Chitinase activities from Shumard oak tissues were determined to study changes in chitinase activities related to water stress. The enzyme extracted in sodium acetate buffer (0.1M, pH 4.5) was assayed by a colorimetric method. In addition, the fungal hyphae of Hypoxylon atropunctatum in xylem tissues of oak were observed through scanning electron microscopy. The enzyme in oak tissues was mainly endochitinase, and optimum pH for enzyme activity was 5. Specific chitinase activities from both of stems held under high relative humidity (ranges of 0.63-1.11 pKatal/$\mu\textrm{g}$ of protein) and stems held under low relative humidity (ranges of 0.41-0.99 pKatal/$\mu\textrm{g}$ of protein) were significantly increased following fungal inoculation with H. atropunctatum. However, there was no significant difference in chitinase activities between tissues held under high and low humidities, which might be due to fungal chitinase. Scanning electron microscopy showed holes in fungal hyphae in the xylem tissues of stems held under high humidity but not in the stems held under ow humidity, suggesting that hyphae might be hydrolyzed by plant hydolases such as chitinase.
A variety of important roles for branched-chain amino acids in metabolic regulation has been suggested. Branched-chain $\alpha$-keto acid dehydrogenase(BCKAD) complex is a rate limiting enzyme in branched-chain amino acid metabolism. The purpose of this study was to examine the effects of exercise on the activity and activity state of branched-chain $\alpha$-keto acid dehydrogenase in rat hert and liver thssues. Forty-eight Sprague-Dawley rats were assigned into three experimental groups : sedentary control, exercised, or exercised-rested. Submaximal exercise(running) for two hours significantly increased basal activity without a change in total activity in both tissues, with a concomitiant increase in activity state of the enzyme complex. At 10 min post-exercise, heart enzyme activity significantly decreased, though not to the control level, while liver enzyme activity remained unchanged. These data suggested that the exercise-induced increase in branched-chain $\alpha$-keto acid decarboxylation in rat tissues may not be the result of enzyme synthesis, but rather is due to increased activity of the BCKAD.
본 연구에서는 붕어를 이용하여 탁도 변화에 따른 여러 조직의 항산화효소 활성을 조사하였다. 탁도 50, 100, 150 NTU에서 사육기간에 따른 붕어 조직의 항산화효소 활성의 변화는 50과 100 NTU에서는 대조구에 비해 큰 변화가 없었으며, 고탁도인 150 NTU에서 비교적 두드러진 차이를 나타냈다. 붕어 조직의 항산화능을 DPPH 소거 활성도에서 보면 아가미, 간 조직에서 높은 활성을 보여 고탁도의 사육 조건에서 장기간 사육될 때 비효소적 항산화작용이 일어나는 것으로 생각된다. 효소적 항산화작용은 50, 100, 150NTU로 탁도가 높아짐에 따라 항산화효소의 활성이 증가하였으며 SOD, CAT및 GST의 활성은 표피를 제외한 모든 조직에서 사육 후 20일까지는 증가한 후 사육 후기로 갈수록 감소하는 경향을 나타냈다. 그러나 GR과 GPX의 활성은 거의 모든 조직에서 활성이 매우 낮으므로 어류의 항산화과정에서 이들 효소의 역할은 매우 적을 것으로 생각된다.
The branched-chain ${\alpha}$-keto acid dehydrogenase (BCKAD) complex is a rate limiting enzyme which catalyzes the oxidative decarboxylation of branched-chain ${\alpha}$-keto acids. Numerous studies have suggested that BCKAD is subject to covalent modification in vitro via phosphorylation and dephosphorylation, which are catalyzed by a specific kinase and phosphatase, respectively. The biggest difficulty in the assay of BCKAD activity is to arrest the interconversion between the active and inactive forms. BCKAD activity was determined from fresh rat heart and liver tissues using homogenizing and assay buffers containing inhibitors of phosphatase and kinase. The results suggest that a radiochemical assay using ${\alpha}$-keto[1-$^{14}C$]-isovalerate as a substrate for the enzyme can be applied as a reliable method to determine in vitro enzyme activity with arrested interconversion between the active and inactive forms of the BCKAD complex.

