Effects of feeding level on nutrient digestibility and enteric methane production in growing goats (Capra hircus hircus) and Sika deer (Cervus nippon hortulorum)

  • Objective: Two experiments were conducted to determine the effects of feeding level on nutrient digestibility and enteric methane ($CH_4$) emissions in growing goats and Sika deer. Methods: Three growing male goats (initial body weight [BW] of $22.4{\pm}0.9kg$) and three growing male deer (initial BW of $20.2{\pm}4.8kg$) were each allotted to a respiration-metabolism chamber for an adaptation period of 7 d and a data collection period of 3 d. An experimental diet was offered to each animal at one of three feeding levels (1.5%, 2.0%, and 2.5% of BW) in a $3{\times}3$ Latin square design. The chambers were used for measuring enteric $CH_4$ emission. Results: Nutrient digestibility decreased linearly in goats as feeding level increased, whereas Sika deer digestibility was not affected by feeding level. The enteric production of $CH_4$ expressed as g/kg dry matter intake (DMI), g/kg organic matter intake, and % of gross energy intake decreased linearly with increased feeding level in goats; however, that of Sika deer was not affected by feeding level. Six equations were estimated for predicting the enteric $CH_4$ emission from goats and Sika deer. For goat, equation 1 was found to be of the highest accuracy: $CH_4(g/d)=6.2({\pm}14.1)+10.2({\pm}7.01){\times}DMI(kg/d)+0.0048({\pm}0.0275){\times}dry$ matter digestibility (DMD, g/kg)-0.0070 (${\pm}0.0187$)${\times}$neutral detergent fiber digestibility (NDFD; g/kg). For Sika deer, equation 4 was found to be of the highest accuracy: $CH_4(g/d)=-13.0({\pm}30.8)+29.4({\pm}3.93){\times}DMI(kg/d)+0.046(0.094){\times}DMD(g/kg)-0.0363({\pm}0.0636){\times}NDFD(g/kg)$. Conclusion: Increasing the feeding level increased $CH_4$ production in both goats and Sika deer, and predictive models of enteric $CH_4$ production by goats and Sika deer were estimated.

  • Objective: To evaluate the effects of the carmine cochineal-resistant spineless cactus genotypes cv. Orelha de Elefante Mexicana (Opuntia) and Miúda (Nopalea) on the intake and digestibility of nutrients, ingestive behavior, performance, and ultrasound measurements of growing lambs. Methods: Thirty-six male (non-castrated) Santa Inês lambs were used, with an average age of 6 months and an initial average weight of 22.0±2.9 kg. They were distributed in a completely randomized design with 3 treatments (Tifton hay, Nopalea and Opuntia) and 12 replications, using initial weight as a covariate. The experimental period was 86 days, with the first 30 days used for the adaptation of the animals to the facilities, diets and management, and the remaining 56 days used for evaluation and data collection. Results: The intake and apparent digestibility of dry matter (DM), organic matter (OM), crude protein (CP), neutral detergent fiber (NDF), total carbohydrates (TC), non-fibrous carbohydrates (NFC), and total digestible nutrients (TDN) showed a significant difference (p<0.05) as a function of the diets, with the Nopalea treatment (p<0.05) increasing DM intake (g/kg and % body weight [BW]), CP, TDN, and TC digestibility, whereas the Tifton hay diet led to a high (p<0.001) neutral detergent fiber corrected for ash and protein (NDFap) g/d intake, NDFap (BW %) and digestibility of said nutrient. There was no effect of treatments (p>0.05) on feeding time, however, rumination time and total chewing time were higher (p<0.05) for animals fed Tifton hay. The performance of the animals was similar (p>0.05). For the ultrasound measurements, Nopalea promoted an increase in the final loin eye area, compared to Tifton hay. Conclusion: The use of spineless cactus variety Miúda leads to the greater intake and digestibility of nutrients. The evaluated carmine cochineal-resistant spineless cactus genotypes are alternatives for semi-arid regions as they do not negatively affect the performance of growing lambs.

