Advanced Lipid Extraction Method for the Determination of the Phospholipase D Activity

  • Phospholipase D is a ubiquitous enzyme that plays an important role in various lipid mediated cellular signaling pathways and produces rare phospholipids, phosphatidylethanol or phosphatidylbutanol, instead of phosphatidic acid with unique catalytic activity transphosphatidylation in the presence of primary alcohols. The reaction products, phosphatidylethanol or phosphatidylbutanol are used as markers of in vitro phospholipase D activity in many studies. For the sensitive detection of the phospholipase D products, we developed an advanced lipid extraction method that facilitates recovery of the compounds. With the new method, the activity change of phosaholipase D by agonists could be detected more easily and the recovery rate was also increased. The increase of detected enzyme activity change was about double fold compared to the conventional lipid extraction method. This method provides selective force for the phospholipase D products in the extraction procedure.

The ascorbic acid oxidizing activity in streptomyces viridochromogenes (Streptomyces viridochromogenes의 ascorbic acid 산화 활성)

  • The competence time of Streptomyces viridochromogenes for aerial mycelium formation was determined. Within 10 hrs after spore inoculation the submerged mycelium was programed to form aerial mycelium, when the former was laid on agar plate. The white aerial mycelium was formed 17-22 hrs after the transfer. Ascorbic acid oxidizing enzyme band on native gel showed chracteristic mobility change during aerial mycelium formation. Total activity of this enzyme did not show any correlation with the differentiation. The asay condition for the crude enzyme was determined. EDTA and $FeCl_{2}$ showed stimulatory effect. Approximate ratio of oxygen consumed to ascorbic acid oxidized was 1:1.

Diurnal Changes of Chlorophyll Fluorescence and Antioxidative Enzyme Activity of the Leaves from Four Subtropical Plants (아열대성 식물 4종의 엽록소형광과 항산화효소 활성의 일주기적 변화)

  • The diurnal changes of chlorophyll fluorescence and antioxidative enzyme activity were investigated in the leaves from four subtropical plant species (Crinum asiaticum var. japonicum Bak., Osmanthus insularis Koidz., Asplenium antiquum Makino and Chloranthus glaber Makino) under the natural habitats in summer and winter. The intensity of chlorophyll fluorescence was lower in O-, I-, J-, P-steps of O-J-I-P transient in winter than summer, and prominent diurnal change was not found in the fluorescence intensity of four subtropical plant species in winter. The activity and isoenzyme pattern of SOD and catalase did irregularly change seasonally and diurnally in four subtropical plant species. In contrast, the peroxidase activity and isoenzyme pattern was different depending on plant species and growth seasons; The activity increased slightly more in winter than in summer in four subtropical plant species, and several isoenzymes appeared in the leaves from C. asiaticum var japonicum, O. insularis and A. antiquum in winter.

Continuous Production of Fructooligosaccharides Using Fructosyltransferase Immobilized on Ion Exchange Resin

  • A continuous production of fructooligosaccharides from sucrose was investigated by fructosyltransferase immobilized on a high porous resin, Diaion HPA25. The optimum pH(5.5) and temperature(55$^{\circ}C$) of the enzyme for activity was unaltered by immobilization, and the immobilized enzyme became less sensitive to the pH change. The optimal operation conditions of the immobilized enzyme column for maximizing the productivity were as follows: 600g/L of sucrose feed concentration, flow rate of superficial space velocity 2.7h-1. When the enzyme column was run at 50$^{\circ}C$, about 8% loss of the initial activity of immobilized enzyme was observed after 30 days of continuous operation, during which high productivity of 1174g/L$.$h was achieved. The kinds of products obtained using the immobilized enzyme were almost the same as those using soluble enzymes or free cells.

