Biological Activity of Ethanol Extracts from Amelanchier asiatica Fruits (채진목(Amelanchier asiatica) 열매 에탄올 추출물의 생리활성)

  • Amelanchier asiatica fruits have been used as a traditional medical food. This research was investigated to assess angiotensin converting enzyme, xanthine oxidase (XOase) and elastase inhibitory activity and antioxidant activities. The content of total phenolic compounds in A. asiatica fruits extracts was 17.6mg/mL. In extracts, the electron donating ability by 1,1-diphenyl-2-picrylhydrazyl free radical scavenging test of A. asiatica fruits extracts was 90.18% at $200{\mu}g/mL$. The 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid radical decolorization of A. asiatica fruits extracts was 98.81% at $200{\mu}g/mL$. The inhibition rate of the antioxidant protection factor was 1.03, and thiobarbituric acid reactive substance was 73.27% at $200{\mu}g/mL$. The XOase inhibition activity of A. asiatica fruits extracts of showed to be 13.19% at $200{\mu}g/mL$. The angiotensin converting enzyme activity was significantly inhibited by A. asiatica fruits extracts as 82.52% inhibitory rate at $200{\mu}g/mL$. Elastase inhibitory activity in the A. asiatica fruits extracts (41.48% at $200{\mu}g/mL$) was higher than vitamin C (12.8% at $200{\mu}g/mL$). These results suggests that A. asiatica fruits extracts have the greatest property as a functional food and functional cosmetic source.

Properties of Aspergillar Xylanase and the Effects of Xylanase Supplementation in Wheat-based Diets on Growth Performance and the Blood Biochemical Values in Broilers

  • Three experiments were conducted to study the property of xylanase and the effects of xylanase in wheat-based diets on growth performance of broilers, respectively. Experiment 1 was performed in vitro to evaluate the effect of different pH and temperature on xylanase activity, and to evaluate the enzymic stability under different conditions. The results indicated that the optimum temperature and pH for xylanase activity were $50^{\circ}C$ and 4.5, respectively. The activity of enzyme solution was reduced rapidly after the treatment of water bath above $60^{\circ}C$ for 10 min. The enzyme was relatively stable at pH 3.5 to 8.0 and deteriorated when incubated at pH below 3.5. In Experiment 2, a total of 378 d-old male Arbor Acres broilers were randomly distributed to 7 different treatments with 6 replicates (9 birds) in each treatment. The treatments were as follows: (1) corn based diet (CS), (2) wheat based diet (WS), (3) WS+ 0.05% xylanase, (4) WS+0.15% xylanase, (5) WS+0.25% xylanase, (6) WS+0.35% xylanase, (7) WS+0.45% xylanase. The results showed that the body weight and feed/gain ratio of the broilers fed wheat-based diets have been significantly improved (p<0.05) compared to that fed corn-based diet in the first 3 wk. With regard to the wheat-based diets, the xylanase supplementation had a tendency to improve the growth performance in first 3 wk. After 3 wk, no significant difference (p>0.05) was found among all these different treatments. The supplementation of xylanase and the type of diets did not affect the feed intake but increased the concentration of triglyceride in serum. In Experiment 3, a total of 360 d-old male Arbor Acres broilers were assigned to 30 groups with 12 birds in each group randomly. These groups were then randomly distributed to 5 different treatments with 6 replicates within each treatment. The broilers of each treatment were fed one of the diets as follows: (1) Corn based diet, (2) White wheat based diet (WW) (3) White wheat based diet+0.25% xylanase, (4) Red wheat based diet, (5) Red wheat based diet+0.25% xylanase. The results showed that the body weight and feed/gain ratio had been significantly improved (p<0.05) by xylanase supplementation in the first 2 or 3 wk. The effect of xylanase in red wheat diet is a little higher than that used in white wheat diet. From the results of the present experiments, it can be concluded that the supplementation of Aspergillar xylanase can improve the performance of the broilers fed the wheat-based diet.

