Rates of Sulfate Reduction and Iron Reduction in the Sediment Associated with Abalone Aquaculture in the Southern Coastal Waters of Korea (남해연안 전복양식장 퇴적물의 황산염 환원과 철 환원에 의한 유기물 분해 특성)

  • We investigated geochemical properties, and microbial sulfate- and iron(III) reduction in sediment influenced by the aquaculture of abalone in the Nohwa-do, southern coastal sea in Korea. Concentrations of ammonium, phosphate, and sulfide in the pore-water were higher at farm sites than at control sites. The differences between the 2 types of sites were most apparent in terms of the weights of abalone and the temperature increase during September. Accordingly, the rates of sulfate reduction at the farm sites during September (61 mmol S $m^{-2}d{-1}$) were 3-fold higher than the sulfate reduction during May (20 mmol S $m^{-2}d{-1}$). In contrast, Fe(III) reduction rates were highest at the control sites in May, but its significance was relatively decreased at farm sites during September when sulfate reduction rates were highest. During September, benthic ammonium flux was 3-fold greater at the farm sites (35 mmol N $m^{-2}d{-1}$) than at the control sites (12 mmol N $m^{-2}d{-1}$), and phosphate flux was 8-fold higher at the farm sites (0.018 mmol P $m^{-2}d{-1}$) than at control sites (0.003 mmol P $m^{-2}d{-1}$). Overall results indicated that the inappropriate operation of a large-scale aquaculture farm may result in excess input of biodeposits and high nutrient fluxes from the sediment, thereby decreasing diversity of the benthic ecosystem and deepening eutrophication in coastal waters.

  • Alzheimer's disease (AD) is the most common cause of dementia in the elderly, and its pathology is characterized by the presence of numerous numbers of senile plaques and neurofibrillary tangles. Several genetic and transgenic studies have indicated that excess amount of $\beta$-amyloid protein (A$\beta$) is produced by mutations of $\beta$TEX>$\beta$-amyloid precursor protein and causes learning impairment. Moreover, $A\beta$ has a toxic effect on cultured nerve cells. To prepare AD model animals, we have examined continuous (2 weeks) infusion of $A\beta$ into the cerebral ventricle of rats. Continuous infusion of $A\beta$ induces learning impairment in water maze and passive avoidance tasks, and decreases choline acetyltransferase activity in the frontal cortex and hippocampus. Immunohistochemical analysis revealed diffuse depositions of $A\beta$ in the cerebral cortex and hippocampus around the ventricle. Furthermore, the nicotine-evoked release of acetylcholine and dopamine in the frontal cortex/hippocampus and striatum, respectively, is decreased in the $A\beta$-infused group. Perfusion of nicotine (50 $\mu\textrm{M}$) reduced the amplitude of electrically evoked population spikes in the CA1 pyramidal cells of the control group, but not in those of the $A\beta$-infused group, suggesting the impairment of nicotinic signaling in the $A\beta$-infused group. In fact, Kd, but not Bmax, values for [$^3H$] cytisine binding in the hippocampus significantly increased in the $A\beta$-infused rats. suggesting the decrease in affinity of nicotinic acetylcholine receptors. Long-term potentiation (LTP) induced by tetanic stimulations in CA1 pyramidal cells, which is thought to be an essential mechanism underlying learning and memory, was readily observed in the control group, whereas it was impaired in the $A\beta$-infused group. Taken together, these results suggest that $A\beta$ infusion impairs the signal transduction mechanisms via nicotinic acetylcholine receptors. This dysfunction may be responsible, at least in part, for the impairment of LTP induction and may lead to learning and memory impairment. We also found the reduction of glutathione- and Mn-superoxide dismutase-like immunoreactivity in the brains of $A\beta$-infused rats. Administration of antioxidants or nootropics alleviated learning and memory impairment induced by $A\beta$ infusion. We believe that investigation of currently available transgenic and non-transgenic animal models for AD will help to clarify the pathogenic mechanisms and allow assessment of new therapeutic strategies.

