) 수는 적었으며, 유산균 (lactobacilus) 수는 많았다. 3. 돼지의 분 중 p-크레졸과 스카톨 농도를 측정한 결과 대조구에서는 각각 33%와 67%가 검출한계 이하로 조사되었으나, 발효사료 구에서는 각각 67%와 100% 검출한계 이하로서 조사되어 발효사료를 섭취한 돼지의 분에서 악취가 감소될 수 있었다. 4. 따라서 임신돈에 청보리사일리지가 첨가된 발효사료를 급여하였을 때 대장의 기능이 개선되어 변비예방과 악취감소의 효과를 기대할 수 있을 것이다.

가축분퇴비와 유기질비료에서 병원성박테리아의 분포도 분석 (Prevalence of Pathogenic Bacteria in Livestock Manure Compost and Organic Fertilizer)

  • 최근에 건강식품의 선호에 따라 신선 채소류 등의 소비가 증가하면서 퇴비등 유기농자재의 안전성에 대한 관심이 점점 증가하고 있다. 본 연구는 우리나라를 경기도, 강원도, 충청도, 전라도, 경상도로 크게 5그룹으로 구분한 후 각 지역에서 국내 축산의 주요 축종인 소, 돼지, 닭의 분뇨를 주원료로 생산, 유통되는 가축분 퇴비 32종과 박(粕) 종류가 주원료인 유기질 비료 28종을 수집하여 총균수, 대장균군 E. coli, Salmonella spp., E. coli O157:H7, S.aureus, L. monocytogenes, C. sakazakii, B. cereus 등 미생물 오염도를 분석하고자 수행되었다. 가축분 퇴비 32종과 유기질 비료 28종의 총균수는 각각 $5.09{\sim}9.68log\;CFU\;g^{-1}$, $3.16{\sim}7.09log\;CFU\;g^{-1}$이었다. 총균수에서는 유기질 비료가 가축분 퇴비보다 더 낮은 미 생물 분포를 보였으며, 특히 가축분 퇴비의 경우에는 총 32종 중 약 88%가 $7log\;CFU\;g^{-1}$ 이상의 수준을 보였다. 가축분 퇴비와 유기질 비료에서 대장균군은 각각 32종 중 4종 (12.5%)에서, 28종 중 4종 (14.2%)에서 검출되었고, E. coli은 각각 32종 중 2종 (6.3%), 28종 중 1종 (3.5%)에서 검출되었다. 대장균군은 가축분 퇴비에서, 그 범위가 $2.77{\sim}4.63log\;CFU\;g^{-1}$, 평균 $3.42log\;CFU\;g^{-1}$ 수준으로 나타났다. 반면 유기질 비료에서, 그 범위는 $1.00{\sim}3.25log\;CFU\;g^{-1}$, 평균 $2.42log\;CFU\;g^{-1}$ 수준으로 나타났다. E. coli은 가축분 퇴비 32종 중 2종에서 $2.62{\sim}2.85log\;CFU\;g^{-1}$ 수준으로 검출되었으며, 유기질 비료에서 $1.85log\;CFU\;g^{-1}$ 수준으로 검출되었다. 가축분 퇴비에서 Salmonella spp., E. coli O157:H7, S. aureus, L. monocytogenes, C. sakazakii 등은 검출되지 않았고, B. cereus는 3종 (9%)에서 검출되었다. 유기질 비료에 Salmonella spp., E. coli O157:H7, S. aureus, L. monocytogenes, C. sakazakii, B. cereus 등은 전혀 검출되지 않았다. 따라서 병원성 미생물을 사멸시키기 위해서는 충분한 부숙 과정이 필요하다고 판단된다.

Cefoperazone(T-1551)의 약리학적 연구 (Pharmacological Studies of Cefoperazone(T-1551))

