Oxidative Stability and Antioxidant Changes in Perilla Seeds and Perilla Oil Affected by UV Irradiation (들깨 및 들기름의 자외선 조사 중 지방질 산화와 산화방지제의 변화)

  • Effects of UV irradiation on lipid oxidation in perilla seeds and perilla oil were evaluated by determining the contents of peroxides, conjugated dienoic acids, and thiobarbituric acid reactive substances, and analyzing fatty acid composition. Tocopherols and polyphenol contents were also determined. Perilla seeds were unroasted or roasted at $180^{\circ}C$ for 20 min, and perilla oil was obtained by pressing the roasted perilla seeds. Lipid oxidation during UV irradiation was higher and faster in perilla oil than that in perilla seeds, with a slight loss of linolenic acid. Unroasted perilla seeds were more oxidation-stable than roasted seeds. Tocopherols and polyphenols were degraded during UV irradiation, with a higher degradation rate observed in unroasted perilla seeds than in roasted ones. Antioxidant concentration dependency of the lipid oxidation during UV irradiation was higher in perilla oil than that in perilla seeds, and the contribution of polyphenols to oxidative stability was higher than that of tocopherols in all samples.

The Hypoglycemic Effect of Complex of Chinese Traditional Herbs (CTH) and Macelignan in Type 2 Diabetic Animal Model (레이저 제2형 당뇨동물모델에서 macelignan과 한약제 열수 추출물의 병용효과)

  • This study investigated the efficacy of macelignan and hot water with Chinese traditional herb (CTH) extract on altering severe diabetic conditions in C57BL/KsJ-db/db mice. Previously, the anti-diabetic effects of macelignan were partly reported as a PPAR $\alpha/\gamma$-dual agonist. Here, we futher studied whether a combination of macelignan and CTH had more beneficial effects or not. The macelignan and CTH compound significantly decreased fasting blood glucose and HbA1c compared to macelignan-treated mice, and also significantly improved postprandial glucose, insulin sensitivity, and plasma lipid profiles (FFA, and TG). On the other hand, insulin levels were not significantly changed compared to the diabetic control group. There were no significant changes in the concentrations of total cholesterol and HDL-cholesterol, but there were changes in HTR and AI. These results suggest that the macelignan and CTH compound ameliorates hyperglycemia and efficiently improves postprandial glucose, insulin sensitivity, and hyperlipidemia compared with macelignan in db/db mice. Moreover, the macelignan and CTH compound seems to be more potent in affecting diabetic complications than macelignan.

Regulation of Acetyl-CoA Carboxylase Gene Expression by Hormones and Nutrients

  • This study was investigated to identify the regulatory mechanism of ACC gene expression by hormones and nutrition. The fragment of ACC promoter I (PI) -220 bp region was recombined to pGL3-Basic vector with luciferase as a reporter gene. The primary hepatocyte from the rat was used to investigate the regulation of ACC PI activity. ACC PI (-220 bp)/luciferase chimeric plasmid was transfected into primary rat hepatocyte by using lipofectin. ACC PI activity was shown by measuring luciferase activity. The addition of insulin, dexamethasone, and triiodothyronine to the culture medium increased the activity of ACC PI by 2.5-, 2.3- and 1.8-fold, respectively. In the presence of 1 $\mu$M dexamethasone, the effects of insulin was amplified about 1.2-fold showing the additional effects of dexamethasone. Moreover the activity of luciferase was increased by insulin, dexamethasone, and triiodothyronine treatment approximately 4-fold. These results indicated that insulin, dexamethasone and thyroid hormone coordinately regulate ACC gene expression via regulation of promoter I activity. On the -220 to +21 region of ACC PI, the addition of the glucose to the culture medium increased the activity of ACC PI. With 25 mM glucose, luciferase activity increased by 7-fold. On the other hand, on the -220 bp region, ACC PI activity was not changed by polyunsaturated fatty acids. Therefore, it can be postulated that there are response elements for insulin, triiodothyronine, dexamethasone, and glucose, but not PUFAs on the -220 bp region of ACC PI.

