Bioremediation of hazardous hydrophobic organic compounds, such as polycyclic aromatic hydrocarbons (PAHs), is a major environmental concern due to their toxic and carcinogenic properties. Bue to their low solubility in water, the compounds are microbiologically persistent. This work investigates optimal conditions to enhance the biodegradation of phenanthrene in water and soil-slurry systems. Biodegradation tests were performed with three different types of supplements: glucose as a general carbon source, salicylate as an enzyme inducer, and Triton X-100 as a surfactant. The tests indicate that glucose and Triton X-100 were not very effective to increase biodegradation rate, even though the number of microorganisms are highly increased in the case of glucose addition. Salicylate accelerated biodegradation of phenanthrene, but the addition above optimal concentration inhibited microbial growth. Salicylate is considered to be an attractive alternative for the successful bioremediation of PAH-contaminated soil.
The fiber-rich fractions including enzyme treated insoluble dietary fiber, alcohol insoluble solid, and water insoluble solid were prepared from the peel of soft ripe persimmon, and to evaluate and compare the yields, proximate compositions, monosaccharide profiles, and functional properties. The results showed that uronic acid was the main sugar followed by glucose, which indicated that all insoluble fibers were mainly composed of pectic substances and cellulose. The presence of xylose and fucose indicated the occurrence of hemicellulose. All fiber-rich fractions were exhibited high yield and functional properties. Thus, the peel of ripe persimmon could be used as fiber supplements.
This study was to investigate lipid peroxidation, antioxidant enzyme activity and DNA damage after exercise, and the protective effect of garlic against exercise-induced oxidative stress. Male Sprague-Dawley rats(4 weeks old) were randomly divided into three groups of 6 rats each; control group(Con) without garlic and exercise, Ex group with exercise alone, and Ex-G group with 2% garlic and exercise. For 4 weeks, rats were given diets containing 15% corn oil and 1% cholesterol with or without garlic. The swimming was selected as a model for exercise performance. Rats swam for 40 min a day, for 5 days a week. Group Ex and Ex-G showed significant lowering in body weight gain and fat accumulation compared to control. No significant changes were observed in levels of plasma cholesterol and triglyceride among three groups, demonstrating that exercise and garlic had no effects on changes of blood lipid. This finding of blood lipid seems to be due to higher plant sterol content in corn oil. The DNA tail moment of lymphocytes showed greater tendency in Ex and Ex-G than in control, but garlic supplements failed to suppress DNA damages. Compared to control, Ex had higher plasma TBARS which was lowered to the control's level in Ex-G with 2% garlic supplementation(p<0.05). Ex-G led to a higher hepatic superoxide dismutase(SOD) activity than control and Ex(p<0.05). Activity of hepatic catalase was also increased in Ex-G, while in Ex it was significantly low(p<0.05). It seemed that TBARS levels were related to the activities of SOD and catalase, and that garlic contributed to increasing the enzyme activities and led to decrease of TBARS. These results demonstrate that lipid peroxidation and DNA damage occur as a consequences of oxidative stress after exercise, and that antioxidant defense against oxidative stress could be enhanced by garlic supplementation through the induction of antioxidant enzymes. However, further investigations should be done on the garlic effect on DNA damage.

The current study examined the effect of quercetin supplements on the lipid-lowering and antioxidant metabolism in ethanol-fed rats. The control $group(E_8)$ received ethanol only diet for 8 wks, whereas the other group$(E_8Q_4)$ received a diet including quercetin supplementation(0.05% wt/wt) for 4 wks while on the ethanol diet for 8wks. The hepatic alcohol dehydrogenase activity was significantly higher in the $E_8Q_4$ group than in the $E_8$ group. Supplementation with quercetin significantly elevated the HDL- cholesterol concentration, the HDL-C/total-C ratio, and lowered the atherogenic index(AI) compared to the $E_8$ group. The hepatic triglyceride and cholesterol contents were significantly lowered by the quercetin supplement compared to those of the control group. The hepatic HMG-CoA reductase and ACAT activities of the $E_8Q_4$ group were significantly lower than those of the $E_8$ group. The overall potential for antioxidant defense was significantly enhanced by the quercetin supplement, as indicated by a decrease in plasma and hepatic TBARS levels. The hepatic GSH-Px and G6PD activities were significantly higher in the $E_8Q_4$ group compared to the $E_8$ group. The current results suggest that dietary quercetin leads to the inhibition of HMG-CoA reductase and ACAT, which in turn lowers cholesterol levels and normalizes antioxidant enzyme activities.
