Prevalence of feline calicivirus in Korean cats determined by an improved real-time RT-PCR assay

  • Feline calicivirus (FCV) is considered the main viral pathogen of feline upper respiratory tract disease (URTD). The frequent mutations of field FCV strains result in the poor diagnostic sensitivity of previously developed molecular diagnostic assays. In this study, a more sensitive real-time reverse transcription-polymerase chain reaction (qRT-PCR) assay was developed for broad detection of currently circulating FCVs and comparatively evaluated the diagnostic performance with previously developed qRT-PCR assay using clinical samples collected from Korean cat populations. The developed qRT-PCR assay specifically amplified the FCV p30 gene with a detection limit of below 10 copies/reaction. The assay showed high repeatability and reproducibility, with coefficients of intra-assay and inter-assay variation of less than 2%. Based on the clinical evaluation using 94 clinical samples obtained from URTD-suspected cats, the detection rate of FCV by the developed qRT-PCR assay was 47.9%, which was higher than that of the previous qRT-PCR assay (43.6%). The prevalence of FCV determined by the new qRT-PCR assay in this study was much higher than those of previous Korean studies determined by conventional RT-PCR assays. Due to the high sensitivity, specificity, and accuracy, the new qRT-PCR assay developed in this study will serve as a promising tool for etiological and epidemiological studies of FCV circulating in Korea. Furthermore, the prevalence data obtained in this study will contribute to expanding knowledge about the epidemiology of FCV in Korea.

대형 상용차량 하이브리드 전동식 조향 시스템 주행 성능평가를 위한 HILS 시스템 개발 (Development of HILS System for Performance Evaluation of a Heavy Commercial Vehicle Hybrid Electric Power Steering System)

  • Most commercial vehicles have adopted the hydraulic power steering system. To reduce fuel consumption and to improve steering controllability, a hybrid electric power steering system is being developed for commercial vehicles. In this study, the HILS (Hardware In the Loop Simulation) system equipped with a commercial vehicle hybrid electric power steering system was developed and the vehicle dynamic performance of a truck with the steering system was evaluated. The hybrid electric power steering system is composed of the EHPS motor pump, column mounted EPS system, and ball nut steering gear box for heavy commercial vehicles. The accuracy of vehicle models equipped with the HILS system was verified with comparisons between the simulation results and field test results. The road reaction forces of the steering system were generated from the vehicle model and verified using field test results. Step steering tests using the verified HILS system were carried out and the performance of a newly developed commercial vehicle hybrid electric power steering system was evaluated.

Analysis of signal cable noise currents in nuclear reactors under high neutron flux irradiation

  • Cables are indispensable in nuclear power plants for transmitting data measured by various types of detectors, such as self-powered neutron detectors (SPNDs). These cables will generate disturbing signals that must be accurately distinguished and eliminated. Given that the cable current is not very significant, previous research has focused on SPND, with little attention paid to cable evaluation and validation. This paper specifically focuses on the quantitative analysis of cables and proposes a theoretical model to predict cable noise. In this model, the reaction characteristics between irradiated neutrons and cables were discussed thoroughly. Based on the Monte Carlo method, a comprehensive simulation approach of neutron sensitivity was introduced and long-term irradiation experiments in a heavy water reactor (HWR) were designed to verify this model. The theoretical results of this method agree quite well with the experimental measurements, proving that the model is reliable and exhibits excellent accuracy. The experimental data also show that the cable current accounts for approximately 0.2% of the total current at the initial moment, but as the detector gradually depletes, it will contribute more than 2%, making it a non-negligible proportion of the total signal current.

  • Today, the research related to the robot is achieved in various part. With the high interest in means of transport, various researches about autonomous mobile robot and next generation transport is continuing. The unicycle robot among these needs much control technique like balance control model and driving model. For autonomous driving of this unicycle robot, from the basic balance control to direction switching control and velocity control are needed. But the environment elements like a gradient and frictional force or unbalanced elements from the structural feature. The unicycle needs the real time balance control so more complex, harder to control. And when functional addition is made, the problem that fall entire reaction velocity or accuracy would be happen. This paper introduces entire dynamics modeling of the unicycle robot and reduced model. And propose the new balance control algorithm using fuzzy controller. Also the evaluation about performance would be made through the test.

