  • The purpose of this study is to show inherent and differentiated beauty by designing costume knitwear using Dancho patterns representing peculiar beauty, thus expressing free and convenient knitwear in the modern sense for women with the image of reasonableness and elegance, proposing knit design using high-class materials for satisfying the demand for the highest. With regard to expression technique of works, I selected computer Jacquard method to indicate effective comparison effect of expression material and made expression with $12{\sim}14$ gauge as base by using computer knitting machine. Through above-mentioned research process and design using Knit CAD system, I obtained following conclusion. First, it is possible to express variously and widely traditional patterns by making the most of the beauty of the orient and by recomposing the patterns of plant patterns representing "feminine image". Second, it is possible to widely represent the sense you can feel from knit by expressing variously diversity of knit structure and Jacquard knitting structure and gauge. Finally, it is possible to express designs more freely and imaginably by using computer Knit CAD system instead of actual knitting of various knit expression technique and materials application.

  • Development of internet had centered on that offer universality of expression. But, graphic processing of expression center has many inconveniences because of method that is not structural in World Wide Web. Announced SVG that is XML application to permit efficient expression and storage of vector graphic expression, share in W3C to supplement these things. Therefore, because using SVG document in this treatise, edit vector graphic through interface that could edit expressed graphic object directly structural edit and screen that is offered in graphic edit system form and designed SVG edit system to express. This system may use usefully to efficient XML graphics processing on the GIS system and Mobile service environment and so on.

Induction of cancer cell-specific death via MMP2 promoterdependent Bax expression

  • Controlled gene expression in specific cells is a valuable tool for gene therapy. We attempted to determine whether the lentivirus-mediated Tet-On inducible system could be applied to cancer gene therapy. In order to select the genes that induce cancer cell death, we compared the ability of the known pro-apoptotreic genes, Bax and tBid, and a cell cycle inhibitor, p21cip1/waf1, and determined that Bax was the most effective. For the cancer cell-specific expression of $rtTA2^S$-M2, we tested the matrix metalloproteinase-2 (MMP-2) promoter and determined that it is highly expressed in cancer cell lines, including SNU475 cells. The co-transduction of two lentiviruses that contain sequences for TRE-Bax and $rtTA2^S$-M2, the expression of which is controlled by the MMP-2 promoter, resulted in the specific cell death of SNU475, whereas other cells with low MMP-2 expression did not evidence significant cell death. Our data indicate that the lentivirus-mediated Tet-On system using the cancer-specific promoter is applicable for cancer gene therapy.

Expression of an Antimicrobial Peptide Magainin by a Promoter Inversion System

  • A method was developed for the controlled expression of an antimicrobial peptide magainin in Escherichia coli. A series of concatemeric magainin genes was constructed with a gene amplification vector, and fused to the 3'end of malE gene encoding the affinity ligand, E. coli maltose-binding protein (MBP). The construct directed the synthesis of the fusion protein with the magainin polypeptide fused to the C-terminus of MBP. The fusion protein was expressed in a tightly regulatable expression system which was under the control of an invertible promoter. The MBP-fused magainin monomer was expressed efficiently. However, the expression level of the MBP-fused magainin in E. coli decreased with the increasing size of multimers possibly because of the transcription and translation inhibition by the multimeric peptides. After purification using an amylose affinity column, the fusion protein was digested by factor Xa at a specific cleavage site between the monomers. The recombinant magainin had an antimicrobial activity identical to that of synthetic magainin. This experiment shows that a biologically active, antimicrobial peptide magainin can be produced by fusing to MBP, along with a promoter inversion vector system.

Facial Expression Control of 3D Avatar by Hierarchical Visualization of Motion Data (모션 데이터의 계층적 가시화에 의한 3차원 아바타의 표정 제어)

  • This paper presents a facial expression control method of 3D avatar that enables the user to select a sequence of facial frames from the facial expression space, whose level of details the user can select hierarchically. Our system creates the facial expression spare from about 2,400 captured facial frames. But because there are too many facial expressions to select from, the user faces difficulty in navigating the space. So, we visualize the space hierarchically. To partition the space into a hierarchy of subspaces, we use fuzzy clustering. In the beginning, the system creates about 11 clusters from the space of 2,400 facial expressions. The cluster centers are displayed on 2D screen and are used as candidate key frames for key frame animation. When the user zooms in (zoom is discrete), it means that the user wants to see mort details. So, the system creates more clusters for the new level of zoom-in. Every time the level of zoom-in increases, the system doubles the number of clusters. The user selects new key frames along the navigation path of the previous level. At the maximum zoom-in, the user completes facial expression control specification. At the maximum, the user can go back to previous level by zooming out, and update the navigation path. We let users use the system to control facial expression of 3D avatar, and evaluate the system based on the results.

