Advances in higher-order chromatin architecture: the move towards 4D genome

  • In eukaryotes, the genome is hierarchically packed inside the nucleus, which facilitates physical contact between cis-regulatory elements (CREs), such as enhancers and promoters. Accumulating evidence highlights the critical role of higher-order chromatin structure in precise regulation of spatiotemporal gene expression under diverse biological contexts including lineage commitment and cell activation by external stimulus. Genomics and imaging-based technologies, such as Hi-C and DNA fluorescence in situ hybridization (FISH), have revealed the key principles of genome folding, while newly developed tools focus on improvement in resolution, throughput and modality at single-cell and population levels, and challenge the knowledge obtained through conventional approaches. In this review, we discuss recent advances in our understanding of principles of higher-order chromosome conformation and technologies to investigate 4D chromatin interactions.

The experiences of patients with tinnitus: a qualitative study (이명 환자의 증상 경험 연구)

  • Purpose: Tinnitus, which is often described as the conscious awareness of a sound without an external auditory stimulus, is one of the most common symptoms in the general population. This study aimed to explore the experiences of individuals living with tinnitus. Methods: In this qualitative exploratory study, data were collected from November 2017 to January 2018 through in-depth individual interviews with 11 tinnitus patients. Verbatim transcripts and field notes were analyzed using content analysis. Results: The extracted content was classified into three core categories of tinnitus experiences: "suffering from constant sound," "being imprisoned surrounded by persistent noise," and "efforts to overcome." Conclusion: This study shows that tinnitus had a severe negative impact on patients' lives. The finding will help healthcare providers understand their patients more deeply.

A Simple, Reliable, and Inexpensive Intraoperative External Expansion System for Enhanced Autologous Structural Fat Grafting

  • External volume expansion of the recipient site by suction has been proposed as a way of improving fat graft survival. The objective of this study was to present an innovative and simple intraoperative external expansion system to enhance small-volume autologous fat grafting (40-80 mL) and to discuss its background and its mechanism of action. In this system, expansion is performed using a complete vacuum delivery system known as the Kiwi VAC-6000M with a PalmPump (Clinical Innovations). The recipient site is rapidly expanded intraoperatively 10 times for 30 seconds each with a negative pressure of up to 550 mm Hg before autologous fat injection. During this repetitive stimulation, the tissues become grossly expanded, developing macroscopic swelling that regresses slowly over the course of hours following the cessation of the stimulus. The system sets various mechanisms in motion, including scar release, mechanical stimulation, edema, ischemia, and inflammation, which provide an environment conducive for cell proliferation and angiogenesis. In order to maintain the graft construct in its expansive state, all patients are encouraged postoperatively to use the Kiwi three times daily for one minute per session over the course of three days. The handling of this system is simple for both the patients and the surgeon. Satisfactory clinical outcomes have been achieved without significant complications.

  • In functional materials, in situ experimental techniques as a function of external stimulus (e.g., electric field, magnetic field, light, etc.) or changes in ambient environments (e.g., temperature, humidity, pressure, etc.) are highly essential for analyzing how the physical properties of target materials are activated/evolved by the given stimulation. In particular, in situ electric-field-dependent X-ray diffraction (XRD) measurements have been extensively utilized for understanding the underlying mechanisms of the emerging electromechanical responses to external electric field in various ferroelectric, piezoelectric, and electrostrictive materials. This tutorial article briefly introduces basic principles/key concepts of in situ electric-field-dependent XRD analysis using a lab-scale XRD machine. We anticipate that the in situ XRD method provides a practical tool to systematically identify/monitor a structural modification of various electromechanical materials driven by applying an external electric field.

A Study for the Analysis of EEG Signals Evoked by Auditory Stimulus using Wavelet Transformations (Wavelet변환을 이용한 청각자극에 의해 유발되는 뇌파의 분석에 관한 연구)

  • We are exposed to the various external stimuli input from the environment, which cause emotional changes based on the characteristics of the stimuli. Unfortunately, there are no quantitative results on relationship between human sensibility and the characteristics of physiological signals. The objective of this study was to quantify EEG signals evoked by auditory stimulation based on the assumption that the analysis of the variability on the characteristics of the EEG waveform may provide the significant information regarding changes in psychological states of the subject. The experiment was devised with seven experimental conditions, which are control and six different types of auditory stimulation. Twenty subjects were used to obtain EEGs while introducing auditory stimulation. Wavelet transformation was employed to analyze the EEG signals. The results showed that the reconstructed signals at the decomposition level revealed the different energy value on the EEG signals. Also, general patterns of EEG signals in rest state compare with negative and positive stimulus were found. This study could be extended to estabilish an algorithm which distinguishes psychophysiological states of the subjects exposed to the auditory stimulation.

