Study on Clinical Effect of Low Level Laser Therapy by Live Blood Analysis (생혈액분석을 통한 저용량 He-Ne 레이저 유침치료에 관한 임상적 연구)

  • Objective : The purpose of this study was to investigate the effect of Intravascular Laser Irradiation of Blood(ILIB) by the live blood analysis. Methods : We had analysed the changing forms of the live blood samples with Ultra Darkfield Microscope before and after Intravascular Laser Irradiation of Blood. Results and Conclusion : 1. Nomal Erythrocytes were increased significantly after ILIB by the live blood analysis. 2. Poikilocytes were decreased significantly after ILIB by the live blood analysis. 3. Target Cells were decreased significantly after ILIB by the live blood analysis. 4. Somatid was improved significantly after ILIB by the live blood analysis. 5. Erythrocyte Aggregtion was improved significantly after ILIB by the live blood analysis.

A Study on the Morphogenesis of Human Fetal Hepatic Tissue (사람태아 간조직의 형태형성에 관한 연구)

  • Hemopoiesis and morphogenesis of the human fetal liver through from 10 to 32 weeks of gestation were investigated by light and electron microscopy. The results obtained were as follows. Hemopoiesis of fetal liver tissue was found from 10 to 32 weeks of gestation, but the hemopoiesis was decreased at 32 weeks of gestation. At the 32 weeks of gestation, matured erythrocytes were observed in the sinusoid, and formation of liver cell cord and portal triad were established. Differentiation of hepatic cell was characterized by the increase of amount of cell organelles within cytoplasm, decrease of hemopoietic cell, morphological change of nuclear envelope from folding form to round form during the developmental period. These results suggest that human fetal liver plays a hematopoietic function until bone marrow and spleen play their function, but morphology of liver at 32 weeks of gestation was differed with structure observed in liver of adult.

  • Gilbert syndrome is the most common inherited disorder of bilirubin glucuronidation. It is characterized by intermittent episodes of jaundice in the absence of hepatocellular disease or hemolysis. Hereditary spherocytosis is the most common inherited hemolytic anemia and is characterized by spherical, osmotically fragile erythrocytes that are selectively trapped by the spleen. The patients have variable degrees of anemia, jaundice, and splenomegaly. Hereditary spherocytosis usually leads to mild-to-moderate elevation of serum bilirubin levels. Severe hyperbilirubinemia compared with the degree of hemolysis should be lead to suspicion of additional clinical conditions such as Gilbert syndrome or thalassemia. We present the case of a 12-year-old boy with extreme jaundice and nausea. The diagnosis of hereditary spherocytosis was confirmed by osmotic fragility test results and that of Gilbert syndrome by genetic analysis findings.

Studies on Lectins From Marine Shells (V) - Isolation and Purification of Letin from Tapes philippinarum. (해양 패류 렉틴성분 연구 (V) - 반지락조개의 렉틴성분 분리정제에 관한 연구)

  • The result of the screening of lectins on 28 species of marine shell fishes(mollusks) showed that 10 species (Tapes philippinarum, Cyclosunetta menstrualis, Neptunea arthritica cumingii, Omphalius pfeifferi carpenteri, Chlorostoma argyrostoma turbinatum, Chiorostoma argyrostoma lischkei, Semisulcosira corea, Neptunea polycosta, Babylonica japonica, Noverita didyma) were present hemagglutinating properties to human A, B, and O group, and animal blood erythrocytes. A new lectin from Tapes philippinarum. was isolated and partially characterized. The lectin was purified by (NH$_4$)$_2$SO$_4$ precipitation and ion exchange chromatography on DE 53 column. Six fractions were obtained from DE 53 column by salt gradient elution but only 0.3M, and 0.4M NaCl fraction had strong lectin activity. On its 0.3M NaCl fraction, purity was identified by polyacrylamide gel electrophoresis. This lectin was inhibited by N-acetyl-D-galactosamine. It seems that two kinds of lectin react as antigen by immunochemical studies.

  • The mutagenic potential of rHu-EPO was evaluated using the short-term genotoxicity tests including Ames, chromosome aberration and micronuclei tests. In Salmonella typhimurium assay, rHu-EPO did not show any mutagenic response in the absence or presence of S9 mix with TA98, TA100, TA1535, and TA1537. In chromosome aberration test, rHu-EPO did not show any significant effect on Chinese Hamster Ovary(CHO) cells compared with control. In micronucleus test using male ICR mice, a dose-dependence increase in the frequency of micronucleuted polychromatic erythrocytes(MNPCEs) was observed in bone marrow cells treated with rHu-EPO. However, it was related to the secondary effect of rHu-EPO and the number of MNPCEs was equal to spontaneous frequency. These results indicate that rHu-EPO does not show any positive response in short-term genotoxicity assays.