  • The pharmacological and microbiological studies of Cefoperazone (T-1551, Toyama Chemical Co., Japan) were conducted in vitro and in vivo. The studies included stability and physicochemical characteristics, antimicrobial activity, animal and human pharmacokinetics, animal pharmacodynamics and safety evaluation of Cefoperazone sodium for injection. 1) Stability and physicochemical characteristics. Sodium salt of cefoperazone for injection had a general appearance of white crystalline powder which contained 0.5% water, and of which melting point was $187.2^{\circ}C$. The pH's of 10% and 25% aqueous solutions were 5.03 ana 5.16 at $25^{\circ}C$. The preparations of cefoperazone did not contain any pyrogenic substances and did not liberate histamine in cats. The drug was highly compatible with common infusion solutions including 5% Dextrose solution and no significant potency decrease was observed in 5 hours after mixing. Powdered cefoperazone sodium contained in hermetically sealed and ligt-shielded container was highly stable at $4^circ}C{\sim}37^{\circ}C$ for 12 weeks. When stored at $4^{\circ}C$ the potency was retained almost completely for up to one year. 2) Antimicrobial activity against clinical isolates. Among the 230 clinical isolates included, Salmonella typhi was the most susceptible to cefoperazone, with 100% inhibition at MIC of ${\leq}0.5{\mu}g/ml$. Cefoperazone was also highly active against Streptococcus pyogenes(group A), Kletsiella pneumoniae, Staphylococcus aureus and Shigella flexneri, with 100% inhibition at $16{\mu}g/ml$ or less. More than 80% of Escherichia coli, Enterobacter aerogenes and Salmonella paratyphi was inhibited at ${\leq}16{\mu}/ml$, while Enterobacter cloaceae, Serratia marcescens and Pseudomonas aerogenosa were somewhat less sensitive to cefoperagone, with inhibitions of 60%, 55% and 35% respectively at the same MIC. 3) Animal pharmacokinetics Serum concentration, organ distritution and excretion of cefoperazone in rats were observed after single intramuscular injections at doses of 20 mg/kg and 50 mg/kg. The extent of protein binding to human plasma protein was also measured in vitro br equilibrium dialysis method. The mean Peak serum concentrations of $7.4{\mu}g/ml$ and $16.4{\mu}/ml$ were obtained at 30 min. after administration of cefoperazone at doses of 20 mg/kg and 50 mg/kg respectively. The tissue concentrations of cefoperazone measured at 30 and 60 min. were highest in kidney. And the concentrations of the drug in kidney, liver and small intestine were much higher than in blood. Urinary and fecal excretion over 24 hours after injetcion ranged form 12.5% to 15.0% in urine and from 19.6% to 25.0% in feces, indicating that the gastrointestinal system is more important than renal system for the excretion of cefoperazone. The extent of binding to human plasma protein measured by equilibrium dialysis was $76.3%{\sim}76.9%$, which was somewhat lower than the others utilizing centrifugal ultrafiltration method. 4) Animal pharmacodynamics Central nervous system : Effects of cefoperazone on the spontaneous movement and general behavioral patterns of rats, the pentobarbital sleeping time in mice and the body temperature in rabbits were observed. Single intraperitoneal injections at doses of $500{\sim}2,000mg/kg$ in rats did not affect the spontaneous movement ana the general behavioral patterns of the animal. Doses of $125{\sim}500mg/kg$ of cefoperazone injected intraperitonealy in mice neither increased nor decreased the pentobarbital-induced sleeping time. In rabbits the normal body temperature was maintained following the single intravenous injections of $125{\sim}2,000mg/kg$ dose. Respiratory and circulatory system: Respiration rate, blood pressure, heart rate and ECG of anesthetized rabbits were monitored for 3 hours following single intravenous injections of cefoperazone at doses of $125{\sim}2,000mg/kg$. The respiration rate decreased by $3{\sim}l7%$ at all the doses of cefoperazone administered. Blood pressure did not show any changes but slight decrease from 130/113 to 125/107 by the highest dose(2,000 mg/kg) injected in this experiment. The dosages of 1,000 and 2,000 mg/kg seemed to slightly decrease the heart rate, but it was not significantly different from the normal control. All the doses of cefoperazone injected were not associated with any abnormal changes in ECG findings throughout the monitering period. Autonomic nervous system and smooth muscle: Effects of cefoperazone on the automatic movement of rabbit isolated small intestine, large intestine, stomach and uterus were observed in vitro. The autonomic movement and tonus of intestinal smooth muscle increased at dose of $40{\mu}g/ml$ in small intestine and at 0.4 mg/ml in large intestine. However, in stomach and uterine smooth muscle the autonomic movement was slightly increased by the much higher doses of 5-10 mg/ml. Blood: In vitro osmotic fragility of rabbit RBC suspension was not affected by cefoperazone of $1{\sim}10mg/ml$. Doses of 7.5 and 10 mg/ml were associated with 11.8% and 15.3% prolongation of whole blood coagulation time. Liver and kidney function: When measured at 3 hours after single intravenous injections of cefoperaonze in rabbits, the values of serum GOT, GPT, Bilirubin, TTT, BUN and creatine were not significantly different from the normal control. 5) Safety evaluation Acute toxicity: The acute toxicity of cefoperazone was studied following intraperitoneal and intravenous injections to mice(A strain, 4 week old) and rats(Sprague-Dawler, 6 week old). The LD_(50)'s of intraperitonealy injected cefoperazone were 9.7g/kg in male mice, 9.6g/kg in female mice and over 15g/kg in both male and female rats. And when administered intravenously in rats, LD_(50)'s were 5.1g/kg in male and 5.0g/kg in female. Administrations of the high doses of the drug were associated with slight inhibition of spontaneous movement and convulsion. Atdominal transudate and intestinal hyperemia were observed in animals administered intraperitonealy. In rats receiving high doses of the drug intravenously rhinorrhea and pulmonary congestion and edema were also observed. Renal proximal tubular epithelial degeneration was found in animals dosing in high concentrations of cefoperazone. Subacute toxicity: Rats(Sprague-Dawley, 6 week old) dosing 0.5, 1.0 and 2.0 g/kg/day of cefoperazone intraperitonealy were observed for one month and sacrificed at 24 hours after the last dose. In animals with a high dose, slight inhibition of spontaneous movement was observed during the experimental period. Soft stool or diarrhea appeared at first or second week of the administration in rats receiving 2.0g/kg. Daily food consumption and weekly weight gain were similar to control during the administration. Urinalysis, blood chemistry and hematology after one month administration were not different from control either. Cecal enlargement, which is an expected effect of broad spectrum antibiotic altering the normal intestinal microbial flora, was observed. Intestinal or peritoneal congestion and peritonitis were found. These findings seemed to be attributed to the local irritation following prolonged intraperitoneal injections of hypertonic and acidic cefoperazone solution. Among the histopathologic findings renal proximal tubular epithelial degeneration was characteristic in rats receiving 1 and 2g/kg/day, which were 10 and 20 times higher than the maximal clinical dose (100 mg/kg) of the drug. 6) Human pharmacokinetics Serum concentrations and urinary excretion were determined following a single intravenous injection of 1g cefoperazone in eight healthy, male volunteers. Mean serum concentrations of 89.3, 61.3, 26.6, 12.3, 2.3, and $1.8{\mu}g/ml$ occured at 1,2,4,6,8 and 12 hours after injection respectively, and the biological half-life was 108 minutes. Urinary excretion over 24 hours after injection was up to 43.5% of administered dose.