Differential Expression and Stability of Endogenous Nuclear Factor E2-related Factor 2 (Nrf2) by Natural Chemopreventive Compounds in HepG2 Human Hepatoma Cells

  • Nuclear factor-E2-related factor 2 (Nrf2) is known as a key regulator of ARE-mediated gene expression and the induction of Phase II detoxifying enzymes and antioxidant enzymes, which is also a common property of many chemopreventive agents. In the present study, we investigated the regulatory role of different chemopreventive agents including sulforaphane (SUL), allyl isothiocyanate (AITC), indole-3-carbinol (I3C), and parthenolide (PTL), in the expression and degradation of Nrf2 and the induction of the antioxidant enzyme HO-1. SUL strongly induced Nrf2 protein expression and ARE-mediated transcription activation, retarded degradation of Nrf2 through inhibiting Keap1, and thereby activating the transcriptional expression of HO-1. AITC was also a potent inducer of Nrf2 protein expression, ARE-reporter gene and HO-1 but had little effect on delaying the degradation of Nrf2 protein. Although PTL and I3C could induce ARE reporter gene expression and Nrf2 to some extent, they were not as potent as SUL and AITC. However, PTL dramatically induced the HO-1 expression, which was comparable to SUL, while I3C had no effect. In addition, when treated with SUL and PTL, inhibition of proteasome by MG132 did not cause additional accumulation of Nrf2, suggesting the involvement of other degradation mechanism(s) in the presence of these compounds such as SUL and PTL. In summary, the results of our current study indicated that different chemopreventive compounds have different regulatory properties on the accumulation and degradation of Nrf2 as well as the induction of cellular antioxidant enzyme HO-1.

Effect of Corni Fructus on Testosterone Deficiency Syndrome in In vitro and In vivo (In-vitro와 In-vivo에서 산수유의 남성갱년기 개선효과)

  • This study was carried out to evaluate the preventive effect of the Corni Fructus (SSU) 50 % EtOH extract (SSU-E50) against bisphenol A (BPA) toxicity in Leydig cells and improving testosterone deficiency syndrome in orchidectomized Sprague-Dawly (SD) rats. Antioxidant properties were measured by radical scavenging activity of SSU-E50 in ABTS assay and DPPH assay. Also, real-time polymerase chain reaction(real-time PCR) was performed to quantify the mRNA expression levels of antioxidant enzyme. SD rats were divided into eight group: normal, sham operation (Sham), orchidectomized (ORX), ORX treated with testosterone 1 mg/kg (Tes. 1), ORX treated with SSU water extract 100 mg/kg (SSU-A 100) and 300 mg/kg (SSU-A 300), ORX treated with SSU 50 % EtOH extract 100 mg/kg (SSU-E 100) and 300 mg/kg (SSU-E 300). On a comparative basis, the SSU showed better activity quenching ABTS with an IC50 value of 0.29 mg/ml and DPPH with an IC50 value of 0.33 mg/ml. Cell viability was evaluated by MTS assay as described not cytotoxic at the highest concentration of $500{\mu}g/ml$. Cytotoxicity of BPA showed in $200{\mu}M$, but definitely survived by treatment with SSU in Leydig cells. In addition, SSU increased the mRNA expression levels of antioxidant enzyme in BPA induced Leydig cells. Superoxide dismutase (SOD) level was slightly increased and malondialdehyde (MDA) level was decreased with SSU-A 100 in in-vivo. These results suggest that Corni Fructus extracts have the greatest property as a natural anti-oxidative and improves testosterone deficiency syndrome source.

