A study of analgesic effect of Zanthoxylum bungeanum Maxim pharmacopuncture

  • Objectives : This study was carried out to evaluate analgesic effects of Zanthoxylum bungeanum Maxim (ZM) pharmacopuncture on formalin-induced pains in Sprague-Dawley (SD) rats and ICR-mice. Methods : The subjects were divided 8 weeks aged rats with constant pain sensitivity into five groups; normal (treated with normal saline at Taegye (KI3) and before injected with normal saline at hindpaw), Con-1 (treated with normal saline at KI3 before injected with formalin at hindpaw), Lido-1 (treated with lidocaine at KI3), ZMWG-1 (treated with Hot water extraction pharmacopuncture of Zanthoxylum bungeanum Maxim at KI3), ZMEG-1 (treated with ethanol extraction pharmacopuncture of Zanthoxylum bungeanum Maxim at KI3). After 35 minutes, we measured ultrasonic vocalization (USV) and enzyme activities of both Aspartate aminotransferase (AST), Alanine aminotransferase (ALT) in rat serum. In addition, Tail flick test is performed by injecting ICR mice at 5 weeks of age. And it classified into 4 groups (Con-2, Lido-2, ZMWG-2, ZMEG-2) according to the kind of drug (normal saline, lidocaine, ZMW, ZME). After each drug injection, we examined the reaction by placing the tail in water at $50^{\circ}C$. Results : ZME had analgesic effects in the early and late phase of USV during the formalin test. There were no significant differences between ZMEG-1 and Lido-1 in early and late phase of USV. Also, No significant differences observed in serum AST and ALT activity in ZMWG-1 and ZMEG-1 compared with Con-1. For tail-flick test, analgesic effect on warmth significantly increased in Lido-2 and ZMEG-2 compare to that of Con-2. Conclusion : ZME pharmacopuncture had analgesic effects on formalin-induced pain without liver toxicity. Also, tail-flick test suggest that ZME pharmacopuncture could be useful technique on analgesic effect on warmth and treatment of pains.

단삼(Salvia Miltiorrhiza) 추출물의 암세포 증식 억제 효과에 관한 연구 (Inhibitory Effect of Salvia miltiorrhiza Extract on Growth of Some Cancer Cells)

  • 본 연구에서는 단 삼(Salvia miltiorrhiza)의 추출물이 인체 결장암세포인 HT-29, 간암세포인 HepG2 및 직장 암세포인 HRT-18의 증식에 미치는 영향을 in vitro에서 확인하였다. 암세포의 배양액에서 단삼의 수용성 추출물과 에탄올 추출물의 암세포 증식억제효과는 농도에 비례하여 에탄올 추출물에서 더 효과적이었다. 이를 토대로 단삼의 에탄올 추출물의 급성독성, 수명연장 및 고형암형 성억제를 살펴보기 위해 in vivo 실험을 하였다. 급성독성실험결과 대조군의 15일 째의 평균체중은 32.3 g 이었고 실험군은 31.6 g으로 정상적인 상태를 유지하였으며, 흰쥐의 육종암세포인 sarcoma-180를 접종한 mouse의 수명연장실험결과 대조군에 비해 61%의 수 명연장 효과가 있음을 관찰하였다. 고형암억제 실험 결과에서도 대조군에 비해 에찬올 추출 물 처리군이 35%의 고형암 형성억제능을 나타내었다. 따라서 단삼의 에탄올 추출물 중에는 in vitro와 in vivo 실험을 통해 항암활성을 갖는 성분이 존재하며, 이성분은 유효한 항암제 로 개발될 수 있으리라 사료된다.

