The nitric oxide (NO) formation and intrinsic nitrosation may be involved in the possible mechanisms of liver fluke-associated carcinogenesis. We still do not know much about the responses of inducible NO synthase (iNOS) induced by Clonorchis sinensis infection. This study was conducted to explore the pathological lesions and iNOS expressions in the liver of mice with different infection intensity levels of C. sinensis. Extensive periductal inflammatory cell infiltration, bile duct hyperplasia, and fibrosis were commonly observed during the infection. The different pathological responses in liver tissues strongly correlated with the infection intensity of C. sinensis. Massive acute spotty necrosis occurred in the liver parenchyma after a severe infection. The iNOS activity in liver tissues increased, and iNOS-expressing cells with morphological differences were observed after a moderate or severe infection. The iNOS-expressing cells in liver tissues had multiple origins.
In order to evaluate the effects of the cell-mediated immunity of the animal in the chronic diseases the guinea pigs and rabbits were inoculated with Mycobacterium bovis. After 6 weeks these animals were sensitized and challenged with 2,4-dinitrochlorobenzene. The cutaneous reactions observed in these animal species were similar each other. Macroscopic and microscopic responses in the animal experimentally infected with M. bovis were markedly reduced compared to those in the control animals. The results indicated that the cell-mediated immunity of the animals was depressed by infection of M. bovis.
To investigate the pathogenicity and virological properties of TJ-89-2 strain of canine parvovirus(CPV -2) that was isolated from the diseased puppies in Korea, seven puppies-were inoculated intraorally with the isolate at the HA titer of 8,192. All of the puppies showed the signs of anorexia, vomiting, diarrhea and died at the 5th to 10th days post inoculation (pi). All of the fecal samples from the puppies revealed significantly high HA titers afterward the 5th days pi. Body temperature and the number of total leucocytes were slightly increased at the early stage of infection, but extremly decreased at the stage of collapse. HI titers of the sera began to increase at the 3rd to 4th days pi, reaching 512 to 1,024 at the 4th to 5th days pi.
The pathogenecity of atypical Aeromonas salmonicida was studied in healthy Oreochromis niloticus. Inoculum at concentration of $1.5{\times}10^8CFU/ml$ and $3{\times}10^8CFU/ml$ was injected into healthy fish through intramuscular and intraperitoneal injections. Experimentally infected Oreochromis niloticus showed ulceration at the dorsal musculature and trunk region in addition to black coloration, congested gills, exophthalmia, and ocular hemorrhage. Congested liver and kidney were recorded in post-mortem examination. Mortality of the experimentally infected Oreochromis niloticus reached 100% after intramuscular injection at concentration of $3{\times}10^8CFU/ml$. Histopathological investigation of infected organs was also performed. There was a focal area of bundles of skeletal musculature showing hyalinization. In addition, hyperplasia, congestion, and fusion were noticed in the gill lamellae. There was also congestion in the blood vessels in the ocular chamber. Severe congestion was also noticed in the central vein of liver associated with focal aggregation of the melanin pigmented cells in the parenchyma. Degenerative changes were noticed in the epithelial cells lining of kidney tubules. Plant extracts carvacrol and its biological precursor cymene were found to be effective in treating experimentally infected Oreochromis niloticus at concentration of 100 or 200 ppm.
The present study was undertaken in an effort to investigate the effects of alcohol on survival of mice and on their humoral and cellular immune responses, The immune responses examined were Arthus and delayed-type hyperrsnesitivity(DTH) reactions to sheep red blood cells(SRBC), contact hypersensitivity to dinitrofluorobenzend(DNFB), antibody response to thymus - dependent SRBC and to thymus -independent polyvinylpyroridone(PVP), and the recovery of Crytococcus neoformans from the liver, spleen, kidney and brain of experimentally infected mice. The administration of ethanol concentrations of 20% or less did not cause any change in survival rates as compared withs saline injected control group. In general, ethanol administration inhibited the Arthus and DTH reactions to SRBC, contact hypersensitivity to DNFB, and antibody response to both SRBC and PVP and it also decreased the resistance of mice to C. neoformans infection. Taken together, the present study stongly suggested that ethanol inhibits immune response and decrease the resistance of mice to C. neoformans infection.
