Effect of Different Types of Dietary Fat on Renal Functions in Aged Rats (식이 지방의 종류가 흰쥐의 노화 과정 중 신장 기능에 미치는 영향)

  • This study was performed to investigated the effect of dietary fat sources on renal senescence in aged rats. Seventeen month old male Sprague-Dawley rats were divided into 3 groups according to urinary protein excretion. Four month old rats were used as a control group. The rats were fed one of three different experimental diets ; 20% beef tallow, 20% corn oil 20% fish oil diet. They were fed experimental diets ad libitum for 16 weeks . The results are summarized as follows. Serum lipid concentrations were higher in aged rats than in control rats, with the beef tallow group showing the highest level, followed by the corn oil and fish oil groups. Old rats showed higher HDL and lower LDL levels than the control groups. Age and dietary fat had no effect on VLDL. GFR for the both age groups were increased with experimental period with the beef tallow group showing the highest value. Urinary protein excretion was also increased with experimental period in both age groups. There was a large increase in urinary protein in old rats that were fed beef tallow and corn oil, but not in old rats fed fish oil. On the contrary , the effect of dietary fat on urinary protein was not found in control groups. There was individual susceptibility in the effect of dietary fat on urinary protein. Old rats fed beef tallow with high initial urinary protein showed highest increase, but , the change was not significant in rats with a low initial value . It was also found that the increase was kept low in rats of the fish oil group with high initial urinary protein. The corn oil group showed in between values. There were no differences in urine and renal tissue concentrations of TXB2 . Aged rats showed a tendency of having higher urinary PGE2 excretion and lower renal cortex content. Since higher PGE2 excretion was reported to be associated with decreased renal function, this might suggest that the aged rats show renal function reduction. Light microscopic examination showed that glomerular segmental sclerosis, mesangial matrix expansion and tubular atrophy were more frequently present in aged rats, and that these changes were more significant in the beef tallow group, followed by corn oil and fish oil groups. The percentage of urinary protein excretion was increased in aged rats in association with increased glomerular sclerosis and mesangial matrix . This change could be partly due to a change in eicosanoids metabolism . Therefore, modification of dietary fat could affect the eicosanoids metabolism in kidney and renal senescence.

Effect of Cnidi rhizoma Water-Extracted Solution on Fat Contents in Plasma, Liver and Adipose, and Fecal Steroids of Fatted Rats (천궁의 열수 추출액이 고지방식이에 의한 흰쥐의 혈장, 간 및 지방조직의 지질함량과 분변 Steroids에 미치는 영향)

  • This study was carried out to evaluate the effect of Cnidi rhizoma (CR) water extract on fat accumulation in fatted rats induced by the oral high fat administration for six weeks. Total cholesterol, neutral lipid and epididymal fat pad of CR groups were lower than the control group. On the other hand, high density lipoprotein(HDL) -cholesterol was somewhat higher In CR than In the control group. In addition, the same result as the above case was obtained when examining adipose for liver and epididymal fat components. During the feeding experiment, level of low density Lipoprotein(LDL) and very low density Lipoprotein (VLDL) increased while HDL was decreased. The degree of LDL and VLDL was decreased however, and HDL was, increased In CH groups as compared with the control group. Insulin and cortisol were higher than the control group, due to the fat accumulation. Experiment group showed the increased excretion as compared with the control group, in the excretion of bile acid and neutral sterol. The liver of the control group observed by the light microscope appeared to the fatty liver, but CR groups showed some improvement of the fatty liver. Based on the above results, it was shown that It Is possible to improve fat accumulation induced, by high fat dietary through using the oral administration of Cnidi rhizoma boiling extract solutions.

Effect of Chlorella vulgaris intake on cadmium detoxification in rats fed cadmium

  • The aim of this study was to investigate if dietary Chlorella vulgaris(chlorella) intake would be effective on cadmium(Cd) detoxification in rats fed dietary Cd. Fourteen-week old male Sprague-Dawley(SD) rats weighing $415.0{\pm}1.6\;g$ were randomly divided into two groups and fed slightly modified American Institute of Nutrition-93 Growing(AIN-93G) diet without(n=10) or with(n=40) dietary Cd(200 ppm) for 8 weeks. To confirm alteration by dietary Cd intake, twenty rats fed AIN-93G diet without(n=10) and with(n=10) dietary Cd were sacrificed and compared. Other thirty rats were randomly blocked into three groups and fed slightly modified AIN-93G diets replacing 0 (n=10), 5 (n=10) or 10% (n=10) chlorella of total kg diet for 4 weeks. Daily food intake, body weight change, body weight gain/calorie intake, organ weight (liver, spleen, and kidney), perirenal fat pad and epididymal fat pad weights were measured. To examine Cd detoxification, urinary Cd excretion and metallothonein (MT) concentrations in kidney and intestine were measured. Food intake, calorie intake, body weight change, body weight gain/calorie intake, organ weight and fat pad weights were decreased by dietary Cd intake. Urinary Cd excretion and MT concentrations in kidney and small intestine were increased by dietary Cd. After given Cd containing diet, food intake, calorie intake, body weight change, body weight gain/calorie intake, organ weights and fat pad weights were not influenced by dietary chlorella intake. Renal MT synthesis tended to be higher in a dose-dependent manner, but not significantly. And chlorella intake did not significantly facilitate renal and intestinal MT synthesis and urinary Cd excretion. These findings suggest that, after stopping cadmium supply, chlorella supplementation, regardless of its percentage, might not improve cadmium detoxification from the body in growing rats.