치즈 숙성 중의 곰팡이 오염 방제 - 현황과 전망 (Prevention of Fungal Contamination during Cheese Ripening - Current Situation and Future Prospects)

  • Molds cause severe cheese deterioration, even though some white and blue molds are used for the manufacture of Camembert and Blue cheese, respectively. The species of Geotrichum, Moniliella, Aspergillus, Penicillium, Mucor, Fusarium, Phoma, and Cladosporium are the main fungi that affect contamination during cheese ripening. Once deteriorated by fungal spoilage, cheese becomes toxic and inedible. Fungal deterioration of cheese decreases the nutritional value, flavor profiles, physicochemical and organoleptic properties, and increases toxicity and infectious disease. Fungal contamination during cheese ripening is highly damaging to cheese production in Korean farmstead milk processing companies. Therefore, these companies hesitate to develop natural and ripened cheese varieties. This article discusses the recent and ongoing developments in the removal techniques of fungal contamination during cheese ripening. There are 2 categories of antifungal agents: chemical and natural. Major chemical agents are preservatives (propionic acid, sodium propionate, and calcium propionate) and ethanol. Among the natural agents, grapefruit seed extract, phytoncide, essential oils, and garlic have been investigated as natural antifungal agents. Additionally, some studies have shown that antibiotics such as natamycin and Delvocid$^{(R)}$, have antifungal activities for cheese contaminated with fungi. Microbial resources such as probiotic lactic acid bacteria, Propionibacterium, lactic acid bacteria from Kimchi, and bacteriocin are well known as antifungal agents. In addition, ozonization treatment has been reported to inhibit the growth activity of cheese-contaminating fungi.

감초 물 추출물 및 Glycyrrhizin이 인체 간 Microsome에서 Cytochrome P450 약물대사효소에 미치는 영향 (Inhibitory Effect of Licorice Ethanol Extracts and Glycyrrhizin on Cytochrome P450 Drug-Metabolizing Enzymes in Human Liver Microsomes)

  • Objective : The aim of present study is to evaluate the inhibitory potential of licorice extract and glycyrrhizin on cytochrome P450(CYP) in human liver microsomes. Methods : Using human liver microsomes, water extract of licorice and glycyrrhizin as an inhibitor were co-incubated with each probe drug representing selective CYP isoform activity. We measured relative metabolic activity in incubation condition compared to that with no extract of licorice using HPLC system. Results : Both water extracts of licorice and glycyrrhizin showed inhibitory effect on CYP-catalyzed reactions. CYP2C19 $(IC_{50}=126.7{\mu}g/ml)$ is most potently inhibited by water extract than other tested CYP isoforms$(IC_{50}>450{\mu}g/ml)$, but glycyrrhizin exhibited potent inhibition on CYP1A2$(IC_{50}=106.9{\mu}g/ml)$ followed by CYP2C9 and CYP2D6. Conclusion: These results indicate that water extract of licorice and glycyrrhizin have inhibitory potential on CYP-catalyzed reaction in human liver microsomes. But the mechanism of inhibition was slightly different between them Water extract of licorice mainly inhibited CYP2C19, and glycyrrhizin primarily inhibited CYP1A2. The inhibition by water extract of licorice and glycyrrhizin on CYP isoforms may cause drug interaction with co-administered drug leading to toxicity or treatment failure.

천정기보단(天精氣保丹)의 자외선에 의한 세포 손상 억제 효과 (Protective Effect of Cheonjeongkibo-Dan UV-Induced Cellular Damage in Human Dermal Fibroblast)

  • In this study, we prepared CheonJeongKiBo-Dan(7 oriental medicinal plants, 7OMP: Astragalus Membranaceus root, Panax Ginseng root, Glycyrrhiza Glabra (licorice) root, Schizandra Chinensis fruit, Polygonatum Odoratum, Rehmannia Glutinosa root, Paeonia Albiflora root) by extracting them in one reactor and studied its efficacies on skin. UV irradiation has been suggested as a major cause of photoaging in skin. In order to investigate protective effects against UV-B induced cellular damage, 7OMP was extracted with 70% ethanol and dissolved in DMSO. The protective effect was detected by MTT assay, reactive oxygen species (ROS) generation, phosphorylation of ATR and p53 in human dermal fibroblast cell system after UV-B irradiation. 7OMP reduced UV-B-induced cellular damage in HDFs cells, and inhibited ROS generation. UV-B-induced toxicity accompanying ROS production and the resultant DNA damage are responsible for activation of ATR, p53 and Bad. In this study, 7OMP hampered phosphorylations of ATR and p53 in human dermal fibroblasts. Therefore, 7OMP may be protective against UV-induced skin photoaging.