This study an analyzes the effects of the P/S ratio of dietary lipids and antioxidant vitamin supplements on serum lipids level and fatty acid profile, the degree of lipid peroxidation, and the antioxidant enzyme activities in the liver of rats treated with 7,12-dimethylbenz($\alpha$) anthracene(DMBA). P/S ratio of dietary lipids was made into 0.5, 1 and 2 by mixing palm oil, soybean oil, sesame oil and perilla oil at 10%(w/w) fat level and n-6/n-3 ratio was fixed to 4. Antioxidant vitamin of $\alpha$-tocopherol or $\beta$-carotene was supplemented in addition to vitamin mixture which was given at 1 % of the standard diet. female Sprague-Dawley strain rats, about 60 days old, were divided into three groups(LP : low P/S ratio(0.5), MP : medium P/S ratio (1.0), HP , high P/S ratio(2.0)) and each group was sub-divided into three groups(S ; standard, T ; tocopherol supplemented, C : carotene supplemented): Two weeks after feeding experimental diets, all groups were treated with a single dose of DMBA(2mg/100g BW) by gastric intubation and fed experimental diet for 9 week. The results were as follows ; 1) Serum total cholesterol(TC) level was not significantly influenced by diet but tended to be lower in HP groups compared to LP and MP groups. Triglyceride level was the highest in LP groups and the lowest in $\alpha$-tocopherol supplemented groups. 2) Thiobarbituric acid reactive substance(TBARS) level, representing lipid peroxidation in hepatic microsome, tended to be increased as the unsaturation of dietary lipids increases. $\alpha$-Tocopherol supplement significantly decreased TBARS level. 3) The activities of superoxide dismutase(SOD) and glutathione peroxidase(GSHPx) in hepatic cytosol showed the tendency to be high with increasing P/S ratio of dietary lipids. SOD activity was not significantly influenced by antioxidant vitamin, but GSHPx activity was significantly increased in $\alpha$-tocopherol supplemented groups. In summary, high polyunsaturated fat diet was effective on reducing the serum level of total cholesterol and triglyceride, while it increased unsaturation and peroxidizability of serum fatty acid. With increasing P/S ratio of dietary lipids, lipid peroxidation was increased in the liver and antioxidant enzyme system was induced to inhibit lipid peroxidation against oxidative damage. $\alpha$-Tocopherol supplement was effective in lowering lipid peoxidation, but $\beta$-carotene supplement did not exhibit antioxidant effect. This trial was conducted to determine the effects of various feed additives on nutrient digestibility, performance and carcass traits of growing-finishing pigs fed diets containing wheat distiller' grains with solubles (WDGS). Seventy-two, individually fed pigs (19.7${\pm}$2.6 kg), were assigned to one of six dietary treatments in a 6${\times}$2 (treatment${\times}$sex) factorial design (N = 12). The control diet was based on wheat and soybean meal while the five experimental diets contained 20% WDGS during the growing period and 12% WDGS during the finishing period. One 20% WDGS diet was unsupplemented while the remaining diets were supplemented with either 0.1% threonine, 5% canola oil, 0.2% enzyme (0.1% Endofeed W containing 1,250 units/g of xylanase and 385 units/g of $\beta$-glucanase and 0.1% Vegpro containing 7,700 HUT/g protease and 75 CMC/g cellulase), or a combination of the three additives at the same levels as those fed separately. The digestibility of dry matter, crude protein and energy were all significantly higher in the control diet than the unsupplemented diet containing 20% WDGS. None of the feed additives improved nutrient digestibility. In addition, none of the additives had any significant effect on gain or feed intake during the growing (19.7 to 43.6) or finishing (43.6 to 114.3 kg) periods or overall (19.7 to 114.3 kg). During the growing period, feed conversion was significantly improved for pigs fed the combination of additives compared with the unsupplemented WDGS diet. During the finishing period and overall, feed conversion was significantly improved for pigs fed 5% canola oil alone or in combination with the other additives. None of the supplements had any effect on carcass traits. These results indicate that WDGS can be successfully used as a partial replacement for soybean meal in diets fed to growingfinishing pigs. However, due to its low energy content, there may be some merit in including high energy ingredients such as canola oil when diets containing WDGS are fed.
