A Cannabinoid Receptor Agonist N-Arachidonoyl Dopamine Inhibits Adipocyte Differentiation in Human Mesenchymal Stem Cells

  • Endocannabinoids can affect multiple cellular targets, such as cannabinoid (CB) receptors, transient receptor potential cation channel, subfamily V, member 1 (TRPV1) and peroxisome proliferator-activated receptor ${\gamma}$($PPAR{\gamma}$). The stimuli to induce adipocyte differentiation in hBM-MSCs increase the gene transcription of the $CB_1$ receptor, TRPV1 and $PPAR{\gamma}$. In this study, the effects of three endocannabinoids, N-arachidonoyl ethanolamine (AEA), N-arachidonoyl dopamine (NADA) and 2-arachidonoyl glycerol (2-AG), on adipogenesis in hBM-MSCs were evaluated. The adipocyte differentiation was promoted by AEA whereas inhibited by NADA. No change was observed by the treatment of non-cytotoxic concentrations of 2-AG. The difference between AEA and NADA in the regulation of adipogenesis is associated with their effects on $PPAR{\gamma}$ transactivation. AEA can directly activate $PPAR{\gamma}$. The effect of AEA on $PPAR{\gamma}$ in hBM-MSCs may prevail over that on the $CB_1$ receptor mediated signal transduction, giving rise to the AEA-induced promotion of adipogenesis. In contrast, NADA had no effect on the $PPAR{\gamma}$ activity in the $PPAR{\gamma}$ transactivation assay. The inhibitory effect of NADA on adipogenesis in hBM-MSCs was reversed not by capsazepine, a TRPV1 antagonist, but by rimonabant, a $CB_1$ antagonist/inverse agonist. Rimonabant by itself promoted adipogenesis in hBM-MSCs, which may be interpreted as the result of the inverse agonism of the $CB_1$ receptor. This result suggests that the constantly active $CB_1$ receptor may contribute to suppress the adipocyte differentiation of hBM-MSCs. Therefore, the selective $CB_1$ agonists that are unable to affect cellular $PPAR{\gamma}$ activity inhibit adipogenesis in hBM-MSCs.

Effects of Green Tea Catechin on Microsomal Phospholipase $A_2$ Activity and Arachidonic Acid Cascade in Rat's Heart When Exposed to Microwave (녹차 Catechin의 마이크로웨이브 조사 흰쥐 심장 조직의 Phospholipae $A_2$ 활성과 Arachidonic Acid Cascade계 개선 효과)

  • The purpose of the present study is to investigate the effects of green tea catechin on microsomal phospholipase $A_2(PLA_2)$ activity and the arachidonic acid (AA) cascade in hearts of microwave exposed rats. Sprague-Dawley male rats weighing $100{\pm}10$ g were randomly assigned to one normal group and three microwave exposed groups. The microwave exposed groups were subdivided into three groups: catechin free diet (MW) group, 0.25% catechin (MW-0.25C group and 0.5% catechin (MW-0.5C) group according to the levels of dietary catechin supplementation. Rats were sacrificed $6^{th}$ day after microwave irradiations (2.45 GHz, 15 min). The heart microsome $PLA_2$ activity in the MW group was 130% greater than that of normal groups, whereas there was no significant difference between normal group and MW-0.25C, MW-0.5C group. The per- centage phosphatidyl ethanolamine (PE) hydrolyzed in the heart microsome in the MW was increased 54% by microwave irra- diation, whereas there was no significant difference between normal group and MW-0.5C group. The percentage phosphatidyl choline (PC) hydrolyzed in the heart microsome in the MW group was increased by 104% and by microwave irradiation, whereas there was no significant difference between normal group and MW-0.5C group. The formation of thromboxane $A_2(TXA_2)$ in the heart microsome was 70% greater in the MW group than in the normal group. However, the MW-0.25C and MW-0.5C group maintained the normal level. The formation of prostacyclin ($PGI_2$) in the heart microsome was 21% lower in the MW group than in the normal group, while that of MW-0.25C and MW-0.5C group were maintained in the normal group. The heart microsomal thiobarbituric acid reactive substances (TBARS) concentrations, as an index of lipid peroxide, were 71% greater in the MW group, as compared with normal group. However, the MW-0.25C and MW-0.5C group were 4.6% and 9.2% lower, respectively, than that of MW group. In conclusion, heart function appeared to be improved by green tea catechin supplementation due to its antithrombus action, which in return controls the AA cascade system.

