Bond Strength of Wafer Stack Including Inorganic and Organic Thin Films (무기 및 유기 박막을 포함하는 웨이퍼 적층 구조의 본딩 결합력)

  • The effects of thermal cycling on residual stresses in both inorganic passivation/insulating layer that is deposited by plasma enhanced chemical vapor deposition (PECVD) and organic thin film that is used as a bonding adhesive are evaluated by 4 point bending method and wafer curvature method. $SiO_2/SiN_x$ and BCB (Benzocyclobutene) are used as inorganic and organic layers, respectively. A model about the effect of thermal cycling on residual stress and bond strength (Strain energy release rate), $G_c$, at the interface between inorganic thin film and organic adhesive is developed. In thermal cycling experiments conducted between $25^{\circ}C$ and either $350^{\circ}C$ or $400^{\circ}C$, $G_c$ at the interface between BCB and PECVD $ SiN_x $ decreases after the first cycle. This trend in $G_c$ agreed well with the prediction based on our model that the increase in residual tensile stress within the $SiN_x$ layer after thermal cycling leads to the decrease in $G_c$. This result is compared with that obtained for the interface between BCB and PECVD $SiO_2$, where the relaxation in residual compressive stress within the $SiO_2$ induces an increase in $G_c$. These opposite trends in $G_cs$ of the structures including either PECVD $ SiN_x $ or PECVD $SiO_2$ are caused by reactions in the hydrogen-bonded chemical structure of the PECVD layers, followed by desorption of water.

Identification of QTLs for Some Agronomic Traits in Rice Using an Introgression Line from Oryza minuta

  • Wild progenitor species provide potential gene sources for complex traits such as yield and multiple resistances to biotic and abiotic stresses, and thus are expected to contribute to sustainable food supplies. An introgression line 'IR71033-121-15' was derived from a wild species Oryza minuta (2n = 48, BBCC, Acc No. 101141) at IRRI. Introgression analysis using 530 SSR and STS markers revealed that at least 14 chromosomal segments distributed over 12 chromosomes had been introgressed from O. minuta. An $F_{2:3}$ population from the cross between IR71033 and Junambyeo (a Korean japonica cultivar) consisting of 146 lines was used for quantitative trait loci (QTL) analysis of 16 agronomic traits. A total of 36 single-locus QTLs (S-QTLs) and 45 digenic epistasis (E-QTLs) were identified. In spite of it's inferiority of O. minuta for most of the traits studied, its alleles contributed positively to 57% of the QTLs. The other QTLs originated from either parent, IR71033 or Junambyeo. QTLs for phenotypically correlated traits were mostly detected on introgressed segments. Fourteen QTLs corresponded to QTLs reported earlier, indicating that these QTLs are stable across genetic backgrounds. Twenty-two QTLs controlling yield and its components had not been detected in previous QTL studies. Of these, thirteen consisted of potentially novel alleles from O. minuta. QTLs from O. minuta introgression could be new sources of natural variation for the genetic improvement of rice.

Changes of Vascular Contraction and Relaxation of Rat aorta under Arsenic Stress (비소 스트레스에 의한 흰쥐 대동맥의 수축과 이완반응의 변화양상)

  • In order to examine whether arsenic, one of environmental stress, contribute to augumentation and relaxation of rat aorta, this study was performed in vivo and in vitro, using intacted or denuded rats aorta ring preparation, respectively. The carotid arterial pressure was recorded on an ink-writing physiograph(Grass Co. 79E) connected to strain gauge. The contractile response of vascular ring with or without endothelium preparation isolated from rat were determined in organ bath and was recorded on physiograph connected to isometric transducer. Vasopressin-,and phenylephrine- induced increase in arterial pressure significantly enhanced in arsenic-treated rats; increase of 19.1%, and 46.6%, respectively. Vascular contractile response was measured in vitro preparations exposed to 0, 0.5, 1, 2 and 4 mM of arsenic for 1, 3, 5 and 8 hours. The dose-vascular responses of phenylephrine augmented by increasing dose of arsenic in the strips exposed to arsenic for 8 hours, and did not augmented for 1, 3, 5 hours. The phenomenon was not affected by strips denuded endothelium. And the response of relaxation of rat aorta induced by nitroprusside was not influenced by arsenic stress, but acetylcholine was a little increased. compared to that of control. There were no significant difference in relaxation between control and arsenic treated rings with endothelium or denuded. All of the results, phenyleprine-induced vascular contraction was significantly enhanced in 4 mM arsenic-treated rat aortic rings compared with control, whether endothelium was present or denuded at 8 hours after arsenic treatment. It may be a mechanism by which long-term arsenic stresses play a role in development of hypertension.

