Synthesis and characterization of a new energy material (guanidinium dinitramide) with crystallization solvent

  • An environmentally favorable (chlorine-free) solid oxidizer, guanidinium dinitramide [GDN; $NH_2C(NH_2)NH_2N(NO_2)_2$], was newly synthesized from guanidine carbonate [$NH_2C(=NH)NH_2{\cdot}1/2H_2CO_3$]. Two different crystalline forms (${\alpha}-type$ and ${\beta}-type$) appeared according to the applied solvents and synthesis conditions. Moisture, during extraction, might become trapped in a crystal between inner molecules. Therefore, despite having the same chemical composition, Raman-IR and TGA-DSC revealed different physical characteristics of the two forms. Peaks of Raman shift near $1000cm^{-1}$ implied different chemical structures. Thermal analysis revealed an exothermic temperature $155.7^{\circ}C$ for ${\alpha}-type$ but one of $191.6^{\circ}C$ for ${\beta}-type$. The caloric value of ${\alpha}-type$ was 536.4 J/g, which was 2.5 times larger than that of the ${\beta}-type$, which was 1310 J/g. While the synthesized GDN of ${\alpha}-type$ showed a steep exothermic decomposition, the ${\beta}-type$ was slowly decomposed after melting through an endothermic process. This work implied that despite of the same molecular formula some different core thermal properties would appear depending on synthesis conditions.

Studies on Solvent Extraction and Flotation Technique Using Metal-Dithizone Complexes(II). Determination of Trace Elements in Water Samples by Solvent Sublation

  • The preconcentration and determination of trace elements in water samples were studied by a solvent sublation utilizing dithizonate complexation. After metal dithizonates were formed, trace amounts of cadmium, cobalt, copper and lead were floated and extracted into small volume of a water-immiscible organic solvent on the surface of sample solution and determined in the solvent directly by GF-AAS. Several experimental conditions as formation condition of metal-dithizonate complexes, pH of solution, amount of dithizone, stirring time, the type and amount of surfactants, N2 bubbling rate and so on were optimized for the complete formation and effective flotation of the complexes. And also four kinds of light solvents were compared each other to extract the floated complexes, effectively. After the pH was adjusted to 4.0 with 5 M HNO3, 8.0 mL of 0.05% acetone solution of dithizone was added to 1.00 L water sample. The dithizonate complexes were flotated and extracted into the upper methyl isobutylketone (MIBK) layer by the addition of 2.0 mL 0.2% ethanolic sodium lauryl sulfate solution and with the aid of small nitrogen gas bubbles. And this solvent sublation method was applied to the analysis of real water samples and good results of more than 85% recoveries were obtained in spiked samples.

  • The use of caffeine as a psychoactive stimulant in tea has been observed to have serious negative effects in humans' systems such as respiratory, nervous, cardiovascular, renal and skeletal systems. This study was carried out to assess the levels of caffeine in 10 different tea brands available in local market in Dutse, Jigawa State, Nigeria. Quantitative analysis of caffeine was performed by a simple and fast UV-Vis spectrophotometric methods using different solvents for extraction. The caffeine content in all the tea samples analyzed in this study were below the maximum allowable limits set by the USFDA. Tea have been associated with adverse health effects and the claims made by manufacturers about the benefits of tea do not highlight risks associated with excessive consumption of a combination of the ingredients contained in tea. Long term effects of tea consumption of children and young people have not been adequately studied. Therefore, it is recommended that further research be carried out on the adverse effects of energy drinks on children. Research is also needed to be done on the effects of the combination of ingredients on health and excessive consumption of those ingredients to children and adolescents. People need to be educated and given proper awareness on the health risks associated with caffeine containing beverages.