Effect of Biologically Active Isomers of Conjugated Linoleic Acid on Immune Response and Body Composition in Mice (Mice에서 CLA의 생물학적 활성이성체의 투여가 면역반응과 체구성에 미치는 영향)

  • Numerous physiological effects are attributed to conjugated linoleic acid(CLA). The purpose of this study is to consider these effects with respect to the cis-9, trans-11 and trans-10, cis-12 CLA isomer. Both isomers are natural products. The c9,t11-CLA isomer is the principal dietary form of CLA, but the concentrations of this isomer and the t10,c12-CLA Isomer in dairy products or beef vary depending on the diet fed to cows or steers, respectively. The influence of dietary CLA isomers on the immune response was examined, body weight and weight ratio of organ to body of Balb/C mice. Mice were divided into four groups of 8 mice. Balb/C mice were fed the experimental diets supplemented with 1% CLA (c9,t11-CLA isomer : t10,c12-CLA isomer = 2:3) (Group 1), 1% CLA (c9,t11-CLA isomer t10,c12-CLA isomer : 1:1) (Group 2), 1% safflower oil (Group 3) or nothing (Control) for 3 weeks. After 3 weeks, serum, gut lumen lavage, fat, liver, spleen and thymus were taken. Measurement of total immunoglobulin were executed using sandwich ELISA. Serum levels of IgA and IgM showed that group fed with t10,c12-CLA isomer significantly were higher than group fed with c9,tl1-CLA isomer. In addition serum level of IgG showed that group fed with t10,c9-CLA isomer significantly were lower than group fed with c9,tl1-CLA isomer. However, no significantly differences were observed in the serum IgE and secretory IgA. Weight ratio of spleen to body showed no significant differences. In weight ratio of liver and thymus to body, tl0,c9-CLA isomer significantly were respectively higher than group fed with c9,t11-CLA isomer. In weight ratio of fat to body, tl0,c9-CLA isomer significantly were respectively lower than group fed with c9,tl1-CLA isomer. In conclusion, t10,c12-CLA isomer produced a situation favorable for immunopotentiative effect and body composition. But it should be protected against hepatomegaly induced lipid accumulation in liver.

Effects of Different Exercise Intensity on FDEIA and Related Mechanisms (운동 유발성 알레르기 질환(FDEIA)에 미치는 영향과 기전분석)

  • Food-dependent exercise-induced anaphylaxis (FDEIA) is a distinct form of food allergy induced by physical exercise. It is typified by the onset of anaphylaxis during exercise, which is preceded by the ingestion of causal food allergens. Diagnosis of FDEIA is heavily dependent on clinical history. To describe the physiopathological mechanism, etiologic factors, and clinical manifestations, we evaluated the spleen index, proliferation assay of lymphocyte, ROS, ASAS, and cytokines levels in sensitized and exercise-trained mice. One-hundred mice were bred in the animal lab at D and P university under controlled conditions [$22{\pm}2^{\circ}C$, RH 45-55%, and a 12-hour photoperiod]. Animals are 7-weeks-old at the time of study and were fed a standard commercial chow diet from 09:00 to 15:00 over the 8-week study period. The mice were allowed access to distilled deionized water ad libitum. Daily food intake and weekly body gains were routinely recorded throughout the experimental period using computing scale (CAS). Mice were divided into the control group (S; control sensitized, n=25), 30 min swim training group (S30, N=25), 50 min swim training group (S50, N=25), and 80 min swim training group (S80, N=25). The results were as follows: Spleen index showed the highest level in the S80 group compared to other groups; this level was exercise-dependent. In proliferation assay of Med and OVA, the S80 group showed the highest level compared to the other groups; this level also was exercise intensity- dependent. Peritoneal ROS and IL-4 showed a statistically significant difference compared to S; however, there was no significant differences in ROS among S30, 50, and 80. From the results, we concluded that FDEIA is correlated with exercise intensity based on the levels of peritoneal ROS and cytokine profiles.

Effects of Dietary Germanium Supplementation on the Meat Quality of Duck (게르마늄 급여가 오리의 육질에 미치는 영향)

  • This study was carried out to investigate the influence of dietary germanium supplementation on the meat quality of duck. Ducks raised for 42 days were slaughtered and the ground meats were stored at 3$^{\circ}C$ for 12 days under 1200 lux. Experimental treatments were divided into 3 kinds of meat from ducks fed germanium as follows; 1) Control(natural water + commercial feed), 2) T1(1~10 days: natural water + commercial feed, 11-42 days: natural water + commercial feed supplemented with 1 % germanium), 3) T2(1~10 days: germanium submersion water + commercial feed, 11~42 days: natural water + commercial feed supplemented with 1 % germanium). The pH of duck meat was not different among the treatments(p>0.05). The germanium treatments exhibited significantly higher crude fat content in both breast and thigh meat(p<0.05). In the fatty acid composition, T2 contained more unsaturated fatty acid than control or Tl. TPA(textural profile analysis) value such as hardness, gumminess, chewiness and adhesiveness decreased in meat from duck fed supplemental germanium. Cholesterol contents of duck meat decreased in dietary supplemental germanium treatment compared to control, but were not significantly different among them(p>0.05). Dietary germanium supplementation affected on lipid oxidation of meat during storage. TBARS of meat from duck fed germanium was lower than that of control(po.05). The CIE L*, b* and h$\^$0/ values of germanium treatments were significantly(p.o5) higher than those of control. Also a* value showed more stable in germanium diet treatment during storage. Therefore, dietary supplemental germanium to duck resulted in light brownish color formation, improvement tenderness and retardation of lipid oxidation of meat during refrigerated storage.

