Effects of Dried Leaf Powders, Water and Ethanol Extracts of Persimmon and Green Tea Leaves on Lipid Metabolism and Antioxidative Capacity in 12-Month-Old Rats (감잎, 녹차의 건분 및 물, 에탄올추출물이 노령쥐의 지방대사와 항산화능에 미치는 영향)

  • The effects of dried leaf powders and water and ethanol extracts of persimmon and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity were investigated in 12-month-old rats. Forty-nine male Sprague-Dawley rats weighing 520$\pm$19g were blocked into seven groups according to body weight. Rats were raised for four weeks with control(no tea leaf powder or extracts) and experimental diets containing either 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) or green tea(Camellia sinensis O. Ktze), or water or ethanol extract from equal amounts of each dried tea powder. Food intakes of all tea diet groups were higher than that of control. Weight gains and food efficiency ratios of all tea diet groups were not significantly different from those of control. All tea diets decreased plasma triglyceride level, especially, green tea powder and persimmon ethanol diets were more effective than other diet. All the tea diet groups showed decrease in liver triglyceride level, and persimmon powder and ethanol extract increased fecal triglyceride excretion. Plasma cholesterol levels of all the tea diet groups were not significantly different from the control, but control. Fecal cholesterliver cholesterol concegroups were significantlntrations of all tea y lower than that of ol excretions of persimmon powder, green tea ethanol extract, persommon ethanol extract and green tea ethanol extract groups were significantly higher than that of control. Plasma and liver thiobarbituric acid reactive substance(TBARS) concentrations of all the tea diet groups were lower than that of control. Especially, plasma TBARS concentrations of green tea powder and persimmon ethanol extract groups were sinificantly low. Red blood cell(RBC) superoxide dismutase(SOD) activities of persimmon ethanol extract and green tea water extract groups were increased, and RBC catalase activities of all experimental groups were not significantly different. RBC glutathione peroxidase(GSH-px) activities of persimmon ethanol extract, persimmon water extract and green tea powder groups were increased. Liver SOD activities of all the tea diet groups except green tea ethanol extract group were higher than that of control. Liver catalase activities of all experimental groups were not significantly different, and liver GSH-px activity of green tea powder group was significantly higher than that of control. In conclusion, dried leaf powders, and water and ethanol extracts of persimmon and green tea were effective in lowering lipid level, inhibiting lipid peroxidation and increasing antioxdative enzyme activities in 12-month-old rat. Green tea leaf powder with high contents of flavonoids and water soluble dietar fiber was most effective in lowering plasma triglyceride, cholesterol and TBARS level.   • The purpose of this study was to make ramyon from Korean cultured wheat by adding with hot water extract powder from safflower seed in order to add the value of it. The cooking quality, instrumental texture and sensory characteristics of ramyon were analyzed. The ramyons with 0.1%, 0.3%, 0.5%, and 0.7% of hot water extract powder(HEP) from safflower seed, control, and those with 3% and 5% of dried powder(DP) from safflower seed were compared. The yield of HEP was 7.8%. Lightness, redness, Max. weight, strength, hardness, adhesiveness, cohesiveness and springiness were measured. As the amounts of HEP and DP was increasing, ramyons smelled stronger and was getting harder and chewier, while became less transparent and had no difference in elasticity and adhesiveness. In overall acceptability, both control and ramyon with HEP had similar points. From three important factors, appearance, color and smell to make ramyon more acceptable, addition of 0.3~0.5% of hot water extract powder from safflower seed was found to be the best. However, further studies on smells are needed to make processed foodstuffs with safflower seed.

Effect of Eisenia Bicyclis and Its Pill on Serum Lipid Status in Rats Fed High Fat Diet (고지방식이 급여 흰쥐에서 대황 및 대황정제환의 혈청 지질 개선 효과)