사과 부위별 항산화 성분 및 항산화 활성 (Antioxidant Component and Activity of Different Part Extracts in Apple (Malus domestica cv. Fuji))

  • To improve the utility of apple, apple flesh (Malus domestica cv. Fuji), pomace and peels were dried and then extracted using solvents (water, 70% and 100% ethanol). The contents of polyphenols, flavonoids, ascorbic acid, ursolic acid and antioxidant activities were measured as extracted from the apple flesh, pomace and peel. The content of polyphenols was the highest in the apple peel when 100% ethanol was extracted. The content of flavonoids was about 3~8 times higher in apple peel than those of flesh and pomace, and 70% ethanol extract was the highest content. Ascorbic acid content was the highest in apple peel as well as flavonoid content. The content of ursolic acid in the apple flesh was not detected in water and ethanol extraction. In the case of apple pomace and peel, ursolic acid was not detected in water extract but it was detected when ethanol was extracted. The content of ursolic acid was highest in the apple peel. In the case of antioxidative activity, DPPH and ABTS radical scavenging ability showed the highest ability when apple peel was extracted with 70% ethanol.

Antibacterial activity of Dioscorea batatas, Morus alba, and Tagetes erecta against E. coli and S. aureus

  • In this study, the anti-bacterial activity of three medicinal herbs such as Dioscorea batatas, Morus alba, and Tagetes erecta, have been applied in oriental medicine against Escherichia coli and Staphylococcus aureus. The antibacterial activity of Dioscorea batatas ethanol extract (DBEE), Morus alba ethanol extract (MAEE), and Tagetes erecta ethanol extract (TEEE) was analyzed by disk diffusion, minimum inhibitory concentration (MIC), and minimum bactericidal concentration (MBC) assays. As a result, three medicinal herbs exhibited antibacterial activity in a dose-dependent manner as well as MAEE and TEEE showed the most potent antibacterial activity followed by disk diffusion assay against E. coli and S. aureus, respectively. MIC against E. coli and S. aureus was 1.6 mg/mL of MAEE and TEEE as well as 0.2 mg/mL of TEEE. MBC against both strains was 5 mg/mL of DBEE and 2.5 mg/mL of TEEE. Consequently, three medicinal herbs in this study showed potent antibacterial activity through the inhibited growth of E. coli and S. aureus, which is considered as a potential candidate for cosmeceutic agents to attenuate the pruritus. In addition, antibacterial and antihistamine effects followed by the extraction solvents should be analyzed in future studies.

남해산 치자(Gardenia jasminoides Ellis fructus) 껍질의 생리활성 및 지질과산화 저해 활성에 미치는 영향 (Effects of Gardenia jasminoides Ellis Peel Extract in Namhae Korea on the Bioactivity Compounds and Lipid Peroxidation Inhibition Activity)

  • 치자 껍질의 phytic acid 함량과 CM (chloroform:methanol, 2:1, v/v), n-부탄올 및 70% 에탄올을 사용한 추출물의 용매(3가지) 별 총 페놀 함량 및 항산화 활성 능력 등을 통하여 치자 껍질의 효용 가치를 검토한 결과, phytic acid 함량은 $4.966{\pm}0.996mg\;PAE/g\;DW$ (dry weight)로 나타났으며, 용매 별 생리 활성은 농도(0.2, 0.4, 0.6 mg/mL)가 높아질수록 유의적으로 증가하였으며(p<0.05), 양성대조군으로 사용된 ascorbic acid, BHA, trolox 보다는 각 농도에서 낮은 활성이 관찰되었다. 치자 껍질의 총 페놀 함량은 CM, n-부탄올, 70% 에탄올 추출물 순으로 CM 추출물에서 $39.74{\pm}0.15mg\;CAE/g$으로 가장 높았으며, nitric oxide (NO) 라디칼 소거능과 지질과산화 저해능은 용매 별로 CM > 70% 에탄올 > n-부탄올, nitrite ($NO_2$) 소거능은 n-부탄올 > CM > 70% 에탄올, ${\beta}-carotene$ 탈색 저해능과 환원력은 CM > n-부탄올 > 70% 에탄올 순의 활성이 나타났다. 이상의 결과, 치자 껍질의 용매 별 총 페놀 함량 순과 항산화력, 환원력은 일치 하였으며, 질소산화물 소거능과 지질과산화 저해능 분석에서는 일치하지 않은 것으로 확인되었다. 이는 질소산화물 소거능과 지질과산화 저해능은 페놀 성분 보다 다른 생리활성물질이 더 큰 영향을 주는 것으로 추정된다. 이에, 치자 껍질은 질소산화물 소거능, 항산화능 및 지질과산화 저해능 등이 우수한 것으로 나타나 기능성 식품 소재로서의 가치가 있는 것으로 판단된다.

