The Biological Activities of Extracts and Fractions of Herbal Plants (수종의 한약재 분획물의 생리활성 효과)

  • Objectives : This study was conducted to identify the antimicrobial anti-oxidative and tyrosinase inhibitory effect of three kinds of herbal plants, Polygonum cuspidatum, Chelidonii herba, Pharbitis nil L. naturally grown across the nation. Methods : To investigate in vitro anti-oxidative activity assay, antimicrobial effect and tyrosinase inhibitory activity, MeOH 80% extract, n-hexane, chloroform, ethyl acetate, n-butanol and water fractions of herbal plants were tested by diphenylpicrylhydrazyl(DPPH) method. Results : Bsutanol fractions of each plants showed about 90% on anti-oxidative effect. In case of tyrosinase activity, ethlyacetate fraction of Polygonum cuspidatum showed a potent inhibition effect. In the antimicrobial effect, ethylacetate fraction of Polygonum cuspidatu, hexane fractions of Chelidonii herba and the methanol 80% extracts of Pharbitis nil L. exhibited inhibition effect significantly. Conclusions : In MeOH 80% extracts, we couldn't found any extracts or fractions which have antimicrobial, anti-oxidative and tyrosinase inhibitory activity, however ethylacetate fractions of Polygonum cuspidatum showed effects commonly in these three assay system.

Production of Water-Solubled Pigment from Mycelial Culture of Cordyceps scarabaeicola KEFC-C252 and Its Antimutagenic Effect (Cordyceps scarabaeicola KEFC-C252의 균사체 배양에 의한 수용성 색소의 생산과 색소의 항돌연변이 효과)

  • Cultural conditions for the production of water-soluble pigment from mycelial culture of Cordyceps scarabaeicola KEFC-C252 and antimutagenic activity of the pigment were investigated. To obtain the maximum productivity of the pigment from mycelial culture of C. scarabaeicola KEFC-C252, the optimized medium was made with 1.5% sucrose, 2.5% yeast extract and initial pH 5.5. C. scarabaeicola KEFC-C252 was cultivated to reach the maximum concentration of the pigment at $26^{\circ}C$ for 108 hrs. C. scarabaeicola KEFC-C252 produced about 1.2 g/liter pigment under the optimized condition. The pigment was isolated from the culture filtrate by ethylacetate extraction, acidic precipitation and crystallization. The isolated pigment was scarlet hexagonal column crystal, and the color of the pigment was changed according to pH of the solution. The pigment showed violet in the alkaline water but showed red color in the acidic water. The pigment showed inhibitory activity against mutagenic activity induced by 4-nitroquinoline N-oxide. Furthermore, the pigment showed inhibitory activity against spontaneous mutation on Salmonella typhimurium TA98 and TAlOO.

Bioactive Phenylpropanoids from Asiasarum sieboldi Roots (세신(細辛)의 생리활성물질(生理活性物質) Phenylpropanoids의 분리(分離))

  • Treatment of ethylacetate extract of Asiasarum sieboldi inhibited the germination and the growth of radish seeds. Two phenylpropanoids were isolated from ethylacetate extract. Their structures were identified as safrole and o-methyleugenol by spectroscopic evidence. From the test to inhibitory effect, o-methyleugenol had inhibited the germination and the growth of radish seeds, while safrole did not. The germination rate and radicle length of radish seeds were decreased to 63.0%, 31.5 % of control at 5mg/ml of o-methyleugenol, respectively. At the same concentration, o-methyleugenol inhibited the hypocotyl growth up to 100%.