Effects of Sotosaja-hwan on the Generation of ROS, RNS, and on the Expression of NF-${\kappa}B$-dependent Proteins in ob/ob Mouse (소도사자환이 ob/ob mouse에서 ROS/ RNS 생성 억제 및 NF-${\kappa}B$ 의존성 단백질에 미치는 영향)

  • Objectives: Peroxynitrite ($ONOO^-$), superoxide anion radical (${\cdot}{O_2}^-$ and nitric oxide (NO) are cytotoxic because they can oxidize several cellular components such as proteins, lipids and DNA. They have been implicated in the aging processes, and age-related diseases such as Alzheimer's disease, rheumatoid arthritis, cancer, diabetes, obesity and atherosclerosis. The aim of this study was to investigate the $ONOO^-$, NO, ${\cdot}{O_2}^-$ scavenging and NF-${\kappa}B$ related anti-inflammatory activities of Sotosaja-hwan in ob/ob mice. Methods: Mice were grouped and treated for 5 weeks as follows. Both the normal lean (C57/BL6J black mice) and control obese (ob/ob mice) groups have received standard chow. The experimental groups were fed with a diet of chow supplemented with 30 and 90 mg Sotosaja-hwan per 1 kg of body weight for 14 days. For this study, the fluorescent probes, namely 2',7'-dichlorodihydrofluorescein diacetate (DCFDA), 4,5-diaminofluorescein (DAF-2) and dihydrorhodamine 123 (DHR 123) were used. Western blotting was performed using anti-phospho-$I{\kappa}B$-${\alpha}$, anti-IKK-${\alpha}$, anti-NF-${\kappa}B$ (p50, p65), anti-COX-2, anti-iNOS, anti-YCAM-1 and anti-MMP-9 antibodies, respectively. Results: Sotosaja-hwan inhibited the generation of $ONOO^-$, NO and ${\cdot}{O_2}^-$ in the lipopolysaccharide (LPS)-treated mouse kidney postmitochondrial fraction in vitro. The generation of $ONOO^-$, NO, ${\cdot}{O_2}^-$ and PGE2 were inhibited in the Sotosaja-hwan-administered ob/ob mice groups. The GSH/GSSG ratio was decreased in the ob/ob mice, whereas the ratio was improved in the Sotosaja-hwan-administered groups. Sotosaja-hwan inhibited the protein expression levels of phospho-$I{\kappa}B$-${\alpha}$, IKK-${\alpha}$, NF-${\kappa}B$ (p50, p65), COX-2, iNOS, YCAM-1 and MMP-9 genes. Conclusions: These results suggest that Sotosaja-hwan is an effective $ONOO^-$, ${\cdot}{O_2}^-$ and NO scavenger and has NF-kB related anti-inflammatory activity in ob/ob mice. Therefore, Sotosaja-hwan might be a potential therapeutic drug against the inflammation process and inflammation-related diseases.

Effects of Food Waste feed and Probiotics Supplement on Productivity in Layer Chicks (남은 음식물 사료와 생균제가 산란계의 생산성에 미치는 영향)

  • The objective of this study was to investigate the effects of food waste and probiotics supplementation on productive performance and egg composition of laying hens. Total 120 layers (Brown Tetran) 22 weeks old were divided into five groups with 4 replicates for each treatments and raised in battery cage. The experimental design was completely randomized. The egg Production rate was significantly increased in layer supplemented with 0%, 0.5%, 1.0% and 1.5% of probiotic supplemented diets compared to control (P<0.05). At feeding with food waste and probiotic supplementation had no effect on the egg shell strength (P>0.05). Haugh unit of the eggs was significantly increased in layers fed with 1.5% of probiotic supplemented diet (P<0.05). The different levels of inclusion of the food waste and probiotic supplementation had not effect on egg shell color, blood spot and albumen index of the eggs (P<0.05). There were not significant differences in over all acceptability, juiciness, texture and color of the eggs from layer fed with different levels of wood waste and probiotic supplementation. The appearance and flavor significantly deteriorated in eggs from layers fed with probiotic diets compared to control (P<0.05).

