Aptamer Based SPREETA Sensor for the Detection of Porphyromonas gingivalis G-Protein

  • We have developed an aptamer that specifically binds to Porphyromonas gingivalis to reduce the cellular damage caused by P. gingivalis infection and applied it as a biosensor. P. gingivalis is one of the major pathogens causing destructive periodontal disease among the periodontal microorganisms constituting complex biofilms. Porphyromonas gingivalis G-protein (PGP) known to play an important role in the transmission of germs was used as a target protein for the screening of aptamer. The aptamer that has binds to the G-protein of P. gingivalis, was screened and developed through the Systemic Evolution of Ligands by Exponential Energy (SELEX) method. Modified-Western blot analysis was performed with the aptamer which consisted of 38 single-stranded DNA to confirm the selectivity. ELONA (enzyme linked oligonucleotide assay) used to confirm that the aptamer was sensitive to PGP even at low concentration of 1 ㎍/ml. For the rapid detection of P. gingivalis, we constructed a surface plasmon resonance biosensor with SPREETA using the PGP aptamer. It was confirmed that PGP could be detected as low concentration as at 0.1 pM, which is the minimum concentration of aptamer sensor within 5 min. Based on these results, we have constructed a SPREETA biosensor based on aptamer that can bind to P. gingivalis G-protein. It can be used as an infection diagnosis system to rapidly diagnose and analyze oral diseases caused by P. gingivalis.

Comparison of Enzymatic Activity and Cleavage Characteristics of Trypsin Immobilized by Covalent Conjugation and Affinity Interaction (공유결합과 친화력결합에 의한 고정화 Trypsin의 효소역가와 절단특성 비교)

  • We investigated the effects of immobilization chemistry on the yield of immobilization and the bioactivity of the immobilized enzymes. Trypsin as a model protein and macroporous polymer beads(Toyopearl AF 650M, Tosho Co., Japan) was used as a model matrix. Four methods were used to immobilize trypsin; covalent conjugation by reductive amination(at pH 10.0 and pH 4.0) and affinity interaction via streptavidin-biotin, and double-affinity interaction via biotin-streptavidin-biotin system. The covalent conjugation immobilized $3{\sim}4$ mg/ml-gel, ca. 3-fold higher than the affinity method. However, the specific activity of the covalently(pH 10.0) and affinity-immobilized trypsin(via streptavidin-biotin) are ca. 37% and 50%, respectively, of that of the soluble enzyme(on the low-molecular-weight BAPNA substrate). When the molecular size of a substrate increased, the affinity-immobilized trypsin showed higher clavage activity on insulin and BSA. This result seemed to indicate the streptavidin-biotin system allowed more steric flexibility of the immobilized trypsin in its interaction with a substrate molecule. To confirm this, we studied the molecular flexibility of immobilized trypsin using quartz crystal microbalance-dissipation. Self-assembled monolayers were formed on the Q-sensor surface by aminoalkanethiols, and gultaraldehyde was attached to the SAMs. Trypsin was immobilized in two ways: reductive amination(at pH 10.0) and the streptavidin-biotin system. The dissipation shift of the affinity-immobilized trypsin was $0.8{\times}10^{-6}$, whereas that of the covalently attached enzyme was almost zero. This result confirmed that the streptavidin-biotin system allowed higher molecular flexibility. These results suggested that the bioactivity of the immobilized enzyme be strongly dependent on its molecular flexibility.

  • 바이오센서는 효소(enzyme), 생분자(biomolecule), 항체(antibody), 세포(cell) 등의 biological agent를 인지 물질(recognition material)로 하여 측정하고자하는 분석 대상(analyte)과 높은 선택성으로 반응을 일으키게 하여 그 결과를 기존의 물리, 화학센서로 감지 해내는 방식이므로 기존의 의료용 화학센서를 대체하는 추세이다. 바이오센서가 기존의 센서와 구별되는 점은 생물질의 선택적인 반응 및 결합을 이용하는 것이므로 바이오센서의 실용화에 있어서 가장 중요한 것은 생체 반응 물질의 고정화 기술과 고정화막의 선택이라 할 수 있다. 일정전압법을 이용한 요소센서는 많이 연구되어 오고 있으나 낮은 농도에서의 감도저하에 따른 단점으로 상용화에 이르지 못하고 있다. 본 논문은 요소센서의 이용하기 위한 고정화막으로 3-mercaptopropionic acid 자기조립 단인층의 전기화학적 특성을 고창하였다. 자기조립 단일층은 직접적인 전자전달로 인하여 낮은 요소 농도에서 뛰어난 강도와 빠른 반응 시간을 보였으며, 특히 다공질 실라콘을 기질로 사용한 경우 평면 전극 보다 약 3배의 감도 증가 효과를 가져왔다. 자기조립 단일층의 표면 분석은 X-ray photoelectron spectroscopy(XPS)를 이용하였다.