Efficient Anaerobic Digestion for Highly Concentrated Particulate Organic Wastewater (고농도 입자성 유기폐수의 고효율 혐기성 소화 공정)

  • It has been mentioned that CSTR (Completely Stirred Tank Reactor) and UASB (Upflow Anaerobic Sludge Blanket) processes, the existing anaerobic processes, have problems in the treatment of highly concentrated particulate organic wastewater (HCPOW). Therefore, this paper discusses the treatment possibility of distillery wastewater which is a typical HCPOW using ADEPT (anaerobic Digestion Elutriated Phased Treatment) process. In the comparison of CSTR and ADEPT, ADEPT produced much higher gas than that of CSTR removing more organic matters and suspended solids in ADEPT process, ADEPT had no effect on the decrease in pH by volatile fatty acids and showed steady pH in spite of relatively short HRT. In the results of removal rate according to recycle ratios between 6Qin and 2Qin in ADEPT, 6Qin showed high removal rate during the operation time. Therefore it appears that ADEPT had an applicability for the treatment of distillery wastewater. ADEPT could be a economical process, due to the short HRT, the energy recovery by the methane production, and the utilization for carbon source of produced organic acid from the ADEPT-acid reactor.

Molecular Structures and Catalytic Mechanism of Bacterial Lipases. (세균성 리파제의 분자구조와 작용기작)

  • Bacteria produce lipases, which can catalyze both the hydrolysis and the synthesis of long chain triglycerides. These reactions usually proceed with high regioselectivity and enantioselectivity, and, therefore, lipases have become very important biocatalysts used in organic chemistry. 3D lipase structures were solved from several bacterial lipases. They have an $\alpha/\beta$ hydrolase fold and a catalytic triad consisting of a nucleophilic serine, and an aspartate or glutamate residue that is hydrogen bonded to a histindine. Active sites are covered with $\alpha$-helical lid structure, of which movement is involved in the enzyme's activation at oil/water interface. Four substrate binding pockets were identified for triglycerides: an oxyanion hole and three pockets accommodating the fatty acids bound at positions sn-1, sn-2, and sn-3. These pockets determine the enantiopreference of a lipase. The understanding of structure-function relationships as well as the development of molecular evolution techniques will enable researchers to tailor new lipases for biotechnological applications.

Isolation and Characterization of Oil Degrading Bacteria from Southern Sea of Korea (남해안 해수로부터 원유 분해 세균의 분리 및 특성)

  • A marine bacterium having a high oil-degrading activity was isolated form the oil-polluted southern sea of Korea, and was identified as Pseudomonas aeruginosa and was named Pseudomonas aeruginosa BYK-2. The optimal tmeperatur, culture time, pH and NaCl concentration for biosurfactant production and cell growth showed $25^{\circ}C$, 48h, 7.0 and 0%(w/v), respectively. After cultivation at $25^{\circ}C$, 180 rpm in 250 mL erlenmeyer flask for 7days, 1%(w/v) arabian light crude oil and bunker C oil which are considered to be hardly degradable compounds were degraded 92.1%(w/w) and 76%(w/w) respectively. And then, cell adherence was measured on various carbon sources. The cell adherence indicated over 80% on hydrocarbons(arabian light crude oil, kuwait curde oil, bunker C oil, n-paraffine, n-hexadecane, n-tetradecane) as carbon sources. Lecithin among fatty acids(oleic acid, olive oil, lecithin) showed highest cell adherence of 91.5%. The cell adherence of sugars(arabinose, trehalose, dextrose, galactose, lactose, fructose, maltose, sorbitol, sucrose) observed to be less than 70% except for arabinose, galactose, sorbitol and sucrose.

The Conservation of the Mourning Clothes from the Kyonggi Provincial Museum (경기도 박물관 소장 상복에 대한 보존처리)

  • The purpose of this study was to clarify the performance of mourning clothes from Konggi Provincial Museum, to identify the fiber of these clothes, and investigate the washing effect by wet cleaning. Shapes of mourning clothes were studied, and some ingredients of non fibrous extracted matter were analyzed by FT-IR spectroscopy to clarify the performance of mourning clothes. Microscope examination, melting test, and stain test were used for the identification of the fiber. SEM was used to confirm the effect of washing after mourning clothes washed by wet cleaning added anionic detergent, sodiumdodecylbenzenesulfonate (LAS). The performance of these clothes was mourning cloth, not mummy cloth as results of analyses to the clothes' shapes and extract ingredients. The extract ingredients were carbohydrate, alkyl alcohol, and aldehyde. They didn't have any nitrogen compounds and fatty acids. The fiber identification showed this fiber was hemp. The effect of washing was high as the surface of fiber was clean and linear after wet washing.