As a novel strategy to remove ${\beta}$-lactam antibiotic residues from fish tissues, utilization of ${\beta}$-lactamase, enzyme that normally degrades ${\beta}$-lactam structure-containing drugs, was explored. The enzyme (TEM-52) selectively degraded ${\beta}$-lactam antibiotics but was completely inactive against tetracycline-, quinolone-, macrolide-, or aminoglycoside-structured antibacterials. After simultaneous administration of the enzyme with cefazolin (a ${\beta}$-lactam antibiotic) to the carp, significantly lowered tissue cefazolin levels were observed. It was confirmed that the enzyme successfully reached the general circulation after intraperitoneal administration, as the carp serum obtained after enzyme injection could also degrade cefazolin ex vivo. These results suggest that antibiotics-degrading enzymes can be good candidates for antibiotic residue depuration.
어류 및 폐류내에 내재하는 내재성 ${\beta}-galactosidase$의 활성도를 분석함으로서 외래유전자의 이식시 기초자료로 활용하고자 어류 6종 및 패류 4종을 대상으로 본 실험을 행하였다. 어류에 있어서의 X-gal 염색 결과 혈청 및 근육을 제외한 모든 조직에서 모두 positive ( + ) 염색 양상을 보였다. 패류에 있어서도 어류와 같은 양상을 보여 근육을 제외한 모든 조직에서 모두 positive 염색 양상을 보였으며 더욱이 참굴에 있어서는 폐각근 및 근육에 있어 매우 진한 염색 반응을 보였다. 근육으로 부터 DNA를 추출한 후 PCR을 수행한 결과 모든 종에서 positive (+) 결과를 나타내었으며, 어패류간 그리고 각 개체간 차이는 없었다. 각 장기별 ${\beta}-galactosidase$의 활성도 측정결과 모든 종에 있어서 혈청의 활성도는 무시할 만한 수준이었고, 근육에서 가장 낮게 나타났다. 미꾸라지와 잉어에서는 신장에서 가장 높은 활성을 보였고, 틸라피아는 소화관에서 가장 높은 값을 보였다. 해산어인 넙치의 경우 돌가지미, 문치가자미의 경우에 비해 간에서 활성이 높았다. 그러나 돌가자미 및 문치가자미인 경우 신장에서 가장 높은 값을 보였다. 패류에 있어서는 소화맹낭에서 가장 높은 활성값을 나타내었고 폐각근 및 외투막에서 매우 낮은 활성값을 나타내었다. 그러나, 참굴에 있어서는 참전복, 피조개 및 진주조개 보다는 상대적으로 폐각근 및 외투근에서 높은 활성을 나타내었다.
The purpose of this study was to probe the influences of krill (Euphausia superba) meal with NaF oral administration on a dose-effect relationship between fluoride levels of krill meal and serum enzyme activity such as alkaline phosphatase (ALP), aspartate aminotransferase (AST), alanine aminotransferase (ALT), and lactate dehydrogenase (LDH) in rats fed experimental diets for 5 weeks. There were no significant decreases in the activities of ALP, AST, ALT, and LDH in sera among krill meal diet groups (KF10, KF20, KF30). However, these groups were significantly (p<0.05) lower enzyme activities than normal diet (ND) plus NaF 10 mg group (NF). The fluoride levels of serum and organ tissues (liver, brain, heart, lung, kidney) in NaF 10 mg groups (NF, KF10, KF20, KF30) were significantly increased by adding krill meal in comparison with normal diet group. The results indicate that a difficult to found toxicity to the tissues from krill meal diet groups.