  • Zinc oxide is, one of metal oxide semiconductor, harmless to human and environment-friendly. It has excellent chemical and thermal stability properties. Wurtzite-zinc oxide is a large band gap energy of 3.37 eV and high exciton binding energy of 60 meV. It can be applied to various fields, such as solar cells, degradation of the dye waste, the gas sensor. The photocatalytic activity of zinc oxide is varied according to the particle shape and change of crystallinity. Therefore, It is very important to specify the additives and the experimental variables. In this study, the zinc oxide were synthesized by using a microwave assisted hydrothermal synthesis. The precursor was used as the zinc nitrate, the pH value was controlled as 11 by NaOH. Surfactants are the ethanolamine, cetyltrimethylammonium bromide, sodium dodecyl sulfate, sorbitan monooleate was added by changing the concentration. The composite particles had the shape of a star-like, curcular cone, seed shape, flake-sphere. Physical and chemical properties of the obtained zinc oxide was characterized using x-ray diffractometer, field emission scanning electron microscopy, thermogravimetric analysis and optical properties was characterized using UV-visible spectroscopy, photoluminescence and raman spectroscopy.

Changes' of glycolipids and phospholipids during maturation of perilla seed(Perilla frutescens) (들깨종자의 성숙과정중 당지방질과 인지방질의 변화)

  • To investigate changes in fatty acid and lipid composition of maturing perilla (Perilla frutescens var. japonica Hara) seeds, Suweon 10 and Jeju varieties were subjected to lipid analysis. The results were summarized as follows; As perilla seeds matured, content of glycolipid and phospholipid decreased. Glycolipid and phospholipid of mature seed were 25.4% and 4.5% of total lipids in Suweon 10 and those from Jeju were 44.5%, 4.0%, respectively. Cerebroside, galatosyl diglyceride and monogalatosyl diglyceride were major constitutents of ether-extractable glycolipids in developing perilla seeds. Monogalatosyl diglyceride, the richest constitutent in the early stage of seed development, decreased rapidly as seeds matured. In ether-extractable glycolipid of mature Suweon 10 seeds, content of linolenic acid was 51.1% which was higher than 19.4% of oleic acid of total acids. However, in Jeju variety, content of oleic acid was 31% which was higher than 18.6% of linolenic acid. Phosphatidyl choline, phosphatidyl ethanolamine, phosphatidyl glycerol and lysophosphatidyl inositol were major constituents of phospholipids. Content of phosphatidyl glycerol in developing seeds showed irregular changes in Jeju but in Suweon 10, it decreased after rapid increase in the early stage of seed development. Olieic acid ranges $28.7{\sim}35.2%$, linolenic acid $8.2{\sim}11.2%$, linoleic acid $16.4{\sim}32.5%$ and Palmitic acid $22.7{\sim}29.9%$ of total fatty acids in methanol-ertractible phospholipid of mature perilla seeds.

Characterization of Transparent TiO2 Power and Thin Films through Sol-Gel Process (졸-겔법을 이용한 투과성 TiO2 분말 및 박막의 특성 연구)

  • Transparent $TiO_2$ thin films prepared by sol-gel process using the modification of titanium(IV) alkoxide showed improved thermal stability and high refraction index. Compared to the pure $TiO_2$ film, the modified $TiO_2$ films show the increased index of refraction under proper condition at pH 2.5. Transparency of these $TiO_2$ thin films were more than 80% in the visible region. It has been demonstrated that the reaction occurs in the amorphous phase: an exchange of phase results in anatase before and after 400$^{\circ}C$, in rutile over 700$^{\circ}$C form the XRD results. The particle sizes, shapes and structures of these nanoclusters in the $TiO_2$ films have been identified through a SEM and XRD. The physical properties and structures of their powders have also been studied through a SEM, XRD, TGA and DSC. The thickness and index of refraction have been determined by the analysis of ellipsometric spectra.