Anti-cancer and Anti-inflammatory Effects of Mung Bean and Soybean Extracts (녹두 및 대두추출물의 항암 및 항염증 활성)

  • The quinone reductase (QR) inducing activities of mung bean and soybean solvent extracts were compared using murine hepatoma cells (Hepa 1c1c7). The mung bean extracts (ethylacetate and ethanol) showed higher chemoprevention index values (7.88-8.22) than those of soybean extracts (2.9-5.2) from four different cultivars. The mung bean extracts also had significantly higher inhibitory effects (47-62% at 100 ${\mu}g$/mL) than the soybean extracts (15-42% at 100 ${\mu}g$/mL) against the production of nitric oxide and prostaglandin E2 in lipopolysccharide stimulated macrophage RAW264.7 cells without cytotoxicity. Among seven recovered fractions of mung bean ethanol extract obtained by C 18 silica flash column chromatography, the most non-polar fraction exhibited the highest chemoprevention index of 10.4.

Inhibitory Activity of the Fruit Extract of Gardenia jasminoides on Monoamine Oxidase (치자추출물의 Monoamine Oxidase 저해활성)

  • We examined the inhibitory activities against monoamine oxidase (MAO) of Gardenia jasminoides in vitro and in vivo methods. Methanolic extract and ethylacetate fraction of Gardenia jasminoides fruit showed a significant inhibitory activity on MAO-A and MAO-B in vitro. The IC$_{50}$ values of each fraction on MAO-A and MAO-B are as fo11owed; total methanol extracts 1.23 and 1.34 mg/ml, EtOAc fraction 0.72 and 0.77 mg/ml. Water-soluble fraction also showed IC$_{50}$ values of 0.81 mg/ml on MAO-B. MAO-A activity was increased by the oral administration of ethanolic extract of G. jasminoides, while MAO-B activity was decreased. The concentration of serotonin of brain tissue administrated of ethanolic extract of G. jasminoides is slightly increased in rat. This tendency is not different from the activity of deprenyl which is a well known MAO inhibitor was used as a positive control. Consequently, we suggest that G. jasminoides may have the effects on the inhibitory activity against MAO This activity of G. jasminoides is considerable for development of functional materials for treatment and control of depression, dementia, Parkinson' disease, stress and promoting exercise, etc.

The Electron Donating Ability, Nitrite Scavenging Ability and NDMA Formation Effect of Solvent Extracts from Yuza (Citrus junos SIEB ex TANAKA) (유자 용매추출물의 전자 공여능, 아질산염 소거능 및 NDMA 생성 억제능)

  • Methanol, ether, ethylacetate and water extracts from flesh and peel of yuna (Citrus junos) were investigated to analyze their ability of electron donating, nitrite scavenging and nitrosodimethylamine (NDMA) formation in model system. The electron donating ability of ynza flesh extract, when it added at 10 mg/ml or over, were more an $50\%$, except ether extract. The high electron donating ability ($99.2{\pm}0.37\%$) was observed in the methanol extract and its effect was similar to BHA and ascorbic acid, when 40 mg/ml of methanol extract was added in reaction solution. Nitrite scavenging ability of all extracts ken yuza was increased in proportion to sample concentration and more than $40\%$ when 1 mg/ml sample was added, under pH 1.2. Also nitrite scavenging ability was higher in peel extracts than flesh extracts of yuza. Inhibition ratio of NDMA formation from flesh methanol extract of yuza was $31.7{\pm}1.25\%$, when 40 mg/ml added under pH 1.2. In all samples, inhibition effects were lower than $20\%$, at pH 4.2 and 6.0.

Anti-thrombosis and anti-oxidative activity of the root of Arctium lappa L. (우엉 뿌리의 항혈전 및 항산화 활성)