Effect of Sodium Chloride Intake Related to the Composition of the Diet (식이조성(食餌組成)에 따른 식염섭취량(食鹽攝取量)에 관(關)한 연구(硏究))

  • Sodium chloride plays an important role as the main condiment at daily meal. It is well known that humans require sodium chloride as an essential nutrient to keep the homeostasis of electrolytes. The amounts of salt intake may be a reflection of geography, culture and food habit rather than necessity. Lee has reported (1962) that Koreans ingest high amounts of sodium chloride in their meals, with an intake of excess carbohydrate (80-90% of total Calories) and low protein in their diet. This includes large amounts of rice, Kimchi and other fermented soybean products common in the Korean diet. This investigation was designed to study the dietary relations of sodium chloride to other nutrients in the Korean diet. Twenty four albino male rats, weighing from 290-300g, were divided into four dietary groups according to the amounts of carbohydrate, protein and fat in the basal diet. Each diet contained a rice powder as a carbohydrate source. Diet I was a control diet, Diet II, low protein, Diet III, low protein and low fat diet and Diet IV, low fat diet. All rats were provided with 3% sodium chloride solution. Diet and salt solution were given ad libitum. The experiment was carried out for 9 weeks during which time the body weight, the food intake, and 3% sodium chloride solution consumption were determined. At the 9th week, the urine was collected the blood sample from the artery of each rat for the analysis of sodium and potassium and other chemical studies. The rats were sacrificed and the kidney, adrenal, liver and spleen were measured, and observed changes of the pathological tissue in the kidney and adrenal. The results were summarized as follows: 1) The growth rate was higher in Diet I than in the other experimental diets (II, III and IV) after 4 weeks. There was no significant difference found between the experimental Diets II, III and IV. 2) The daily food intake was greater in the experimental diets II, III and IV than in the control diet. However, there was no difference among the high carbohydrate diets Diet II, III and IV. 3) The daily water (3% sodium chloride solution) intake was also greater in the Diets II, III and IV, than in the control diet. However, there was no difference between Diets II, III and IV. 4) The concentration of sodium and potassium in the blood were within the normal range in all diets. 5) The amount of sodium chloride in the urine was significantly greater in Diets II, III and IV than in the control diet. Diets II, III, IV had a larger amount of sodium solution consumption. 6) Observation of pathological tissue in the experimental diets found a cell proliferation in the glomerlulus of the kidney, while such change was not found in the control diet.

  • Lactic acid is widely used in the food, chemical and pharmaceutical industries, and there is an increasing demand for lactic acid as the raw material of poly lactic acid(PLA), which is a biodegradable polymer. Lactic acid production has been changing over from production by synthesis to production by fermentation, since the fermentation process is more nature friendly and economic. However, the fermentation method generates excess water and impurities with high boilers. The presence of high boilers and non volatility of lactic acid makes the separation of lactic acid very difficult job. Also, the purification-separation process requires the many investment costs and energy costs. Reactive distillation concept was also introduced for the process, giving higher selectivity and yield compared to the convention method. We introduce a new highly integrated process, reactive diving wall column, to reduce the capital and energy cost for producing a pure lactic acid. The reactive dividing wall column combines reactive distillation and dividing wall column. We compared capital and energy consumption required for the purification of lactic acid the between the proposed design structures. And we examined the effect of major process variables on the process performance and determined optimal process.

Evaluation of Phytotoxicity of Food Waste During Composting (부숙과정중 음식물찌꺼기의 식물독성 평가)

  • This study was to investigate the changes of phytotoxicity of food waste during composting at an aerated static pile for 80 days. The concentration of sodium in the final compost increased by more than 30% and almost of the sodium was a water soluble type which corresponds to about 94% of the total concentration. Major volatile fatty acid(VFA) produced during the composting was acetic acid and also somewhat of butyric acid. High concentration of acetic acid during the composting was kept up for a month and then decreased progressively. It suggests that the phytotoxicity by the VFAs produced during composting of food waste could be removed through a stabilization process in a period of about 40 days. From the statistical analysis between germination index(G.I.) and VFAs. or G.I. and sodium, the phytotoxicity of food waste samples was more dependent in the changes of acetic acid concentration than that of sodium concentration. Considering low G.I. value in the final food waste compost which is matured completely, however. the agricultural utilization of the food waste compost may be to have a big problem due to excess sodium of the compost which is impossible to remove by composting and seriously inhibits growth of plants.