Comparison of Lipid Components in Wild and Cultured Bastard (천연 및 양식산 넙치의 지방질성분)

  • The lipid components of cultured bastard(Paralichthys olivaceus) were analyzed and compared with those of wild one and flounder(Pleuronichthys cornutus). Total lipid(TL) content of dorsal muscle in wild and cultured bastard, flounder was 2.0%, 1.6%, and 1.9%, respectively. Wild bastard and flounder were higher in neutral lipid(NL) and glycolipid(GL) contents, while lower in phospholipid(PL) content than those of cultured one. The NL was mainly consisted of triglyceride(70.7-72.6%), and PL was mainly occupied by phosphatidylcholine(53.6-58.3%), phosphatidyl-ethanolamine(25.9-29.5%). In fatty acid composition of TL, cultured bastard showed higher content in polyenes such as docosahexaenoic acid. and lower in monoenes such as palmitoleic, oleic acid than those of wild one. Flounder was higher in Percentage of arachidonic, eicosapentaenoic acid compard with bastard. The major fatty acids of these fishes were palmitic, docosahexaenoic, oleic, eicosapentaenoic and palmitoleic acid in order. These fatty acid composition of NL, GL and PL fractions were not significantly different among wild and cultured bastard, flounder.

Effect of Anti-Phospholipid Antibodies and Phospholipids on In Vitro Fertilization and In Vitro Development of Mouse Oocytes (항인지질 자가항체 및 각종 인지질의 처리가 Mouse 난자의 체외수정 및 초기 배발생에 미치는 영향)

  • Anti-phospholipid antihodies (aPL) have important roles in various pregnancy complications such as recurrent miscarrige, growth retardation, placental abruption and stillbirth. However, their biological actions on preimplantation development of oocytes are still unclear. In this study, we investigated whether either aPL containing sera or phospholipids could affect in vitro fertilization and development of mouse oocytes. Sera used in this study were collected from three patients and the presence of aPL in the sera was confirmed by enzymatic-linked immunosorbent assay. When mouse oocytes were cultured in a serum-free, Chatot, Ziomek and Bavister (CZB) medium (Experiment 1), addition of aPL-containing sera (10%) to CZB medium did not. significantly (P>0.05) influence sperm penetration of oocytes. However, development to the blastocyst stage was significantly (P<0.05) inhibited by serum addition, and formation of morulae (16-23% vs. 58%) and blastocysts (0-4% vs. 21%) was markedly reduced compared with no addition (Experiment 2). In Experiment 3, pronuclear stage embryos were cultured for 96 h in GZB medium supplemented with 1 $\mu$g /ml phosphatidyl ethanolamine, 1 $\mu$g/ml phosphatidyl inositol or 1 $\mu$g /ml phosphatidyl choline. No increase in embryo development was found after addition of the phospholipids to CZB medium. These results suggest that 1) aPL have an inhibitory role in preimplantation development of mouse embryos, and that 2) the action of aPL may be related to a specific phospholid (s) rather than the tested phospholipids in the present study.

Changes in Lipid Components of Gae-bul, Urechis unicinctus, During Hot-air Drying (개불 건조중의 지방질성분의 변화)

  • Changes in lipid components of Gae-bul, Urechis unicinctus, during hot-air drying ($40^{\circ}C$, 7 hrs) were studied. Raw sample contained 1.3% total lipid (TL) which consisted of 35.1% neutral lipid (NL), 18.0% glycolipid (GL) and 46.9% phospholipid (PL), and dried sample contained 5.3% TL which consisted of 51.8% NL, 20.5%GL and 27.7% PL. There were about 40% decrease in PL content and a slight increase in NL content during drying. The NL of raw sample mainly consists of triglyceride (TG, 39.8%), free sterol (FS, 39.6%), free fatty acid (FFA, 12.2%). and also identified diglyceride (DG), monoglyceride and esterified sterol and hydrocarbon in less quantify. The percent of TG and FS decreased, while that of FFA and DG increased during drying. And main components in the PL were phosphatidyl choline (PC,45.6%), phosphatidyl ethanolamine (PE,34.8%), and followed by phosphatidyl serine (PS) and an unknown substance. In the components of PL, PE, PS and PC decreased slightly in order during drying. And major fatty acids of raw and dried samples were generally 16:0, 18: 1, 18:3, and 20:5. The content of the polyenoic acid such as 20:5 decreased. while the saturated acid increased slightly during drying.