  • This study was conducted to investigate the effect of nondigestable substances and calcium such as oligosaccharide, agar, saponin, tannin and calcium on the reduction of lipid status in rats fed high fat diet. In order to make the observation, the lipid content in plasma, liver and the feces, and bile acid excretion were measured of r 4 weeks. the results obtained from this research are as follows. Concentration of total lipid in plasma seemed highest in the control group and were significantly lower in groups oil-gosaccharide, agar and calcium-tannin, compared to the control group. Concentration of total cholesterol in plasma was significantly lower in groups oligosaccharide, agar, calcium and calcium-saponin, compared to the control group. The improvement in lipid status seems to be insignificant with oligosaccharide, showed increase in total bile acids excretion in feces and decrease in total cholesterol in plasma, compared to the control group. These evidence seems to indicate improvement of the plasma lipid status by calcium and agar supplementation.

EFFECTS OF DIETARY FULL-FAT SOYBEAN LEVELS ON GROWTH PERFORMANCE AND FEED UTILIZATION AND PHOSPHORUS EXCRETION OF CARP (Cyprinus carpio)

  • In order to estimate the nutritive value of roasted full-fat soybean (FFS) in carp diet, growth performance and excretion of protein and phosphorus were examined using carps having mean body weight of 111 g. Growth trial was conducted for 4 weeks using the fishes fed 5 diets (Control, $F_{24}S_{13}$, $F_{16}S_{27}$, $F_8S_{40}$ and $F_0S_{56}$) containing 32%, 24%, 16%, 8% and 0% of fish meal (F) and 0%, 13%, 27%, 40% and 56% of full-fat soybean, respectively. A total of 800 fishes were allotted randomly by groups of 40 to 5 treatments with 4 replicates per treatment. Fishes were fed to station eight times daily. Feeding trial was conducted for 4 weeks. As dietary FFS increased from 0% to 56%, weight gain of fish decreased from 91 g to 39 g and feed conversion ratio increased from 1.06 to 1.95. Protein efficiency ratio (PER) was highest (2.35) in control group which had the highest protein intake, while PER significantly decreased with decrease in protein intake as dietary FFS level increased. Although dietary protein and energy levels were maintained constant, protein excretion per kg weight gain varied from 273 g to 579 g for the groups control and $F_0S_{56}$, respectively. However, the value for control group was not significantly different to those for the groups $F_{24}S_{13}$ and $F_{16}S_{27}$. On the other hand, the highest protein retention efficiency was found in group fed the diet $F_{24}S_{13}$. Fish fed the diet $F_8S_{40}$ excreted the lowest phosphorus (P) based on kg weight gain showing the highest P retention efficiency of 62%. P excretion per kg feed intake was in the range of 5 g to 10 g for the groups $F_8S_{40}$ and $F_{24}S_{13}$, respectively. The present results indicated that as dietary FFS level increased, growth performance and feed utilization decreased while excretion of protein and phosphorus increased. Therefore, it was concluded that more than 25% substitution by FFS for fish meal could exert negative effects on growth and feed utilization of carp.

  • Anti-lipogenic effects of tannic acid on 3T3-L1 adipocytes as well as on rats fed high fat diet (HFD) were investigated. Tannic acid stimulated lipolysis through suppression of the leptin secretion and an increase of glycerol release in a dose dependent manner in 3T3-L1 adipocytes. For animal study, the rats were fed either HFD or HFD supplemented with 1%(w/w) tannic acid (HFDT) for 12 weeks, respectively. Body weight gain, liver weight, and visceral fat mass in rats fed HFDT were significantly decreased compared to those of rats fed HFD. The lipid profiles of HFDT group were significantly decreased compared with HFD group in the serum and liver, whereas fecal total cholesterol excretion was increased in HFDT group. These results suggest that anti-lipogenic effect of tannic acid in 3T3-L1 adipocytes and in rats fed HFD may be due to the stimulation of lipolysis and the reduction of lipid levels.