  • In order to investigate whether Eisenia bicyclis (EB) as a functional food material improves serum lipid status, supplementation of EB powder or EB extract to the high fat/cholesterol diet was tested in 6-week old male Sprague-Dawley rats for four weeks. In the first experiment, four kinds of seaweed powder -Eisenia bicyclis (EB), Undaria pinnatifida (Up), Laminaria japonica (LJ), and Ecklonia stolonifera (ES) were compared with alginic acid (ALG) as well as control. In the second experiment, EB water extract and EB ethanol extract and EB pill containing EB and LJ powder were compared with the EB powder. Amounts of dietary fiber contained in experimental diets were adjusted to provide 5% of diets. Serum total cholesterol was significantly lower only in rats fed LJ or EB powder, but HDL cholesterol was higher in rats fed UP, LJ, ES or EB powder. Ratios of HDL-/total cholesterol of all seaweed groups including ALG were significantly higher than that of the control group. Serum triglyceride was lower in LJ, EB nd ES powder groups than the control group. In comparison of EB extracts, triglyceride level was decreased in rats fed only EB water extract as compared to control rats, but HDL cholesterol was elevated with both water and ethanol extracts. Serum levels of total cholesterol, triglyceride were decreased and HDL cholesterol was increased by the EB pill. Serum levels of thiobarbituric acid-reactive substances were lower in rats fed ES powder, and EB powder, EB water extract or EB pill, as compared to the control rats. Supplementation of EB powder and EB pill reduced serum level of GOT and GPT, respectively, as compared to the control group. In conclusion, EB and EB pill improve serum lipid status and may be utilized as ingredient of functional foods for the purpose of improving serum lipid profile and inhibiting peroxidation of lipids.

  • This study was performed to investigate the effects of dried leaf powders and ethanol extracts of Perilla frutescens(perilla leaf), Artemisia princeps var. orientalis(mugwort) and Aster scaber(chamchui) on lipid metabolism and antioxidative capacity in rats. Forty-nine male Sprague-Dawley rats weighing 105.9$\pm$1.7g were blocked into seven groups according to body weigth and raised for four weeks with diets containing 5% dried powders of perilla leaf, mugwort and chamchui, or ethanol extracts from the same amount of each dried leaf powder. Food intake was higher in the control and chamchui powder groups than the other remaining groups. Weight gain was not significantly different among all experimental groups. Food efficiency ratio was highest in the perilla leaf powder group. Plasma total lipid and cholesterol, liver total lipid and triglyceride levels were highest in the perilla leaf powder group. In contrast, dried powders of mugwort and chamchui showed hypolipidemic effects in plasma and liver. Plasma and liver TBARS levels in both dried powder and ethanol extract groups of the three plants were lower than control. There was also no significant difference between corresponding dried powder and ethanol extract groups. Catalas and GSH-Px activities in erythrocyte and liver were not different among all the experimental groups. However, SOD activities were significantly different among the esperimental groups. In erythrocyte. SOD activiteis of all dried powder and ethanol extract groups except the perilla leaf powder group were higher than control, and the chamchui power group showed the highest activity among them. In liver, all the plant groups showed higher SOD activities than control. There was no significant difference between corresponding dried powder and ethanol extract groups. In conclusion, the dried chamchui powder and liver. All the dried plant powder and ethanol extract were effective in decreasing the TBARS levels of liver and particularly plasma. Among the antiocidative enzymes, SOD activity was most responsive to the experimental diets. All the plant groups showed increases in liver SOD activities and there were more increments in dried powder groups. Since the effects of dried powder groups on lipid metabolism were better than those of extract groups, it is plausible that the high dietary fiber level in dried powders was effective. Antioxidative effects were not significantly different between corresponding dried power and extract groups, and it was thought that ethanol extracts from three plants also had similar antioxidative effects as dried powderers.

  • This experiment was designed to investigate the effect of the CMT and MCMT hot water extract & ultra-fine powder on microglia and memory deficit model. The effects of the CMT and MCMT hot water extract on expression of IL-l${\beta}$, IL-6, TNF-${\alpha}$, NOS-II, COX-2, IL-10, TGF-${\beta}$1 mRNA and production of IL-lP, IL-6, TNF-a, NO, ROS in BV2 microglial cell line treated by lipopolysacchaide(LPS) ; serum glucose, uric acid, AChE activity of the memory deficit mice induced by scopolamine , behavior of the memory deficit mice induced by scopolamine and were investigated, respectively. The CMT and MCMT hot water extract suppressed the expression of IL-1${\beta}$, IL-6, TNF-${\alpha}$, NOS-II, COX-2 mRNA, production of IL-l${\beta}$, IL-6, TNF-${\alpha}$, NO, ROS and increased the expression of IL-10, TGF-${\beta}$l mRNA in BV2 microglial cell line treated by LPS. The MCMT hot water extract & ultra-fine powder increased glucose, decreased uric acid and AChE significantly in the serum of the memory deficit mice induced by scopolamine. The CMT and MCMT hotwater extract & ultra-fine powder groups showed significantly inhibitory effect on the scopolamine-induced impairment of memory in the experiment of Morris water maze. According to the above result, it is suggested that the CMT and MCMT hot water extract & ultra-fine powder might be usefully applied for prevention and treatment of dementia.