LC-MS/MS를 이용한 수산물 중 콜리스틴 분석법 개발 (Development of Analytical Method for Colistin in Fish and Shrimp using Liquid Chromatography Mass Spectrometry)

  • LC-MS/MS를 이용하여 폴리펩타이드계 동물용의약품인 콜리스틴에 대한 시험법을 확립하여 정량성 및 정밀성을 확보하였으며, 확립된 시험법의 적용성 검증을 위해 국제식품규격위원회 기준에 따라 특이성, 정확성, 직선성, 정밀성, 검출한계, 정량한계 등을 검증하였다. 콜리스틴 표준용액을 잔류허용기준의 농도에 따라 검량선을 작성한 결과 0.99 이상의 직선성을 확인 할 수 있었으며, 본 실험에서의 평균 회수율은 85.9~107%이었다. 또한, 분석오차는 11.8% 이하로 정확성 및 재현성이 우수하였으며, 검출한계는 0.02 mg/kg, 정량한계는 0.05 mg/kg이었다. 또한, 실험실간 교차검증을 통하여 신뢰성을 확보하였다. 확립된 분석법은 양식 수산물 중 잔류할 수 있는 동물용의약품인 콜리스틴에 대한 안전관리에 활용 할 수 있을 것으로 판단된다.

Comparative Assessment of Diagnostic Performances of Two Commercial Rapid Diagnostic Test Kits for Detection of Plasmodium spp. in Ugandan Patients with Malaria

  • Prompt diagnosis of malaria cases with rapid diagnostic tests (RDTs) has been widely adopted as an effective malaria diagnostic tool in many malaria endemic countries, primarily due to their easy operation, fast result output, and straightforward interpretation. However, there has been controversy about the diagnostic accuracy of RDTs. This study was conducted to evaluate the diagnostic performances of the 2 commercially available malaria RDT kits, RapiGEN Malaria Ag Pf/Pv (pLDH/pLDH) and Asan $EasyTest^{TM}$ Malaria Ag Pf/Pv (HRP-2/pLDH) for their abilities to detect Plasmodium species in blood samples collected from Ugandan patients with malaria. To evaluate the diagnostic performances of these 2 RDT kits, 229 blood samples were tested for malaria infection by microscopic examination and a species-specific nested polymerase chain reaction. The detection sensitivities for P. falciparum of Malaria Ag Pf/Pv (pLDH/pLDH) and Asan $EasyTest^{TM}$ Malaria Ag Pf/Pv (HRP-2/pLDH) were 87.83% and 89.57%, respectively. The specificities of the 2 RDTs were 100% for P. falciparum and mixed P. falciparum/P. vivax infections. These results suggest that the 2 RDT kits showed reasonable levels of diagnostic performances for detection of the malaria parasites from Ugandan patients. However, neither kit could effectively detect P. falciparum infections with low parasitaemia (<$500parasites/{\mu}l$).

  • 새로 개발되거나 혹은 기존의 어떤 진단검사를 다양한 임상상황에 적용하기 위해서는 먼저 이들 검사법의 진단적 정확도를 추정하는 연구가 반드시 선행되어야 한다. 진단의 정확도에 대한 추정치를 모른다면 검사결과를 해석하는 것이 불가능하기 때문이다. 특히 동일한 개체에서 감염부위별로 3개 이상의 시료를 얻어 진단검사를 적용하는 경우 각 시료의 검사결과는 독립적인 측정시료가 아니라 개체내에서 연관성이 매우 높은 종속적인 시료에 해당 한다. 즉 동일한 개체에서 얻은 시료일수록 검사결과에서 유사한 반응을 보이며 이 경우 분석의 단위는 각각의 개체가 아니라 검사부위가 되는데 이는 의학연구에서 매우 흔하다. 본 연구에서는 Helicobacter 균에 의한 감염을 검출하기 위하여 동일한 개로부터 위의 해부학적 구조상 pyloric antrum, body 및 cardia의 생검시료에 대하여 urease 검사와 PCR 검사를 적용하여 각 검사의 진단적 정확도를 추정하였다. urease 검사의 민감도와 특이도는 0.74 (95% 신뢰구간: 0.64-0.84)와 0.87 (95% 신뢰구간: 0.67-1.00)이었으며 PCR 검사의 민감도와 특이도는 0.95 (95% 신뢰구간: 0.89-1.00)와 0.90 (95% 신뢰구간: 0.70-1.00)로 두 검사의 특이도는 높은 것으로 나타났다. 그러나 PCR 검사에 비하여 urease 검사의 경우 가음성 (false negative)의 가능성이 높기 때문에 진단결과에 대한 신중한 해석이 요구된다.

