Oenothera biennis, commonly known as evening primrose, a potential source of natural bioactive substances: flavonoids, steroids, tannins, fatty acids and terpenoids responsible for a diverse range of pharmacological functions. However, whether extract prepared from aerial part of O. biennis (APOB) protects skin against oxidative stress remains unknown. To investigate the protective effects of APOB against oxidative stress-induced cellular damage and elucidated the underlying mechanisms in the HaCaT human skin keratinocytes. Our results revealed that treatment with APOB prior to hydrogen peroxide ($H_2O_2$) exposure significantly increased viability, and the highest DPPH radical-scavenging activities and reducing power of HaCaT cells. APOB also effectively attenuated H2O2-induced comet tail formation and inhibited the $H_2O_2$-induced phosphorylation levels of the histone ${\gamma}H2AX$, as well as the number of apoptotic bodies and Annexin V-positive cells. In addition, APOB exhibited scavenging activity against intracellular reactive oxygen species (ROS) accumulation and restored the mitochondrial membrane potential loss by $H_2O_2$. Moreover, $H_2O_2$ enhanced the cleavage of caspase-3 and degradation of poly (ADP-ribose)-polymerase (PARP), a typical substrate protein of activated caspase-3, as well as DNA fragmentation; however, these events were almost totally reversed by pretreatment with APOB. Furthermore, APOB increased the levels of heme oxygenase-1 (HO-1), which is a potent antioxidant enzyme, associated with the induction of nuclear factor-erythroid 2-related factor 2 (Nrf2). According to our data, APOB is able to protect HaCaT cells from $H_2O_2$-induced DNA damage and cell death through blocking cellular damage related to oxidative stress through a mechanism that would affect ROS elimination and activating the Nri2/HO-1 signaling pathway.

Physicochemical characteristics of rice milled by newly designed abrasive milling machine were studied. Crude protein and crude lipid contents were 7.81∼8.50% and 1.07∼0.95%, respectively, and they showed a tendency to decrease with milling times. However, they showed higher values than that of polished rice. Major fatty acids of crude lipid were palmitic acid, oleic acid, and linoleic acid. Whiteness of tested samples was higher than brown rice but lower than polished rice. The remain rate of rice germ of four pass-through and polished one time sample (FPS) was 39.33%, and it was higher than that of polished rice (4.00%). One thousand grain weight of samples over than three pass-through was significantly different from brown rice (p<0.05). FPS had smooth surface similar to polished rice and it showed some residual rice germ by scanning electron microscopy. Sensory evaluation showed that the taste of three pass-through rice, four pass-through rice, and FPS was not significantly different from polished rice, whereas their color, odor, texture, and overall acceptability had lower score than polished rice but higher than brown rice. Therefore, we estimate that rice ground over three pass-through by newly designed abrasive milling machine might be a good source having nutritive value.

A feeding experiment for 160 days was conducted to evaluate effects of dietary soybean meal (SBM) on the apparent digestibility, ammonia excretion and body composition of rainbow trout (Oncorhynchus mykiss). Seven diets containing $0\~70\%$ SBM were formulated based on iso-nitrogenous and iso-caloric basis of $44\%$ crude protein and approximately 4,200 Kcal/kg of gross energy. To evaluate the nutritional utilization of the SBM diets at the end of 60 and 120-day rearing with test diets, digestion rates of protein and lipid of SBM diets were measured. Fish fed the diets containing above $46\%$ SBM showed higher apparent digestibility for protein, but lower for lipid than did fish fed the control diet in both trials. Protein and fat contents in the carcass were similar for all experimental fish, except for fish fed $58\%$ and $70\%$ SBM which showed lower fat content than the others, but composition of fatty acid and amino acids were not affected by dietary SBM levels. Gill and urinary post-prandial ammonia ($NH_3-N$) excretions were measured at 12 and 24 hours after single feeding of the experimental diets. Excretions of $NH_3-N$ measured after 24 hours were almost 1 times higher than those measured after 12 hours. Total excretion fer the 24 hours by fish fed $70\%$ SBM was $15\%$ higher than that of fish fed the control diet. Results of present study may suggest that the rainbow trout which were fed above $34%\;or\;46\%$ of SBM diet showed a decrease gradually in lipid bioavailability compared to the control group.