The effect of aluminum coating to corrugated packaging on quality characteristics of paprika during storage (골판지 포장재에 알루미늄 코팅이 파프리카의 저장 중 품질특성에 미치는 영향)

  • The objective of this study was to investigate the effect of corrugated packaging coated with aluminum (Al) on quality characteristics of red paprika during storage at $25^{\circ}C$. Characteristics such as weight loss, hardness, total phenolic content, antioxidant activities, polygalacturonic acid (PG) activity, and oxidative enzyme activities (polyphenol oxidase and peroxidase activities) of paprikas, packed in corrugated packaging with or without Al-coating were compared as a function of storage time. Al coating inside of corrugated box was found to inhibit PG activity, resulting in prevention of weight loss and maintenance of hardness of paprika during storage, compared to the control sample. This may be due to Al coating treatment that enhance moisture-proof property and hinder gas transmission of corrugated packaging. Furthermore, paprika in Al-coated-corrugated packaging was lower in oxidative enzyme activities than the control, which caused higher total phenolic content and antioxidant activities during storage. As a results, the Al coated-corrugated packaging can be used as a functional packaging material to extend the shelf-life and improve the storage quality of paprika by preventing their respiration and transpiration.

The Studies on Hydrolysis of the Silk Fibroin by Proteolytie Enzyme, Bombyx mori II. Relation between the Fibroin Hydrolysis of Different Cocoon Layers and Physical Property of Silk Fiber (견 Fibroin의 효소분해에 관한 연구 II-견층부위별 Fibroin 분해율 차이와 견사물성과의 관계-)

  • The study was carried out to investigate the variations in the fibroin hydrolysis of different cocoon shell layers, and the relationship between the hydrolysis and silk physical property. The obtained results are as follows: 1. The fibroin hydrolyzing ratio was highest at the inner layer of cocoon shell and. next in order. at the middle layer and at the outer layer. 2. The fibroin hydrolyzing ratio of abnormal cocoons was higher than normal cocoon and it was different among them, tile highest was double coroon, thin shell cocoon and perforated cocoon in order. 3. The sericin content and fibroin hydrolyzing ratio of Jam 111 and Jam 112 was higher than that of Jam 111 $\times$ Jam 112. 4. The fibroin hydroiyzing ratio of the cocoons fed with the artificial diets was increased at the inner layer. The sericin content of those lessened at the inner layer, however, it slightly increased at the most inner layer more than at the inner layer. 5. The breaking strength of the degummed silk fiber of different cocoon layers was reduced at the inner layer. The breaking Strength of abnormal silk fiber was less than that of normal silk fiber. 6. A negative correlation (r= -0.8) )was approved between fibroin hydrolyzing ratio and breaking strength of silk fiber, and the regression line was Y=-0.29x+5.07.

Processing Conditions for Protein Enriched Jeung-Pyun (Korean Fermented Rice Cake) (전통증편의 단백질보강에 관한 연구)

  • To improve the food quality of traditional Jeung-pyun(Korean fermenfed rice cake), effect of mixed ingredients on the quality of protein enriched product was studied. Changes were evaluated in chemical and textural properties of protein enriched Jeung-pyun altered by fermentation time, temperature, and steaming time. The maximum volume wxpansion was noted in dough mixed with rice flour(100 g), soy flour(25 g), sucrose(20 g), Tak-ju(50 ml) and water(10 ml), and then fermented at $35^{\circ}C$ for 3 hours. Soy protein isolate(SPI) enriched rice dough had a maximum expanded volume when 15g(w/w) of SPI, 20g(w/w) of sucrose, 60ml(v/w) of Tak-ju and 50ml(v/w) of water added into 100g rice flour, and then fermented at $30^{\circ}C$ for 2 hours. Steaming after fermentation made the smaller volume of bulk $45^{\circ}C$-50% of the initial volume be showed just after fermentation. Protein enriched Jeung-pyun prepared under the maximal volume endowing conditions showed the best protein quality(protein digestibility and protein efficiency ratio) and starch structure which was susceptible towards enzyme reaction. The improved starch quality of protein enriched Jeung-pyun could be confirmed by reducing power, gelatinization degree and amylose content. Jeung-pyun riched with soy flour at 20% level or with SPI at 5% showed the best overall quality by sensory and textural property but it had a problem in browning.

