Anticariogenic Properties of the Ethanol Extract of Pomegranate (Punica granatum) Husk (석류피 에탄올 추출물의 항치아우식 활성)

  • Dental caries is a major worldwide oral disease problem. Streptococcus mutans (S. mutans) plays an important role in the information of dental plaque and it is being noticed as major causative bacteria of dental caries. Therefore, the development of more effective, substantial and safe preventive agents against dental caries is strongly required. In the present study, inhibitory effects of the ethanol extract of the husk of pomegranate (Punica granatum L.) on the growth, acid production, adhesion and water-insoluble glucan synthesis of Streptococcus mutans (S. mutans) were examined. The ethanol extract of pomegranate husk (250 - 4000 ${\mu}$g/ml) significantly lowered the growth of S. mutans in a dose dependent manner. The acid production of S. mutans were inhibited by the presence of ethanol extract of pomegranate husk (500 - 4000 ${\mu}$g/ml) significantly. The ethanol extract of pomegranate husk (5000 - 4000 ${\mu}$g/ml) also significantly lowered the adherence of S. mutans. In water-insoluble glucan synthesis assay, 1000 - 4000 ${\mu}$g/ml of the ethanol extract of pomegranate husk significantly inhibited the formation of water-insoluble glucan. These results suggest that pomegranate husk may inhibit the caries-inducing properties of S. mutans. Further studies are necessary to clarify the active constituents of pomegranate husk responsible for such biomolecular activities.

In the present study, inhibitory effects of the ethanol extract of Dianthus superbus Linne (D. superbus) on the growth, acid production, adhesion and water-insoluble glucan synthesis of S. mutans were examined. The ethanol extract of D. superbus (0.5 - 4 mg/ml) significantly lowered the growth of S. mutans in a dose dependent manner. The acid production of S. mutans were inhibited by the presence of ethanol extract of D. superbus(1 - 4 mg/ml) significantly. The ethanol extract of D. superbus (0.25 - 4 mg/ml) also significantly lowered the adherence of S. mutans in a dose dependent manner. In water-insoluble glucan synthesis assay, 0.25 - 4 mg/ml of the ethanol extract of D. superbus significantly inhibited the formation of water-insoluble glucan. These results suggest that D. superbus may inhibit the caries-inducing properties of S. mutans. Further studies are necessary to clarify the active constituents of D. superbus responsible for such biomolecular activities.

  • In the present study, Inhibitory effects of the ethanol extract of Saussurea lappa (S. lappa) on the growth, acid production, adhesion and water-insoluble glucan synthesis of Streptouccus mutans (S. mutans) were examined. The growth and acid production of S. mutans were Inhibited by the presence of ethanol extract of S. lappa (0.5-4 mg/ml) significantly. The ethanol extract of S. lappa (0.25-4 mg/ml) also significantly lowered the adherence of S. mutans in a dose dependent manner. In water-insoluble glucan synthesis assay, 2-4 mg/ml of the ethanol extract of S. lappa significantly inhibited the formation of water-insoluble glucan. These results suggest that S. lappa may inhibit the caries-inducing properties of S. mutans. Further studies are necessary to clarify the active constituents of S. lappa responsible for such biomolecular activities.

  • The hot water extracts of Paeonia japonica, Paeonia moutan, Eucommia ulmoides, Bupleurum falcatum, Cornus officinalis were freeze dried and dissolved with 70% ethanol. These extracts were fractionated with ethanol soluble fraction(ESF) and ethanol insoluble fraction(EIF). To compare oxidative stability effect of these extracts with that of BHA, linoleic acid with 3,000ppm extracts was incubated at 35$^{\circ}C$. Induction period of BHA was 9 days but that of Paeonia japonica ESF and Scutellaria baicalensis ESF was 12, 9 days respectively. And the higher antioxidant activity was observed for ESF than EIF of these extracts.