LPS로 유도된 대식세포에서 수치 치자의 항염효과 (Anti-inflammatory Effect of the Processed Gardeniae Fructus in LPS-induced Macrophages)

  • Objective : Herbal processing is one of the traditional techniques used in Korean medicine to increase the effectiveness of herbs or reduce their toxicity. In this study, Gardeniae Fructus processed with ginger juice and alcohol was prepared to evaluate the anti-inflammatory effect on lipopolysaccharide (LPS)-induced macrophages. Methods : The processing of Gardeniae Fructus was performed by adding 40 % ginger juice or 10% alcohol to the total weight of Gardeniae Fructus and then roasting at 150℃ for 5 minutes. Cell viability was determined by 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. To detect nitric oxide (NO) production, culture media were mixed with Griess reagent and measured the absorbance at 540 nm. Prostaglandin E2 (PGE2) and pro-inflammatory cytokines were detected by enzyme-linked immunosorbent assay (ELISA). Western blot was applied to monitor protein expression levels. Results : LPS-induced NO, PGE2 and inflammatory cytokines were decreased by the treatment of normal or processed Gardeniae Fructus ethanol extracts (GFE). Compared to normal GFE, the processed GFE showed a stronger inhibitory effect on the production of NO and PGE2. These inhibitory effect of GFE was due to the suppression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) mediated from the inhibition of nuclear factor kappa B (NF-κB). Furthermore, processed GFE showed more suppressive effects on the expression of iNOS, COX-2 and IκBα proteins than normal GFE. Conclusion : From these results, it was concluded that GFE had an improved anti-inflammatory effect compared to normal GFE. These results provide an objective evidences for the use of herbal processing in Korean medicine.

돌외 잎 추출물의 콜라겐 합성 증진 성분 규명 (Constituents of Collagen Synthesis Activation from the Extracts of Gynostemma pentaphyllum Leaves)

  • 본 연구에서는 주름개선 화장품 원료를 개발하기 위하여 돌외 잎 70% 에탄올 추출물을 제조하여 섬유아세포 HDFn에 대하여 콜라겐 합성능을 측정하였다. 그 결과 돌외 추출물이 세포 독성 없이 농도 의존적으로 콜라겐 합성을 촉진함을 확인하였다. 돌외 추출물로부터 용매 분획 및 크로마토그래피 분리과정을 거쳐 2개의 활성 성분을 분리하였다. NMR 데이터 및 문헌치로 확인한 결과 이들은 플라보노이드 배당체인 Ombuine 3-O-rutinoside(1) 및 Quercetin 3-O-rutinoside(2)로 규명되었다. 섬유아세포의 type I procollagen 생합성에 미치는 영향을 분석한 결과 상기 화합물은 농도 의존적으로 콜라겐 합성을 촉진함을 알 수 있었다. 본 실험 결과는 화합물 1과 2를 함유한 돌외 추출물의 주름개선 화장품 소재로서의 개발 가능성을 보여주고 있다.

회초리말(Mutimo cylindricus)의 항산화, 항염 및 미백 활성 (The Antioxidation Effect of Mutimo cylindricus Extract and Its Influence on Cell Bioactivity)

  • 본 연구는 갈조류 중 하나인 회초리말의 기능성 화장품 원료로서 기능성을 알아보기 위하여, 항산화 실험 및 세포실험을 실시하였다. 항산화능 측정을 위해 회초리말을 70% 에탄올로 추출물을 제조하였다. 항산화 측정에는 DPPH와 ABTS 법을 사용하였으며, 각 실험에서 EC50값이 2.040과 2.182 mg/mL로 나타났다. 폴리페놀 함량과 환원능 측정에서는 103 mg gallic acid/g extract의 폴리페놀 함량과 134 mg ascorbic acid/g extract의 환원능이 나타났다. 세포실험에서는 MTT 법을 사용하여 해당 추출물이 세포독성이 없음을 나타내었으며, 100 ㎍/mL 농도에서 항염능과 미백능을 나타내었다. 결과적으로 수송나물 추출물은 미백 및 항염능을 가진 화장품 소재로서 사용가능함을 확인하였다.