The rumen microbial ecosystem is coming to be recognized as a rich alternative source of genes for industrially useful enzymes. Recent advances in biotechnology are enabling development of novel strategies for effective delivery and enhancement of these gene products. One particularly promising avenue for industrial application of rumen enzymes is as feed supplements for nonruminant and ruminant animal diets. Increasing competition in the livestock industry has forced producers to cut costs by adopting new technologies aimed at increasing production efficiency. Cellulases, xylanases, ${\beta}$-glucanases, pectinases, and phytases have been shown to increase the efficiency of feedstuff utilization (e.g., degradation of cellulose, xylan and ${\beta}$-glucan) and to decrease pollutants (e.g., phytic acid). These enzymes enhance the availability of feed components to the animal and eliminate some of their naturally occurring antinutritional effects. In the past, the cost and inconvenience of enzyme production and delivery has hampered widespread application of this promising technology. Over the last decade, however, advances in recombinant DNA technology have significantly improved microbial production systems. Novel strategies for delivery and enhancement of genes and gene products from the rumen include expression of seed proteins, oleosin proteins in canola and transgenic animals secreting digestive enzymes from the pancreas. Thus, the biotechnological framework is in place to achieve substantial improvements in animal production through enzyme supplementation. On the other hand, the rumen ecosystem provides ongoing enrichment and natural selection of microbes adapted to specific conditions, and represents a virtually untapped resource of novel products such as enzymes, detoxificants and antibiotics.

Anaerobic rumen fungi have been regarded as good genetic resources for enzyme production which might be useful for feed supplements, bio-energy production, bio-remediation and other industrial purposes. In this study, an expressed sequence tag (EST) library of the rumen anaerobic fungus Neocallimastix frontalis was constructed and functional genes from the EST library were analyzed to elucidate carbohydrate metabolism of anaerobic fungi. From 10,080 acquired clones, 9,569 clones with average size of 628 bp were selected for analysis. After the assembling process, 1,410 contigs were assembled and 1,369 sequences remained as singletons. 1,192 sequences were matched with proteins in the public data base with known function and 693 of them were matched with proteins isolated from fungi. One hundred and fifty four sequences were classified as genes related with biological process and 328 sequences were classified as genes related with cellular components. Most of the enzymes in the pathway of glucose metabolism were successfully isolated via construction of 10,080 ESTs. Four kinds of hemi-cellulase were isolated such as mannanase, xylose isomerase, xylan esterase, and xylanase. Five $\beta$-glucosidases with at least three different conserved domain structures were isolated. Ten cellulases with at least five different conserved domain structures were isolated. This is the first solid data supporting the expression of a multiple enzyme system in the fungus N. frontalis for polysaccharide hydrolysis.

The purpose of this study was to evaluate the antioxidant and anticholinesterase activities of yellow and white bitter yams from South Western Nigeria using methanolic extraction and simulated gastrointestinal digestion models. The phenolic compounds in the bitter yam varieties were evaluated by high performance liquid chromatography with a diode array detector (HPLC-DAD). The total phenolic content of the bitter yams was measured by the Folin-Ciocalteu method, reductive potential by assessing the ability of the bitter yam to reduce $FeCl_3$ solution, and the antioxidant activities were determined by the 2,2-diphenyl-1-picrylhydrazyl radical ($DPPH^{\cdot}$) scavenging activity, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical cation ($ABTS^{{\cdot}+}$) scavenging activity, nitric oxide radical ($NO^{\cdot}$) scavenging ability, hydroxyl radical scavenging ability, and ability to inhibit $Fe^{2+}$-induced lipid oxidation. The HPLC-DAD analysis revealed the presence of some phenolic compounds in the studied bitter yam varieties, with varying degree of quantitative changes after cooking. The antioxidant indices (total phenolic content, total flavonoid content, reducing power, $DPPH^{\cdot}$ scavenging activity, $ABTS^{{\cdot}+}$ scavenging activity, and $NO^{\cdot}$ scavenging activity) were higher in the simulated gastrointestinal digestion model compared to the methanolic extract, with the in vitro digested cooked white bitter yam ranking higher. Similarly, the in vitro digested yams had a higher inhibitory action against lipid oxidation compared to the methanolic extracts, with the cooked white bitter yam ranking high. The methanolic extracts and in vitro enzyme digests showed no acetylcholinesterase inhibitory abilities, while methanolic extracts and the in vitro enzyme digest displayed some level of butyrylcholinesterase inhibitory activities. Therefore the studied bitter yams could be considered as possible health supplements.
In this study, enzyme (thermoase) hydrolysis was applied to the porcine blood order to increase the iron content and solubility. It was confirmed that content of iron was increase up to 158.11 mg/100 g porcine powders after 0.2% thermoase treatment at $60^{\circ}C$ during 4 hr. The solubility of porcine blood powders was higher than other enzyme (various protease), temperature, reaction time. This optimized conditions were also worked to the in vitro iron bioavailability rate increasement, the bioavailability of hydolyzed porcine powders was 3-fold higher than that of an iron supplement on the market. These results indicate the possibility of porcine blood powder in iron supplements market as natural material. Also utilizing of reduced porcine blood will be possible to improve environmental issues.