Efficient Biotinylation of Nitrocellulose Membrane for Immuno-Filtration Capture Assay

  • We investigated biotinylation of nitrocellulose membrane for immuno-filtration capture assay. In order to enhance the efficiency of biotinylation, nitrocellulose membranes were pretreated with several chemicals for the purpose of suitable protein absorption through surface modification. As a signal generating enzyme, urease was used and the concentration of avidin was optimized for the efficient binding kinetics between urease-biotin in liquid phase and biotinylated membrane in solid phase. For effective biotinylation, bovine serum albumin-biotin complexes could be immobilized at a concentration of $370\;{\mu}g$/stick ($4.4\;cm^2$). Among tested chemicals, polylysine (0.25%) showed a significant effect in biotinylation. Polylysine is thought to enhance surface area by extending unbound residues into solution. Time of treatment over 30 min and higher molecular weight of polylysines (58,100 dalton) showed positive effect on the enhancement of biotinylation. The result from this study may be useful for developing a new biosensor and other biofunctional membranes for examining molecular recognition.

Chitosan-Cu-salen/Carbon Nano-Composite Based Electrode for the Enzyme-less Electrochemical Sensing of Hydrogen Peroxide

  • Cu-Salen complex was prepared and attached into chitosan (Cs) polymer backbone. Nanocomposite of the synthesized polymer was prepared with functionalized carbon nano-particles (Cs-Cu-sal/C) to modify the electrode surface. The surface morphology of (Cs-Cu-sal/C) nanocomposite film showed a homogeneous distribution of carbon nanoparticles within the polymeric matrix. The cyclic voltammogram of the modified electrode exhibited a redox behavior at -0.1 V vs. Ag/AgCl (3 M KCl) in 0.1 M PB (pH 7) and showed an excellent hydrogen peroxide reduction activity. The Cs-Cu-sal/C electrode displays a linear response from $5{\times}10^{-6}$ to $5{\times}10^{-4}M$, with a correlation coefficient of 0.993 and detection limit of $0.9{\mu}M$ (at S/N = 3). The sensitivity of the electrode was found to be $0.356{\mu}A\;{\mu}M^{-1}\;cm^{-2}$.

Assembly of Laccase over Platinum Oxide Surface and Application as an Amperometric Biosensor

  • Laccase could be successfully assembled on an amine-derivatized platinum electrode by glutaraldehyde coupling. The enzyme layer formed on the surface does not communicate electron directly with the electrode, but the enzymatic activity of the surf ace could be followed by electrochemical detection of enzymatically oxidized products. The well-known laccase substrates, ABTS (2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid)) and PPD (p-phenylenediamine) were used. ABTS can be detected down to 0.5 ${\mu}M$ with linear response up to 15 ${\mu}M$ and current sensitivity of 75 nA/ ${\mu}M.$ PPD showed better response with detection limit of 0.05 ${\mu}M$, linear response up to 20 ${\mu}M$, and current sensitivity of 340 nA/ ${\mu}M$ with the same electrode. The sensor responses fit well to the Michaelis-Menten equation and apparent $K_M$ values are 0.16 mM for ABTS and 0.055 mM for PPD, which show the enzymatic reaction is the rate-determining step. The laccase electrode we developed is very stable and more than 80% of initial activity was still maintained after 2 months of uses.

  • Rice is one of the most important cereals in the world. Japanese people use about 9 million tons of rice per you. We use rice for cooked rice as staple foods and for processing, such as rice wine (sake), rice crackers and miso fermentation, etc. Palatability, eating quality, of rice is evaluated by the sensory test and various kinds of physicochemical measurements. Japanese National Food Agency started the storage of 1.5 million tones of rice in 1996. We carried out the storage test using high quality rices since 1995 until 1996. As indices for the quality deteriorations of rice grains during the storage, germination ratio, enzyme activities, fat acidity, physical properties of cooked rice were clarified to be useful. We applied colorimetric method for the measurements of fat acidities in the place of titration method. Processing suitabilities of rice differ depending on the products. Low amylose rice is more suitable for soft rice crackers and high amylose rice is preferred more for rice noodle. Pre-cooked rice products, such as frozen cooked rice, retort-pouched rice and aseptic rice, are increasing recently in Japan. In addition to above-mentioned physico-chemical tests, NIR spectroscopy,“Midometer”and“Taste sensor”are novel and useful to evaluate eating quality and processing suitabolities. Recently, rice wholesalers and retailers have been obligated to display the name of cultivar, location of cultivation and the year of production of rice grains which they sell by the Japanese Agricultural Standard Law (JAS). In order to detect the dishonest labeling of rice cultivars, we developed new cultivar identification method based on DNA polymorphism.