  • Single-cell gel electrophoresis (comet assay) was used to detect DNA single strand break in blood cells from several marine fish species. Three fish species were collected from Georgia coastal area. Mummichog, Fundulus heteroclitus showed higher DNA damage than sea bass, Lateolabrax japonicus and trout, Oncorhynchus masou masou under the same experimental conditions. Mummichogs had more alkaline-labile sites on their DNA than other fish species. The comet assay with mummichog blood cells at pH 12.5 showed a dose-response curve with the increasing concentrations of hydrogen peroxide. While the isolated leucocytes showed no increase of DNA damage after in vitro exposure to 2-methyl-1,4-naphthoquinone (MNQ), erythrocytes showed dose-dependent DNA damage. These results indicate that the comet assay can be applied successfully as a bioassay using erythrocyte for environmental monitoring.

Genotoxicity Study of AS6, a Triterpenoid Derivatives

  • To assess the genotoxicity of AS6, several classical toxicological tests were performed. In Ames test, AS6 did not show any transformation of revertant with or without S-9 metabolic activating system, indicating the lack of mutagenic effect of the compound. To assess clastogenic effect, in vivo micronucleus and in vitro chromosomal aberration assays were performed using male ICR mice and Chinese hamster lung (CHL) fibroblast cells, respectively. Chromosomal aberration was not induced regardless of the presence of S-9 metabolic activating system. In addition, AS6 did not cause any increase in the incidence of micronucleated polychromatic erythrocytes at any of the dose levels, suggesting little clastogenicity in vitro or in vivo. Taken together, these results demonstrate that AS-6 has no mutagenic effect in our test system.

  • Biological response modifiers (BRMs) can alter interactions between the immune system and cancer cells to boost, direct, or restore the body's ability to fight disease. Mice with ethylnitrosourea- (ENU) induced leukemia were here used to monitor the therapeutic efficacy of lipopolysaccaride (LPS), Bacillus Calmette Guerin (BCG) and sheep erythrocytes (SRBC). Flow cytometry based CD34+ positivity analysis, clonogenicity, proliferation and ultrastructure studies using scanning electron microscopy (SEM) of stem cells in ENU induced animals with and without BRMs treatment were performed. BRMs improved the stem-stromal relationship structurally and functionally and might have potential for use as an adjunct in human stem cell therapy.

  • The effects of kanendomycin on the potassium permeability in the rabbit erythrocyte membrane are investigated and the results are summarized as follows. 1. Kanendomycin causes the efflux of $K^+\;and\;influx\;of\;Na^+$ across the rabbit erythrocyte membrane. 2. Osmotic resistance of kanendomycin treated erythrocytes is diminished. This diminution of osmotic resistance is more pronounced by increasing concentration of kanendomycin and longer incubation time. But osmotic resistance is rather increased in the presence of lower concentration of kanendomycin. 3. Cysteine and glutathione have little influence on $K^+$ efflux induced by kanendomycin. 4. EDTA has no effect on the increase in $K^+$ outflux by kanendomycin while PCMB augments slightly on $K^+$ outflux. 5. Kanendomycin inhibits $Ca^{++}$ binding competitively to rabbit erythrocyte membrane fragments.

Moyamoya-Like Vasculopathy in Neurosarcoidosis

  • A 31-year-old man presented with dull headache and memory disturbance lasting for one week. Computed tomographic scans revealed acute hydrocephalus. The cerebrospinal fluid contained 53 leukocytes/$mm^3$, with a mononuclear preponderance and no erythrocytes. Magnetic resonance imaging revealed hydrocephalus and leptomeningeal enhancement. Magnetic resonance angiography and digital subtraction angiography showed supraclinoid occlusion of the right internal carotid artery, which resembled unilateral moyamoya disease. Neuroendoscopic biopsy of a lesion in the septum pellucidum revealed noncaseating granulomas, which was consistent with sarcoidosis. The patient was successfully managed with intravenous methylprednisolone and ventriculoperitoneal shunting. To our knowledge, this is the first case of moyamoya-like vasculopathy associated with neurosarcoidosis.