The Effect of Ginseng Saponin on Yeast Alcohol Dehydrogenase (인삼 사포닌이 효모 Alcohol Dehydrogenase에 미치는 영향)

  • Yeast alcohol dehydrogenates and ginseng saponin interaction has been investigated to understand the non-specific enzyme stimulating effect of the saponin of Panax ginseng C.A. Meyer. It was confirmed that several amphiphiles such as sodium dodecyl sulfate(SDS), Triton X-100, sodium taurodeoxycholate (Na-TDC) as well as ginseng saponin mixture and purified ginseng glycosides lowered Km values of yeast alcohol dehydrogenase (ADH) for ethanol and NAD in the presence of the above amphiphiles suggesting that the surface activity of the amphiphiles might play a significant role in the ADH catalyzed reactions. Conformational change of yeast alcohol dehydrogenase in the presence of the above amphiphiles at their optimal concentration for the maximum activity was studied. Circular dichroism (C.D) spectrum of yeast ADH showed that the conformational change of the enzyme occurred in the presence of above amphiphiles. Fluorescence data also showed that the hydrophobic area increased in the presence of above amphiphiles. Examination of the interaction between ADH and ginseng saponin using radioactive saponin showed that there might be a very weak interaction between them. From the above results, it was concluded that the non-specific enzyme stimulating effect of the saponin might be due to the change of polarity of the enzyme solution in the presence of the saponin.

Change of ROS Generation and Antioxidant Enzyme Activity of Flavonol Quercetin in the Presence of Vitamin E, L-Ascorbit acid, Reduced Glutathione on the B16F10 Murine Melanoma Cells (B16F10 세포에서 Quercetin과 Vitamin E, L-Ascorbic acid, 환원형 글루타치온과의 병용 투여에 의한 활성산소종 발생과 항산화 효소의 활성 변화)

  • It has been known that quercetin, a bioflavonoid widely distributed in fruits and vegetables as dietary-derived flavonoid and exert significant multiple biological effects such as antioxidant and anti-inflammatory, anti-tumor effects. In addition, it has been shown to have a chemoprotective role in cancer, though complex effects on signal transduction involved in cell proliferation and angiogenesis. The present study investigated whether quercetin can enhance antioxidant enzyme activity (glutathione peroxidase: GPx, superoxide dismutase: SOD, catalase: CAT) and regulate the reactive oxygen species (ROS) generation in the presence of vitamin E, L-ascorbic acid, reduced glutathione (GSH) on B16F10 murine melanoma cells. After 48h treatment of cells with quercetin in the presence of vitamin E, L-ascorbic acid, GSH, we measured the cytotoxicities by MTT assay. The cells exhibited a dose-dependent inhibition in their proliferation in the presence of vitamin E, L-ascorbic acid, GSH respectively. We also investigated the effects of antioxidant enzyme activity and ROS generation. The antioxidant enzyme activity of quercetin in the presence of vitamin E was stronger than GSH, L-ascorbic acid, the same treatments decreased ROS generation in B16F10 murine melanoma cells. Taken together, these result demonstrate that the antioxidant effect of quercetin can enhanced in the presence of vitamin E and it might plays an important role in anti-oxidative effects.

Changes in the Quality Characteristics of Cheonggukjang prepared with Hazelnut (헤이즐넛 첨가에 따른 청국장의 품질 특성 변화)

  • This research was conducted to investigate the changes in quality characteristics of cheonggukjang fermented with the addition of hazelnut (10, 20, 30 and 40%) including; water content, pH, hydrophilic and lipophilic substances, color, viscosity and angiotensin converting enzyme inhibition activity. There was no significant change in pH with the addition of hazelnut. The water content significantly decreased with the addition of hazelnut. Hazelnut was also found to brighten the color of cheonggukjang. L-value and b-value increased with the addition of cheonggjuang. There was an insignificant change in the a-value. There was a slight decrease in the content of hydrophilic with addition of hazelnuts. Where there was more than 20% addition of hazelnut to soybean, the viscous substance content in cheonggukjang decreased significantly. Angiotensin converting enzyme inhibitory activity increased proportionally to the amount of hazelnut added. It was identified that the addition of 40% of hazelnut made its angiotensin converting enzyme inhibitory activity 10% point higher than that of control. These results suggests that the addition of hazelnut makes it possible to produce cheongkukjang of excellent angiotensin converting enzyme inhibitory activity.

