Alpha-linolenic acid (ALA) is one of n-3 polyunsaturated fatty acids and found mainly in the chloroplasts. Many studies have been reported that intracellular reactive oxygen species (ROS) in mammalian oocytes were reduced by supplementation of ALA in in vitro maturation (IVM) medium. Based on these reports, we expected that ALA acts as an antioxidant during IVM of porcine oocytes. Therefore, the objective of this study was to investigate the antioxidant effect of ALA supplementation during IVM in porcine oocytes. The cumulus-oocyte complexes (COCs) were incubated in IVM medium containing $200{\mu}m$$H_2O_2$ or $H_2O_2$ with $50{\mu}m$ ALA for 44 h. Nuclear maturation stage of oocytes was evaluated using aceto-orcein method. For measurement of oxidative stress state, intracellular ROS and glutathione (GSH) levels were measured using carboxy-DCFDA and cell tracker red, respectively. In results, oocytes in metaphase-II (MII) stage development was significantly reduced in $H_2O_2$ group compared to non-treated control group $61.84{\pm}1.42%$ and 80.00%, respectively; p<0.05) and it was slightly recovered by treatment of ALA ($69.76{\pm}1.67%$; p<0.05). The intracellular GSH levels was decreased in $H_2O_2$ groups compared with control groups, but it was enhanced by ALA treatment (p<0.05). On the contrary, $H_2O_2$ treatment increased intracellular ROS level in oocytes and $H_2O_2$-induced ROS was decreased by treatment of ALA (p<0.05). Our findings suggested that ALA treatment under oxidative stress condition improve oocyte maturation via elevated GSH and reduced ROS levels in oocytes. Therefore, these results suggest that ALA have an antioxidative ability and it could be used as antioxidant in in vitro production system of porcine embryo.

It is necessary to recycle and reuse waste paper for preservation of the global environment. Instead of a solid deinking agent, liquid deinking agents have been made using various fatty acids with EO/PO added for higher deinking effect. The removal efficiency of ink on waste paper was evaluated by the ERIC and brightness properties after processes of recycling pulping (sample composition ONP: OMG = 5:5, concentration 15%, temperature $45^{\circ}C$, pulping time 20 min.) and flotation (concentration 1%, temperature $45^{\circ}C$, air flow 7 L/min, time 8 min.). Also, the ink reattachment of paper was evaluated by hyperwashing. The results show that it is effective for mass-preparation to saponify oleic acid with KOH at $80^{\circ}C$ and to add EO/PO. This deinking agent was used in a paper plant, and we evaluated the brightness, color, and ERIC. The deinking agent showed excellent performance.

We hypothesized that supplementing finishing diets with palm oil would promote adipogenic gene expression and stearoyl-CoA desaturase (SCD) gene expression in subcutaneous (s.c.) and intramuscular (i.m.) adipose tissues of feedlot steers. Eighteen Angus and Angus crossbred steers were assigned to three groups of 6 steers and fed a basal diet (control), with 3% palm oil, or with 3% soybean oil, for 70 d, top-dressed daily. Tailhead s.c. adipose tissue was obtained by biopsy at 14 d before the initiation of dietary treatments and at 35 d of dietary treatments. At slaughter, after 70 d of dietary treatment, tailhead s.c. adipose tissue and i.m. adipose tissue were obtained from the longissimus thoracis muscle. Palm oil increased plasma palmitic acid and soybean oil increased plasma linoleic acid and ${\alpha}$-linolenic acid relative to the initial sampling time. Expression of AMP-activated protein kinase alpha ($AMPK{\alpha}$) and peroxisome proliferator-activated receptor gamma ($PPAR{\gamma}$) increased between the initial and intermediate biopsies and declined thereafter (p<0.03). SCD gene expression did not change between the initial and intermediate biopsies but declined by over 75% by the final period (p = 0.04), and G-coupled protein receptor 43 (GPR43) gene expression was unaffected by diet or time on trial. Soybean oil decreased (p = 0.01) $PPAR{\gamma}$ gene expression at the intermediate sample time. At the terminal sample time, $PPAR{\gamma}$ and SCD gene expression was less in i.m. adipose tissue than in s.c. adipose tissue (p<0.05). $AMPK{\alpha}$ gene expression was less in s.c. adipose tissue of palm oil-fed steers than in control steers (p = 0.04) and CCAAT enhancer binding protein-beta ($CEBP{\beta}$) gene expression was less in s.c. and i.m. adipose tissues of palm oil-fed steers than in soybean oil-fed steers (p<0.03). Soybean oil decreased SCD gene expression in s.c. adipose tissue (p = 0.05); SCD gene expression in palm oil-fed steers was intermediate between control and soybean oil-fed steers. Contrary to our original hypothesis, palm oil did not promote adipogenic gene expression in s.c. and i.m. adipose tissue.

