Lernergrammatik fur den DaF-Unterricht in Korea (독일어 교육을 위한 학습자 문법)

  • DaF-Lehrende in Korea erfahren oft, dass koreanische DaF-Lernende $\"{u}ber$ ein gutes Regelwissen im Deutschen $verf\"{u}gen$, aber oft nur weinh verstehen und sprechen $k\"{o}nnen$. Das bedeutet, dass das Auswendiglernen von grammatischen Regeln und ihr tatsachliches Anwenden zweierlei sind. Wenn Grammatikkompetenz mit der fremdsprachlichen Kompetenz nicht gieichzusetzen ist, stellt sich die Frage, wie den Lemern durch die Grammatik ein $selbst\"{a}ndiges$ Erlernen von erammatischen Eigenschaften bis hin zur freien Anwendung des Gelernten in den Kommunikationssituationen $erm\"{o}glicht$ werden kann. $F\"{u}r$ die Antwort auf die Frage werden im vorliegenden Artikel Lornergrarnmatik und ihre didaktischen Prinzipien vorgestelit. In der Lornergrarnmatik handelt es sich um die Beschreibung des Regelsystems einer Sprache $f\"{u}r$ Unterrichtszwecke. Das ist mehr als eine Sprachbeschreibung. Sie $ber\"{u}cksichtigt$ dabei den Sanzen Lern- und Lehrverlauf von Regeln und auch die didaktiche Frage, wie man ein Reselsystem aufteilen, darstellen und im Unterricht $pr\"{a}sentieren$ kann

Development of a Recombinant Streptomyces griseus with sprA and sprB Genes for Proteolytic Enzyme Production (Streptomyces griseus IFO13350 유래 sprA 및 sprB 유전자를 이용한 Pretense 생산균주 개발)

  • Pronase, a protease produced for commercial purpose by Streptomyces griseus, was composed of serine protease, alkaline protease, aminopeptidase and carboxypeptidase complex, and it has been widely used as anti-inflammatory drugs for human therapy. In this study, we developed a new integration vector, pHJ101 derived from pSET152, containing strong promoter, ermE, to overexpress a certain protease gene. Specific PCR primers for cloning of sprA (a gene for S. griseus protease A) and sprB (a gene for S. griseus protease B) genes were designed from the basis of nucleotide sequence in databases and amplified by PCR. Plasmid pHJ201 and pHJ202 were constructed by inserting of amplified each gene in a vector pHJ101. S. griseus HA and S. griseus HB were respectively obtained by conjugal process of a parent strain, S. griseus IFO 13350 with the recombinant Escherichia coli harboring plasmid pHJ201 or pHJ202. When protease activity was measured in flask cultivation, produced protease levels of S. griseus HA and S. griseus HB increased about 5.3 times and 5 times, respectively, more than that of parent strain. And, the constructed integrating plasmid pHJ101 was applicable for overexpression of a certain gene in Streptomyces sp.

Effects of the Essential Oil of Nelumbo nucifera Flower on Glioma Cells (련화향(蓮花香) 정유액이 glioma cell에 미치는 효과)

  • Objective : Herb medicines are potential sources of useful edible and medicinal plants. They are used as a drug because of their various biological activities such as immunomodulatory, antiviral, and antitumor functions. Nelumbo nucifera have been applied in Chinese herbal prescriptions to improve tissue inflammation. However, it has not been elucidated on the effect of the flower of Nelumbo nucifera in cells. Method : In the present study, to examine the effect of that on glioma cells, U87, the essential oil was extracted from the flower of Nelumbo nucifera (NN essential oil). U87 cells were exposed to different concentrations of 2-40 ug/ml of NN essential oil in ethanol. Cell viability was measured by MTT assay at 24 h. To find out the intracellular target signal molecule(s) for this antiproliferative activity of NN essential oil, phosphorylation of Akt, ERM, MAPK or p38 proteins were examined by Western blot analysis. To study long term effect of NN essential oil in U87 cells, the image of cells treated with NN essential oil for 4 days were obtained. Results and Conclusion : NN essential oil was shown to exhibit antitumor activity in glioma cells, at a broad range of concentrations of 10-40 ug/ml. The phosphorylation of Akt and Endoplasmic Reticulum Matrix (ERM) proteins which known to be involved in the cell death, were gradually decreased to 2 hours after addition 20 ug/ml of NN essential oil. However, the phosphorylation of mitogen-activated protein (MAPK) and p38 was found to increase in NN essential oil treated cells. NN essential oil treated cells showed decreased glioma cell number. These results provide a possible NN essential oil-induced inhibitory signal for tumor cell proliferation that is initiated by the decrease in Akt activity. Moreover, it is likely that the activation of p38 is required for the NN essential oil-induced inhibition of tumor proliferation.

