Effects of Endurance Exercise and Ginsenoside Rb1 on AMP-Activated Protein Kinase, Phosphatidylinositol 3-Kinase Expression and Glucose Uptake in the Skeletal Muscle of Rats (지구성 운동과 Ginsenoside Rb1가 쥐 골격근의 AMP-Activated Protein Kinase(APMK), Phosphatidylinositol 3-Kinase(PI3K) 발현 및 Glucose Uptake에 미치는 영향)

  • This study investigated the effects of endurance exercise and ginsenoside $Rb_1$ on AMP-activated protein kinase (AMPK), phosphatidylinositol 3-kinase (PI3K) protein expression and glucose uptake in the skeletal muscle of rats. A total of 32 rats were randomly divided into four groups: CON (Control group, n=8), Ex (Exercise group; 25 m/min for 1 h, 6 days/week, 2 weeks, n=8), $Rb_1$ (Ginsenoside $Rb_1$ group; n=8), and $Rb_1/Ex$ ($Rb_1$+Exercise group, n=8). The $Rb_1$ and $Rb_1/Ex$ groups were incubated in ginsenoside $Rb_1$ (KRBP buffer, $100{\mu}g/mL$) for 60 min after a 2-week experimental treatment. After 2 weeks, the expression of phosphorylated $AMPK{\alpha}$ $Thr^{172}$, total $AMPK{\alpha}$, the p85 subunit of PI3K, pIRS-1 $Tyr^{612}$, and pAkt $Ser^{473}$ were determined in the soleus muscle. Muscle glucose uptake was measured using 2-deoxy-D-[$^3H$] glucose in epitroclearis muscle. Muscle glucose uptake was significantly higher in the three experimental groups (Ex, $Rb_1$, $Rb_1/Ex$) compared to the CON group (P<0.05). The expression of $tAMPK{\alpha}$ and $pAMPK{\alpha}$ $Thr^{172}$ was significantly higher in the Ex, $Rb_1$, and $Rb_1/Ex$ groups compared to the CON group (P<0.05). The expression of pAkt $Ser^{473}$ was significantly higher in the $Rb_1$ group compared to the CON and EX groups. However, the expression of pIRS-1 $Tyr^{612}$ and the p85 subunit of PI3K were not significantly different between the four groups. Overall, these results suggest that ginsenoside $Rb_1$ significantly stimulates glucose uptake in the skeletal muscle of rats through increasing phosphorylation in the AMPK pathway, similar to the effects of exercise.

The Effect of Aerobic Dancing and Ca Supplementation on Ca Metabolism in Postmenopausal Women (에어로빅 운동과 칼슘보충이 폐경이후 여성의 칼슘대사에 미치는 영향)

  • This study was designed to investigate the effects of aerobic dancing and/or Ca supplemen-tation for six months on Ca metabolism in postmenopausal women. The subjects were healthy 29 women aged from 60 to 70 years old. They were divided into four groups ; placebo and Ca supplementation group with and without exercise. The amount of Ca supplemented was 500mg a day. The frequency of doing exercise was three times a week and it took 40 minutes to complete once all the course of aerobic dancing programmed for old women. The results were summarized as follows : 1) Bone density of the second lumbar spine and ward's triangle measured after experiment decreased significantly in control group as compared with pre-experimental level(P<0.05). On the contrary it showed a tendency to increase or maintained the pre-experimental level after experiment in other groups. 2) Urinary Ca/creatinine and P/creatinine ratios did not show any significant differences among groups before and after experiment. But urinary Ca/creatinine ration decreased signifi-cantly in Ca group after experiment(P<0.05) 3) Serum Ca and P levels did not show any significant differences among groups before and after experiment. But serum Ca level in creased significantly in Ca group after experiment(P<0.05) 3) Serum Ca and P levels did not show any significant differences among groups before and after experiment. But serum Ca level increased significantly in Ca group after experiment (P<0.05). 4) Serum PTH level also did not show any significant differences among groups before and after experiment. But serum calcitonin level decreased significantly in Ca group after experiment(P<0.05). The above results showed that it will be difficult to prevent degenerative bone loss without regular exercise and/or Ca supplementation in postmenopausal women having Korean usual diets.