Exposure to cold may affect growth performance in accordance with the metabolic and immunological activities of animals. We evaluated whether ambient temperature affects growth performance, blood metabolites, and immune cell populations in Korean cattle. Eighteen Korean cattle steers with a mean age of 10 months and a mean weight of 277 kg were used. All steers were fed a growing stage-concentrate diet at a rate of 1.5% of body weight and Timothy hay ad libitum for 8 weeks. Experimental period 1 (P1) was for four weeks from March 7 to April 3 and period 2 (P2) was four weeks from April 4 to May 1. Mean ($8.7^{\circ}C$) and minimum ($1.0^{\circ}C$) indoor ambient temperatures during P1 were lower (p<0.001) than those ($13.0^{\circ}C$ and $6.2^{\circ}C$, respectively) during P2. Daily dry matter feed intake in both the concentrate diet and forage groups was higher (p<0.001) during P2 than P1. Average daily weight gain was higher (p<0.001) during P2 (1.38 kg/d) than P1 (1.13 kg/d). Feed efficiency during P2 was higher (p = 0.015) than P1. Blood was collected three times; on March 7, April 4, and May 2. Nonesterified fatty acids (NEFA) were higher on March 7 than April 4 and May 2. Blood cortisol, glucose, and triglyceride concentrations did not differ among months. Blood CD4+, CD8+, and CD4+CD25+ T cell percentages were higher, while CD8+CD25+ T cell percentage was lower, during the colder month of March than during May, suggesting that ambient temperature affects blood T cell populations. In conclusion, colder ambient temperature decreased growth and feed efficiency in Korean cattle steers. The higher circulating NEFA concentrations observed in March compared to April suggest that lipolysis may occur at colder ambient temperatures to generate heat and maintain body temperature, resulting in lower feed efficiency in March.

An isotope dilution liquid chromatography tandem mass spectrometry was developed as a primary method for the quantitative analysis of cholesterol in infant formula. Cholesterol-d4 was used as an internal standard and spiked into the infant formula sample. In order to release cholesterol out of cholesteryl ester, which is cholesterol bound to fatty acids in infant formula, saponification was carried out. Saponification conditions were optimized with heating temperature, reaction time and the concentration of KOH. The optimum conditions were as follows; heating temperature was 70 ℃, reaction time was 180 min and the concentration of KOH was 0.8 mL of 8 M KOH for about 0.1 g infant formula sample. Extraction of cholesterol out of sample solution was carried out with hexane uisng liquid-liquid extraction. Chromatographic analysis was carried out using Phenomenex Kinetex C18 column. Mobile phase was 0.1% acetic acid in methanol/water (v/v, 99/1) and flow rate was 0.3 mL/min. Cholesterol and cholesterol-d4 were monitored at mass transfer m/z 369/259 and 373/263 respectively. Reproducibility of the method was evaluated to be 0.23% of the measurement result. The expanded uncertainty of the measurement result of cholesterol in infant formula was approximately 1.9% at a 95% confidence level. NIST standard reference material having certified values of cholesterol in infant formula, was analyzed in order to verify this method. The ID-LC/MS/MS results were well agreed with the certified values of NIST SRM within the uncertainty.

We measured the influence of antifungal antagonists Bacillus subtilis AH18 and Bacillus licheniformis K11 on soil microbial community in microcosms. Both antifungal antagonists were confirmed to suppress hot pepper phytophthora blight. Phospholipid fatty acids (PLFA) were analyzed to investigate the soil microbial community. B. subtilis AH18 changed the total PLFA composition and bio-indicators of PLFA, compared with other treatments. B. subtilis AH18 decreased the proportion of bacteria and gram negative/gram positive bacteria, and increased the fungi/bacteria and anaerobic/aerobic microorganisms. In addition cy19:0/18:$1{\omega}7c$, which means adaptation to unfavorable environmental conditions, was increased by the application of B. subtilis AH18. On the other hand the inoculation of B. licheniformis K11 or combined inoculation of both antifungal strains did not affect soil microbial community. The suppression of phytophthora blight and preservation of indigenous soil microbial community may be achieved by the combined application of B. subtilis AH18 and B. licheniformis K11.