In commercial greenhouses, senescent flower petals or flowers of vegetables such as tomato, strawberry, hot pepper and zucchini squash were blighted to be removed from fruits within five days after spraying of Trichoderma harzianum YC459 (TORY), a biocontrol agent for the gray mold rot of vegetables caused by B. cinerea The mechanism for selective colonization of senescent floral tissues by T. harzianum YC459 was elucidated using fresh and senescent (Hays and 14days after flowering, respectively) floral tissues of zucchini squash (Cucurbita moschata Duchesne). The spores of T. hrzianum YC459 were produced more on agar and liquid culture media supplemented with 5% dry powder of senescent floral tissues than fresh tissues during 15days. Mycelial growth was also much better in the media with senescent tissues than with fresh tissues. Enzyme activities of amylase, polygalacturonase and cellulase in the liquid media which might be involved in the colonization of tissues by T. harzianum YC459 were compared. The activities of three enzymes were much higher in the media with senescent floral tissues than with fresh floral tissues reaching to the maximum during 9 to 12days of incubation. Based on the results, the removal of senescent floral tissues, a possible inoculum source of the pathogen, may be another mechanism for biocontrol of gray mold rot of vegetables by T. harzianum YC459.
We assessed the effect of Se-methylselenocysteine (MSC) treatment, at a dose of 0.75 mg/rat/day for 1 or 2 weeks, on the activities of antioxidant systems in Sprague-Dawley rat tissues. Significant changes in glutathione and antioxidant enzyme activities, with different patterns among tissues, were evidenced. Glutathione content and its reduction state in the liver, lung, and kidney were elevated upon MSC treatment, whereas they were significantly lowered in the spleen. Among the tissues exhibiting glutathione increase, there were different enzymatic responses: $\gamma$-glutamylcysteine ligase activity, the rate-limiting enzyme in the glutathione synthesis pathway, was increased in the liver, whereas the activities of the enzymes associated with glutathione recycling, namely, glutathione peroxidase, glutathione reductase, and glucose 6-phosphate dehydrogenase, were significantly increased in the lung and the kidney. The superoxide dismutase activity was decreased in all tissues upon MSC treatment, whereas catalase activity was increased in all tissues but the liver. Lipid peroxidation level was transiently increased at 1 week in the lung and the kidney, whereas it was persistently increased in the spleen. The increase was not evident in the liver. The results indicate that the MSC treatment results in an increase in the antioxidant capacity of the liver, lung, and kidney principally via an increase in glutathione content and reduction, which appeared to be a result of increased synthesis or recycling of glutathione via tissue-dependent adaptive response to oxidative stress triggered by MSC. The spleen appeared to be very sensitive to oxidative stress, and therefore, the adaptive response could not provide protection against oxidative damage.
본 연구는 Cd에 노출시킨 붕어를 통하여 각 조직에 축적된 Cd 양과 항산화효소 활성을 통해 생리적인 변화 및 아가미와 신장 조직에서의 미세구조 변화를 조사하였다. Cd을 처리한 실험군 붕어 조직 내 Cd의 함량은 아가미 조직에서 가장 높았고 근육 조직에서 가장 낮았다. 또한 모든 조직에서 Cd의 함량은 노출기간이 길어질수록 증가하여 노출 20일에 가장 높았다. 항산화효소의 활성은 아가미와 근육 조직에서 높았으며, 신장과 간 조직에서 낮았다. 모든 조직에서 SOD의 활성이 가장 높았으며, SOD와 CAT의 활성은 10일에 높은 활성을 보이고 감소하는 유사한 결과를 보였다. Cd에 20일간 노출시킨 붕어의 아가미 조직은 대조군 아가미 조직에 비하여 노출기간이 길어질수록 새변 사이에 점액세포의 수가 증가하였으며, 이차새변에서는 부종, 상피세포의 박리와 새변과 새변이 융합한 형태도 관찰되었다. 또한 아가미 조직의 미토콘드리아와 핵에서는 막의 손상이 확인되었다. 신장 조직은 대조군에서 보우만 주머니 안에 사구체가 꽉 찬 정상구조였으나 노출기간이 길어짐에 따라 사구체가 수축하여 보우만 주머니 안의 공간이 넓은 형태로 관찰되었다.