  • It has been well established that S. mutans is the major etiological agent in dental caries, one of the most common oral diseases worldwide. The present study was designed to investigate the effect of Saururus chinensis (S. chinensis) ethanol extracts on the growth, acid production, biofilm formation, adhesion, and insoluble glucan synthesis of S. mutans. The ethanol extracts of S. chinensis showed concentration dependent inhibitory activity against the growth and acid production of S. mutans, and produced significant inhibition at the concentration of 0.025, 0.05, 0.1, 0.2 and 0.4 mg/ml compared to the control group. The extracts markedly inhibited S. mutans adherence to HA treated with saliva, and cell adherence was repressed by more than 80% at the concentration of 0.05 mg/ml and complete inhibition was observed at the concentration of 0.4 mg/ml. On the activity of glucosyltransferase which synthesizes water insoluble glucan form sucrose, ethanol extract of S. chinensis showed more than 10% inhibition over the concentration of 0.025 mg/ml. The synthesis of insoluble glucan was decreased in the presence of 0.025 ${\sim}$ 0.4 mg/ml of the ethanol extract of S. chinensis. Our research strongly suggested S. chinensis was a promising natural product for the prevention of dental caries.

Inhibitory Effects of Galla Chinensis Extract on Cariogenic Properties of Streptococcus mutans (오배자 Galla Chinensis 추출물이 Streptococcus mutans의 우식활성 억제에 미치는 영향)

  • Streptococcus mutans (S. mutans) is one of the most important bacteria in the formation of dental plaque and dental caries. S. mutans adheres to an acquired pellicle formed on the tooth surface, and aggregates with many oral bacteria, and initiates plaque formation by synthesizing glucan from sucrose, which is catalyzed by glucosyltransferases. S. mutans metabolizes the dietary sugar to the organic acids. The organic acids demineralize tooth surface and result in dental caries. Galla Chinensis have been traditionally used for stopping bleeding of gingiva, removing edema and halitosis, drainage, fixing the teeth and as an antiphlogistic agent. In previous reports, antibacterial effects of Galla Chinensis have been investigated whereas anti-cariogenic effects is still not examined enough. Therefore we tested effects of ethanol extracts of Galla Chinensis on the cariogenic properties such as the growth, acid production, adhesion, and insoluble glucan synthesis of S. mutans. In the result, ethanol extracts of Galla Chinensis showed the inhibition of S. mutans growth and organic acids production over 0.031 mg/ml concentrations. The adhesion of S. mutans to Saliva-coated Hydroxyapatite beads S-HAs has decreased with the increase of concentration of ethanol extracts of Galla Chinensis. And it seems to have adhesion inhibitory effect in concentration of over 0.25 mg/ml. It gives us the result that Galla Chinensis have anti-caries effects. But ethanol extract of Galla Chinensis didn't have inhibitory effect on insoluble glucan synthesis. Preliminary phytochemical analysis of the ethanol extract of Galla Chinensis showed strong phenolic compounds, medium steroids & terpenoids and glycosides, and weak organic acids and peptides. These results suggest that the ethanol extracts of Galla Chinensis may have anti-cariogenic properties, which may be able to be related with strong phenolic compounds.

  • Streptococcus mutans is considered one of the primary etiologic agents of dental caries. we studied the effect of the ethanol extracts of Tribuli fructus (T. fructus) on the growth, biofilm formation, acid production, adhesion and insoluble glucan synthesis of S. mutans. The ethanol extracts of T. fructus showed concentration dependent inhibitory activity against the growth and acid production of S. mutans, and produced significant inhibition at the concentration of 0.025, 0.05, 0.1, 0.2, 0.3, 0.5 mg/ml compared to the control group. In the biofilm assay, the ethanol extracts of T. fructus inhibited formation of biofilm synthesized by S. mutans at the concentration of 0.05 mg/ml. The extracts markedly inhibited S. mutans adherence to HA treated with saliva, and cell adherence was repressed by more than 50% at the concentration 0.05 mg/ml. On the activity of glucosyltransferase which synthesizes water insoluble glucan form sucrose, ethanol extract of T. fructus showed more than 10% inhibition over the concentration of 0.025 mg/ml. Hence, we conclude that T. fructus might be a candidate of anticaries agent. Further studies are necessary to clarify the active constituents of T. fructus responsible for such biomolecular activities.

