Preparation of Commercial Agarose from Jeju Seaweed, Gelidium amansii using DMSO Extraction and EDTA Washing (제주산 우뭇가사리(Gelidium amansii)로부터 DMSO 추출과 EDTA 수세법에 의한 상용화 아가로스 제조)

  • Agar was prepared from Gelidium amansii collected from Jeju Island, South Korea. This agar preparation has high gel strength and low sulfate content compared with G. amansii agar from Morocco. Accordingly, agarose was made from the Jeju agar through the consecutive refining processes of dimethyl sulfoxide (DMSO) extraction and ethylene diamine tetra acetic acid (EDTA) washing. The physicochemical properties of the resulting agarose were compared with those from agarose prepared using only DMSO extraction. Consecutive DMSO extraction and EDTA washing more strongly affected the physicochemical properties of the agarose (purified agarose) compared with the use of DMSO extraction alone. These properties were similar to those of commercial agarose used for electrophoresis. In DNA electrophoresis, the separation and movement speed of the purified agarose were similar to those of the commercial agarose. In a $^{13}C$ NMR analysis, the purified agarose exhibited the same carbon peak as the commercial agarose. When observed under scanning electron microscopy, the agar had an even and smooth surface without irregularities or pores, and the purified agarose had a wide surface area with a large number of pores; the commercial agarose had an irregular surface that would allow the solvent to easily permeate. These results illustrate that the physicochemical properties of agarose prepared from DMSO extraction and EDTA washing were more effective than those observed after DMSO extraction alone; thus, these processes used in succession will be useful in agarose industries.

A Study on the Improvement Methods of a Tourist Resort with Traditional Oriental Medicine in Sancheong-gun, Gyeongsangnam-do (경남 산청군 전통한방 휴양관광단지의 개선방안 연구)

  • This study attempts to propose ways for activated methods of Sancheong tourist resort with Traditional Oriental Medicine(the following this resort) by examining the placement plan, space plan and characteristics of this resort. The results of this study are as follow. First, the accessibility has to be improved for safe and quick transportation between this resort and nearby areas. The curved roads must be replaced by straight roads at least partially. Second, an experiencing space like herb farm is necessary to activate the function of the tourist resort in the aspect of the arrangement planning for this resort. Third, the roads within the complex must be improved into Ring Roads for the circulation of cars to use the space in the tourist resort in a reasonable and positive way. Fourth, various places for tourism must be developed in the rear for sightseeing tours. Moreover, the development of tourism area together with the residents must be achieved with the active participation of the local people in the festival.

Development of Ecotechnology for the Biotope creation in Korean Rural Area -With a Special Reference to Dragonfly Biotope- (농촌지역에서의 생물서식공간 조성기술의 개발(1) -잠자리 서식환경 조성을 중심으로-)

  • Owing to the urbanization, the industrialization and joining to WTo, there are lots of fallow and abandoned farmlands in Korea. Those results have occurred inefficient land use and destruction of agricultural ecosystem with the secondary succession and invasion of alien weeds. Using these fallow and abandoned farmlands, Korean government tries to create the ecological parks for the nature conservation, amenity, education and research, etc. For these purposes, materials on the ecological park construction were gathered from abroad such as England, Germany, Switzerland and Japan and also field trips to those countries were executed in April,1996. Accordingly, the goal for the ecological parks could be set suitably to Korean situation as of Japanese styles which are characterized by man-made biotopes. For a case study, dragonfly was selected as an indicator of the rehabilitation of water-side ecosystem, arid a site was selected for the dragonfly biotope under the criteria of selection to the Experimental Farm of Korea University including near abandoned farmlands (Dogok-ri, Wabu-eup, Namyangju city Kyunggi-do). Natural and cultural factors of this site were inventoried and analyzed, and then with the results dragonfly biotope was planned to the site with the detailed ecotechnology.

