Characterization of an Anti-gout Xanthine Oxidase Inhibitor from Pleurotus ostreatus

  • We selected Pleurotus ostreatus from among several edible mushrooms because it has high anti-gout xanthine oxidase (XOD) inhibitory activity. The maximal amount of XOD inhibitor was extracted when the Pleurotus ostreatus fruiting body was treated with distilled water at $40^{\circ}C$ for 48 hr. The XOD inhibitor thus obtained was purified by Sephadex G-50 gel permeation chromatography, ultrafiltration, $C_{18}$ solid phase extraction chromatography and reverse-phase high-performance liquid chromatography with 3% of solid yield, and its XOD inhibitory activity was 0.9 mg/mL of $IC_{50}$. The purified XOD inhibitor was a tripeptide with the amino acid sequence phenylalanine-cysteine-histidine and a molecular weight of 441.3 Da. The XOD inhibitor-containing ultrafiltrates from Pleurotus ostreatus demonstrated dose-dependent anti-gout effects in a Sprague-Dawley rat model of potassium oxonate-induced gout, as shown by decreased serum urated levels at doses of 500 and 1,000 mg/kg, although the effect was not as great as that achieved with the commercial anti-gout agent, allopurinol when administered at a dose of 50 mg/kg.

  • The nature of work which is changing has increased the importance of HRD within organization. Also, these changes are making HRD shift its paradigm : from the segregated, closed, operational & administrative, dependent, utilitarian view to the integrative, open, strategic & facilitative, interdependent, generative view. The shifts of the perspective on HRD cause HRD professionals rethink the purpose and definition of HRD, and the role & competency needed to HRD in 21st century. The findings which are drawn through literature review and researcher’s intuition based on a long field experience are as follows. First, The function of HRD has recently been much more important then that of the past. Second, There is no consensus over the conceptual identity of HRD. Nonetheless, it is apparent that the preferred activities of the HRD function is performance improvement. Third, It is too difficult that we should define clearly the role & competency of HRD because of diverse perspectives on them. But we find much evidence converging to support the fact that the role & competency of HRD should be extended from the sole focus on training & development to adding the additional role & competency of change agent, who includes planning, implementing, and managing change.

Pseudomonas putida Strain 17 Isolated from Replant Soil Promotes Tomato Growth and Inhibits Conidial Germination of Soilborne Plant Pathogens

  • The induction of growth promotion on numerous crops by rhizobacteria is a well documented phenomenon. In case of tomato (Lycopersicon esculentum), fruit yield is higher in replant soil than that in fresh soil. To investigate what kind of rhizobacterium is involved, microbial community in rhizosphere and on rhizoplane of tomato plants from each soil was analyzed by dilution plating on selective media. Many Gram-negative bacteria and actinomycetes were isolated from tomato in replant soil. One Gram-negative rhizobacterium isolated was identified as Pseudomonas putida based on its biochemical characteristics, fatty acid methyl ester analysis and 16S rDNA sequence. This bacterium designated strain 17 inhibited the growth of Pseudomonas corrugata, and increased growth of tomato seedlings. In addition, its culture filtrate inhibited conidial germination of plant-pathogenic fungi such as Fusarium oxysporum f. sp. radicis-lycopersici, F. oxysporum f. sp. cucumerinum, and Nectria radicicola. Scanning electron microscopy revealed strain 17 colonized and persisted on the epidermal surfaces of tomato radicles and roots. These results suggest that P. putida strain 17 may serve as a biological control agent to suppress multiple soil-borne diseases for tomato plants. Increased microbial populations that suppress deleterious microorganisms including pathogens could be one of the major factors in increased tomato yield in replant soil.

Differential Responses of Soybean Cultivars to Cercospora sojina Isolates, the Causal Agent of Frogeye Leaf Spot in Korea

  • During the summer of 2005, specimens of soybean cultivars (Daepung, Daewon, Hwanggeum and Taegwang) showing frogeye leaf spot (FLS) were obtained from various areas in Korea. Eight isolates identified as Cercospora sojina were inoculated on the adaxial leaf surfaces of 63 Korean soybean cultivars; the disease responses to each isolate were evaluated 14 days and 21 days after inoculation. Based on the disease responses (resistant or susceptible) of the cultivars by the isolates, a set of cultivars (Anpeong, Bogwang, Cheongdu No. 1, Cheongja No. 3, Dachae, Daemang, Jangwon, Namhae, Sowon, Taegwang) were selected and inoculated with seven isolates for further testing pathogenic variance. Interestingly, 6 out of 7 tested C. sojina isolates revealed differential ability in infecting different soybean cultivars. This result may indicate the possibility of new race occurrence or pathogenic variation; this also presents evidence for prevalent FLS occurrence during humid and hot weather in Korea.

Tencel FR/Cotton 복합원단의 물리적 특성 및 난연성에 관한 연구 (Study on the Physical Properties and Flame Retardancy of Tencel FR/Cotton Complex Knit Fabrics)

  • This study analyzed the physical properties and flame retardancy of Tencel FR/Cotton complex knit fabrics in order to satisfy two aspects of eco-friendliness and functionality. The flame retardant (FR) treatment of complex knit fabrics was applied by a pad-dry-cure method for additional functionality. Tensile strength, extension, bursting strength, LOI, and flame retardancy were measured by the KS (Korean Standard) K manual. The hand value knit fabrics were measured by KES-FB system. Subsequently, tensile strength and extension of wale and tensile strength of course increased in tandem with the Tencel FR yarn content. Tencel FR/Cotton complex knit fabrics were suitable for summer-weight and for baby clothes through the KES-FB system measurements. The bursting strength of Tencel FR/Cotton complex knit fabrics decreased as the contents of the Tencel FR increased; in addition, LOI increased as the contents of Tencel FR increased. This was due to the Tencel FR flame resistance function; however, the tensile strength decreased. The optimum fiber content of Tencel FR/cotton content was 1:1. The optimum conditions of flame retardant treatment were a treatment temperature $130^{\circ}C$ and the concentration of finishing agent and assistance binder (AR4260) was 35% and 1%, respectively.