Potential antioxidant and lipid peroxidation inhibition of Phyllanthus acidus leaf extract in minced pork

  • Objective: This study investigated the effect of extraction solvents on antioxidant bio-active compounds as well as potential antioxidant and lipid peroxidation inhibition of Phyllanthus acidus (P. acidus) leaf extract in minced pork. Methods: The effect of various solvent systems of water, 25%, 50%, 75% (v/v) ethanol in water and absolute ethanol on the extraction crude yield, total phenolic content, total flavonoid content and in vitro antioxidant activities of P. acidus leaves was determined. In addition, antioxidant activities of the addition of crude extract from P. aciuds leaves at 2.5 and 5 g/kg in minced pork on 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging, 2,2′-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical cation decolorization, reducing power and inhibition of lipid peroxidation (thiobarbituric acid reactive substances; TBARS) were determined. Moreover, sensory evaluation of the samples was undertaken by using a 7-point hedonic scale. Results: The results showed that the highest crude yield (2.8 g/100 g dry weight) was obtained from water which also had the highest recovery yield for total phenolic content, total flavonoid content and the strongest antioxidant activity. The addition of crude water extract from P. acidus leaves was more effective in retarding lipid peroxidation and higher antioxidant activity than control and butylated hydroxytoluene in minced pork. In particular, the samples containing P. acidus extract had no significant effect on the sensory scores of overall appearance, color, odor, texture, flavor, and overall acceptability compared to the control. Conclusion: Water solvent was an optimally appropriate solvent for P. acidus leaf extraction because of its ability to yield the highest amount of bio-active compounds and in vitro antioxidant property. Particularly, P. acidus crude water extract also strongly expressed the capacity to retard lipid oxidation, radical scavenging, radical cation decolorization and reducing power in minced pork. The results of this study indicated that P. acidus leaf extract could be used as natural antioxidant in the pork industry.

토양(土壤) , 수(水) , 농작물중(農作物中)에 있어서 잔류농약분석(殘溜農藥分析)을 위한 수증기(水蒸氣) 증류법(蒸溜法)의 응용(應用)에 관한 연구 (Studies on the Application of Steam Distillation for the Analysis of Pesticide Residues in Soil, Water and Crops)

  • 토양(土壤), 수(水), 농작물(農作物)중 잔류(殘留)하는 각종(各種) 농약(農藥)의 분석(分析)을 위한 수증기증류법(水蒸氣蒸溜法)의 응용성(應用性) 및 정밀도(精密度)등에 대하여 연구검토(硏究檢討)하였다. 수증기증류(水蒸氣蒸溜)에 의하여 추출(抽出)이 가능(可能)했던 농약(農藥)은 PCNB, ${\alpha}-BHC$, ${\beta}-endosulfan$, IBP, diazinon, fenitrothion, fenthion, alachlor, butachlor, pretilachlor, metolachlor, pendimethalin, benthiocarb, molinate등이었으며 simazine, atrazine, nitrofen등은 불가능(不可能)하였다. 농약(農藥)의 증류(蒸溜)까지 요(要)하는 유출액양(溜出液量)은 농약(農藥)의 종류(種類)에 따라 크게 달랐으며, 일정량(一定量)의 유출액(溜出液)을 취(取)한 후 냉각관(冷却管)을 acetone으로 씻어 줌으로써 증류시간(蒸溜時間)을 단축(短縮)시킬 수 있었다. 수증기증류법(水蒸氣蒸溜法)은 소량(小量)의 유기용매(有機溶媒)로 농약(農藥)의 추출(抽出)이 가능(可能)했으며, 분석대상물질(分析對象物質)에 따라서는 기기분석시(機器分析時) 방해물질(妨害物質)이 검정(檢定)되기는 했지만 많은 경우 농축(濃縮)과 정제과정(精製過程)을 생략(省略)할 수도 있어 매우 유용(有用)한 방법(方法)이라 하겠다.