This study was to investigate the chemical compositions of germinated (GSS)- and ungerminated (UGSS)-safflower (Carthamus tinctorius) seeds. GSS had higher amount of sugar and crude fiber than UGSS, but less amounts of protein and lipid. Levels of $\alpha$-tocopherol and essential amino acids of GSS were higher than those of UGSS, although there are no difference in fatty acid composition between GSS and UGSS. Among the nine phenolic compounds detected, five phenolic compounds, except for two lignans and two flavonoids, were found in both GSS and UGSS. Four serotonin derivatives accounted for about 80 per cent of total phenolic compounds, and levels of five phenolic compounds decreased slightly with germination. These results suggest that germination may enhance the functionality of safflower seed by increasing nutritional compositions and by decreasing phenolic compounds with bitter taste and cathartic effects.

Data are limited on biological mechanisms underlying the associations of sleep insufficiency with obesity and dyslipidemia. To explore these mechanisms, we investigated appetite-regulating hormones, stress-related hormones, and cardiometabolic indicators in association with sleep fragmentation, which is a type of sleep disorder. In an experimental study, we randomly allocated 40 Wistar male rats aged 7 weeks into four groups; rats with ad libitum sleep and ad libitum intake (Control), those exposed to sleep fragmentation (SF), those with diet restriction (DR), and those exposed to sleep fragmentation and diet restriction (SF+DR). Amongst them, 13-day chronic sleep fragmentation was applied to the SF and SF+DR groups while 50% reduction in chow intake was applied to the DR and SF+DR groups for 13 days. After these experiments, blood lipid and lipoprotein profiles, leptin, ghrelin, adiponectin, cortisol, epinephrine, and norepinephrine levels were compared among the four groups. In the results, the SF group showed the highest levels of serum ghrelin (P<0.001) and the lowest levels of serum adiponectin (P<0.01). All experimental groups showed higher levels of low density lipoprotein-cholesterol (LDL-C) than the Control (P<0.001). LDL-C levels and the ratio of LDL-C and high density lipoprotein-cholesterol were positively correlated with ghrelin levels (P<0.05) in the SF group, but not in the DR and SF+DR groups. In the SF group, the highest levels of serum free fatty acids were also observed and correlated with lower levels of serum adiponectin, which reflects insulin resistance (P<0.05). Based on these findings, we suggest that chronic sleep fragmentation may induce disturbances in lipid metabolism and appetite-regulating hormones independent of food intake, and these metabolic disturbances may be worse due to insulin resistance related to overeating, which is indicated by elevated ghrelin levels in sleep fragmentation. For persons with sleep insufficiency, anti-atherogenic dietary interventions may be recommended to prevent cardiovascular disease.
Purpose : In recent years, many countries have adopted newborn screening programs that use tandem mass spectrometry (MS/MS) to screen and the number of diseases screened has also increased. We began screening for inherited metabolic disorders using MS/MS in April, 2001. Our goal was to determine the overall prevalence of metabolic disorders and to assess the effectiveness of newborn screening by MS/MS in Korea. Methods : From April, 2001 to December, 2007, we screened newborns and high risk groups using MS/MS. Acylcarnitines and amino acids were extracted and butylated and were introduced into the inlet of MS/MS. Confirmatory testing including a repeat newborn screening, and urine organic acid and plasma amino acid analysis were performed on a case-by-case basis. Results : The total number of screened subjects 284,933 which comprised 251,799 neonates and 33,134 high risk subjects. The recall rate was 0.4% (1158 tests) and true positive cases were 117 (0.04%). Confirmed metabolic disorders (newborn/high risk group) were as follows; 78 (25/53) amino acid disorders, 27 (16/11) organic acid disorders, and 12 (5/7) fatty acid oxidation disorders. The estimated prevalence of inherited metabolic diseases in newborns was 1:5,000 and that in the total group was 1:2,000. Conclusion : Newborn screening by MS/MS improved the detection of many inherited metabolic disorders. We therefore propose that all newborns be screened by a MS/MS national program and followed-up using a systemic organization strategy.
