Cholesterol-lowering Effect of Rice Protein by Enhancing Fecal Excretion of Lipids in Rats

  • The aim of this study was to investigate the effects of isolated protein from white rice on lipid metabolism in a hypercholesterolemic animal model. Male Sprague-Dawley rats were divided into three groups and fed either a normal diet or a high-cholesterol diet (HCD) containing either casein or isolated rice protein for 4 weeks. Compared with rats fed a HCD with casein, the total cholesterol (TC) level in the plasma was significantly reduced in the rats fed rice protein. However, no significant differences were observed in the triglycerides, high-density lipoprotein (HDL), and glucose levels among the experimental groups. Hepatic total lipids and TC levels were significantly decreased by supplementation with rice protein. In addition, rice protein significantly increased the levels of TC and bile acids in the feces. These results suggest that rice protein may improve HCD-induced hypercholesterolemia by enhancing fecal excretion of cholesterol.

Hesperidin Lowers Activities of 3-Hydroxy-3-methylglutaryl-CoA Reductase and Acyl-CoA:Cholesterol Acyltransferase in Rats Fed High -Cholesterol Diet

  • In this study, a potential mechanism through which the hesperidin might work on the effect was examined in vivo. Male rats were fed a high cholesterol synthetic diet (1%, wt/wt) with hesperidin (0.1%, wt/wt) for 42 days. Activity of hepatic HMG-CoA reductase was significantly lowered by the hesperidin supplement compared to the control. Hesperidin did not significantly alter plasma or hepatic lipids, but tended to lower those lipid levels. Hesperidin also subsequently reduced the fecal neutral sterols compared to the control(253.3mg/d vs.521.9 mg/d). The inhibition of HMG-CoA reductase resulting from the hesperidin supplementation could count for the reduction in fecal neutral sterols that appears to compensate for the decreased cholesterol biosynthesis. The dose of hesperidin in a high choles-terol diet should apparently be more than 0.1% to exhibit the hypocholesterolemic response in these rats. It remains to be determined whether the observed alterations in cholesterol metabolism are specific to the rat or also could be applied to the humans.

Supplementary effect of Soybean oil and Rice germ oil on Lipid Metabolism in Insulin dependent Diabetic Mice (대두유와 쌀눈기름의 급여가 인슐린 의존형 당뇨 마우스의 지질대사에 미치는 영향)

  • This study was carried out to investigate the supplementary effects of soybean oil and rice gem oil compared with lard on lipid metabolism of insulin dependent diabetic mice. Streptozotocin-induced diabetic mice were fed three kinds of experimental diets with 20% lipid from lard(L), soy bean oil(SBO) and rice gem oil(RGO) for 7 weeks, respectively. Diet intake, body weight, organs weights and lipids levels of serum, liver and feces were measured. There was no significant difference in diet intake, body and organs weights among experimental groups. But the concentrations of serum triglyceride of SBO and RGO groups, and of serum total cholesterol were lower in RGO group than in the other groups. The hepatic total lipid and total cholesterol levels of RGO group were significantly lower than those of the other groups. The contents of total lipid and total cholesterol excreted in feces of SBO and RGO groups were higher than those of L group, and the significance was shown only in RGO group. These results suggested that soy bean oil and rice germ oil can reduce serum triglyceride and total cholesterol levels and hepatic total lipid concentration of insulin dependent diabetic mice compared with lard as a animal fat source by increasing fecal lipid excretion of these groups. But the significant reducing effects on serum and liver lipid levels were shown only in RGO group, and we need to investigate the hypolipidemic effect of this oil by supplementary level and period.

Effects of Dehulled Defatted Soy Flour on Cholesterol Metabolism and Gastronitestinal Physiology in Rats (거피 탈지 콩가루 식이가 흰쥐의 콜레스테롤 대사와 소화생리에 미치는 영향)

  • This study was carried out to explore the hypocholesterolemic effect of dehyulled defatty soy flour and its possible mechanisms including endocrine status, cholesterol biosynthesis, and fecal excretion in rats. Animals fed casein were used as control and each phospholipid compared with casein feeding. Cholesterol concentrations in all lipoprotein fraction were significantly lower in defatted soy flour group compared with casein-fed control. Defatted soy flour feeding also significantly lowered hepatic total lipid, cholesteol and TG, and increaed fecal bile acid excretion by 270% compared with casein feeding. Defattd soy flour feeding had no significant effect on plasma thyroid hormone levels and hepatic 3-hydroxy-3-methyl glutary coenzyme A(HMG-CoA) reductase activity. However, plasma T4 concentration was slightly elevated and HMG-CoA reductase activity was suppressed in defatted soy flour group. These metabolic alterations partially explain the reduced plasma and hepatic cholesterol levels of rats fed defatted soy flour.