Questionnaire Study on the Difficulties and Improvements of the 6th Industrialization Dairy Farm (설문을 통한 6차산업형 목장경영의 애로사항과 개선방안에 관한 연구)

  • This study was conducted to investigate the difficulties of dairy farms in practicing 6th industrialization and methods for overcoming these difficulties. A qustionnaire survey was carried out to examine the present states of farms, recognition of the farmstead milk-processing market situation, possibility of farmstead milk processing for reducing the raw milk surplus, assessment of government policies, and difficulties dairy farmers confront in realizing the 6th industrialization. Farm sizes, types, and human resources organizations varied between farms. Most farmers were producing yogurt and/or fresh (string or barbecue) cheeses, which were marketed through 'Visit and Purchase' channel. Farmers who answered the questionnaire were relatively positive about the current situation of farmstead milk processing, expecting to be involved in the disposal of excess raw milk. Nevertheless, they responded negatively about current relevant policies, citing the main difficulties caused by 'excessive regulation'. Other barriers to successful '6th industrialization' are difficulties in marketing and lack of funds. Approximately 19% of dairy farms practicing the '6th industrialization' use automatic milking system (AMS) and 38.46% of dairy farmers whose milking depends on conventional milking system intend to introduce AMS in the future. Positive expectations of AMS adoption were mostly related to 'lack of time and labor', 'exhibiting for tourism', and 'succession of dairying'.

A Study on the Types and Determinants of Young Farmers: Focusing on Young Farmers in Muan-gun, Jeollanam-do (청년농업인 유형화 및 결정요인 분석: 전남 무안군 청년농업인 중심으로)

  • Based on Muan-gun, Jeollanam-do, this study explores how to mitigate the disappearance of rual areas. The study surveyed 95 young farmers in Muan-gun to assess their farming practices and the challenges they face. We further employ factor analysis and cluster analysis classify young farmers in Muan-gun, facilitating the identification of tailored policies or initiatives aimed at fostering and supporting young farmers. The results are summarized as follows. First, Muan County does not have any ordinances or original projects specifically designed to support young farmers. Second, the succession rate of farmland among young farmers in Muan County is 41.1%, which is comparable to the national rate of 43.7%. This indicates that approximately 40% of young farmers in Korea have inherited farmland, a critical foundation for agricultural activities. Third, despite accumulating farming experience, young farmers have not seen any improvement in local living conditions, and rather their difficulties have intensified. Fourth, this study conducted a factor analysis using 21 variables, resulting in the selection of seven common factors for cluster analysis. Consequently, young farmers in Muan County were categorized into three groups. The multinomial logit analysis revealed that the typology of young farmers is influenced by indicators such as cultivated area, farming experience, demand for smart farms, farm income, and farming type (rice cultivation or other). Therefore, to attract young farmers and prevent the decline of rural areas, policy efforts should focus on minimizing entry barriers to farming infrastructure, such as access to farmland, and improving local settlement conditions.

Vegetation Structure and Management Planning of Yongha Gugok in Woraksan National Park (월악산국립공원 용하구곡의 식생구조 및 관리방안)

  • This study was conducted to suggest vegetation management plan for Gugok landscape maintenance and improvement by deducing the vegetation landscape factors inherent in Yongha Gugok and understanding vegetation structure through the investigate of existing vegetation and plant community structure of Yongha valley in Woraksan National Park. There were broad and flat rocks, natural layered stones, clear water, light stones, stream, valleys, waterfalls, Pinus densiflora and Acer pseudosieboldianum as a result of deducing natural factors on poetry. There were P. densiflora and A. pseudosieboldianum appeared as one of main vegetation landscape elements. The actual vegetation analysis results were as followed. The natural vegetation occupied 67.5% and it was classified as P. densiflora community, Quercus variabilis community, Q. variabilis-P. densiflora community, Q. variabilis-Q. serrata community, Q. serrata community, Q. mongolica community, Q. mongolica-P. densiflora community, Deciduous broad-leaved tree community. The artificial vegetation(18.7%) was classified as Q. serrata community-Larix kaempferi community, Q. mongolica- Castanea crenata community, L. kaempferi community, L. kaempferi-C. crenata community, fruticeta, L. kaempferi-Q. mongolica community. The grassland area(2.0%) was classified as Miscanthus sinensis community, Phragmites communis community, and other areas were classified as landscape tree planting area, farm, orchard, residential area. The representative vegetation were P. densiflora community, Q. variabilis-Q. serrata community, L. kaempferi community, Deciduous broad-leaved tree community in Yongha Gugok. The species diversity index of Shannon was 0.6274~0.9908 on the whole. Yongha Gugok, as a symbol of succession on confucianism and reverence for nature, should be preserved natural valley landscape being clean and wijungchuksa at the end of Joseon Dynasty and Japanese Colonial era. In this historical and cultural Gugok, vegetation landscape management plan is needed to landscape maintenance with P. densiflora community, density control with L. kaempferi community. And it is considered when natural disasters and artificial damages happened, P. densiflora-oriented vegetation restoration plan should be applied in order to restore.