Effects of Ginseng Saponin on the Cytokine Gene Expression in Human Immune System (인삼 사포닌이 인간면역계 사이토카인 유전자의 발현에 미치는 영향)

  • In order to investigate the Immunomodulatory effects of ginseng, we have studied the effects of ginseng saponin on the proliferation and cytosine gene expression of human pheripheral blood mononuclear cell (PBMC). In the PBMC proliferation assay, total saponin exhibited proliferation inhibition on the PBMC or phytohemagglutinin(PHA)-stimulated PBMC in a dose-dependent fashion. Immunomodulatory effects of ginseng were further investigated using the cytokine gene expression as the indicators. In the reverse transcription-polymerase chain reaction (RT-PCR) test, interleukin (IL)-1, IL-2, IL-3, IL-4, IL-6, IL-13, granulocyte macrophage-colony stimulating factor, tumor necrosis factor (TNF), migration inhibitory factor and transforming growth factor genes were expressed in the PHA-stimulated PBMC 48 hrs after cell culture. Among expressed cytokines, total saponin could increase the expression of IL-1 and TNF of PBMC without stimulation of PHA. All of ginsenosides, $Rb_1$, $Rb_2$, $Rg_1$, Rc, Re, incresed TNF gene expression. Especially, Rb2 (20 g/ml) showed most prominent effect on TNF gene expression and it also slightly increased IL-1 gene expression of PBMC.

Realtime Facial Expression Data Tracking System using Color Information (컬러 정보를 이용한 실시간 표정 데이터 추적 시스템)

  • It is very important to extract the expression data and capture a face image from a video for online-based 3D face animation. In recently, there are many researches on vision-based approach that captures the expression of an actor in a video and applies them to 3D face model. In this paper, we propose an automatic data extraction system, which extracts and traces a face and expression data from realtime video inputs. The procedures of our system consist of three steps: face detection, face feature extraction, and face tracing. In face detection, we detect skin pixels using YCbCr skin color model and verifies the face area using Haar-based classifier. We use the brightness and color information for extracting the eyes and lips data related facial expression. We extract 10 feature points from eyes and lips area considering FAP defined in MPEG-4. Then, we trace the displacement of the extracted features from continuous frames using color probabilistic distribution model. The experiments showed that our system could trace the expression data to about 8fps.

  • In this paper, both an approximate expression and an exact expression for the contribution factor of an element to the natural frequency of the finite element discretized system of a structure in general and a membrane in particular have been derived from the energy conservation principle and the finite element formulation of structural eigenvalue problems. The approximate expression for the contribution factor of an element is used to predict and determine the elements to be removed in an iteration since it depends only on the quantities associated with the old system in the iteration. The exact expression for the contribution factor of an element makes it possible to check whether the element is correctly removed at the end of an iteration because it depends on both the old system and the new system in the iteration. Thus, the combined use of the approximate expression and the exact expression allows a considerable number of elements to be removed in a single iteration so that the efficiency of the evolutionary structural optimization method can be greatly improved for solving the natural frequency optimization problem of a structure. A square membrane with different boundary supports has been chosen to investigate the general evolutionary path for the fundamental natural frequency of the structure. The related results indicated that if the objective of a structural optimization is to raise the fundamental natural frequency of the structure to an optimal value, the general evolutionary path during its optimization is that the elements are gradually removed along the direction from the area surrounded by the contour of the highest value to that surrounded by the contour of the lowest value.

Expression of Anti-breast Cancer Monoclonal Antibody in Transgenic Plant

  • BACKGROUND: Plant expression system for mass production of recombinant proteins has several advantages over other existing expression systems with economical and safety issues. Breast cancer is a cancer originating from breast tissue and comprises almost 25% of all cancers in women world widely. Lewis-Y antigen is difucosylated oligosaccharide and is carried by glycoconjugates at cancer cell surface. In this study, the anti-breast cancer mAb BR55, which recognizes the epitope Lewis-Y, was expressed in the plant expression system. METHODS AND RESULTS: We have developed plant system for production of mAb BR55 with or without KDEL (the ER retention signal). This ER retention signal was attached to C-terminus of protein to help retain the recombinant glycoprotein carrying oligomannose glycans and enhance glycoprotein accumulation. PCR analysis was performed and confirmed the presence of recombinant genes. Western blot validated that the recombinant proteins mAb BR55 with or without KDEL were expressed in transgenic plants, moreover, the expression level of the mAb BR55 with KDEL was higher compared to the mAb BR55 without KDEL. CONCLUSION: These results indicate that KDEL fusion is a good way to produce proteins and plant can be an ideal expression system to obtain proteins and enhance accumulation of proteins.
