Expression of peroxisome proliferator-activated receptor (PPAR)-${\alpha}$ and PPAR-${\gamma}$ in the lung tissue of obese mice and the effect of rosiglitazone on proinflammatory cytokine expressions in the lung tissue

  • Purpose: We investigated the mRNA levels of peroxisome proliferator-activated receptor (PPAR)-${\alpha}$, PPAR-${\gamma}$, adipokines, and cytokines in the lung tissue of lean and obese mice with and without ovalbumin (OVA) challenge, and the effect of rosiglitazone, a PPAR-${\gamma}$ agonist. Methods: We developed 6 mice models: OVA-challenged lean mice with and without rosiglitazone; obese mice with and without rosiglitazone; and OVA-challenged obese mice with and without rosiglitazone. We performed real-time polymerase chain reaction for leptin, leptin receptor, adiponectin, vascular endothelial growth factor (VEGF), tumor necrosis factor (TNF)-${\alpha}$, transforming growth factor (TGF)-${\beta}$, PPAR-${\alpha}$ and PPAR-${\gamma}$ from the lung tissue and determined the cell counts and cytokine levels in the bronchoalveolar lavage fluid. Results: Mice with OVA challenge showed airway hyperresponsiveness. The lung mRNA levels of PPAR${\alpha}$ and PPAR-${\gamma}$ increased significantly in obese mice with OVA challenge compared to that in other types of mice and decreased after rosiglitazone administeration. Leptin and leptin receptor expression increased in obese mice with and without OVA challenge and decreased following rosiglitazone treatment. Adiponectin mRNA level increased in lean mice with OVA challenge. Lung VEGF, TNF-${\alpha}$, and TGF-${\beta}$ mRNA levels increased in obese mice with and without OVA challenge compared to that in the control mice. However, rosiglitazone reduced only TGF-${\beta}$ expression in obese mice, and even augmented VEGF expression in all types of mice. Rosiglitazone treatment did not reduce airway responsiveness, but increased neutrophils and macrophages in the bronchoalveolar lavage fluid. Conclusion: PPAR-${\alpha}$ and PPAR-${\gamma}$ expressions were upregulated in the lung tissue of OVA-challenged obese mice however, rosiglitazone treatment did not downregulate airway inflammation in these mice.

Experimental infection of white spot syndrome virus (WSSV) in wild crab, Gaetice depressus

  • To confirm the possibility of a wild marine crab, Gaetice depressus, as a carrier for white spot syndrome virus (WSSV) and to develop an alternative experimental model for WSSV in winter season, the susceptibility of the crab to WSSV was assessed by artificial challenge and subsequently tested for infection by PCR assay. The results revealed that the crabs were as highly susceptible as penaeid shrimps. WSSV caused 100% mortality in G. depressus within 16 days after intramuscular injection. The presence of WSSV in the moribund crabs was confirmed by PCR and was found in gills and muscle tissue. These results suggest that G. depressus can be naturally infected by WSSV via moribund shrimps, and can act as a potential carrier of WSSV. In addition, G. depressus can be used as an alternative experimental animal for WSSV.

Effects of Fermented Soybean Meal on Immune Response of Weaned Calves with Experimentally Induced Lipopolysaccharide Challenge

  • The present study was conducted to evaluate the effects of fermented soybean meal (FSBM) on the level of cortisol hormone and immune-related serum proteins in weaned calves after experimentally induced lipopolysaccharide (LPS) challenge. Holstein neonatal calves (n = 21; 8 males and 13 females, BW = $42.2{\pm}6.15$ kg) were randomly allocated to one of two dietary treatments: SBM (control calf starter having soybean meal (SBM) as a main protein source) and FSBM (substitute SBM in control diet with FSBM) groups. All calves were fed milk replacer using an automatic milk-feeder according to step-down milking method and weaned at 7 weeks old. Experimental diets were given to calves ad libitum throughout the experimental period. For LPS challenge, all calves except negative control animals given phosphate buffered saline (PBS), were injected subcutaneously with Salmonella typhimurium LPS on day 7 (D7) after weaning (D0). No significant difference in growth performance and milk intake was observed between SBM and FSBM calves. Feeding FSBM diet resulted in significantly (p<0.05) higher LPS-specific IgG at D12 and D19 and LPS-specific IgA at D19 in peripheral blood. Calves fed with FSBM diet also had significantly (p<0.05) higher concentration of serum haptoglobin (Hp) at D8. Overall concentration of cortisol in FSBM group was considerably lower than that of SBM group. Results from current study indicate that FSBM may provide beneficial effects in alleviating weaning stress and enhance immune status of weaned calves.

Effects of Chromium Propionate Supplementation on Growth Performance, Serum Traits and Immune Response in Weaned Pigs

  • This study investigated the effects of chromium propionate on growth performance, serum traits and immune response in weaned pigs. Twenty-four 4 wk-old crossbred weanling pigs (initial body weight about 9.52${\pm}$0.48 kg) were randomly allotted into one of two groups, a control group (basal diet), chromium propionate group (diet supplemented with 200 ${\mu}g$ $kg^{-1}$ (ppb) of chromium propionate). This experiment was conducted over nine weeks. Escherichia coli lipopolysaccharide (LPS) 100 ${\mu}g$ $kg^{-1}$ BW was used as the stress-inducing agent in the middle (4 wks) and final (8 wks) periods. The experimental results indicated that chromium propionate had no effect on growth performance (p>0.05). Chromium propionate supplementation reduced the percentage of LDL+VLDL (low and very low-density lipoprotein) and increased HDL (high-density lipoprotein), but did not affect other serum traits. Pigs supplemented with chromium propionate had higher antibody titers specific for sheep red blood cells (SRBC) and serum total globulin relative to the control during the final period (p<0.05). A challenge with LPS increased white blood cells in the chromium propionate group in both experimental periods (p<0.05). The chromium propionate group exhibited higher IgG and $\gamma$-globulin than the control during the middle experimental period (p<0.05). Moreover, the PHA (phytohemagglutinin) challenge result in the chromium propionate group was better than the control group (p=0.056). Greater neutrophil activity was displayed than in the control (p<0.05). This suggests that chromium propionate supplementation benefited the weaned pigs in lipoprotein and immune response.