Qualitative and quantitative analysis of the saponins in Panax notoginseng leaves using ultra-performance liquid chromatography coupled with time-of-flight tandem mass spectrometry and high performance liquid chromatography coupled with UV detector

  • Background: Panax notoginseng leaves (PNL) exhibit extensive activities, but few analytical methods have been established to exclusively determine the dammarane triterpene saponins in PNL. Methods: Ultra-performance liquid chromatography coupled with time-of-flight mass spectrometry (UPLC/Q-TOF MS) and HPLC-UV methods were developed for the qualitative and quantitative analysis of ginsenosides in PNL, respectively. Results: Extraction conditions, including solvents and extraction methods, were optimized, which showed that ginsenosides Rc and Rb3, the main components of PNL, are transformed to notoginsenosides Fe and Fd, respectively, in the presence of water, by removing a glucose residue from position C-3 via possible enzymatic hydrolysis. A total of 57 saponins were identified in the methanolic extract of PNL by UPLC/Q-TOF MS. Among them, 19 components were unambiguously characterized by their reference substances. Additionally, seven saponins of PNL-ginsenosides Rb1, Rc, Rb2, and Rb3, and notoginsenosides Fc, Fe, and Fd-were quantified using the HPLC-UV method after extraction with methanol. The separation of analytes, particularly the separation of notoginsenoside Fc and ginsenoside Rc, was achieved on a Zorbax ODS C8 column at a temperature of $35^{\circ}C$. This developed HPLC-UV method provides an adequate linearity ($r^2$ > 0.999), repeatability (relative standard deviation, RSD < 2.98%), and inter- and intraday variations (RSD < 4.40%) with recovery (98.7-106.1%) of seven saponins concerned. This validated method was also conducted to determine seven components in 10 batches of PNL. Conclusion: These findings are beneficial to the quality control of PNL and its relevant products.

곤달비(Ligularia stenocephala) 잎 분획물의 항산화활성 평가 (Study on Antioxidant Effects of Fractional Extracts from Ligularia stenocephala Leaves)

  • 본 연구는 곤달비(Ligularia stenocephala)의 생리 효능을 체계적으로 밝히기 위한 기초 자료를 얻고자 80% 에탄올 추출물과 순차적 분획물에 대하여 항산화 및 항균 효과를 검증하였다. 곤달비 에탄올 추출물의 추출수율은 21.36%이었으며, 이에 대하여 용매별로 분획한 수율은 water 분획물이 60.35%로 가장 높았으며, n-butanol, n-hexane, ethyl acetate 순이었다. 곤달비 추출물 및 분획물의 총 폴리페놀 함량은 ethyl acetate 분획물에서 254.00 mg/g으로 가장 높게 나타났다. DPPH radical 소거활성을 측정한 결과, 곤달비 ethyl acetate 분획물에서 $IC_{50}$ 값이 0.28 mg/mL로 높은 활성을 나타내었다. 0.5 mg/mL 농도에서의 ABTS 라디칼 소거활성은 ethyl acetate 분획물에서 96.67%의 활성을 보였다. FRAP 활성 역시 1 mg/mL 농도에서 ethyl acetate 분획물이 2.70 mM의 가장 높은 활성을 나타내었다. 항균활성은 B. cereus 균주에 대해 ethyl acetate 분획물 5 mg/disc에서 10.2 mm의 저해환을 나타내었다. 이상의 결과에서 곤달비의 추출물 및 분획물 중 ethyl acetate 분획물의 경우 높은 항산화 활성과 약간의 항균활성을 나타내었으며 천연물소재로서 식품첨가물 및 화장품 원료로의 가능성이 있는 것으로 사료된다.