Development of high yield rice of long grain type adaptable to South-East Asia tropical region

  • The long grain rice varieties adaptable to South-east Asia tropical regions were tried to develop in Cambodian Agriculture Research and Development Institute (CARDI), Cambodia. The final goal is to develop rice varieties which can culture in diverse environmental conditions of tropical regions of South-east Asia under climate change. We collected and evaluated for agronomic traits of 131 rice germplasm from Cambodia, China, India, Indonesia, Myanmar, Philippines and Vietnam in CARDI. We selected core germplasm including leading varieties of target countries and made 813 F1 cross combinations between leading varieties of each country and promising germplasm of high yield potential, resistance to biotic/abiotic stresses, aromatic rice, and so on. Out of 607 F1s evaluated to heading date, plant type, agronomic traits, and grain type, 106 F1s selected and advanced to F2 populations. 106 F2 populations were evaluated to major agronomic traits, grain type and yield-component traits, and selected 2,560 plants in 62 F2 populations. During six seasons in 2014~2016, the lines of F3 subsequent-generation were cultured a total of 6,256 lines. In yield trial for promising lines in F5 generation, the growth duration from sowing to harvesting was 97~114 days. These lines were 88~129 in number of grain per panicle, an average of 84.6% in the range of 79.3~91.9% in the percentage of ripened grain and 17.5~22.8g in 1000-brown rice weight. The rough rice yields were in the range of 4.33~6.06 ton/ha with an average of 5.23 ton/ha. The yield was increased to 5~47% than Chulsa and 12~41% than IR66. Five lines, KR54-28-1, KR55-14-2, KR57-5-2, KR67-57-2 and KR128-19-1 were 5.33~6.06 ton/ha in rough rice yield. These high yield potential lines would be evaluated to adaptability in Cambodia, Laos, Myanmar and Vietnam during 2017.

  • The phytoplankton ecology of estuarine waters was investigated in the Kyeonggi Bay from May 1981 to September 1982 on monthly basis. In this study area, a total of 228 phytoplankton species was identified. Among these taxa, the most dominant species are diatoms in this area. Tychopelagic plankton occupies 40.4% of total species. The percentage of tychopelagic plankton density ranged from 10.2% in September to 92.7% in March of monthly standing crops. From late autumn to early spring, the percentage values are more than 72%. They play an important role from late autumn to early spring in this estuarine plandton community. These tychopelagic planktons are induced from benthic diatoms. Because the bottom shear stresses generated by the tides and winds are stronger than the adhesive and tractive force of benthic diatoms, most of benthic diatoms must be resuspended into tychopelagic suspensions during autumn and winter. Paralia sulcata is the most important tychopelagic plankton as an indicator species of water mixing in the eastern coastal area of Yellow Sea. This species seems to have even broader tolerance to the environmental stress than Skeletonema costalum, and tends to fill the gaps in winter, when the phytoplandton is relatively unsuccessful. Skeletonema costatum and Chaetoceros debilis are dominant in other seasons. Typical blooms of phytoplankton occur in spring and early autumn, The first bloom is started by Skeletonema costatum in early May, second peak is formed by various diatom population in September.

Water relations of plants under environmental stresses: role of aquaporins

  • Effects of low temperature ($8^{\circ}C$) on the hydraulic conductivity of young roots of a chilling-sensitive (cucumber; Cucumis sativus L.) and a chilling-resistant (figleaf gourd; Cucurbita ficifolia Bouche) crop have been measured at the levels of whole root systems (root hydraulic conductivity, $Lp_r$) and of individual cortical cells (cell hydraulic conductivity, Lp). In figleaf gourd, there was a reduction only in hydrostatic $Lp_r$ but not in osmotic $Lp_r$ suggesting that the activity of water channels was not much affected by low root temperature (LRT)treatment in this species. Changes in cell Lp in response to chilling and recovery were similar asroot level, although they were more intense at the root level. Roots of figleaf gourd recovered better from LRT treatment than those of cucumber. In figleaf gourd, recovery (both at the root and cell level) often resulted in Lp and $Lp_r$ values which were even bigger than the original, i.e. there was an overshoot in hydraulic conductivity. These effects were larger forosmotic (representing the cell-to-cell passage of water) than for hydrostatic $Lp_r$. After a short term (1 d) exposure to $8\;^{\circ}C$ followed by 1 d at $20\;^{\circ}C$, hydrostatic $Lp_r$ of cucumber nearly recovered and that of figleaf gourd still remained higher due to the overshoot. On the contrary, osmotic $Lp_r$ and cell Lp in both species remained high by a factor of 3 as compared to the control, possibly due to an increased activity of water channels. After pre-conditioning of roots at LRT, increased hydraulic conductivitywas completely inhibited by $HgCl_2$ at both the root and cell levels. Different from figleaf gourd, recovery from chilling was not complete in cucumber after longer exposure to LRT. It is concluded that at LRT, both changes in the activity of aquaporins and alterations of root anatomy determine the water uptake in both species. To better understand the aquaporin function in plants under various stress conditions, we examined the transgenic Arabidopsisand tobacco plants that constitutively overexpress ArabidopsisPIP1;4 or PIP2;5 under various abiotic stress conditions. No significant differences in growth rates were found between the transgenic and wild-type plants under favorable growth conditions. By contrast, overexpression of PIP1;4 or PIP2;5 had a negative effect on seed germination and seedling growth under drought stress, whereas it had a positive effect under cold stress and no effect under salt stress. Measurement of water transport by cell pressure probe revealed that these observed phenotypes under different stress conditions were closely correlated with the ability of water transport by each aquaporin in the transgenic plants. Together, our results demonstrate that PIP-type aquaporins play roles in seed germination, seedling growth, and stress response of Arabidopsis and tobacco plants under various stress conditions, and emphasize the importance of a single aquaporin-mediated water transport in these cellular processes.