  • The roots and rhizome of Acanthopanax koreanum are used as folk medicine to ameliorate hepatitis in Korea. The ability of A. koreanum to protect mice from fulminant hepatitis induced in mice by D-galactosamine and lipopolysaccharide was evaluated.   • It is noted that Streptococcus mutans (S. mutans) triggers dental caries establishment by two major factors: the synthesis of organic acids, which demineralize dental enamel, and the synthesis of glucans, which mediate the attachment of bacteria to the tooth surface. Therefore, it is noted that the development of a more effective, substantial and safe preventive agent that works against dental caries and periodontal disease is required at this time. For this reason, the present study was designed to investigate the effect of croton seed ethanol extracts on the growth, acid production, adhesion, and insoluble glucan synthesis of S. mutans. In this case, the ethanol extract of croton seed showed concentration dependent inhibitory activity against the growth, acid production and adhesion of S. mutans. Especially, it is important to note that it has produced significant inhibition at the concentration of 0.1 and 0.2 mg/ml as compared to the control group. Moreover, these results suggest that the application of croton seed extract may be considered to be a useful method for the prevention of dental caries.

Effect of Dried Powders or Ethanol Extracts of Garlic Flesh and Peel on Lipid Metabolism and Antithrombogenic Capacity in 16-Month-Old Rats (마늘 육질과 껍질의 건분 및 에탄올추출물이 노령흰쥐의 지방대사와 항혈전능에 미치는 영향)

  • This study was performed to investigate effect of dried powders and ethanol extracts of garlic flesh and peel on lipid metabolism and antithrombogenic capacity in 16-month-old rats. Forty Sprague-Dawley male rats weighing 618.1$\pm$6.5 g were blocked into five groups according to body weight and raised for 3 months with control and experimental diets containing 5% (w/w) of dried powders of garlic flesh or peel, or ethanol extracts from equal amount of each dried powder and control diet. Plasma and liver total lipids, triglyceride and total cholesterol, and plasma HDL-cholesterol, throm-boxane $B_2$ (TX $B_2$), 6-keto-prostaglandin $F_{1a}$ (6-keto-PG $F_{1a}$) concentrations were measured. Total, insoluble and soluble dietary fibers contents were highest in peel powder followed by fresh powder, and those in ethanol extracts of flesh and peel, especially soluble, very low. Plasma and liver total lipids, triglyceride, and total cholesterol concentrations were lower in all the garlic experimental groups compared to Especially, flesh and peel powder lowered plasma total lipids, triglyceride and total cholesterol concentrations markedly, and flesh powder and flesh ethanol extract lowered liver total lipids, triglyceride and total cholesterol concentration remarkably. Plasma TX $B_2$ concentrations in garlic experimental groups were lower than that of control group, and 6-keto-PG $F_{1a}$ concentrations. In garlic experimental groups were higher than that of control group. Flesh ethanol extract group showed the lowest TX $B_2$ and the highest 6-keto-PG $F_{1a}$ concentrations among experimental groups, so TX $B_2$/6-keto-PG $F_{1a}$ ratio in flesh ethanol extract group was significantly lower than that of control group. Moreover, clotting time was significantly increased in flesh ethanol extract group as compared to control group. In conclusion, intakes of dried powders and ethanol extracts of garlic flesh and peel were effective in lowering lipid levels of liver and plasma. And also flesh ethanol extract diet was most effective in antithrombogenic activity among garlic experimental groups as TX $B_2$/6-keto-PG $F_{1a}$ ratio in flesh ethanol extract group was significantly lower and clotting time was significantly increased in this group as compared to control group.ntrol group.