  • 이 논문에서는 소프트웨어 부품이 되는 컴포넌트를 설계하기 위한 요소 중 컴포넌트간 상호작용에 중점을 두어 이를 체계적으로 분류한 후, 이에 맞는 도해적 표기방법(graphical notation)과 이를 이용한 설계 방법을 제시하고 실제 설계 사례를 통해 이를 검증하며, 컴포넌트 설계 및 모델링을 효과적으로 표현할 수 있는 방법에 대해 논의하였다. 컴포넌트는 일반 클래스와는 달리 그 규모가 크며 인터페이스와 이를 이용한 컴포넌트간의 상호작용에 대한 표현이 매우 부족하다. 따라서 이 연구에서는 컴포넌트 표현기법을 제시하기 위하여 UML을 확장하였고 이를 이용하여 실험적으로 설계해 보고 그 효용성을 검토하였다.

Content Protective Multi-Agent Platform for MsMu Service and Pattern-Based Content Management

  • Recent research on mobile Internet protocol television and digital right management (DRM) interconnections has focused on multimedia technologies designed to enhance content scalability and adaptive content distribution. However, due to the architectural and scalable limitations, recent systems are not flexible and securable with respect to their adaptive content distribution and protective policy management. Therefore, we propose a content protective multi-agent platform that provides secure multimedia services, correlation management, pattern-based management, and multi-source multi-use (MsMu)-based services. Our architecture, supported by DRM, lets us create a rich set of MsMu-based content protection and seamless multimedia services through the extension of one source multi-use (OsMu)-based content services. We have verified our platform, which provides scalable and securable services with a 17% lower service response time by using a testbed.

Bacterial Mixture from Greenhouse Soil as a Biocontrol Agent Against Root-Knot Nematode, Meloidogyne incognita, on Oriental Melon

  • The biological control efficacy of a greenhouse soil bacterial mixture of Lactobacillus farraginis, Bacillus cereus, and Bacillus thuringiensis strains with antinematode activity was evaluated against the root-knot nematode Meloidogyne incognita. Two control groups planted in soil drenched with sterile distilled water or treated with the broad-spectrum carbamate pesticide carbofuran were used for comparison. The results suggest that the bacterial mixture is effective as a biocontrol agent against the root-knot nematode.

Occurrence of Bacterial Stem Rot of Ranunculus asiaticus Caused by Pseudomonas marginalis in Korea

  • In December 2016, stem rot symptoms were observed on Persian buttercup (Ranunculus asiaticus) plants in Chilgok, Gyeongbuk, Korea. In the early stage of the disease, several black spots appeared on the stem of infected plants. As the disease progressed, the infected stem cleaved and wilted. The causal agent was isolated from a lesion and incubated on Reasoner's 2A (R2A) agar at $25^{\circ}C$. Total genomic DNA was extracted for phylogenetic analysis. Based on the 16S rRNA gene analysis, the isolated strain was found to belong to the genus Pseudomonas. To identify the isolated bacterial strain at the species level, the nucleotide sequences of the gyrase B (gyrB) and RNA polymerase D (rpoD) genes were obtained and compared with the sequences in the GenBank database. As the result, the causal agent of the stem rot disease was identified as Pseudomonas marginalis. To determine the pathogenicity of the isolated bacterial strain, it was inoculated into the stem of healthy R. asiaticus plant, the inoculated plant showed a lesion with the same characteristics as the naturally infected plant. Based on these results, this is the first report of bacterial stem rot on R. asiaticus caused by P. marginalis in Korea.

타우린의 뼈 형성 작용과 관련질환의 경감 (Taurine in Bone Formation and Alleviation of Its Diseases)

  • Taurine, amino acid, chemically known as 2-amino ethane sulphonic acid was discovered more than two hundred years ago from ox bile. it is widely distributed in both mammals and nonmammals. It is found in considerably high amount in hUl11an: a normal adult of 70 kgs contains about 70 grams of taurine. Taurine with this much concentration, is involved in almost all life processes. Its deficiency causes several abnormalities in major organs like brain, eye and heart. Taurine-bone interaction is latest addition to its long list of actions. In bone cells, taurine is also found in high concentration. Taurine is found to help in enhancing the bone tissue formation which is evidenced by increased matrix formation and collagen synthesis. Besides stimulating the bone tissue formation, it also inhibits the bone loss through inhibiting the bone resorption and osteoclast formation. Thus, taurine acts as a double agent. In addition to these two major actions of taurine in bone, it also has beneficial effect in wound healing mld bone repair. Taurine possess radioprotective properties, too. As it is a naturally available molecule, it can be used as a preventive agent. Taurine has a potential to replace bisphosphonates which are currently in use for the inhibition of bone loss but this needs in depth study. As taurine is involved in bone formation and inhibition of bone loss, a detailed study can make it a single marker of bone metabolism. All these taurine-bone interaction is a symbol of their deep involvement but still require further extension to make taurine as a choice for tile sound bone health.