Ginsenoside Rg1 supplementation clears senescence-associated β-galactosidase in exercising human skeletal muscle

  • Background: Ginsenoside Rg1 has been shown to clear senescence-associated beta-galactosidase (SA-${\beta}$-gal) in cultured cells. It remains unknown whether Rg1 can influence SA-${\beta}$-gal in exercising human skeletal muscle. Methods: To examine SA-${\beta}$-gal change, 12 young men (age $21{\pm}0.2years$) were enrolled in a randomized double-blind placebo controlled crossover study, under two occasions: placebo (PLA) and Rg1 (5 mg) supplementations 1 h prior to a high-intensity cycling (70% $VO_{2max}$). Muscle samples were collected by multiple biopsies before and after cycling exercise (0 h and 3 h). To avoid potential effect of muscle biopsy on performance assessment, cycling time to exhaustion test (80% $VO_{2max}$) was conducted on another 12 participants (age $23{\pm}0.5years$) with the same experimental design. Results: No changes of SA-${\beta}$-gal were observed after cycling in the PLA trial. On the contrary, nine of the 12 participants showed complete elimination of SA-${\beta}$-gal in exercised muscle after cycling in the Rg1 trial (p < 0.05). Increases in apoptotic DNA fragmentation (PLA: +87% vs. Rg1: +133%, p < 0.05) and $CD68^+$ (PLA:+78% vs. Rg1:+121%, p = 0.17) occurred immediately after cycling in both trials. During the 3-h recovery, reverses in apoptotic nuclei content (PLA:+5% vs. Rg1 -32%, p < 0.01) and increases in inducible nitrate oxide synthase and interleukin 6 mRNA levels of exercised muscle were observed only in the Rg1 trial (p < 0.01). Conclusion: Rg1 supplementation effectively eliminates senescent cells in exercising human skeletal muscle and improves high-intensity endurance performance.

  • Scientific training, diet, and ergogenic aids are widely used to overcome the limits of humans' physical abilities and to achieve excellent sports records. The adoption of nutritional strategies is important for athletes to perform at their highest level, and one of the main factors determining endurance ability is increased fat metabolism. A ketogenic diet (high fat, low carbohydrates) has thus been proposed as an alternative strategy to maximize fatty acid oxidation during prolonged periods of exercise. However, studies have shown mixed results regarding the ergogenic value of a ketogenic diet. For this reason, exogenous ketone supplements (EKS, ingestible forms of ketone bodies, ketone esters, and/or salts) have been suggested to obtain nutritional ketosis, an acute transient increase in circulating acetoacetate (AcAc) and b-hydroxybutyrate (bHB) concentrations, without limiting carbohydrate intake. Some studies have suggested the beneficial effects of EKS on the performance of endurance exercises by providing an additional fuel substrate for peripheral tissues, such as cardiac and skeletal muscles, sparing carbohydrates/increasing fat oxidation and post-exercise recovery by increasing glycogen resynthesis in the liver/muscle, attenuating protein degradation, stimulating protein synthesis in the skeletal muscle, etc. However, many studies have failed to observe the beneficial effects of EKS as an ergogenic aid. As such, this review summarizes the theoretical basis of, as well as the proposed and proven effects of EKS on exercise performance and recovery to date.

The effect of Galwhahaejungtang and exercise on variation of lipid, BUN and Creatinine in rat with alcohol administration (갈화해성탕(葛花解醒湯) 투여 및 운동 부하가 알코올 섭취 후 흰쥐의 혈청 지질, BUN, Creatinine 함량에 미치는 영향)