  • Caseinomacropeptide (CMP) is a glycopeptide of 64 amino acid residues derived from the C-terminal of mammalian milk K-casein. Recently, human CMP (hCMP) was produced from the recombinant yeast Saccharomyces cerevisiae. In this study, the antiobesity activity of the recombinant hCMP (rhCMP) was investigated in vivo using Sprague-Dawley rats. The rhCMP did not affect the rats fed with a normal fat diet (fat content, $5.0\%$), but decreased the body weight gain of the rats fed with a high fat diet (fat content, $20\%$) by up to $19\%$. Autopsies revealed that the weights of the liver, kidney and adipose tissues decreased when the rats were given the rhCMP, which also reduced the lipid concentrations in the plasma and liver, but enhanced the fecal excretion of lipids. These results suggest that rhCMP prevent the accumulation of lipid by stimulating its fecal excretion, so could be used as a food supplement to alleviate the obesity problem caused by a high fat diet.

Hypocholesterolemic Effect of Tangerine Pulp, Sea Tangle or Prickly Pear Cactus on Lipid Level, Intestinal Cholesterol Absorption, Platelet Aggregation and Liver Tissue in Hypercholesterolemic Rats (감귤박, 다시마, 손바닥 선이장 분말을 함유한 식이의 급여가 고콜레스테롤혈증 흰쥐의 체내 지질수준과 장내 콜레스테롤 흡수, 혈소판 응집성 및 간 조직에 미치는 영향)

  • This study done to investigate the hypocholesterolemic effect of ow dietary fiber in a high cholesterol diet in aspect of cholesterol absorption and excretion using Sprague Dawley rats. After feeding diets containing 0.5% cholesterol and 5% tangerine pulp, sea tangle or prickly pear cactus for four weeks, we measured the levels of plasma and liver cholesterol and triglyceride, fecal neutral sterols and fecal radioactivity after ingestion 14(sup)C-cholesterol. We also examined platelet aggregation and histological change in liver tissues in association of hypercholesterolemia. The liver to body weight ratio was significantly(p<0.01) lower in rats fed prickly pear cactus than in other groups. The levels of plasma total cholesterol, LDL-cholesterol and triglyceride were decreased significantly(p<0.01) in the prickly pear cactus group compared with the control, while there was no difference in the liver levels of total cholesterol and triglyceride among groups. Fecal corprostanol and cholesterol were significantly(p<0.01) higher groups of control and prickly pear cactus compared to the other two groups. Radioisotope excretion after ingestion of 14(sup)C-cholesterol was higher in the control group than in tangerine pulp group or sea tangle group, with the highest in prickly pear group. Radioisotope excretion was the highest during the 2nd and 3rd days in all groups. Hematocrit and platelet aggregation were decreased in all fiber groups compared to the control, but not statistically different. Microscopic examination showed that cholesterol diet cause a fat accumulation in the liver and prickly pear cactus decreased the fat accumulation. Result indicates that prickly pear cactus has hypocholesterolemic effect by decreasing absorption and increasing excretion of cholesterol, thereby protective effect on fatty liver. Control group fed diet containing high cholesterol and low fiber seems to have a self control system in cholesterol absorption and excretion preventing hypercholesterolemia.   • This study is conducted to evalute the effect of ginseng fraction component (ginseng extract solution, GES; ginseng protein, GP; ginseng saponin, GSA; ginseng residue, GR) upon hyperlipidemia and fatty liver induced by high fat administration. In doing so, the serum, liver and epididymal adpoid tissue have been examined for lipid components level and lipoprotein fraction. Feces bile acid and neutral sterol excretion have been also measured. 1'he results obtained from this study are as follows. 1. Serum, liver, epididymal lipid components of GP and GSA group were significantly lower than the controlgroup. 2. During the feeding experiment, VLDL and LDL increase while HDL decrease in all group. However the degree of VLDL and LDL increase and HDL decrease were signficantly small in GP and GSA group compared with control group. 3. In the excretion of bile acid and neutral sterol, all experiment group showed increased excretion in the comparison of control group.

Effect of Corn Peptide on the Lipid Metabolism in Rats (옥수수 펩타이드가 흰쥐의 지질 대사에 미치는 영향)

  • This study was conducted to investigate the effects of corn peptide(CP) on lipid metabolism. Sprague-Dawley(S.D) male rats were assigned to three dietary groups {control diet(CD), high fat diet (HFD) & high fat com peptide diet(FCD)} and fed 4 weeks to examine the effects of CP. There were no significantly different in cholesterol concentrations in the liver among the groups. However, triglyceride(TG) concentrations of the FCD & CD significantly lower than the HFD. Fecal excretion of neutral steroids & bile acids of the FCD significantly higher than the CD & HFD. Serum total cholesterol TG & LDL-cholesterol concentrations of the FCD & CD significantly lower than the HFD. These results suggest the improvement of lipid composition in serum by CP might be inhibit of lipid absorption in intestine & increment of neutral steroids & bile acids excretion in feces.