Three freshwater snail species of the family Lymnaeidae have been reported from Korea, Radix auricularia coreana, Austropeplea ollula and Fossaria truncatula. Out of 3 lymnaeid snail species, A. ollula was naturally infected with the Echinostoma cinetorchis cercariae (infection rate : 0.7%). In the experiments with the laboratory-bred snails, F. truncatula as well as A. ollula was also susceptible to the E. cinetorchis miracidia with infection rates of 25% and 40%, respectively. All of three lymnaeid snail species exposed to the E. cinetorchis cercariae were infected with the E. cinetorchis metacercariae. It is evident that A. ollula acts as the first molluscan intermediate host of E. cinetorchis in Korea, and F. truncatula may be a possible candidate for the first intermediate host of this intestinal fluke. Also, three lymnaeid snail species targeted were experimentally infected with E. cinetorchis metacercariae.
Murine mycoplasmosis, caused by Mycoplasma (M.) pulmonis, is a prominent disease in rodent animals. The aim of this study was to develop a sensitive and specific PCR assay to detect M. pulmonis in animals and to assess the suitability of this assay for the detection of mycoplasmal infection in rats experimentally infected with M. pulmonis. A new PCR assay using the M. pulmonis-specific primer pairs MPul-F and MPul-R was developed. The primers and probe for the assay were designed from regions in the 16S rRNA gene that are unique to M. pulmonis. The novel PCR assay was very specific and sensitive for M. pulmonis, detecting the equivalent of 5 pg of target template DNA. It detected only M. pulmonis and no other Mycoplasma species or other bacterial species. The newly developed PCR assay also effectively detected M. pulmonis infection in rats. These results suggest that this PCR assay using M. pulmonis-specific primer pairs of MPul-F and MPul-R will be useful and effective for monitoring M. pulmonis infection in animals.
More than 1,500 clams of Corbicula fluminea, the most favorable food source of freshwater bivalves in Korea, were collected from 5 localities to examine cercarial and metacercarial infection with Echinostoma cinetorchis. Although 3 clams infected with suspicious E. cinetorchis metacercariae out of 200 specimens collected at Kangjin, Chollanam-do were detected, no cercarial and metacercarial infections with E. cinetorchis were observed in field-collected Corbicula specimens. In the susceptibility experiments with laboratory-reared clams, those infected with miracidia of E. cinetorchis did not release their cercariae up to 60 days after infection. To confirm the identity of second intermediate host of E. cinetorchis experimentally, a total of 30 clams were exposed to the cercariae from Segmentina hemisphaerula that had been infected with miracidia of E. cinetorchis. The clams were susceptible to cercariae of E. cinetorchis with an infection rate of 93.3%. Metacercariae from clams taken more than 7 days after cercarial exposure were fed to rats (S/D strain), and adult worms of E. cinetorchis, characterized by 37-38 collar spines on the head crown, were recovered from the ileocecal regions. This is the first report of C. fluminea as a possible second intermediate host of E. cinetorchis.

Chronic viral infection has been reported to cause a range of hepatic lesion, including hepatic fibrosis, cirrhosis and hepatocellular carcinoma (HCC) in a wide variety of animal species. Woodchucks (Marmota monax) chronically infected with woodchuck hepatitis virus (WHV) develop similar progressive hepatic inflammatory and neoplastic lesions that are remarkably similar to those associated with HBV infection in humans. Twenty two-month-old offspring from woodchucks (Marmota monax) experimentally infected with woodchuck hepatitis virus, were purchased. One randomly chosen animal was autopsied. The liver exhibits marked cirrhotic changes characteristic of the pre-transformation phase of WHV. We believe that this may represent a new suitable and cost-effective model for the disease processes associated with hepadnaviruses in a number of other species, most notably Hepatitis B virus infection in man.
Trichinella spiralis is one of the important zoonotic parasites with a wide variety of vertebrates hosts in nature. The purpose of this study were to analyze ESP(Excretory-Secretory Protein) antigen, to evaluate ELISA for the serological diagnosis of Trichinosis, and to survey T. spiralis infection in finishing pigs using the pepsin digestion method and ELISA in Korea. In the analysis of ESP antigen by SDS-PAGE and Western blot, 4 major bands (70, 55, 52.6, and 49 kDa) were revealed from the ESP antigen. Predilection sites of T. spiralis were the diaphragm, the tongue, masseter muscles, intercostal muscle, and hindlimb in orders in the experimentally infected rats. Sera from 581 swine were tested by ELISA with ESP antigen. The 54 (9.3%) sera were suspected as positive reactors, however, these 54 sera were determined as false positives by the use of Western blotting. This study demonstrated that the ELISA was not suitable for the examination of T. spiralis in pork. The diaphragm muscle samples of 251 finishing pigs were tested by the method of pepsin-digestion for the presence of Trichinella larvae, however, T. spiralis was not detected from the samples. We could not find out T. spiralis infection in pig in Korea pork.