  • The lipid compositions of corns produced in Korea were analyzed. Free and bound lipids of the corn kernels were fractionated, quantitated and compared by silicic acid column, thin layer and gas liquid chromatography. Corn kernels contained 5.02% total lipids, which is consisted of 4.09% free lipids and 0.93% bound lipids. Free lipids comprised of 89.61% neutral lipids, 3.75% glycolipids and 6.40% phospholipids, while bound lipids contained 14.26% neutral lipids, 46.06% glycolipids and 37.18% phospholipids. In the neutral lipids of free lipids, triglycerides were predominant (67.68%) and minor components such as esterified sterols, free sterols, free fatty acids, 1,3-diglycerides, 1,2-diglycerides were present. But in the neutral lipids of bound lipids, esterified sterols were not present and the contents of triglycerides were lower (47.68%) and free fatty acids were higher than those of free lipids. Among the phospholipids in free and bound lipids, phosphatidyl choline and phosphatidyl serines and phosphatidyl inositols were also present as minor components. The major fatty acids in the three lipid classes were linoleic, oleic and palmitic acids.

Phospholipase $A_2$ excreted from the cells of hyperthermophilic microbes (초호열성균이 생성하는 phospholipase $A_2$에 관한 연구)

  • We checked the presence of phospholipase $A_2(PLA)_2$ which could split the ester bond at the position 2 in the glycerol backbone of glycerophospholipids, in the cells of hyperthermophiles of Pyrococcus horikoshii and Sulfolobus acidocaldarius. The results obtained are as follows; (1). Pyrococcus horikoshii cells were grown in obligate anaerobic conditions at $95^{\circ}C$ and they needed sulfur as energy source instead of oxygen, while Sulfolobus acidocaldarius species grew well in the aerobic medium (pH 2.5) containing yeast and sucrose at $75^{\circ}C$. (2). Pyrococcus horikoshii cells produced phospholipase $A_2$ in the cell culture media although this species did not show lipase activity at least in the pH range of 1.5 ${\sim}$ 3.5. Sulfolobus acidocaldarius cells produced lipase hydrolyzing triacylglycerols such as triolein, but did not split any kind of phospholipids used as substates. (3). The compound of 1-decanoyl-2-(p-nitrophenylglutaryl) phosphatidylcholine was not suitable for a substrate in this experiment, though frequently used as a subtrate for checking presence of phospholipase $A_2$, for its decomposi-tion in this experiment. The L-${\alpha}$-phosphatidylcholine-${\beta}$-[N-7-nitrobenz-2-oxa-1, 3-diazol]aminohexanoyl-${\gamma}$-hexadecanoyl labelled with a fluorescent material, did not show any migration of acyl chains in the molecule during the reaction with phospholipase $A_2$ under a hot condition. (4). Phospholipase $A_2$ in the cells of Pyrococcus horikoshii, showed the optimum activity at $pH6.7{\sim}7.2$ and $95{\sim}105^{\circ}C$, respectively, and was activated by addition of calcium chloride solution. Andthe phospholipase $A_2$ specifically hydrolyzed glycero-phospholipids such as phosphatidyl choline, phosphatidyl ethanolamine, phosphatidyl serine and phosphatidyl inositol, but could not split phospholipid containing ether bonds in the molecule such as DL -${\alpha}$-phosphatidylcholine-${\beta}$-palmitoyl-${\gamma}$-O-hexadecyl, DL-${\alpha}$-phosphati- dylcholine-${\beta}$- oleoyl-${\gamma}$-O-hexadecyl, DL-phosphatidylcholine-dihexadecyl.