  • Galwhahaejungtang has been used to alcoholic disease. Object of This study is examining the effect of Galwhahaejungtang and exercise on variation of total cholesterol, HDL-cholesterol, triglyceride, free fatty acid, BUN, Creatinine in rat with alcohol administration. METHODS AND MATERIALS Fifty four rats were divided into four groups. 1. The group of rat with alcohol administration and no treatment 2. The group of rat with alcohol and Galwhahaejungtang administration 3. The group of rat with alcohol administation and exercise 4. The group of rat with alcohol, Galwhahaejungtang administration and exercise Serum of rat were obtained after 2 weeks and 4 weeks. RESULTS 1. The level of total cholesterol was decreased in the group of rat with Galwhahaejungtang administration and exercise after 2 weeks. After 4 weeks, the level of total cholesterol was significantly decreased in the group of rat with Galwhahaejungtang administration and exercise. 2. The level of HDL-cholesterol was significantly increased in the group of rat with exercise after 2 weeks. After 4 weeks, the level of HDL-cholesterol was significantly increased in the group of rat with no treatment and with Galwhahaejungtang administration. 3. The level of try glyceride was significantly decreased in the group of rat with exercise after 2 weeks. After 4 weeks, the level of tryglyceride was significantly decreased in the group of rat with Galwhahaejungtang administration. 4. The level of free fatty acid was significantly decreased in the group of rat with Galwhahaejungtang administration after 2 weeks. After 4 weeks, the level of free fatty acid was significantly decreased in the group of rat with Galwhahaejungtang administration and with Galwhahaejungtang administration and exercise. 5. The level of BUN was significantly decreased in the group of rat with Galwhahaejungtang administration and exercise after 2 weeks. After 4 weeks, the level of BUN was significantly decreased in the group of rat with exercise. 6. The level of creatinine was significantly decreased in the group of rat with Galwhahaejungtang administration and exercise. After 4 weeks, the level of creatinine was significantly decreased in the group of rat with exercise and with Galwhahaejungtang administration and exercise. CONCLUSION Based on these results, it is considered that Galwhahaejungtang administration and exercise has an accelerating effect on alcohol metabolism.

The Relationships Among Body Fat Distribution, Blood Pressure, Blood Lipids and Exercise in Healthy Men and Women

  • A variety of studies show that a centraized rather than a generalized pattern of subcutaneous fat distribution is more directly associated with disorers of carbohydrate and lipid metabolism, as possibly hypertension, This study was an attempt to observe the relationship of body fat distribution, blood lipids, blood pressure and exercise in 85 healthy men and women. Within this group there was a gradation of fat distribution progressing from LBSO and UBSO defined on the basis of WHR. This paper reports the relationship of body fat distribution defined by WHR to plasma glucose, lipids, blood pressure, and exercise in this population. Although the obesity indices(RBW and BMI) were slightly higher in the UBSO group, significant differences existed between UBSO and LBSO groups with respect to WHR blood pressure, and total cholesterol concentration in female. WHR values were substantially different and this was primarily due to greater degrees of differences in waist as opposed to hips circum ference. Although no significant differences existed between UBSO and LBSO groups with respect to age, body weight, and hips circumference, energy intake, total cholesterol, LDL-cholesterol and WHR values were substantially different in male. Positive, significant correlations were found between WHR and both systolic and diastolic blood pressure and between WHR and the total plasma cholesterol concentration and age. When 26 pairs of exercise and nonexercise groups were matched according to sex, age and body weight, blood pressure and blood lipids were significantly lower in the exercise groups than those in the nonexercise groups. In conclusion, these findings suggest that an altered blood lipid profiles will manifast in men and women with upper body obese. Furthermore these findings suggest that exercise and physical activity may be beneficial for controlling blood lipids and blood pressure in healthy adults.

This study known that existed hormone(epinephrine)-immune system with interaction. Exercise were altered insulin binding and MAP Kinase signaling increased into immune cells. This review suggested that the high rate of glutamine utilization by cells of the immune system serves to maintain a high intra cellular concentration of the intermediates of biosynthetic pathways such that optimal rates of DNA, RNA and protein synthesis can be maintained. In the absence of glutamine, lymphocytes do not proliferate in vitro: proliferation increase greatly as the glutamine concentration increase. Glutamine is synthesized in skeletal muscle. Skeletal muscle and plasma glutamine levels are lowered by sepsis, injury, bums, surgery and endurance exercise and in the overtrained athlete. The study of result show that production of ET-1 is markedly increased tissue specifically in the heart by exercise without appreciable changes in endothelin-converting enzyme and endothelial receptor expressions, suggest that myocardial ET-1 may participate in modulation of cardiac function during exercise. Conclusionally, this study indicate that improvement of 'Enhancer Action' in RNA genes changed by exercise or sports. Moreover exercise was effect on Central Dogma with DNA makes RNA makes Protein. This study is expected to contribute the area of sports science, medicine, hereafter more effort is required to establish the relation between gene alters and exercise amount.