Concurrent Electrocatalysis and Sensing of Hydrazine and Sulfite and Nitrite Ions using Electrodeposited Gold Nanostructure-Modified Electrode

  • Concurrent electrocatalysis and sensing of hydrazine, sulfite ions, and nitrite ions in a mixture were studied using electrodes modified by electrodeposited Au nanostructures (NSs). The ${\beta}$-cyclodextrin-mixed silicate sol-gel composite was drop-casted on the electrode surface and nucleation guided by ${\beta}$-cyclodextrin occurred, followed by the electrodeposition of Au NSs. The additive, ${\beta}$-cyclodextrin, played an evident role as a structure-directing agent; thus, small raspberry-like Au NSs were obtained. The modified electrodes were characterized by surface characterization techniques and electrochemical methods. The Au NSs-modified electrodes effciently electrocatalyzed the oxidation of toxic molecules such as hydrazine and sulfite and nitrite ions even in the absence of any other electron transfer mediator or enzyme immobilization. Well-resolved oxidation peaks along with decreased overpotentials were noticed during the electrooxidation process. The fabricated Au nanostructured electrode clearly distinguished the electrooxidation peaks of each of the three analytes from their mixture.

The senstivity characteristics of cholesterol sensor by immobilization methods of the enzyme (효소 고정화 방법에 따른 콜레스테롤 센서의 감도 특성)

  • 최근 콜레스테롤 센서는 전극 상에 효소를 고정화 하는 방식을 이용하여 센서의 집적도를 높이는 시도가 이루어지고 있다. 이러한 전극 상의 효소고정화 방식으로 entrapment, cross liking, covalently binding 등이 있다. 본 논문에서는 이러한 효소 고정화 방식-전도성 고분자인 P3MT를 사용하여 entrap시키는 방법과 silanization을 이용한 covalent bonding 시키는 방법-에 따른 전기화학 센서의 감도 특성에 관한 연구를 수행하였다. 전도성 고분자를 사용한 고정화 방법은 cyclic voltammograms으로 scan rate 10 mA/s, potential 0.5-1.3V의 조건하에서 P3MT를 Polymerization하고, 효소 고정화를 위해 chromoampermeter로 potential 0.6V에서 900초 동안 수행하였다. silanization을 이용한 covalent bonding 시키는 방법은 nitric acid로 Pt 전극표면을 산화시키고, APTER로 silanization 공정을 시행하였다. 효소 고정화를 위해 전해질로는 0.1M Phosphate buffer solution을 사용하여 cyclic voltammograms으로 scan rate 50 mA/s 전위 0.0-0.7V의 조건 하에서 수행하였다. 이 결과 전도성 고분자를 이용한 고정화 방법에서의 senstivity가 0.89 ${\mu}A/mM{\cdot}cm^2$이고, silanization을 이용한 효소 고정화 방법에서는 1.51 ${\mu}A/mM{\cdot}cm^2$였다. 이처럼 후자의 방법에서 더 좋은 감도 특성이 나타났다. 따라서, silanization을 이용한 고정화 방법이 센서 제작 방식으로 더 적합하다고 사료된다.

Determination of Biogenic Amines using an Amperometric Biosensor with a Carbon Nanotube Electrode and Enzyme Reactor (Carbon Nanotube 전극과 효소반응기로 구성된 Amperometric Biosensor를 이용한 Biogenic Amines 검출)

  • Biogenic amines are synthesized by microbial decarboxylation for the putrefaction or fermentation of foods containing protein. Although biogenic amines such as histamine, tyramine, and putrescine are required for many physiological functions in humans and animals, consumption of high amounts of biogenic amines can cause toxicological effects, including serious gastrointestinal, cutaneous, hemodynamic, and neurological symptoms. In this study, a novel amperometric biosensor wasdeveloped to detect biogenic amines. The biosensor consisted of a working electrode, a reference electrode, a counter electrode, an enzyme reactor with immobilized diamine oxidase, an injector, a peristaltic pump and a potentiostat. A working electrode was fabricated with a glassy carbon electrode (GCE) by coating functionalized multi-walled carbon nanotubes (MWCNT-$NH_2$) and by electrodepositing Prussian blue (PB) to enhance electrical conductivity. A sensor system with PB/MWCNT-$NH_2$/GCE showed linearity in the range of $0.5 {\mu}M{\sim}100 {\mu}M$ hydrogen peroxide with a detection limit of $0.5 {\mu}M$. The responses for tyramine, 2-phenylethylamine, and tryptamine were 95%, 75%, and 70% compared to that of histamine, respectively. These results imply that the biosensor system can be applied to the quantitative measurement of biogenic amines.