The combined effect of extraoral vibratory stimulus and external cooling on pain perception during intra-oral local anesthesia administration in children: a systematic review and meta-analysis

  • This study aimed to assess the combined use of extraoral vibratory stimulation and extraoral cooling in reducing the pain (subjective and objective) of dental local anesthesia administration in children. PubMed, Cochrane Central Register of Controlled Trials, and Ovid SP databases were searched up to July 2021. Article titles were screened and full-text evaluations of the selected articles were performed. Finally, seven studies (391 children, aged 4 - 12 years) were included in this qualitative and quantitative analysis. The pooled data determined the combined effect of extraoral vibration and extraoral cooling as a single measure. Extraoral vibration or cooling alone were not compared. The measured primary and secondary outcomes were pain perception and subjective and objective pain, respectively. When compared with the control, extraoral vibration and cooling resulted in significant differences in the mean combined data for the variables, pain perception, and pain reaction. Children's subjective pain as measured by pain scores were reduced when extraoral vibration and cooling was used during local anesthesia administration (mean difference -3.52; 95% confidence interval [-5.06 - 1.98]) and objective pain (mean difference -1.46; 95% confidence interval [-2.95 - 0.02] ; mean difference -1.93; 95% confidence interval [-3.72 - 0.14]). Within the confines of this systematic review, there is low-quality evidence to support the use of combined extraoral vibration and cooling for reducing pain (subjective and objective) during intraoral local anesthesia administration in children.

  • This paper critically examines the role performance of local distributors within the Stimulus-Organism-Response (SOR) model, while also considering the moderating influence of market competition on the organism (O) and response (R) elements. Adopting a holistic approach, the SOR model provides a comprehensive framework for analyzing how external stimuli, including distributive, procedural, and interaction unfairness, interact with internal psychological processes, such as perceived unfairness, to shape the long-term orientation of importing agents. Moreover, this study acknowledges the pivotal role of market competition in the operational context of local distributors. It posits that competitive market dynamics play a crucial role in intensifying the relationship between behavioral factors and the long-term orientation of distributors, thereby revealing contingent effects within the SOR model. Through the exploration of these dynamics, this study contributes to a comprehensive understanding of the interplay among external stimuli, internal psychological processes, and market competition within the SOR framework, advancing our knowledge in this field.

A Relationship Between the EMG Silent Period and Muscle Fatigue of the Masticatory Muscle (저작근의 근전도 휴지기와 근육피로와의 관계)

  • Silent period(SP) is transient stops of muscle activity that are induced by mechanical or electrical stimulus and the duration of silent period is a important parameter that have been associated with symptoms of masticatory dysfunction. Muscle fatigue is induced by sustained muscular contractions. It is associated with the external manifestations as the inability to maintain a desired force output, muscular tremor, and localized pain. Muscle fatigue is a parameter that have been measured or monitored the deterioration of a performance of muscles. On the study of relationship between SP and masticatory muscle fatigue, Nagasawa suggested that SP increases up to 3 min. from the beginning of clenching when the subjects performed sustained contractions at 50% maximum clenching level. In this paper, in order to evaluate a relationship between SP and muscle fatigue, 10 SPs per 1 minute are measured at 10%, 20%, 50% maximum clenching level. We used the median frequency in order to quantify the degree of muscle fatigue. However, the results shows that the duration of silent period was not significantly affected by differing levels of muscle fatigue. Therefore, we suggest that the SP is not increased or decreased as the results of muscle fatigue, also the origin of the SP generation mechanism is discussed.

Investigation of Visual Perception Under Zen-Meditation Based On Alpha-Dependent F-VEPs

  • Variation of brain dynamics under Zen meditation has been one of our major research interests for years. One issue encountered is the inaccessibility to the actual meditation level or stage as a reference. In this paper, we propose an alternative strategy for investigating the human brain in response to external flash stimuli during Zen meditation course. To secure a consistent condition of the brain dynamics when applying stimulation, we designed a recording of flash visual evoked potentials (F-VEPs) based on a constant background EEG (electroencephalograph) frontal $\alpha-rhythm$ dominating activities that increase significantly during Zen meditation. Thus the flash-light stimulus was to be applied upon emergence of the frontal $\alpha-rhythm$. The alpha-dependent F-VEPs were then employed to inspect the effect of Zen meditation on brain dynamics. Based on the experimental protocol proposed, considerable differences between experimental and control groups were obtained. Our results showed that amplitudes of P1-N2 and N2-P2 on Cz and Fz increased significantly during meditation, contrary to the F-VEPs of control group at rest. We thus suggest that Zen meditation results in acute response on primary visual cortex and the associated parts.