Background: Secretory phospholipase $A_2$ ($sPLA_2$) are a group of extracellular enzymes that release fatty acids at the sn-2 position of phospholipids. Group IIA $sPLA_2$ ($sPLA_2$-IIA) has been detected in the inflammatory fluids, and its plasma level increases in the inflammatory disease. This study examined the effect of $sPLA_2$-IIA on mouse macropahges in order to investigate the potential mechanism of $sPLA_2$-induced inflammation. Materials and Methods: Wild type $PLA_2$ and mutant H48Q $PLA_2$ were purified from HEK293 cells transfected with the corresponding plasmids, and the $PLA_2$ activities were measured using 1-palmitoyl-2-[1-$^{14}C$]linoleoyl-3-phosphatidylethanolamine as substrates. The TNF-${\alpha}$ and IL-6 released in the supernatants were determined by ELISA. In addition, the TNF-${\alpha}$ and IL-6 mRNA were analyzed by RT-PCR. Results: $sPLA_2$-IIA stimulated the production of TNF-${\alpha}$ and IL-6 in a dose- and time-dependent manner. In addition, the effect of $sPLA_2$-IIA on cytokine production from the macrophage was found to be associated with the accumulation of their specific mRNA. The mRNA levels of TNF-${\alpha}$ and IL-6 peaked at 2 and 6 hours in a time-dependent manner, respectively. Conclusion: In conclusion, the production of proinflammatory cytokine might be mediated by the binding of $sPLA_2$-IIA to the receptors.
An experiment was conducted to examine the effect of a high environmental temperature on the performance, conformation, and carcass characteristics of pigs and the influence of diet. Thirty-six three-way crossed castrated male pigs with average initial body weight of 50.4 kg were used in the experiment. The pig were allocated to the following treatments: two environmental temperatures (20 and $30^{\circ}C$)${\times}$three dietary energy levels (2,980, 3,300 and 3,600 kcal/kg)${\times}$three protein levels (12.8, 15.2 and 17.2%). Daily weight gain was lighter (p<0.01) and feed: gain ratio lower (p<0.05) in pigs at $30^{\circ}C$ than for pigs at $20^{\circ}C$ The pigs at $30^{\circ}C$ were significantly taller with deeper bodies (p<0.05) and significantly longer (p<0.05) both vertically horizontally in the planum nasal when kept at $30^{\circ}C$. The width of body and the circumference of the neck were greater (p<0.05) at $20^{\circ}C$. The lean meat of the loin, middle section, ham, and hind section were significantly greater (p<0.05) in pigs kept at $30^{\circ}C$ and the belly was significantly heavier. The total unsaturated free fatty acids were significantly higher (p<0.05) in the body fat of pigs kept at $20^{\circ}C$ than in that of pigs at $30^{\circ}C$. The results indicated that when pigs are given very restricted same amounts of feed, they may need less energy to maintain their body temperature under moderately high environmental temperature ($30^{\circ}C$); consequently, their performance is better than that of pigs under optimum environmental temperature. The results showed very clearly that weight gain of pigs increased with increasing of dietary protein and energy content. The increasing of dietary protein content seemed more significant when content increasing to 17.2% compared with the 12.8 and 15.2% protein content treatments. The increasing of dietary energy content was more significant when content increasing to 3,600 compared with the other low energy content treatments.