  • Hepatic fibrosis refers to the scarring of liver tissue, often resulting from chronic liver injury or inflammation. It is characterized by excessive deposition of extracellular matrix proteins, impairing liver function and potentially progressing to cirrhosis if left untreated. To improve the liver functions, Cordyceps militaris, a species of parasitic fungus known for its medicinal properties, is used in the form of extract. It has been traditionally used in Chinese medicine to boost energy, improve stamina, and support overall health. In this study, we investigated the hepatoprotective effects of Cordyceps militaris extract powder in a liver injury model induced by hepatic fibrosis. Sprague-Dawley (SD) rats were administered Dimethylnitrosamine (DMN) to induce liver injury, and the hepatoprotective effects of Cordyceps militaris extract powder intake were assessed by comparing changes in liver enzyme levels and histological observations. Rats injected with DMN were orally administered Cordyceps militaris extract powder at doses of 0, 125, 250, and 500 mg·kg-1·day-1 for three weeks. After three weeks of treatment, no significant differences were observed in hematological, clinical chemical, organ weight, gross examination, or microscopic examination between the DMN-alone group and the Cordyceps militaris extract powder-treated group. In conclusion, hepatoprotective effects against DMN-induced liver injury in SD rats treated with Cordyceps militaris extract powder were not observed under this study condition.

Optimization of Microencapsulation of Inonotus obliquus Extract Powder by Response Surface Methodology and Its Application into Milk

  • This study was carried out to optimize microencapsulation conditions for Inonotus obliquus extract powder with mediumchain triacylglycerol (MCT) using response surface methodology (RSM), and to determine the possibility of adding encapsulated I. obliquus extract to milk toward improving the functionality of milk. and to investigate the effects of added encapsulated I. obliquus extract on the physicochemical and sensory properties of the product. The microencapsulation efficiency of I. obliquus extract powder was investigated with respect to three variables (ratios of core material to coating material, amounts of water added, and spray pressure) in RSM. The optimal conditions for microencapsulation of I. obliquus extract powder were obtained from the ratio of core material to coating material (2.92:10) and the amount of water added (0.5 mL); the spray pressure had an insignificant effect on the microencapsulation (p<0.05). Adding encapsulated I. obliquus extract into milk did not significantly affect either color (L, a and b values) or thiobarbituric acid (TBA) values. Sensory test revealed that there were significant improvements in brownish, herb flavor, and bitterness scores for encapsulated I. obliquus extract-added milk as compared with those for uncapsulated I. obliquus extract-added milk. Based on the data obtained from the present study, it is concluded that microcapsules of I. obliquus extract powder could be applicable to milk without remarkably adverse effects on physicochemical and sensory properties

  • The components of Red-ginseng radix extract solution for herb- acupuncture were analyzed by HPLC. According to the Medical Product Safety Administration Guidelines for safety assessment, mice and rats were used for acute toxicity test. The results were summarized as follows; 1. In the Saponin contents(%) of Red-ginseng radix extract, Ginsenoside $Rb_1$ Saponin was 0.27% in raw material, 1.67% in extract powder and Ginsenoside Rc Saponin was 0.16% in raw material, 1.12% in extract powder and Ginsenoside Rd Saponin was 0.08% in raw material, 0.54% in extract powder. 2. There were no abnormal findings in acute toxicity test treated with Red-ginseng radix extract solution for herb-acupuncture and $LD_{50}$ could not be measured.

Quality Characteristics of Muffins with Added Acorn Jelly Powder and Acorn Ethanol Extract Powder (도토리묵가루 및 추출물을 첨가한 머핀의 품질특성)

  • In this study, muffins were made with the addition of 50% acorn jelly powder and 0, 0.1, and 0.5% acorn ethanol extract powder. The moisture content of muffins decreased with an increase in the acorn ethanol extract powder concentration. Baking loss rate decreased with an increase in powder concentration. The lightness and yellowness of muffins decreased as the concentration of powder increased. Redness increased as the concentration of powder increased. The hardness of muffins increased, and cohesiveness decreased with an increase in powder concentration. Gumminess, and chewiness decreased by the addition of powder. The results of a sensory test showed that the texture quality of the muffin increased as the amount of added acorn increased. The taste of muffins increased with an increase in the powder concentration, but 1.0% acorn ethanol extract powder decreased. In terms of the overall acceptability, 0.1% acorn ethanol extract powder had the highest preference level.
