진달래꽃 탄화수소류의 곡자에 의한 분해 (The Degradation of hydrocarbons in Petal of Azalea by Gokja)

  • Petal of Azalea(Rhododendron mucronulatum Turcz) was incubated with Gokja at 3$0^{\circ}C$ for seven days and the essential oil components of petal of Azalea before and after incubated were analyzed using a GC/MSD. Ten or more essential oil components including n-heneicosane n-tricosane n-tetreacosane n-pentacosane n-heptacosane n-nonacosane and n-hentriacontane were identified from the petal of Azal-ea before incubated while oxygen-containng compounds including (E)-heptenal 2-ethoxy-1 -hexanol n-hexadecanoic acid methyl ester 9, 12-octadecadienoic acid methyl ester 9,12,15-octadecatrienoic acid methyl ester, n-octadecanoic acid methyl ester n-eicosanoic acid methyl ester and 9-docosaenoic acid methyl ester as well as n-alkanes such as n-tricosane that n-pentacosane were identified from the petal of Azalea after incubated. These results suggest that n-alkanes in petal of Azalea might be degraded and some oxygen-containing compounds such as aldehyde, esters and /or acids might be produced when pet-al of Azalea is incubated with Gokja.

Synthesis of Carbobenzoxy-alanyl-thiaarginine (thialysine) benzyl ester and kinetic Studies with Trypsin

  • Carbobenzoxy-alanyl-thiaarginine benzyl ester and carbobenzoxy-alanyl-thialysine benzyl ester were synthesized in solution. Kinetic studies were carried out using three different analytical methods, semi-classical method, progress curve analysis and competitive spectrophotometry. In competitive spectrophotometry, carbobenzoxy-valyl-glycyl-arginyl-p-nitroaniline was used as a detector. Kinetic constants such as $K_m$ and $V_{max}$ measured by competitive spectrophotometry are almost the same as those values measured by semi-classical method. Colorimetric Ellman's assays showed the thio-peptido mimetics to be a suitable substrates for trypsin. Kinetic studies with trypsin gave $K_m$ of 2.33 mM and $k_{cat}$ of $1.50{\times}10^5\;min^{-1}$ for carboxy-alanyl-thiaarginine benzyl ester and $K_m$ of $3.41{\times}10^{-3}\; Mm\; and\; k_{cat}\; of\; 520{\times}102\; min^{-1}$ for carbobenzoxy-alanyl-thialysine benzyl ester, respectively. Kinetic constants $(K_m=2.04{\times}10^{-2}\; mM, K_{cat}=4.42{\times}10^3 \;min^{-1})$ for natural substrate, carbobenzoxy-alanyl-lysine benzyl ester, were also evaluated by competitive spectrophotometry in order to compare the mode of binding on trypsin.

  • 한국 토양균 중에서 esterase 저해제를 생산하는 균주인 Streptomyces strain DMC-498을 oatmeal yeast extract 배지에서 3일간 액내 배양하였을 때 최고의 저해작용을 나타내었으며 균사체의 메탄올 추출물로부터 저해제를 분리하였다. linoleic acid가 저해작용이 있음을 확인하였으며 50% 저해작용을 나타내는 linoleic acid의 양은 $0.045\;\mu\textrm{g}/ml$이었다. 활성이 보다 큰 compound A는 분자량이 500 이상이며 산소를 2개이상 함유하고 있는 환상구조의 지방족 화합물로 생각된다. 이것을 상경적 저해작용을 나타내었다. DMC-498 균주가 함유하는 지질의 주성분은 isostearic acid, isostearic acid methyl ester, linoleic acid methyl ester 및 oleic acid methyl ester이었다.

  • 순무 뿌리로부터 활성 물질을 분리 동정 하기 위하여 80% MeOH 수용액으로 추출하고 이를 여과, 감압 농축하여 MeOH 추출물을 얻었다. 이를 EtOAc분획, n-BuOH분획, $H_{2}O$분획으로 나누었으며, EtOAc분획과 n-BuOH분획에 대해 silica gel 및 ODS column chromatography를 실시하여 5종의 화합물을 분리 정제하였다. $^{1}H-NMR$, $^{13}C-NMR$, DEPT spectrum 및 Mass spectrum 등을 통하여 palmitic acid methyl ester(1), linolenic acid methyl ester(2), linoleic acid methyl ester(3), ${\beta}-sitosterol$(4), daucosterol(5)으로 구조를 결정하였다.

