치주인대섬유모세포들은 완전탈구 된 치아의 성공적인 재식을 위한 중요한 요소이다. 따라서, 외상으로 인해 완전탈구된 치아의 보존을 위한 보관액을 선택하는 것이 중요하다. 성장인자들과 호르몬들은 치주인대섬유모세포들의 생존을 위한 치료적 제제로 고려되고 있다. Epidermal growth factor(EGF)는 다른 조직에서 재생과 상처 치유 과정의 중요한 역할인자로 대두되고 있다. 따라서, 완전탈구 된 치아를 위한 치료적 적용을 위해 EGF의 세포 증식에 미치는 영향을 평가하였다. 또한 EGF와 tri-iodothyronine(T3)의 혼합액, EGF와 Heparin-binding epidermal growth factor-like growth factor(HB-EGF)의 혼합액이 세포 증식에 미치는 상승 효과를 평가하였다. 치주인대섬유모세포의 세포증식은 EGF 농도가 증가함에 따라 증가하였고, 10 ng/ml 농도에서 최대 세포증식을 보였다. EGF는 상처치유분석에서 상처 치유촉진과 이동성을 보여주었다. EGF에 T3와 HB-EGF를 첨가한 혼합액에서 배양한 세포는 EGF만 처리한 경우보다 세포 증식이 상승되었다. EGF와 T3 혼합액의 상승효과 기전을 유추하기 위해서 RT-PCR로 EGF 수용기의 발현을 확인하였고, T3가 EGF 수용기 발현을 증가시켰음을 확인하였다. 따라서 EGF와, EGF와 T3, EGF와 HB-EGF의 혼합액은 완전탈구된 치아의 치료에 있어 유용한 선택이 될 것이다.
Stimulatory effects of epidermal growth factor (EGF) and transforming growth $factor-{\alpha}$($TGF-{\alpha}$) on the growth of squamous cancer cell lines established from human oral cancer tissue with moderate differentiation were studied in vitro. After culturing in serum-free media for 24 hours, growth factors-EGF only, $TGF-{\alpha}$ only and EGF, $TGF-{\alpha}$ together-were added to the media and numbers of cells were analyzed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay and compared with the control at 96, 144 hours. Each of EGF and $TGF-{\alpha}$ showed statistically significant stimulatory effects on the growth of cells respectively. Dose-dependent relationship of the stimulatory effects were not clearly demonstrated. The effects of EGF were higher than those of $TGF-{\alpha}$ and combinative administration showed higher effects than those of single uses. In conclusion, EGF may play an important and major role in differentiation and growth of human oral squamous cancer cells. $TGF-{\alpha}$, produced from cells activated by EGF, also can stimulate the cell growth and could be an alternative ligand for EGF receptor.
개화시기의 수술로부터 수집한 백합화분을 aluminum oxide 미세입자와 섞어 교반에 의해 상해를 발생시켰다. 이어서 이들 화분은 signal peptide-fused epidermal growth factor (EGF) DNA를 지니는 Agrobacterium 세포로 vacuum infiltration을 시켰으며 24 hr 화분신장을 통한 배양을 실시하였다. 이들 신장 화분에서의 EGF mRNA 및 단백질 발현은 성공적으로 확인되었으며 이는 cDNA blot hybridization 및 immuno-blotting의 분석결과이다.
본 연구는 성장인자 EGF, IGF-1, EGF+IGF-1 이 소 난포란의 체외성숙에 미치는 영향을 규명하기 위하여 실시하였다. 소 난포란의 체외성숙 시 EGF를 농도별 (10, 50 및 100ng/m1)로 첨가하여 실험한 결과 통계학적 유의성은 인정되지 않았으나 24시간 배양 후 성숙율은 대조군에 비하여 높은 경향을 나타내었다. 소난포란의 체외성숙 시 EGF와 IGF-1을 병용 처리하여 실험한 결과, EGF 또는 IGF-1 단독처리 군에 비하여 병용처리군이 유의적으로 높은 체외성숙율을 나타내었다(p〈0.05). 따라서 소난포란의 체외성숙율은 EGF와 IGF-1에 의하여 영향을 받으며 특히 EGF와 IGF-1의 상호작용에 의하여 더 유효한 효과를 나타내는 것으로 사료된다.
Using a methylotrophic yeast Hansenula polymorpha, a heterologous gene expression and secretion system was developed for the production of hEGF(human Epidermal Growth Factor) which has been shown to promote epithelial cell proliferation and to inhibit gastric acid secretion. The hEGF gene was chemically synthesized according to the preferred codon usage in H. polymor- pha and expressed under the control of the strong and inducible methanol oxidase(MOX) promoter. The mating factor $\alpha$ pre-pro leader sequence of Saccharomyces cerevisiae was employed for hEGF to be secreted into the extracellular medium. This expression cassette was stably integrated into the host chromosomal DNA. Mature hEGF was efficiently expressed and secreted into the extracel- lular medium. About 24 mg/l of hEGF was detected in the cuture supernatant of a transformant with pA-EGF3 under the suboptimal culture conditions.