Production and Properties of Mannanase by a Bacillus amyloliquefaciens Isolate (Bacillus amyloliquefaciens 분리균의 Mannanase 생산성과 효소특성)

  • In the acidic LB plate, a bacterial strain was isolated from homemade soybean paste as a producer of the extracellular mannanase. The isolate YB-1402, which was a Gram-positive rod-shaped bacterium with spore, has been identified as Bacillus amyloliquefaciens on the basis of its 16S rDNA sequence and biochemical properties. Maximum mannanase productivity of the isolate YB-1402 was reached approximately 150 U/ml in LB broth supplemented with konjac (3.0%). The molecular mass of YB-1402 mannanase was estimated to approximately 38.0 kDa by zymogram of the culture filtrate on SDS-PAGE. The mannanase of culture filtrate was the most active at $55^{\circ}C$ and pH 5.5. The mannanase activity was completely maintained after pre-incubation at pH 3.0 to 10.0 for 1 h. The predominant products resulting from the mannanase hydrolysis were mannose, mannobiose and mannotriose for LBG or mannooligosaccharides. The enzyme could hydrolyze mannooligosaccharides larger than mannobiose.

Improvement of the Thermostability of Xylanase from Thermobacillus composti through Site-Directed Mutagenesis

  • Thermostability is an important property of xylanase because high temperature is required for its applications, such as wood pulp bleaching, baking, and animal feedstuff processing. In this study, XynB from Thermobacillus composti, a moderately thermophilic gram-negative bacterium, was modified via site-directed mutagenesis (based on its 3D structure) to obtain thermostable xylanase, and the properties of this enzyme were analyzed. Results revealed that the half-life of xylanase at $65^{\circ}C$ increased from 10 to 50 min after a disulfide bridge was introduced between the ${\alpha}$-helix and its adjacent ${\beta}$-sheet at S98 and N145. Further mutation at the side of A153E named XynB-CE in the C-terminal of this ${\alpha}$-helix enhanced the half-life of xylanase for 60 min at $65^{\circ}C$. Therefore, the mutant may be utilized for industrial applications.

The Effects of Dictamni Radicis Cortex on the iNOS Expression and Proinflammatory Cytokines Production (백선피의 iNOS발현과 염증성사이토카인의 생성에 미치는 영향)

  • The aim of the present study is to investigate the cytokine production inhibitory effect of a Dictamni Radicis Cortex (DRC). DRC has been commonly used as important medicinal herb in China and it used to control eczema, atopic dermatitis, fever and inflammatory diseases. Inflammation, such as a bacterial infection in vivo metabolites, such as external stimuli or internal stimuli to the defense mechanisms of the biological tissue a variety of intracellular regulatory factors deulin inflammatory TNF-${\alpha}$, IL-$1{\beta}$, IL-6, IL-8, such as proinflammatory cytokines, prostagrandin, lysosomal enzyme, free radicals are involved in a variety of mediators. The present study was designed to determine the effect of the DRC on proinflammatory factors such as NO, iNOS expression and TNF-${\alpha}$, IL-$1{\beta}$, IL-6 in lipopolysaccharide (LPS) - stimulated RAW264.7 cells. The cell toxicity was determined by MTS assay. To evaluate of anti-inflammatory effect of DRC, amount of NO was measured using the NO detection kit and the iNOS expression was measured by reverse transcriptase polymerase chain reaction (RT-PCR). And proinflammatory cytokines were measured by ELISA kit. As a result, the DRC reduced NO, iNOS expression and TNF-${\alpha}$, IL-$1{\beta}$, IL-6 production without cytotoxicity. Our results suggest that the DRC may have an anti-inflammatory property through suppressing inflammatory mediator productions.