  • Recently, the number of the elderly has increased according to the improvement of socioeconomic status and the efficient medical care system. In spite of the development of medicine, the elderly suffers from the various health problems caused by weakness of musculoskeletal system, cardiopulmonary function and immunologic dysfunction. Regular rhythmic exercise program is known to be one of the effective tools to enhance the health condition in the elderly. However, there has been few studies to evaluate the comprehensive effects of rhythmic movement program on the elderly. This study was focused to evaluate the indices of cardiopulmonary function, life satisfaction, calcium, phosphorous, osteocalcin and deoxypyridinoline which are the essential factors of health problems in the elderly women. Twenty six subjects, aged between 68 and 72, who can do the ordinary activities and do not have cardiovascular dysfunction and mental disorder, participated in this study. They were divided into two groups: 13 in the experimental group and 13 in the control group. The experimental group participated in the rhythmic movement program at the welfare center located in G-city. The program were consisted of three sessions a week during 10 weeks. Each session had three parts: warming up(10 minutes), main exercise(40 minutes), finishing(10 minutes). Heart rate, blood pressure and peripheral arterial oxygen saturation were measured for the evaluation of cardiopulmonary function. Serum calcium, phosphorous, osteocalcin and urine deoxypyridinoline were measured as the indices of bony metabolism. Data were analyzed with mean, standard deviation, $x^2$-test, t-test, paired t-test using SPSS PC+ program. The results of this study were as follows. 1) Heart rate of the experimental group showed significant decrease following the rhythmic movement program. Peripheral arterial oxygen saturation of the experimental group showed significant increase following the program. 2) The degree of life satisfaction of the experimental group showed significant increase following the program. 3) Calcium showed significant decrease following the program but remained within normal range. There was no significant difference of phosphorous between two groups. 4) Osteocalcin, the index of bone formation, showed no significant change following the program, but significant increase in the experimental group comparing with the control group. 5) Deoxypyridinoline, the index of bone resorption, in urine of the experimental group showed significant decrease following the program. In conclusion, the rhythmic exercise program in the elderly showed the improvement of physiologic function and favorable effects on life satisfaction and bony metabolism. According to the above results, the regular rhythmic movement program can be strongly recommended for the improvement of health in the elderly women.

  • The aim of this study was to investigate the effect of combine exercise on the fall injury related index and myokine in elderly women. Sarcopenia leads to a loss of strength, alter on to a decreased functional status, impaired mobility, a higher risk of falls, and eventually an increased risk of mortality. Aerobic exercise characterized by rhythmic and repetitive movements of large muscles, for sustained periods that depends primarily on the use of oxygen to meet energy demands through aerobic metabolism, and that is structured and intended to generate improvements in cardiopulmonary fitness, body composition, and cardiorespiratory health. Resistance training has performance in the elderly. As combined exercise therapy can be used to enhance muscle functions and cardiopulmonary functions, it is being highlighted as an effective health management methods for the aged. The myokine has been regarded an important factor of exercise how muscle communicate adipose tissue, bone and muscle to exert beneficial effects at the whole body level.

The Effect of an Obesity Control Program on Body Composition and Blood Lipid Level among Middle-aged Obese Women in a Health Center (중년비만여성을 위한 보건소 비만관리 프로그램 효과)

  • Purpose: The Purpose of this study was to examine an obesity control program using dietary consult and physical exercise among middle-aged obese women in a health center. Method: This study used a pretest-posttest design. The subjects were 63 middle-aged women with over BMI $25(kg/m^2)$ or %BF 28 who participated in the obesity control program for 12 weeks. Results: After the program for 12 weeks, BMI(p= .02) and TC(p= .00) decreased significantly compared with those before the program. Conclusion: Obesity control programs in health centers can decrease the degree of obesity in middle-aged obese women. Therefore health centers should develop strategies for their members' continuous participation in such an obesity control program.