Effects of titrated extract of Centella asiatica (TECA) and epidermal growth factor (EGF) isolated from the urine of pregnant horse on the proliferation of human epidermal keratinocyte in culture were studied. An increase in the number of keratinocyte was observed with the treatment of TECA at the concentration ranges from 1 $\mu\textrm{g}$/mι to 100 $\mu\textrm{g}$/mι. Effects of low molecular weight EGF (LEGF) and high molecular weight EGF (HEGF) on the proliferation of keratinocyte in culture were also studied. The number of keratinocyte in culture was significantly increased with LEGF and HEGF respectively at the concentration of 10 ng/mι. Simultaneous treatment of the keratinocyte with LEGF, HEGF and TECA led to the increased proliferation of keratinocytes resulting 96% of the effect of a positive control, EGF isolated from mouse submaxillary glands.
인유 초유로부터 지방을 제거하여 skim milk을 얻었다. 이것을 ultrafiltration을 한 후 gel filtration을 거쳐 EGF fraction 을 획득하였으며, 이를 SDS-PAGE로 확인하였다. hish performance liquid chromatography(HPLC)에 의해 EGF fraction을 분석한 결과, standard EGF 분석과 비교했을 때 31.545분대와 유사한 31.185분대에 peak가 나타난 것을 인하였다. 분리한 fraction 중 EGF 함유량을 측정하기 위해 immunoassay를 실시한 결과 건조중량 10 mg/mL 중에 약 10ng의 EGF가 포함되어 있는 것으로 측정되었다. SDS- PAGE후 실시한 western blot을 통하여 분리한 분획의 분리도를 확인하였다. 세포성장에 미치는 영향을 살펴본 결과, fibroblast cell인 BALB/3T3의 경우에는 건조중량 1 mg/mL 농도의 fraction 처리후 약 95%의 성장률을 보였으며, epithelial cell 인 IEC-6의 경우에는 약 71%의 성장률을 보였다.

Polymeric nanoparticles composed of polystyrene (PS) as core and poly(methacrylic acid) (PMA) as corona were prepared by the dispersion copolymerization. The potential of the nanoparticles as carriers for recombinant human epidermal growth factor (EGF) was investigated. The nanoparticles showed monodispersity and good water-dispersibility. The loading content of EGF to the nanoparticles was very high due to electrostatic interaction between EGF and nanoparticles. EGF was released as a pseudo-zero order pattern after initial burst effect. The nanoparticles were sufficient for A431 cells proliferation.

Among the various human growth factors, epidermal growth factor (hEGF, consisting of 53 amino acids) has various effects on cell regeneration, stimulation of proliferation, migration of keratinocytes, formation of granulation tissues, and stimulation of fibroblast motility, which are important for wound healing. Owing to their multiple activities, EGFs are used as pharmaceutical and cosmetic agents. However, their low productivity, limited target specificity, and short half-life inhibit their application as therapeutic agents. To overcome these obstacles, we fused the collagen-binding domain (CBD) of Vibrio mimicus metalloprotease to EGF protein. About 18 or 12 amino acids (aa) (of the 33 total amino acids), which were essential for collagen-binding activity, were combined with the N- and C-termini of EGF. We constructed, expressed, and purified EGF (53 aa)-CBD (18 aa), EGF (53 aa)-CBD (12 aa), CBD (18 aa)-EGF (53 aa), and CBD (12 aa)-EGF (53 aa). These purified recombinant proteins increased the numbers of cells in treated specimens compared with non-treated specimens and control hEGF samples. The collagen-binding activities were also evaluated. Furthermore, CBD-hybridized hEGF induced phosphorylation of the EGF receptor. These results suggested that these fusion proteins could be applicable as small therapeutic agents in wound tissue healing.
Human epidermoid carcinoma A431 cells have an extraordinarily large number of epidermal growth factor (EGF) receptors, and their growth is inhibited by EGF, which results in growth arrest at the Gl phase. In order to investigate the EGF-mediated inhibition mechanism, the expression level of DNA topoisomerase (topo) II was analyzed after EGF treatment. As a result, it was shown that EGF treatment lowered the amount of 170 kDa topo II (topo $II{\alpha}$) but not 180 kDa (topo $II{\beta}$). However, the A431 cell variant resistant to EGF was not sensitive to EGF treatment. These results suggest that EGF-induced growth arrest of A431 cells may be closely related to the depletion of topo $II{\alpha}$.