This study investigated the physicochemical characteristics and antioxidant activities of Daehak waxy corns harvested at different times. Daehak waxy corns were harvested at 4 days before suitable time (BST), suitable time (ST) and 4 days after suitable time (AST). As harvest time was delayed, weight, length and width of kernel, the crude ash, lipid and protein contents significantly increased. Fructose, glucose and sucrose contents of BST were higher than those of ST and AST. Copper, sodium, calcium and potassium contents of BST in Daehak waxy corns increased on different times, and contents of ferrous and zinc did not show any significantly difference. The major fatty acids were oleic acid (48.58~49.96%), palmitic acid (19.50~20.64%) and palmitoleic acid (18.70~23.77%) in Daehak waxy corns with different harvest times. The highest total polyphenol content and total antioxidant capacity of EtOH extracts from AST were 104.45 mg/100 g and 20.53 mg AA eq/100 g, respectively. There was a high correlation between total polyphenol content and $IC_{50}$ value of electron donating ability of EtOH extracts from Daehak waxy corns harvested at different times, or total antioxidant capacity (p<0.01).

This study was conducted to develop a simple flood frequency questionnaire (FFQ) based on the results of contributions of specific floods to absolute intake and between-person variance in nutrients using semi-quantitative FFQ with 93 flood items. The subjects were 554 healthy adults for development of a simple FFQ, and 37 students for a validation test of a developed simple FFQ. The contribution of specific floods to 80% absolute nutrient intake was measured by assessing their percentage in total consumption of a nutrient. To assess the contributions of floods to the between-person variance in the intake of each specific nutrient, stepwise multiple regressions were performed. The number of floods necessary to account for the respective 80% of absolute intake was 11-36, depending on the nutrient, while flower floods (5-16 floods) were required for the corresponding percentage of between-person variation for all nutrients. Important floods for between-person variance include Tangsuyuk (pork) and snacks for energy and fat, fish for protein and polyunsaturated fatty acids (PUFA) and snacks for carbohydrates. Spearman correlation coefficients between 93-itemed FFQ and 63-itemed FFQ ranged from 0.91 for vitamin A to 0.99 for fat in the population data used in developing a simple FFQ. Also, the correlation coefficients between the two FFQs were 0.82-7.92 in the population for the validation test. This study suggests that useful information on dietary intake could be obtained using a simple semi-quantitative FFQ in a large-scale dietary survey in Korea.
Conjugated linoleic acid (CLA) is a collective term for positional and geometric isomers of octadecadienoic acid in which the double bonds are conjugated. CLA has anticancer activity in a variety of animal cancer models, and cis-9, trans-11 (c9t11) and trans-10, cis-12(t10c12) CLA are the most predominant isomers present in the synthetic preparations utilized in these animal studies. To compare the ability of c9t11, t10c12 and an isomeric mixture of CLA to inhibit TSU-Prl cell growth, cells were incubated in a serum-free medium with various concentrations of these fatty acids. The isomeric mixture inhibited cell growth in a dose-dependent manner (1-3 $\mu$M) with a 41 $\pm$ 1% inhibition observed at 3 $\mu$M concentration after 48 hours. T10c12 also inhibited cell proliferation in a dote-dependent manner, However, the efficacy and potency of this isomer was much greater than that of the isomeric mixture with a 49 $\pm$ 2% inhibition observed at 0.3 $\mu$M concentration after 48 hours. By contrast, c9t11 slightly increased cell proliferation. To determine whether the growth-inhibiting effect of CLA is related to the changes in production of insulin-like growth factors (IGF) and IGF-binding proteins (IGFBP) by these cells, serum-free conditioned media were collected. Immunoblot analysis of conditioned media using a monoclonal anti-IGF-II antibody showed that both the isomeric mixture and t10c12 inhibited secretion of both mature 7,500 Mr and higher Mr forms of pro IGF-II, whereas c9t11 had no effect. Ligand blot analysis with 125I-IGF-II revealed the presence of two types of IGFBPs : 24,000 Mr IGFBP-4 and 30,000 Mr IGFBP-6. The production of IGFBP-4 slightly decreased at the highest concentrations of the isomeric mixture and t10c12. These results indicate that CLA inhibits human prostate cancer cell growth, an effect largely due to the action of t10c12. The growth inhibition may result, at least in part, from decreased production of IGF-II and IGFBP-4 by these cells.
The purpose of this study is to search the optimal solubilization technology which could be applied to sewage sludge, and is to find the characteristics of biological hydrogen production when the sludge treated by optimal method was used as a sole substrate. As a result of the test, treatment technology mixed with alkali and ultrasonic treatment was very powerful tool for treating sewage sludge with high solubilization, and its ratio and elution rate of organic material was 0.9 and 0.076 $min^{-1}$, respectively. When the sludge treated by above optimal technology was used, 4.4 ml $H_2/g$ VSS of hydrogen was produced. Finally, When the sludge treated by above optimal technology was used, 13.4 ml $H_2/g$ VSS of hydrogen was produced under optimum pH.