Isolation and Characterization of an Agarase-Producing Bacterial Strain, Alteromonas sp. GNUM-1, from the West Sea, Korea

  • The agar-degrading bacterium GNUM-1 was isolated from the brown algal species Sargassum serratifolium, which was obtained from the West Sea of Korea, by using the selective artificial seawater agar plate. The cells were Gram-negative, $0.5-0.6{\mu}m$ wide and $2.0-2.5{\mu}m$ long curved rods with a single polar flagellum, forming nonpigmented, circular, smooth colonies. Cells grew at $20^{\circ}C-37^{\circ}C$, between pH 5.0 and 9.0, and at 1-10% (w/v) NaCl. The DNA G+C content of the GNUM-1 strain was 45.5 mol%. The 16S rRNA sequence of the GNUM-1 was very similar to those of Alteromonas stellipolaris LMG 21861 (99.86% sequence homology) and Alteromonas addita $R10SW13^T$(99.64% sequence homology), which led us to assign it to the genus Alteromonas. It showed positive activities for agarase, amylase, gelatinase, alkaline phosphatase, esterase (C8), lipase (C14), leucine arylamidase, valine arylamidase, ${\alpha}$-chymotrypsin, acid phosphatase, naphthol-AS-BI-phosphohydrolase, ${\alpha}$-galactosidase, ${\beta}$-galactosidase, ${\beta}$-glucosidase, catalase, and urease. It can utilize citrate, malic acid, and trisodium citrate. The major fatty acids were summed feature 3 (21.5%, comprising $C_{16:1}{\omega}7c/iso-C_{15:0}$ 2-OH) and C16:0 (15.04%). On the basis of the variations in many biochemical characteristics, GNUM-1 was considered as unique and thus was named Alteromonas sp. GNUM-1. It produced the highest agarase activity in modified ASW medium containing 0.4% sucrose, but lower activity in rich media despite superior growth, implying that agarase production is tightly regulated and repressed in a rich nutrient condition. The 30 kDa protein with agarase activity was identified by zymography, and this report serves as the very first account of such a protein in the genus Alteromonas.

Nocardioides tritolerans sp. nov., Isolated from Soil in Bigeum Island, Korea

  • A Gram-positive strain designated as MSL-$14^T$ isolated from a soil sample collected from Bigeum Island, Korea, was subjected to polyphasic taxonomy. The isolate was strictly aerobic. Cells were short rods and motile. Optimum growth temperature and pH was 28$^{\circ}C$ and 7.0, respectively. It was characterized chemotaxonomically as having a cell-wall peptidoglycan type based on LL-2,6-diaminopimelic acid and MK-$8(H_4)$ as the predominant menaquinone. The major fatty acids were iso-$C_{16:0}$, $C_{17:1}$ omega8c, and $C_{18:1}$ omega9c. The G+C content was 67.6 mol%. Phylogenetic analysis based on the 16S rRNA gene sequence revealed that strain MSL-$14^T$ is affiliated to the genus Nocardioides and formed a distinct lineage within the genus. MSL-$14^T$ showed highest sequence similarity to Nocardioides aestuarii JCM $12125^T$, having a similarity of 96.5%. Based on the 16S rRNA gene sequence divergence and phenotypic characteristics, it is proposed that strain MSL-$14^T$ should be classified as representing a novel member of the genus Nocardioides, for which we propose the name Nocardioides tritolerans sp. novo The type strain is strain MSL-$14^T$ (=KCTC $19289^T$=DSM $19320^T$).

Comparision of Chemical Components of Ligusticum chuanzxiong HORT and Cnidium officinale MAKINO (토천궁과 일천궁의 화확성분 비교)

  • Chemical components of domestic Ligusticum chuanxiong HORT and Cnidium officinale MAKINO were analyzed. Proximate analysis of each species showed crude protein 18.5% and 11.1%, crude lipid 5.9% and 6.2%, crude fiber 6.4% and 6.5%, crude ash 5.4% and 6.0%, and carbohydrate 63.7% and 70.2%, respectively. Contents of potassium which was found to be the most abundant mineral in both species were 1.8% and 3.0%, and those of sucrose were 0.4% and 0.3% respectively while neither fructose nor glucose were detected in each species. Major fatty acids in Ligusticum chuanxiong HORT and Cnidium officinale MAKINO were linoleic acid (60.7% and 61.2%), oleic acid (19.9% and 21.3%), palmitic acid (11.4% and 11.3%)(respectively) but there was no significant difference between two species. Glutamic acid was revealed as the most abandant amino acid in both species with 2.5% in Ligusticum chuanxiong HORT and 1.6% in Cnidium officinale MAKINO. Ligusticum chuanxiong HORT and Cnidium officinale MAKINO also were shown to be contained 0.0009% and 0.0005% vitamin $B_1$, and 0.6% and 0.7% tannin, respectively.