WETTABILITY AND DRUG DELIVERY OF FUNCTIONALLY GRADED NANO-MICRO POROUS TITANIUM SURFACE

  • STATEMENT OF PROBLEM: It is known that an anodic oxidation technique, one of the methods for the implant surface treatment, remarkably increased surface area, enhanced wettability and accelerated the initial bone healing. Purpose: This study was performed to evaluate the wettability of anodized titanium surface which has a nanotubular structure, to assess osseointegration after the placement of implant with nano-size tubes on tibia of rats and to analyze quantitatively transferable rhBMP-2 on each surface. MATERIAL AND METHOD: Four different kinds of surface-treated titanium discs (polished (machined surface) group, micro (blasting surface) group, nano (anodizedmachined surface) group, and nano-micro (anodized-blasting surface) group) were fabricated (n=10). Three different media were chosen to measure the surface contact angles; distilled water, plasma and rhBMP-2 solution. After a single drop (0.025 $m{\ell}$) of solution, the picture was taken with the image camera, and contact angle was measured by using image analysis system. For the test of osseointegration, 2 kinds of anodized surface (anodized-machined surface, anodized-blasting surface) implants having 2.0 mm in diameter and 5.0 mm in length inserted into the tibia of Wistar rats. After 3 weeks, tibia were harvested and the specimens were stained with hematoxylin and eosin for histological analysis. To test the possibility of drug delivery, after soaking sample groups in the concentration of 250 ng/$m{\ell}$l of rhBMP-2 for 48 hours, the excess solution of rhBMP-2 were removed. After that, they were lyophilized for 24 hours, and then the rhBMP-2 on the surface of titanium was resolved for 72 hours in PBS. All the extracted solution was analyzed by ELISA. One-way analysis of variance (ANOVA) was performed on the data. RESULTS: The wettability is improved by anodic oxidation. The best wettability was shown on the nano-micro group, and it was followed by nano group, micro group, and polished group. In the histological findings, all implants showed good healing and the new bone formation were observed along the implant surface. After 3 days, nano-micro group delivered the most amount of rhBMP-2, followed by nano group, micro group, and polished group. CONCLUSION: It indicated that anodic oxidation on blasting surface produce functionally graded nano-micro porous structure and enhance hydrophilicity of the surface and osseointegration. The findings suggest that the nano-micro porous structure could be a useful carrier of osteogenic molecules like rhBMP-2.

The Optimum Methionine to Methionine Plus Cystine Ratio for Growing Pigs Determined Using Plasma Urea Nitrogen and Nitrogen Balance

  • The objective of this study was to determine the optimum ratio of methionine to methionine plus cystine for growing pigs. A nitrogen balance trial was conducted using a total of 21 barrows (Large WhiteLandrace) over two replicates. The initial body weight was $20.36{\pm}1.22kg$ (mean${\pm}$SD) in the first replicate and $23.54{\pm}1.02kg$ (mean${\pm}$SD) in the second. For each replicate, the 21 pigs were randomly assigned to one of seven dietary treatments with three observations per treatment. The diets included a methionine and cystine-deficient basal diet with all other essential nutrients meeting nutrient requirements and six diets formulated with graded levels of DL-methionine (0.00, 0.03, 0.06, 0.10, 0.13, 0.16%) and $L-Cystine{\cdot}HCl{\cdot}H_2O$ (0.19, 0.15, 0.11, 0.07, 0.04, 0.00%). This resulted in ratios of methionine to methionine plus cystine of 41.3, 29.6, 35.3, 41.2, 46.0, 51.6 and 57.5%. Each experimental period lasted 12 days consisting of a seven-day adaptation period followed by a five-day total collection of urine and feces. During the collection period, pigs were fed 900 g/day for the first replicate and 1,200 g/day for the second replicate. The feed was provided in three equal portions at 0800, 1500, and 2200 h daily. Pigs had ad libitum access to water after feeding. There was a linear (p<0.01) and quadratic (p<0.01) effect on daily gain and feed conversion as the ratio of methionine to methionine plus cystine increased. Pigs receiving the diets providing a methionine to methionine plus cystine ratio of 51.6% had the best daily gain and feed conversion. Plasma urea nitrogen was also lowest for this treatment. Nitrogen retention increased (p<0.01) as the relative proportion of methionine increased up to 51.6% and then a downward trend occurred at 57.5%. The quadratic regression model, as well as one- and two- slope regression line models, were used to determine the optimum ratio of methionine to methionine plus cystine. Eliminating the 35.3% methionine to methionine plus cystine treatment resulted in $R^2$ values in excess of 0.92. The optimal ratio of methionine to methionine plus cystine was estimated to be 54.15% for nitrogen retention and 56.72% for plasma urea nitrogen.