Optimum stocking density of juvenile abalone, Haliotis discus hannai in recirculating culture system (순환여과 사육시스템에서 참전복의 적정 사육밀도)

  • The effects of different stocking densities on the growth of juvenile abalone, Haliotis discus hannai and water quality in the recirculating system with rotating biological contactor, were assessed. The trials were conducted using total weight 32 g juveniles abalone for 60 days at stocking densities of 5, 10, 15 kg/$m^2$. The animals were fed sufficient amounts of the pellet diet of 30.4% protein during the experimental period. Daily feeding rate (DFR) and survival rate (SR) of 5 kg/$m^2$ showed 0.76% and 85.5% those were not significantly differed 10 kg/$m^2$ showing 0.75% and 96.0% (P>0.05). DFR and SR of 15 kg/$m^2$ showing 0.38% and 31.2% were significantly lower than 5 and 10 kg/m2 (P<0.05). Daily growth rate (DGR) and feed efficiency (FE) showed the highest as 0.19% and 24.2% in 5 kg/$m^2$ the lowest as 0.05% and 14.3% in 15 kg/$m^2$ (P<0.05). According to, these results was an inverse relationship between growth and stocking density. The optimum density of juvenile abalone from this experiment is 5 to 10 kg/$m^2$ in the recirculating system.

Effect of Paprika and Zooplankton Supplementation in the Diets on Carotenoid Deposition and Skin Pigmentation of the Rose Bitterling Rhodeus ocellatus (사료내 Paprika와 동물플랑크톤 혼합첨가가 흰줄납줄개 Rhodeus ocellatus의 Carotenoid 축적 및 체색에 미치는 영향)

  • This study was conducted to evaluate the effect of dietary supplementation of paprika powder as carotenoid source with three different zooplankton on carotenoid deposition and skin pigmentation of rose bitterling Rhodeus ocellatus. Two replicate groups of fish (initial average weight 0.8 g) were fed the five experimental diets containing 5% paprika (P5) and mixture of 5% paprika with 3% three zooplankton of water flea Moina macrocopa (P5M3), Artemia nauplius Artemia sp. (P5A3) and rotifer Brachionus plicatilis (P5R3) and without supplementation of carotenoid source (Con) for 10 weeks. Survival of fish among the treatments was not affected by dietary supplementation. Total carotenoid contents of whole body in fish fed the diets containing 3% zooplankton was higher than that of fish fed other diets. Especially, the highest total carotenoid contents of whole body was observed in fish fed the P5A3 diet. Similarity, Lightness, $a^*$ and $b^*$ values of the fish fed the diets containing 3% zooplankton tended to be higher than that in fish fed control and P5 diets. The apparent color intensity of skin and fin in fish fed the diets containing paprika with Artemia nauplius and rotifer was better than those of fish fed other diets. The results of this study suggest that dietary supplementation of paprika with Artemia nauplius and rotifer can improve skin pigmentation of rose bitterling.

Effect of acute heat stress on feed intake and plasma concentrations of tumor necrosis factor-α and free amino acids in growing pigs

  • Exposure to heat stress (HS) has negative effects on pig production and health. Plasma concentrations of amino acids (AAs) can be used as indicators of HS. Therefore, this study was conducted to evaluate the effect of acute HS on feed intake, water drink, and plasma AAs in pigs. A total of 6 growing pigs (n = 6, 3 boars and 3 gilts) were raised in thermal neutral (TN; 25℃) conditions for the 5-d adaptation period as a control. After the adaptation, pigs were exposed to HS at 33℃ (HS33) for 24 h. All the pigs were fed the same diet formulated to meet or exceed predicted requirements during the whole experimental period. Blood samples were collected after the adaptation and heat treatment to verify the AAs. Measurements were the average daily feed intake (ADFI), average daily water intake (ADWI), water loss, tumor necrosis factor (TNF)-α, nitrate, total nitric oxide, and AAs in the plasma samples. Data were analyzed using the PROC GLM of SAS. HS33 had a lower ADFI (p < 0.05) and a tendency for an increased ADWI and water loss compared to TN. The level of TNF-α was significantly decreased (p < 0.05) in HS33. HS33 had a lower concentration of histidine and sarcosine compared to TN; however, there were no differences in the levels of nitrates and total nitric oxide. In conclusion, the results of this study show the differential effect of HS on the plasma concentration of AAs. We expect that the changed AAs could be potential indicators of HS.