HFC-404a와 Polyol ester 오일 혼합물의 증기압 측정 (Measurement of Vapor Pressure of HFC-404a and Polyol ester Mixture System)

  • 기존의 CFC 계열 및 HCFC 계열 냉매들이 오존층 붕괴와 지구온난화의 원인들 중의 한가지로 밝혀지면서 이것을 대체할 수 있는 냉매의 개발은 주요한 연구과제였다. 이제 몇 가지 대체 냉매들이 개발되면서 이것들 이 냉동시스템에 사용될 때 발생하는 상용성 (Compatibility)이 연구될 필요가 있다. 본 연구에서는 대체냉매의 상용성 연구의 일환으로 1,1,1,2-Tetrafluoroethane(HFC-134a) +1,1,1-Trifluoroetane(HFC-143a) + Pentafluoroethane (HFC-125)의 혼합대체냉매 HFC-404a 와 냉매 윤활유의 하나인 POE 오일 혼합물의 증기압을 측정하였다. 측정온도범위는 냉매 시스템의 운전온도를 고려하여, 263.15K 에서 323.15 K로 하였으며 오일농도범위는 0 mass% 에서 90 mass% 까지로 하였다. 측정결과 273.15K 이하의 온도에서 오일 농도 30 mass%까지 증기압에 대한 오일의 영향은 미미하였으며 50 mass% 이상에서는 증기압이 급격히 떨어지는 것이 밝혀졌다. 측정자료를 이용하여 HFC-404a와 POE 오일 혼합물의 증기압을 예측할 수 있는 모형을 개발 하기 위하여 Rault 모델과 Flory-Huggins 모델을 사용하여 측정치와 비교하였다. 그리고 보다 정확하고 실용적으로 증기압을 예측할 수 있는 경험식을 도출하였다.

Cellular growth and fatty acid content of Arctic chlamydomonadalean

  • Arctic microalgae thrive and support primary production in extremely cold environment. Three Arctic green microalgal strains collected from freshwater near Dasan Station in Ny-Alesund, Svalbard, Arctic, were analyzed to evaluate the optimal growth conditions and contents of fatty acids. The optimal growth temperature for KNF0022, KNF0024, and KNF0032 was between 4 and 8℃. Among the three microalgal strains, KNF0032 showed the maximal cell number of 1.6 × 107 cells mL-1 at 4℃. The contents of fatty acids in microalgae biomass of KNF0022, KNF0024, and KNF0032 cultured for 75 days were 37.34, 73.25, and 144.35 mg g-1 dry cell weight, respectively. The common fatty acid methyl esters (FAMEs) analyzed from Arctic green microalgae consisted of palmitic acid methyl ester (C16:0), 5,8,11-heptadecatrienoic acid methyl ester (C17:3), oleic acid methyl ester (C18:1), linoleic acid methyl ester (C18:2), and α-linolenic acid methyl ester (C18:3). KNF0022 had high levels of heptadecanoic acid methyl ester (26.58%) and heptadecatrienoic acid methyl ester (22.17% of the total FAMEs). In KNF0024 and KNF0032, more than 72.09% of the total FAMEs consisted of mono- and polyunsaturated fatty acids. Oleic acid methyl ester from KNF0032 was detected at a high level of 20.13% of the FAMEs. Arctic freshwater microalgae are able to increase the levels of polyunsaturated fatty acids under a wide range of growth temperatures and can also be used to produce valuable industrial materials.