Analysis of Genotype and Phenotype of Erythromycin Resistance in Enterococci spp. Isolated from Raw Milk Samples (원유시료에서 분리한 장구균의 에리스로마이신 내성 유전자형 및 표현형 분석)

  • The aim of this study was to investigate the erythromycin resistance patterns of Enterococci sp. present in cow milk. A total 110 erythromycin resistant Enterococci were isolated from milk samples; E. faecalis (n=101), E. avium (n=7), and E. durans (n=2). The minimum inhibitory concentration of erythromycin against 110 Enterococci were determined. The 66.3% of Entercocci (n=73) showed high level resistance (${\geq}64$ mg/ml). Among 110 isolates, 86.3% (n=95) showed $cMLS_B$ phenotype and 13.6% (n=15) showed $iMLS_B$ The aim of this study was to investigate the erythromycin resistance patterns of Enterococci sp. present in cow milk. Distribution Characteristics of Polycyclic Aromatic Hydrocarbons in the Sediments of Kwangyang Bay in Korea (광양만 연안 퇴적토 중의 다환방향족탄화수소류의 분포특성)

  • The concentrations of 16 priority PAHs (US EPA standard) were analyzed in the surface sediments obtained from 23 sampling sites near Kwangyang Bay in Korea. There was a local variability in the total PAHs ranged from 0.01 to 171.39 mg/kg, with a mean value of $8.13{\pm}24.8mg/kg$. The major pollution sources of PAHs near Kwanyang Bay were Taeindo, Sueo stream and Wallae stream, whose concentrations were 114.81, 38.37 mg/kg and 19.05 mg/kg, respectively. It showed that PAHs concentrations were increased with the decrease of particle size and with the increase of organic carbon contents in three fractioned sediments. From the analysis of PAHs source using LMW/HMW, Phe/Ant, and Fla/Pyr, pyrolysis by-products were mostly showed in Kwangyang Bay and some place showed the mixure of pyrolysis by-products, and crude oil by-products. Besides, the toxic effects assessment on benthic ecosystem for three major pollution sources showed that the PAHs concentration of Taindo which was mainly accumulated with carcinogenic PAHs exceeds ERM value and the PAHs of Sueo and Wallae streams are the degree of ERL value.

Heavy Metal Contamination in Surface Sediments from Masan and Jinhae Bay, Southeast Coast of Korea (남해 동부해역 임해공단 연안퇴적물의 중금속 오염: 마산만 및 진해만)

  • Concentrations of selected heavy metals (Al, Fe, Mn, Cr, Cu, Ni, Zn, Pb, As and Cd) in surface sediments from 96 sites in Masan and Jinhae Bay were studied in order to understand metal contamination. Results show that the surface sediments were mainly enriched by Cu (18-294 ppm), Zn (67-568 ppm), Pb (10-120 ppm) and Cd (0.2-3.5 ppm). The coastal zone of Masan Bay was significantly more contaminated than the non-coastal zone, and spatial distribution pattern suggested additional sources of heavy metal input in the coastal area. The enrichment ratio and geoaccumulation index ($I_{geo}$) have been calculated and the relative contamination levels assessed in the study area. The enrichment ratios of Cu, Zn, Pb and Cd in Masan Bay have been observed to be relatively high. $I_{geo}$ results reveal that the study area is not contaminated with respect to Fe, Mn, Cr and Ni; moderately to strongly contaminated with Cu, Zn and Pb; and strongly to strong contaminated with Cd. The high contents of Cu, Zn, Pb and Cd in the study area result from anthropogenic activities in the catchment area. Based on the eight different sediment quality guideline values from USA (ERL, ERM), Canada (TEL, PEL), Australia/New Zealand (ISQG-high, ISQG-low) and Hong Kong (ISQV-low, ISQV-high), sediment quality of Masan and Jinhae Bay was also assessed and characterized.