  • This study purposes on continuing experimental research of furniture form creation focused on the importance and necessity for a-formal concept for designers to drift away from general acknowledgement and practice routine to design more radical forms for meeting various needs of late industrialized society, which have not been satisfied with formal and universal designs. This study covers presenting new points of view on research for designers to look for different solutions to widen their consideration in the process of furniture form development and to set up academic data base with the case studies of designers in anti-construction concept to define their experimental challenges for new design form creation, on the basis of overall research on the effects and roles of a-formal approach in the furniture design form creation.

Immune response and efficacy of pigeon pox virus vaccine and fowl pox virus vaccine in chickens

  • The humoral immune response of chicken vaccinated with fowl and pigeon pox virus vaccines was determined with the protective potentiality of the two vaccines in field condition of Bangladesh. Different aged Fayoumi chicks were subjected for the study. To assess the relationship with better immune response among experimental groups, the average percentage of 'take reaction' was examined and recorded to 97.77% in group A, 93.33% in group B and 100.0% in group C. The level of immune status induced by different vaccinated group was measured by passive hemagglutination (PHA) microplate test method. The mean PHA titer levels after primary vaccination were $33.06{\pm}14.13$ in group A, $32.0{\pm}14.81$ in group B, and $33.0{\pm}13.66$ in group C. Following booster vaccination, the mean PHA titer levels in prior of challenge were increased to $55.46{\pm}14.64$ in groups A and C, and $46.93{\pm}16.52$ in group B. The recorded PHA titer levels of each group at two weeks after challenge were significantly increased to $106.66{\pm}31.22$, $93.86{\pm}33.04$ and $110.93{\pm}29.29$, respectively. The PHA titer levels after vaccination and challenge were significantly increased compared to pre-vaccination titer levels (P<0.01). Although the PHA titer levels among three groups administrated different vaccine combinations in prior of challenge were significantly varied (P<0.01), it was observed that all of the vaccinated chicks were highly protected against challenge infection.

Differential Excretion of Urinary Eosinophil Protein X after Methacholine Challenge Test in Children with Asthma (천식 환아에서 요중 Eosinophil Protein X의 메타콜린 기관지 유발 시험 전·후의 변화)

  • Purpose : Eosinophil is one of the important inflammatory cell involved in the airway inflammation in childhood asthma. It has been demonstrated that markers of eosinophil activation, including eosinophil cationic protein or eosinophil protein X(EPX), are increased in childhood asthma. Furthermore, they are related to disease activity and are assumed to be helpful in monitoring the treatment effect as urinary EPX(U-EPX) can be obtained easily and in a noninvasive way in children of all ages. Methods : Twenty-five children(22 male and three female) aged $11.87{\pm}3.82$ years with stable asthma were challenged with methacholine and urine was collected from each child during the following periods; before methacholine challenge test(MCT); 0-3 hr after the end of MCT; 4-7 hr after the end of MCT; and 8-24 hr after the end of MCT. Bronchial reactivity was determined by using Dosimeter( Jeager, Germany) with serially diluted methacholine from 0.05 to 25.0 mg. The $FEV_1$ less than 80% of baseline value were classified into positive MCT. U-EPX was measured with a sensitive and specific radioimmunoassay(Pharmacia & Upjohn AB, Uppsala, Sweden). Results were expressed as ${\mu}gEPX/mmol$ creatinine. Results : An early airway response after MCT was associated with an increase of U-EPX excretion for 0-3 hr after methacholine inhalation in comparison with beseline values. Most subjects showed a small increase in U-EPX excretion during late asthmatic response for 4-7 hr, which then decreased to normal level in 8-24 hr. Also, a tendency for a higher increase of U-EPX was associated with a lower threshold of methacholine challenge and a longer duration of asthma. Conclusion : Measurement of EPX in urine is a noninvasive and easy method to assess the severity of airway inflammation in asthmatic children. It may be a helpful index of the events underlying the airway inflammatory responses during nonspecific bronchial challenge, and in monitoring asthma management.

  • The current understanding of the neurobiology of anxiety is generally based on experimental animal model, empirical effective psychopharmacological agents, chemical and naturalistic challenge paradigms, and psychoendocinological assessment. This article focuses on reviewing neuroanantomical, neuroendocinological and neurofunctional research of anxiety disorder. In the decade ahead, we anticipate that extension of current research and the new integrated approach promise novel insight into mechanism of anxiety.

  • An expeliment was carried out to measure the cellular immune response in guinea pigs by sensitizing the animals with 2, 4-dinitrochlorobenzene(DNCB). The guinea pigs could be sensitized with one application of DNCB. The sensitizing and challenge dose was standardized. The histological response was characteristic of a delayed hypersensitivity reaction.

  • We present a study of transition metal compounds using density functional theory (DFT), and density-corrected density functional theory(DC-DFT). By replacing the self-consistent density with that obtained from Hartree-Fock calculation, i.e., HF-DFT, the abnormality driven by self-interaction error is removed in several important cases. We discuss when and how HF-DFT works by examining 3d orbital dimers using approximate functionals and by comparing the results from self-consistent-DFT and HF-DFT with experimental values.