Distribution Characteristics of Alien Plants by Wetland Types in the Ecologically Outstanding Wetlands of South Korea (국내 생태우수습지의 유형별 외래식물상 현황 및 특성)

  • Wetlands are vulnerable to biological invasion by alien species, because they function as sinks that accumulate excess water, sediments, nutrients, and other contaminants from the surrounding watersheds by disturbance. In this study, to understand the status and characteristics of the alien plants based on the type of wetlands, we classified 24 ecologically outstanding wetlands and analyzed the status of alien flora. A total of 130 alien plants were found in the wetlands, accounting for 11% of the total plant species. Among them, the Asteraceae species was the most diverse, with 40 species. Erigeron annuus and Oenothera ordorata had the highest frequency of occurrence. The species richness of alien plants in the riverine and lacustrine wetlands (average: 30 species) was higher than that in the mountainous palustrine wetlands (average: 10 species). The same results were found in the naturalization index, urbanization index, and ratio of annuals and biennials, which indicate the degree of artificial interference. In the cluster analysis, the riverine and lacustrine wetlands were combined, and only the mountainous palustrine wetlands were separated. The number of alien plants is remarkably low in the mountainous palustrine wetlands, and it is considered to be the influence of Erigeron strigosus, Symphytum officinale, and Bilderdykia convolvulus, not found in the other types of wetlands. In particular, invasive alien plants such as Aster pilosus, Ambrosia trifida, Sicyos angulatus, Ambrosia artemisiifolia var. elatior were found intensively in the riverine wetlands. Therefore, it is considered that a methodical management is urgently required considering the dispersal of alien plants in the riverine and lacustrine wetlands with high artificial interference.

Study of Blast Ground Vibration & Noise Measurements In-situ and Effect Analysis for Numerical Analysis, Rational Blasting Design at an Eel Farm (양만장의 발파 진동소음 현장측정과 수치해석을 통한 영향검토 및 합리적인 발파설계 연구)

  • The vibration or/and noise generated by blast operations might cause not only structural damage to properties but mentally also to humans and animals. For that reason, maximum permitted vibration and noise levels are set by sensitivities of structures and they are used for the management of blast vibration. It is known that the fish lived in water are more sensitive to vibration than land animals, and thus the adverse impact of the blasting on fish farms should be very concerned. This study investigated the vibration and noise levels at a large eel farm located some 840 meters of the blasting site through the large real-scale experiments of blastings, prior to conducting the actual blasting. As a result, it was found that the noise met the requirement to be within maximum permitted level, while the ground vibration exceeded the permitted vibration. Accordingly, the impact of the excess vibration was investigated by an existing empirical method and verified by a new three dimensional numerical analysis. In this study, such an inspection process was briefly described, and a method was suggested for the examination of possible adverse effects from blasting on vibration-sensitive structures like the eel farm. The study also introduced a design method that controls the blast effects - ground vibration and noise.

New Methods for Assessing Liquefaction Potential Based on the Characteristics of Material (재료의 역학적 거동특성에 기초한 액상화 평가방법)

  • The purpose of this study is to develop and utilize new assessment of liquefaction potential based on DSC(disturbed state concept) and dissipated energy concept. The term liquefaction has suddenly loses its shear strength and behaves like a fluid. Liquefaction has been a source of a major damage during severe earthquake. In this study, the cyclic undrained behavior of Joomoonjin strand is investigated by using an automates triaxial testing device(C. K. Chan type). In order to assess liquefaction potential of saturated strand, DSC method and energy method are applied for the experimental data. The use of DSC method and energy method to define the liquefaction potential is verified through laboratory testis of cyclic triaxial test on saturated sand specimens. Based on the analytical results of DSC method, the relationship between the factor affecting liquefaction characteristics(Dr) and physical properties of the saturated santa(fs and D.) is found. Based on the analytical results of energy method, it is found that the initial liquefaction of rand is related to the significant change in the dissipated energy. Finally, it is shown that the DSC method and energy method can capture the liquefaction mechanism.