Effect of Soy Isoflavone Supplementation on the Bone Mineral Density and Antioxidant Enzyme Activity in Postmenopausal Women (일부 폐경 후 여성에서 골밀도와 항산화효소 활성에 대한 대두 이소플라본 보충 효과)

  • To elucidate the effect of soy isoflavone supplementation on bone mineral density and antioxidant enzyme activity in 60 postmenopausal women residing in Iksan area were recruited. There were 31 participants in the treatment group and 29 in the control group. The treatment group consumed isoflavone extract capsules daily (which contained 90 mg of soy isoflavones) for 12 weeks. The study compared before and after isoflavone intake in the following areas. Physical examination, diet survey, bone mineral density (BMD) and antioxidant enzyme activity (superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase (CAT) and total antioxidant capacity (TA)). The average age of the treatment group was 64.6 years and that of the control group was 66.5 years. There were no significant differences between the two groups in terms of height, weight and body mass index. Both groups maintained a regular diet pattern in terms of their average daily nutrient intake. There were no significant differences between the treatment group (23.3 mg) and the control group (24.0 mg) in terms of daily isoflavone intake based on diet. Isoflavone supplementation of 12 weeks did not resulted in any significant changes in BMD or parameters of antioxidant enzyme activity, implying the necessity of more intensive intervention for a substantial change. In conclusion, this study revealed that antioxidant enzyme activity holds a significant relationship with the bone mineral density in postmenopausal women and further systematic research on dose and period of isoflavone supplementation is needed to clearify the positive effect of isoflavone on BMD and/or blood antioxidant capacity in postmenopausal women.

Increase in Hepatic DT-Diaphorase Activity by Chronic Administration of Panax ginseng Extract to Mice (생쥐에서의 인삼추출액의 장기간 투여에 의한 간장 DT-Diaphorase 활성의 증가)

  • Effects of chronic administration of ginseng extracts (30 or 150 mg/kg/day for 52 days, p.o.) to mice on the activities of DT-diaphorase and glutathione S-transferase (GST) in the liver and the brain were studied. The DT-diaphorase activity in the liver was increased over 2-fold at the dose of both 30 and 150 mg/kg/day, while there was no change in the activity of the enzyme in the brain. The GST activity in the liver was increased in a dose-dependent fashion up to 142% of the control value at the dose of 150 mg/kg/day. while there was no change in the activity of the enzyme in the brain. The ginseng-induced increase in the activities of these hepatic phase II drug-metabolizing enzymes which are involved in the detoxification of carcinogens, is suggested to underlie, at least in part, the anticarcinogenic activity of Panax ginseng.

Hydrolysis Characteristics of Amylase from Alkaline-Tolerant Bacillus sp. on the Raw Starch (알칼리 내성 Bacillus sp.가 생산하는 Amylase의 생전분 분해 특성)

  • The raw starch hydrolysis by amylase prepared from alkaline-tolerant Bacillus sp. were investigated. Degree of hydrolysis(%) of 5%(w/v) raw rice, corn and potato starch by this enzyme were about 40, 25 and 20%, respectively. The hydrolysis action on raw starch by change of blue value was similar to the action pattern of exo ${\beta}$-amylase. The hydrolysis products of rice starch were mainly glucose and maltose. Oligosaccarides were also detected. From the above results, this enzyme was considered as exo type ${\alpha}$-amylase. This enzyme activity on the raw starch and the gelatinized starch were 28.40 and 86.60 IU/mg protein, respectively, and the ratio of raw starch-digesting activity to gelatinized starch-digesting activity (raw starch digestivity) was about 32%. The Km values for the raw and the gelatinized starch were 4.22 and 3.0mg/mL, respectively, and the VmaX values were 0.20 and 0.31mg/mL/min, respectively.