Development of transgenic cucumbers expressing Arabidopsis Nit gene (애기장대 Nit유전자 발현 오이 형질전환체 개발)

  • To produce transgenic cucumber expressing Nit gene coffering abiotic resistance, the cotyledonary-node explants of cucumber (cv. Eunsung) were inoculated with A. tumefaciens transformed with pPZP211 or pCAMBIA2300 carrying Nit gene, that has cis-acting element involved in resistance to various abiotic environmental stresses. After co-cultivation, the procedures of selection, shoot initiation, shoot elongation, and plant regeneration were followed by cotyledonary-node transformation method (CTM, Jang et al. 2011). The putative transgenic plants were selected when shoots were grown to a length greater than 3 cm from the cotyledonary-node explants on selection medium supplemented with 100 mg/L paromomycin as a selectable agent. The confirmation of transgenic cucumber was based on the genomic PCR, Southern blot analysis, RT-PCR, and Northern blot analysis. A 105 shoots (4.12%) selected from the selection mediums were obtained from 2,547 explants inoculated. Of them, putative transgenic plants were only confirmed with 45 plants (1.77%) by genomic PCR analysis. Transgenic plants showed that the Nit genes integrated into each genome of 39 plants (1.53%) by Southern blot analysis, and the expression of gene integrated into cucumber genome was only confirmed at 6 plants (0.24%) by RT-PCR and Northern blot analysis. These results lead us to speculate that the genes were successfully integrated and expressed in each genome of transgenic cucumber.

Control of Bermudagrass (Cynodon dactylon) Causing Weedy in Zoysiagrass matrella Merr (금잔디에 잡초성 버뮤다그래스 방제)

  • Bermudagrass (Cynodon dactylon.) is one of the most difficult weedy species to control in turfgrass because it's high tolerant to various environmental and management stresses. This experiment was performed to find the integrated weed management including cultural practices to suppress bermudagrass in Zoysiagrass matrella (L) Merr. As results, two sequential applications of Fluazifop-P-butyl 0.05 ml $m^{-2}$ + Triclopyr-TEA 0.5 ml $m^{-2}$ and Fenoxaprop-P-ethyl 0.1 ml $m^{-2}$ + Triclopyr-TEA 0.5 ml $m^{-2}$ applied on 20 days intervals were evaluated the primary option for bermudagrass suppression and turfgrass injury was acceptable in zoysiagrass. In both treatments, turf injury was observed during 30days after the first application and almost recovered at 40days. While Fenoxaprop-Pethyl 0.1 ml $m^{-2}$ + Triclopyr-TEA 0.5 ml $m^{-22}$ were lightly phytotoxic to zoysiagrass in chlorophyll content test, there was no growth inhibition of zoysiagrass. Verticut practice (4 mm depth) just before herbicides application where zoyisagrass is contaminated with bermudagrass was not helpful to reduce turf injury in this experiment. However, alone verticut management was utilized to decrease about 12-14% bermudagrass population. Thus the application of Fenoxaprop-P-ethyl 0.1 ml $m^{-2}$ + Triclopyr-TEA 0.5 ml $m^{-2}$ which are permitted for turfgrass after zoysiagrass is perfectly recovered from turf injury by verticut practice should be utilized for bermudagrass reduction in zoysiagrass.

Miscanthus EST-originated Transcription Factor WRKY Expression in Response to Low Temperature in Warm-season Turfgrasses (억새 EST 정보 유래 전사요소 WRKY의 난지형 잔디의 저온 발현 반응성)

  • Whole genome transcriptomes from Miscanthus species were sequenced and analyzed, which provided 50 different types of transcription factor (TF) involving various developmental processes or environmental stresses. Among the explored TF, WRKY gene family was the major type and one of the WRKY genes, MSIR7180_WRKY4, induced under low temperature environment was selected to investigate how the Miscanthus-originated MSIR7180_WRKY4 TF responds when exposed to low temperature in four warm-season turfgrasses (Z. matrella 'Semil', bermudagrass, St. Augustinegrass, and seashore paspalum). The MSIR7180_WRKY4 was expressed higher during low temperature period in Bermudagrass, but the expression was enhanced in St. Augustinegrass. In contrast, the gene in 'Semil' cultivar was barely expressed and relatively less expressed, but repressed gradually in seashore paspalum, which seems to allow two turfgrasses stay-green longer in the fall season. The results indicate that bermudagrass and St. Augustinegrass adapt to low temperature quickly, but relative tolerance to low or cold temperature at the molecular level needs to be further investigated at different physiological stages and the corresponding genes systematically.