Effects of Dried Leaf Powders, Water and Ethanol Extracts of Persimmon and Green Tea Leaves on Lipid Metabolism and Antioxidative Capacity in 12-Month-Old Rats (감잎, 녹차의 건분 및 물, 에탄올추출물이 노령쥐의 지방대사와 항산화능에 미치는 영향)

  • The effects of dried leaf powders and water and ethanol extracts of persimmon and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity were investigated in 12-month-old rats. Forty-nine male Sprague-Dawley rats weighing 520$\pm$19g were blocked into seven groups according to body weight. Rats were raised for four weeks with control(no tea leaf powder or extracts) and experimental diets containing either 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) or green tea(Camellia sinensis O. Ktze), or water or ethanol extract from equal amounts of each dried tea powder. Food intakes of all tea diet groups were higher than that of control. Weight gains and food efficiency ratios of all tea diet groups were not significantly different from those of control. All tea diets decreased plasma triglyceride level, especially, green tea powder and persimmon ethanol diets were more effective than other diet. All the tea diet groups showed decrease in liver triglyceride level, and persimmon powder and ethanol extract increased fecal triglyceride excretion. Plasma cholesterol levels of all the tea diet groups were not significantly different from the control, but control. Fecal cholesterliver cholesterol concegroups were significantlntrations of all tea y lower than that of ol excretions of persimmon powder, green tea ethanol extract, persommon ethanol extract and green tea ethanol extract groups were significantly higher than that of control. Plasma and liver thiobarbituric acid reactive substance(TBARS) concentrations of all the tea diet groups were lower than that of control. Especially, plasma TBARS concentrations of green tea powder and persimmon ethanol extract groups were sinificantly low. Red blood cell(RBC) superoxide dismutase(SOD) activities of persimmon ethanol extract and green tea water extract groups were increased, and RBC catalase activities of all experimental groups were not significantly different. RBC glutathione peroxidase(GSH-px) activities of persimmon ethanol extract, persimmon water extract and green tea powder groups were increased. Liver SOD activities of all the tea diet groups except green tea ethanol extract group were higher than that of control. Liver catalase activities of all experimental groups were not significantly different, and liver GSH-px activity of green tea powder group was significantly higher than that of control. In conclusion, dried leaf powders, and water and ethanol extracts of persimmon and green tea were effective in lowering lipid level, inhibiting lipid peroxidation and increasing antioxdative enzyme activities in 12-month-old rat. Green tea leaf powder with high contents of flavonoids and water soluble dietar fiber was most effective in lowering plasma triglyceride, cholesterol and TBARS level.   • This study was conducted to evaluate the effect of whole egg supplementation on the blood lipid profiles and cholesterol levels of C57BL/6 mice. Sixty-six mice were divided into two groups: normal-diet supplemented and high-cholesterol diet supplemented. Lyophilized whole egg powder was mixed with the two diets at 2 and 10%: normal diet only, normal diet with 2 and 10% whole egg powder, high cholesterol diet only, high cholesterol diet with 2 and 10% whole egg powder. The mixed diets were fed for 5 wk and feeding condition (body weight change, feed intake, and feed efficiency ratio (FER)), blood lipid profiles (total cholesterol (TC), triglyceride (TG), high density lipoprotein-cholesterol (HDL-C), low density lipoprotein cholesterol, hepatic and fecal lipids (TG, TC)), and fecal bile acids were determined. No significant differences were found in body weight gain or FER after whole egg supplementation in both the normal and high-cholesterol diet fed groups. In the normal-diet fed mice, HDL-C increased significantly in the 2 and 10% whole-egg powder groups. In the high-cholesterol diet fed mice, administering 10% egg powder increased the atherogenic index compared to the control. Furthermore, administration of whole egg powder increased fecal bile acids dose dependently (p<0.05). These results indicate that administering 2% hen whole egg powder did not affect blood lipid profiles and was more beneficial for health by increasing HDL-C and aiding in the excretion of cholesterol by fecal bile acids than those in the control.

Effect of Marine Protein Supplementation on Lipid Profile of Growing Rats Compared to Soybean Protein and Casein

  • Although beneficial effects of dietary plant proteins on lipid metabolism are well documented, not much information exists on the influence of different seafood proteins on the lipid metabolism. The present study evaluated the effect of 2 marine proteins (tuna protein and scallop ovary proteins) in comparison to casein and soy protein in male Wistar rats. The concentration of total lipids in the plasma of rats fed experimental diets was significantly lower from that of control (278.2 mg/dL) group (p<0.05); and, the liver lipid content was not significantly different (p>0.05). Fecal excretion of cholesterol and bile acids was significantly higher in marine proteins and soy protein fed groups compared to casein only fed control (6.1 and 6.4 mg/day, respectively) group (p<0.05). No significant differences were observed in the mRNA concentrations of different transcriptional factors (p>0.05).