Characterization of S. aureus Showing MLS Resistance with Foggy D Shape (fMLS) (안개 D형 저지원을 갖는 MLS 내성 황색 포도상 구균의 분포와 내성 기전)

  • A new type of macrolides, lincosamides and streptogramin B antibiotics (MLS) resistance, showing the characteristic phenotype growing within the inhibition zone area around the clindamycin disk, was identified among clinically isolated Staphylococcus aureus. We named the phenotype as MLS resistance with foggy-D shape (fMLS). The average frequency of MLS isolates was 9%. All of fMLS isolates have only erm(A) for the resistance determinant. The growth pattern of the challenged MLS isolate looks intermediate phase between the patterns of inducible MLS resistance and constitutive MLS resistance.

The Study of MLS-Resistant Gram Positive Cocci Isolated In a Korean Hospital (임상 분리 그람 양성 구균에 대한 MLS계 항생물질의 내성)

  • A total of 398 strains of Staphylococcus aureus (99), coagulase-negative staphylococci (CNS) (99), enterococci (100), Streptococcus pyogenes (18) and Streptococcus agalatiae (82) were in Korean tertiary hospital from Dec. 2004 to Mar, 2005. When minimal inhibitory concentrations (MIC), phenotypes and genotypes were determined, 70% of S. aureus, 65.7% of CNS, 78% of enterococci and 37.8% of S. agalatiae were resistant to erythromycin and 95.7% of erythromycin-resistant (EMR) S. aureus, 92.3% of EMR CNS and all of EMR enterococci and S. agalatiae had erm of the methylase gene or msr(A) of the efflux gene.

Enhanced Clavulanic Acid Production in Streptomyces clavuligerus NRRL3585 by Overexpression of Regulatory Genes

  • We constructed four recombinant plasm ids to enhance the production of clavulanic acid (CA) in Streptomyces clavuligerus NRRL3585: (1) pIBRHL1, which includes ccaR, a pathway-specific regulatory gene involved in cephamycin C and CA biosynthesis; (2) pIBRHL2, containing claR, again a regulatory gene, which controls the late steps of CA biosynthesis; (3) pGIBR containing afsR-p, a global regulatory gene from Streptomyces peucetius; and (4) pKS, which harbors all of the genes (ccaR/ claR/ afsR-p). The plasmids were expressed in S. clavuligerus NRRL3585 along with the $ermE^*$ promoter. All of them enhanced the production of CA; 2.5-fold overproduction for pIBRHL1, 1.5-fold for pIBRHL2, 1.6-fold for pGIBR, and 1.5-fold for pKS compared to the wild type.

Characterization of Dephosphocoenzyme A Kinase from Streptomyces peucetius ATCC27952, and Its Application for Doxorubicin Overproduction

  • Dephosphocoenzyme A (CoaE) catalyzes the last step in the biosynthesis of the cofactor coenzyme A. In this study, we report the identification and application of CoaE from Stretomyces peucetius ATCC27952. After expression of coaE, the protein was found to have a molecular mass of 28.6 kDa. Purification of the His-tagged fused CoaE protein was done by immobilized metal-affinity chromatography, and then in vitro enzymatic coupling assay was performed. The increasing NADH consumption with time shed light on the phosphorylating activity of CoaE. Furthermore, the overexpression of coaA and coaE independently under the $ermE^*$ promoter in the doxorubicin -producing wild type strain, resulted in 1.4- and 1.5-fold enhancements in doxorubicin production, respectively. In addition, the overexpression of both genes together showed a 2.1-fold increase in doxorubicin production. These results established a positive role for secondary metabolite production from Streptomyces peucetius.