Fifteen crossbred cattle bulls of about 22-24 months age (mean body weight ranged from 291.0 to 298.1 kg) were randomly divided into 3 groups of 5 animals following randomized block design. Animals in group 1 were fed concentrate mixture containing wheat bran 97%, mineral mixture 2% and salt 1% plus ad libitum chopped green sugarcane tops (SCT) as a roughage source. Animals in group 2 were fed concentrate mixture containing lentil chuni 97%, mineral mixture 2% and salt 1% while group 3 concentrate contained wheat bran 48.5%, lentil chuni 48.5%, mineral mixture 2% and salt 1% plus ad libitum chopped green SCT to meet their maintenance requirements. During metabolism trial period, the mean total intake and digestibility of DM in groups 1, 2 and 3 was $8.09{\pm}0.41$, $8.41{\pm}0.49$ and $7.86{\pm}0.16kg/d$ and 60.24, 63.24 and 65.05%, respectively. The intake of CP (p<0.05) and EE (p<0.01) was significantly higher in group 2 and group 1, respectively as compared to remaining 2 groups, which were comparable. Digestibility of CP and EE was significantly (p<0.01) higher in group 3 and that of CP was comparable with group 2. The intake and digestibility of total carbohydrates, NDF, ADF, Hemi-cellulose and cellulose were similar among 3 groups. The mean body weight changes in group 1, 2 and 3 were 173.4, 253.4 and 203.4 g/d, respectively, which was significantly (p<0.05) higher in group 2. The total nitrogen (N) and phosphorus (P) intakes were significantly higher in group 2 and group 1, respectively. The total volatile fatty acids, ammonia-N, trichloro-acetic acid precipitable nitrogen (TCA ppt.-N; p<0.05), total-N concentrations and pH were significantly (p<0.01) higher in group 3, however, pH and TCA ppt.-N was comparable with group 2 and 1, respectively. From the results, it may be deduced that the feeding of chopped green SCT supplemented with a concentrate mixture having both wheat bran (48.5%) and lentil chuni (48.5%) has shown a better performance with respect to intake and digestibility of nutrients and growth as well.

The present study was conducted with twenty-four Korean native (Hanwoo) steers to observe the effect of mixed oil and monensin supplementation and duration of feeding on c9,t11-CLA content in plasma and fat tissues. The steers were randomly assigned to three groups of eight animals each according to body weight. Hanwoo steers in the control group were fed the commercial concentrate for the late fattening stage. The other groups of steers were fed the same diet as control steers, but the concentrate was supplemented with high-$C_{18:2}$ oil mixture (soybean oil, sunflower oil, safflower oil) and fish oil at 6% level of concentrate (DM basis), and monensin (20 ppm). The second and third group of steers was fed the oil mixture supplemented diet with monensin for the last 10 weeks and 20 weeks, respectively, prior to being slaughtered. The oil mixture consisted of 45% soybean oil, 20% sunflower oil, 20% safflower oil and 15% fish oil. Average daily gain (p<0.049) and feed efficiency (p<0.018) of the steers fed the diet supplemented with oil mixture and monensin (OM-M) for 20 weeks were higher than those of the other groups of steers. Dressing percent, fat thickness and longissimus muscle area were not affected by the OM-M supplementation and duration of its feeding. The OM-M supplementation increased the content of total-cholesterol (p<0.0001-0.0007) and HDL-cholesterol (p<0.0001) in the plasma of steers compared to the control diet. The steers fed the OM-M diet had a higher proportion of c9,t11-CLA in plasma (p<0.048-0.044) than the control steers. Feeding the OM-M diet for 20 weeks increased the proportion of CLA in intramuscular (p<0.015), intermuscular (p<0.039) and subcutaneous (p<0.001) fat tissues compared with both steers fed the control diet and the OM-M diet for 10 weeks. Increased (p<0.007) proportion of total unsaturated fatty acids in steers fed the OM-M diet for 20 weeks compared to those in control steers was related to the increased (p<0.001) $C_{18:2}$ and decreased (p<0.001) $C_{18:0}$ proportions in subcutaneous tissue.