Development of Phytosterol Ester-added Cheddar Cheese for Lowering Blood Cholesterol

  • This study was carried out to investigate the effect of phytosterol ester addition on lowering blood cholesterol in cholesterol-reduced Cheddar cheese. For cholesterol removal, separated cream was treated with 10% ${\beta}$-cyclodextrin at 800 rpm, then blended with remaining skim milk and homogenized with 1,000 psi at $70^{\circ}C$. Experimental cheeses were manufactured by five different levels of phytosterol addition. After the cholesterol reduction process by ;${\beta}$-cyclodextrin, the cholesterol removal rate was in the range of 91.0 to 92.1%. Amount of short-chain free fatty acid and free amino acids increased with an increase of phytosterol ester, and those were significantly different from that of control in all ripening periods. All rheological properties also increased with an increase of phytosterol ester during ripening period. In sensory analysis, the scores of rancid, bitterness Cheddar flavor and off-flavor intensities increased significantly, while texture was decreased during ripening in phytosterol ester-added groups. Total blood cholesterol was reduced by 18% when rats were fed Cheddar cheese treated with 8% phytosterol. The present study indicated that phytosterol ester addition resulted in a profound lowering effect of blood with cholesterol-reduced Cheddar cheese.

Vegetable Oil Esters에 따른 Offset 잉크의 물성 변화에 관한 연구 (The Variation of Offset Ink Properties according to the Vegetable Oil Estersr)

  • According as gradually increasing the demand for eco-friendly at the printing process, it has been progressed fairly development. Especially, the inks are used by soy oil beginning of ink industry for preventing environment, it is possible to make eco-friendly inks with vegetable ester. So it is not necessary to use petroleum-based solvents at all for preventing environment. These eco-friendly inks have a benefits they are able to use the renewable resource. But basically vegetable oils have that reduce the VOC(Volatile Organic Compound) and high viscosity, high solubility properties. So if the vegetable oils use in the ink, set off problem occur on the paper because of slow drying time. In case of vegetable ester, it has similar the molecular weight and kinetic viscosity with hydrocarbon solvent, it is able to control the power of dilution about the resin. So, it has benefit that solve the problem of the existing eco-friendly inks. In this study, different types of ester were made by six types of vegetable oils and used ester in the varnishes and inks properties are comparison with hydrocarbon solvent based ink. By considering the intrinsic properties of vegetable oil, ester used to analyze the changes in ink properties, using ester varnish is applied to study the rheology characteristics and emulsification with inks.

폴리에스테르 수지를 이용한 콤포스트 백 분해에 관한 연구 (Decomposition of Compost Bag Using Polyester Resin)

  • 본 연구에서는 폴리에스테르 비닐과 고밀도 폴리에틸렌(HDPE)의 분해에서 수분 함량, pH, 연소시 무게에 대하여 조사하였다. 폴리에스테르 비닐은 30일 동안의 음식물 쓰레기의 미생물에 의하여 분해가 이루어졌으나, 고밀도 폴리에틸렌(HDPE)은 분해가 전혀 이루어지지 않았다. 폴리에스테르계 생분해성 비닐은 음식물의 미생물에 의하여 10일후에 급속하게 분해가 일어났으며, 그 결과 무게가 감소함을 나타내었다. 연소반응의 $300^{\circ}C{\sim}600^{\circ}C$ 사이에서는 완전연소가 이루어졌으며, 음식물 속의 미생물에 의한 분해로 인하여 pH는 4.26에서 7.6으로 증가하였다. 그리고 poly ester vinyl은 60일 동안의 운전에서 90% 이상의 분해가 이루어졌다.

식물성유지의 에스테르화반응에서 초음파에너지 효과 분석 (The Effect of Ultrasonic Energy on Esterification of Vegetable Oil)

  • 본 연구에서는 식물성 오일의 에스테르화 반응에 초음파에너지를 도입하여 지방산 methyl ester를 제조하였다. 초음파 에너지가 에스테르화 반응에 도입될 경우 반응온도의 가열효과뿐만 아니라 교반효과를 동시에 가져올 수 있어 반응시간 단축 및 지방산 methyl ester 수율의 증가를 가져올 수 있다. 그러나 초음파에너지가 에스테르화 반응에 도입될 때 연속적인 초음파에너지의 조사로 인한 반응온도의 계속적인 상승이 문제점으로 확인되었다. 따라서 본 연구에서는 cooling system을 이용하여 반응온도의 상승을 억제하여 실험을 수행한 결과 기존공정보다 빠른 30 min 만에 93%의 지방산 methyl ester 수율을 확인하였다.