Effect of Saponin Fraction from Platycodon grandiflorum on Clinical Chemical Changes in TCDD (2,3,7,8-Tetrachlorodibenzo-ρ-dioxin)-induced Rat Toxicity

  • This study was carried out to investigate the protective effect of crude saponin from Platycodon grandiflorum on Clinical chemical parameters in male rats acutely exposed to 2,3,7,8-tetrachlorodibenzo-ρ-dioxin (TCDD). Crude saponin was prepared from Korean Platycodon grandiflorum with Diaion HP-20 adsorption chromatography after extraction of 80% ethanol at 75℃. The crude saponin was confirmed by thin layer chrmatography. When compared with ginseng saponins, the crude saponin had both a few number of saponins and a broad distribution. Forty male rats (200±20 g) were divided into 4 groups. Normal control (NC) group received vehicle and saline; TCDD-treated (TT) group received TCDD (40 ㎍/kg, single dose) intraperitoneally; Platycodon grandiflorum saponin (PG5 and PG10) groups received crude saponin 5 mg/kg and 10 mg/kg (p.o), respectively, for 2 weeks before 1 week of TCDD-exposure. Increase of body weight was retarded greatly by TCDD-exposure. Body weight of animals in TT group was significantly decrease after 2 days of TCDD-exposure. However, body weights of animals in PG groups increased through the experimental perimental period, although the increasing rate was slower than that of NC group. Increases in contents of blood glucose, total cholesterol and triglyceride (TG) and activities of amylase, lipase, AST, ALT and LDH by toxic action of TCDD were significantly attenuated by crude saponin from Platycodon grandiflorum (P<0.05). In conclusion, these results suggest that crude saponin prepared from Korean Platycodon grandiflorum might be a member of useful protective agents against TCDD, which is one of the environmental hormones.

가상 검색 및 시험관 시험을 이용한 총명탕 중 주성분들에 대한 약물작용 및 대사 예측 (In silico Prediction and In vitro Screening of Biological Activities and Pharmacokinetics for the Major Compounds in Chong Myung Tang)

  • Chong Myung Tang is consisted of three medicinal herbs (Acori Graminei Rhizoma, Polygalae Radix and Hoelen cum Radix). It has been used as a medicine for the purpose of learning and memory improvement. In this paper, Chong Myung Tang was screened the biological activities for Alzheimer's disease. The extract (70% ethanol) of Acari Graminei Rhizoma (1 mg/ml) showed that acetylcholinesterase (AChE) and amyloid beta ($A{\beta}$) peptide aggregation inhibitory potency are 43.1% and 76.5%, respectively. The extract of Polygalae Radix showed inhibitory activity against $A{\beta}_{1-42}$ peptide aggregation (51.5%). To predict the drug-likeness, oral absorption ability; blood-brain barrier (BBB) penetraion rate, mutagenecity and carcinogenicity; in silico screening was performed against 16 compounds in the three medicinal herbs. According to the results, all compounds have appropriate chemical structures as medicines. The six compounds in Acori Graminei Rhizoma and the five compounds in Hoelen cum Radix showed excellent oral absorption rate and BBB penetration rate. The four compounds in Polygalae Radix showed excellent oral absorption rate, but their BBB penetration was presented low rate. And, the extract of Hoelen cum Radix didn't show AChE and $A{\beta}_{1-42}$ peptide aggregation inhibitory activities in vitro. Therefore, their activity in brain may be other mechanism. According to all of the results, in silico prediction technology is convenient and effective to determine biological active compounds in medicinal herbs.