The effects of black garlic (Allium satvium) extracts on lipid metabolism in rats fed a high fat diet

  • BACKGROUD/OBEJECTIVES: The mechanism of how black garlic effects lipid metabolism remains unsolved. Therefore, the objectives of this study were to determine the effects of black garlic on lipid profiles and the expression of related genes in rats fed a high fat diet. MATERIALS/METHODS: Thirty-two male Sqrague-Dawley rats aged 4 weeks were randomly divided into four groups (n=8) and fed the following diets for 5 weeks: normal food diet, (NF); a high-fat diet (HF); and a high-fat diet + 0.5% or 1.5% black garlic extract (HFBG0.5 or HFBG1.5). Body weights and blood biochemical parameters, including lipid profiles, and expressions of genes related to lipid metabolism were determined. RESULTS: Significant differences were observed in the final weights between the HFBG1.5 and HF groups. All blood biochemical parameters measured in the HFBG1.5 group showed significantly lower values than those in the HF group. Significant improvements of the plasama lipid profiles as well as fecal excretions of total lipids and triglyceride (TG) were also observed in the HFBG1.5 group, when compared to the HF diet group. There were significant differences in the levels of mRNA of sterol regulatory element binding protein-1c (SREBP-1c), acetyl-CoA carboxylase (ACC), fatty acid synthase (FAS), and glucose-6-phosphate dehydrogenase (G6PDH) in the HFBG1.5 group compared to the HF group. In addition, the hepatic expression of (HMG-CoA) reductase and Acyl-CoA cholesterol acyltransferase (ACAT) mRNA was also significantly lower than the HF group. CONCLUSIONS: Consumption of black garlic extract lowers SREBP-1C mRNA expression, which causes downregulation of lipid and cholestrol metahbolism. As a result, the blood levels of total lipids, TG, and cholesterol were decreased.

Effects of Dietary Calcium and Sodium Levels on Lipid Metabolism in Hyperlipidemic/Hypercholesterolemic Rats (고지혈증 모델 흰쥐에서 칼슘과 소디움 섭취수준이 체내 지질대사에 미치는 영향)

  • The effects of dietary Ca and Na levels on lipid metabolism in hyper lipidemic/hypercholesterolemic rats were examined. In Expt. 1, normal rats were divided into six groups and fed high fat(15%, w/w)/cholesterol(1%, w/w) diet containing two levels of Na, low (0.05) or high(1.5%) and three levels of Ca, low(0.1%), normal (0.5%), or high(1.5%) for 8 weeks. In Expt. 2, hyperlipidemia / hypercholesterolemia rats were induced by feeding high fat / cholesterol diet for 4 weeks. They were divided into four groups and fed the high fat / cholesterol diet, containing two levels of Na, low or high and two levels of Ca, low or high for 4 weeks. In Expt. 1, total lipid and total cholesterol contents in serum and liver were significantly lower in rats fed high Ca diet than in rats fed normal or low Ca diet regardless of dietary Na levels. Serum TG was the highest in rats fed low Ca and low Na diet. In Expt. 2, Serum total lipid, TG, and total cholesterol levels decreased by 24, 35, 26% respectively in rats fed high Ca diet regardless of dietary Na levels. Serum total lipid level tended to increased in rats fed low Na diet. The total lipid and TG contents in liver slightly decreased in rats fed high Ca diet. Another observation was that high Ca intake significantly faciliated the fecal lipid and cholesterol excretion regardless of dietary Na levels. There results suggest that the hypolipidemidc/hypocholesterolemic effects of high Ca diet could be partly due to increase in lipid and cholesterol excretion and these effects may be independent of dietary Na levels.

Effect of Young Barley Leaf on Lipid Contents and Hepatic Lipid-Regulating Enzyme Activities in Mice Fed High-Fat Diet (보리순이 고지방을 급여한 마우스의 지질 함량과 간조직의 지질대사 관련 효소활성에 미치는 영향)

  • This study was conducted to investigate the effects of powdered young barley leaf and its water extract on body weight and lipid metabolism in high-fat fed mice. Male mice were divided into normal group, high-fat (HF) group, high-fat group supplemented with powdered young barley leaf (HF-YBL) and high-fat group supplemented with water extract of the powdered young barley leaf (HF-WYBL). The powdered young barley leaf or its water extract was added to a standard diet based on 1% dried young barley leaf (1 g YBL/100 diet and 0.28 g WYBL/100 g diet) for 8 weeks. Supplementation of YBL and WYBL significantly reduced body weight and epididymal adipose tissue weight in high-fat fed mice. Food intake and daily energy intake were significantly lower in the YBL group than in the HF group. After 8 weeks, plasma triglyceride and cholesterol concentrations were significantly higher in the HF group than in the Normal group; however, both YBL and WYBL significantly lowered those of the high-fat fed mice. The ratio of HDL-cholesterol/total cholesterol of the YBL and WYBL groups were significantly elevated compared to that of HF group. Both YBL and WYBL significantly increased fecal excretion of triglyceride in high-fat fed mice, whereas they did not affect fecal cholesterol concentration. The triglyceride levels of liver, adipose tissue and heart were significantly lower in the YBL and WYBL groups than in the HF group. Supplementation of WYBL also lowered the kidney triglyceride and heart cholesterol concentrations compared to those of HF group. Hepatic lipid regulating enzyme activities, fatty acid synthase, HMG-CoA reductase and acyl-coenzyme A: cholesterol acyltransferase, were significantly lower in the YBL and WYBL groups than in the HF group. Accordingly, these results suggest that YBL and WYBL improve plasma and organ lipid levels partly by increasing fecal lipid excretion and inhibiting fatty acid and cholesterol biosynthesis in the liver.