Ultrasound Breast Elastographic Evaluation of Mass-Forming Ductal Carcinoma-in-situ with Histological Correlation - New Findings for a Toothpaste Sign

  • Background: It is unclear as to whether the size ratio elastographic technique is useful for assessing ultrasound-detected ductal carcinoma-in-situ (DCIS) masses since they commonly lack a significant desmoplastic reaction. The objectives of this study were to determine the accuracy of this elastographic technique in DCIS and examine if there was any histopathological correlation with the grey-scale strain patterns. Materials and Methods: Female patients referred to the radiology department for image-guided breast biopsy were prospectively evaluated by ultrasound elastography prior to biopsy. Histological diagnosis was the gold standard. An elastographic size ratio of more than 1.1 was considered malignant. Elastographic strain patterns were assessed for correlation with the DCIS histological architectural patterns and nuclear grade. Results: There were 30 DCIS cases. Elastographic sensitivity for detection of malignancy was 86.7% (26/30). 10/30 (33.3%) DCIS masses demonstrated predominantly white elastographic strain patterns while 20/30 (66.7%) were predominantly black. There were 3 (10.0%) DCIS masses that showed had a co-existent bull's-eye sign and 7 (23.3%) other masses had a co-existent toothpaste sign, a strain pattern that has never been reported in the literature. Four out of 4/5 comedo DCIS showed a predominantly white strain pattern (p=0.031) while 6/7 cases with the toothpaste sign were papillary DCIS (p=0.031). There was no relationship between the strain pattern and the DCIS nuclear grade. Conclusions: The size ratio elastographic technique was found to be very sensitive for ultrasound-detected DCIS masses. While the elastographic grey-scale strain pattern should not be used for diagnostic purposes, it correlated well with the DCIS architecture.

멀티모달 실감 경험 I/O 인터랙션 시스템 개발 (Development for Multi-modal Realistic Experience I/O Interaction System)

  • 본 연구는 단순 입력 기반 유니모달 인터랙션의 한계를 극복하고 단순 입력 방식이 아닌 멀티모달 기반 사용자의 행위, 의도, 및 집중도를 활용하여 실감적이고 몰입도를 향상시키는 인터랙션 시스템을 제안하는데 그 목적이 있다. 본 연구의 선행연구에서 기존 문헌연구를 토대로 메타분석방법을 활용하여 인터랙션을 위한 3차원 동작 인식 기술의 정확도를 분석하여 최종적인 센서 기반 인터랙션 방법이 선정되었고, 직관적 제스쳐 인터랙션 요소를 추출하여 본 시스템에 반영하였다. 또한 생리반응을 이용한 집중력 판단 기술을 개발하여 사용자 의도를 판단하는 연구를 진행하였다. 본 연구에서 제안하는 시스템은 3부분으로 나눌 수 있다. 선행연구에서 선정된 인터랙션 요소들을 적용하여 가속도(Accelator) 센서와 연성(Flexible) 센서를 활용하여 손 동작을 인식하는 시스템을 구현하였고, 동공 인터랙션을 통한 안경형 시선 추적기를 구현하여 인터랙션이 가능하게 하였으며, 심혈관 반응과 피부 온열 반응을 측정하여 사용자의 의도를 반영한 시스템을 최종 구현하였다. 실감형 디지털 엔터테인먼트 플랫폼 기술 개발을 위한 기초 연구로서 활용이 가능할 것으로 판단된다.

128 채널 뇌파를 이용한 사이버멀미 평가법 개발 (Development of a Method of Cybersickness Evaluation with the Use of 128-Channel Electroencephalography)

  • 가상현실 기술이 발전하면서 다양한 영역에서 여러 목적으로 활용되고 있다. 하지만 사용자로부터 메스꺼움, 어지러움 등과 같은 멀미 증상에 대한 부작용도 함께 보고되고 있다. 이런 멀미 증상을 사이버멀미라고 말하며, 사용자에게 불편을 야기시켜 불쾌감과 스트레스와 같은 부정적 영향을 줄 수 있으며, 현재 사이버멀미의 발생 원인과 해결책에 관한 공식적인 표준이 없다. 본 연구에서는 이런 한계점을 극복하기 위해 정량적, 객관적 사이버멀미 평가법을 제안하였다. 10명의 20대 남성 대상으로 이동과 회전을 하는 가상현실을 경험하게 하면서 128채널의 뇌파를 측정하였다. 11개 영역에서 Delta와 Alpha 상대 파워를 계산하고, ROC curve를 통해 area under the ROC curve (AUC)가 가장 높은 값을 가지는 다중회귀모형을 도출하였다. 이렇게 도출한 다중회귀모형은 11개의 설명변수로 피험자가 응답한 사이버멀미의 정도와 비교하여 95.1 % (AUC = 0.951)의 정확성으로 사이버멀미를 구분하는 것이 가능함을 알 수 있었다. 이러한 결과를 정리하면 본 연구에서는 128 채널의 뇌파를 통해 멀미에 대한 객관적 반응을 확인하였으며, 뇌의 특정부위에서 반응이 있는 것으로 나타났다. 본 연구 결과와 분석법을 활용하면 추후 사이버멀미 관련 연구에 도움이 될 수 있을 것으로 기대된다.