  • To investigate anti-thrombosis and anti-oxidation activities of the root of Arctium lappa L (RALL), which has been used as foodstuff and oriental medicine in Korea, the ethanol extract and its subsequent organic solvent fractions of the RALL were prepared. The yield of ethanol extraction was 10.94%, and the content of total polyphenol and total sugar of ethanol extract were 5.01 and 694.53 mg/g, respectively. The fraction yields of n-hexane, ethylacetate (EA), butanol and water residue were 1.62, 0.42, 5.98 and 85.38%, respectively. In anticoagulation activity assay, the ethanol extract of RALL did not show significant changes in thrombin time (TT), prothrombin time (PT) and activated partial thromboplastin time (aPTT), whereas the EA fractions showed 13 folds extended TT, PT, and aPTT respectively. Interestingly, the water residue showed strong activation effect against blood clotting factors with shortened aPTT, which might provide the evidence of coagulation agent of RALL in folk remedy. In anti-platelet aggregation assay, the activity of the ethanol extract and its fractions were comparable to that of aspirin. Especially the EA fraction showed 2-folds higher inhibitory activity than aspirin. In anti-oxidation activity assay, the EA fraction also showed strong in DPPH, ABTS and nitrite scavenging activity, and reducing power activity. The extract and fractions of RALL have ignorable hemolytic activity against human RBC up to 0.5 mg/mL concentration. Our results suggest that the EA fraction of RALL have potentials as safe and novel anti-thrombosis agent.

Antimicrobial Activity of Coriandoer (Coriandrum sativum L.) Extract (고수(Coriandrum sativum L.) 추출물의 항균활성)

  • To develop natural food preservatives, ethanol and water extracts were prepared from the coriander (Coriandrum sativum L.) and antimicrobial activities were examined against 10 microorganisms which were food borne pathogens and/or food poisoning microorganisms and food-related bacteria and yeasts. Ethanol extract exhibited antimicrobial activities for the microorganisms tested, but not on lactic acid bacteria and yeast. Especially, minimum inhibitory concentrations (MIC) for Bacillus subtilis and Bacillus cereus were as low as 0.25mg/mL. antimicrobial activity of the ethanol extract was not destroyed by the heating at 121$^{\circ}C$ for 15min and not affected by pH. The ethanol extract of coriander exhibiting high antimicrobial activities was fractionated in the order of hexane, chloroform, ethylacetate and butanol fractions to test antimicrobial activity. The highest antimicrobial activity adjust bacteria tested was found in the ethylacetate fraction.

Antioxidative Substances in Mulberry Leaves (뽕잎(桑葉)에 함유된 항산화성 물질)

  • Antioxodative substances in Mulberry leaves were examined. Antioxidative substances in Mulberry leaves were extracted by 80% methanol agueous solution. Antioxidative activity of extract was determined by examining hydrogen donating ability on 1,1-diphenyl-2-picrylhydrazyl (DPPH) and the inhibitory effect on the formation of the peroxide from Linoleic acid in the test tube at $50^{\circ}C$. Antioxidative substance were, then, separated and indentified by thin layer chromatography(TLC), UV-Vis spectrum and High performance liquid chromatography(HPLC) methods. Hydrogen donating ability on DPPH and antioxidative ability on linoleic acid of the extracted antioxidative substance were higher than those of 100ppm butylated hydroxy toluene(BHT). The extracted antioxidative substances were separated by TLC using ethylacetate : chloroform : formic acid : water(8 : 1 : 1 : 1 v/v) as a solvent, and a spot at Rf=0.35 was detected. The spot was scraped from the plate, and extrated by methanol. The extract was analyzed by UV-Vis spetra and HPLC, and chlorogenic acid was identified as a antioxidative substance.

Isolation and Evaluation of an Antitumor Constituent from Pyrolae Herba (녹수초의 항암활성물질의 분리 및 항암력 평가)

  • The cytotoixic effect of Pylorae Herba (Pyrola japonica Klenze) against L1210 and K-562 cells was studied in vitro. The methanolic extract of Pylorae Radix was add ed to the culture of L1210 cells and K-562 cells for the cytotoxic activity and the ED50 values of hexane, ethylacetate, buthanol and water fractions from methanolic extract were determined using MTT (3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazoliumbromide) assay. The active constituent isolated by bioassay guided fractionation followed by purification gave rise to a yellow needle crystal and was clarified to be chimaphilline by the comparison with the published data. The average life spans with it were not prolonged significantly on tumor growth in hybrid female mouse (BDF1-KIST) inoculated subcutaneously with P388 cells.
