식물의 긴비암호화 RNA들의 생물학적 기능 (The Biological Functions of Plant Long Noncoding RNAs)

  • 차세대 염기서열 분석기술의 발달로 대량의 전사수준의 단위체들이 발견되었는데, 이것들 중 대부분의 전사체들은 비암호화 RNA들이다. 이들 중 긴 비암호화 RNA (lncRNA)들은 200개 이상의 뉴클레오티드를 가지며 단백질로 번역이 되지 않는 기능적 RNA 분자들이다. 식물의 lncRNA들은 RNA Pol II, Pol III, Pol IV, Pol V에 의해 전사체가 만들어지고, 전사 후 이들 lncRNA들은 추가적인 splicing과 polyadenylation 과정이 일어난다. 식물의 lncRNA들은 그 발현수준이 매우 낮고, 조직 특이적으로 발현 되지만, 이들 lncRNA들은 외부자극에 의해 강한 발현이 유도된다. 환경스트레스를 포함한 각기 다른 외부자극에 의해 많은 식물 lncRNA들의 발현이 유도되었기 때문에 이들 lncRNA들은 식물의 다양한 생물학적 기능과 식물의 생장발달 과정에 있어 새로운 조절 인자로 고려되고 있다. 특히 후성유전학적인 유전자 억제, 크로마틴 변형, 타겟모방, 광형태형성, 단백질 재배치, 환경스트레스 반응, 병원균 감염등에 관련하여 기능을 한다. 또한 어떤 lncRNA들은 short RNA들의 전구체로 역할도 한다. 최근 식물에서 많은 lncRNA들이 분리 동정되었지만, 이들 lncRNA들의 생리학적인 기능에 대한 현재의 이해는 여전히 제한적이고, 이들의 세부적인 조절 메커니즘 연구는 지속적으로 이루어져야 할 것이다. 이 총설에서는 식물 lncRNA들의 생합성 및 조절 메커니즘과 현재까지 밝혀진 분자수준에서의 중요한 기능들을 요약 정리하였다.

Glutathione is the Major Defensive Mechanism against Oxidative Stress in Human Embryonic Stem Cell

  • Embryonic stem (ES) cells, derived from preimplantation embryo, are able to differentiate into various types of cells consisting the whole body, or pluripotency. In contrast, terminally differentiated cells do not usually alter their nature but frequently die or transform if they are exposed to inappropriate external stimulations. In addition to the plasticity, ES cells are expected to be different from terminally differentiated cells in very many ways, such as patterns of gene expressions, ability and response of the cells in confronting environmental stimulations, metabolism, and growth rate. As a model system to differentiate these two types of cells, human ES cells (MB03) and terminally differentiated cells (HeLa), we examined the ability of these two types of cells in confronting a severe oxidative insult, that is $H_2O$$_2$. Approximately 1$\times$10$^4$ cells were plated in 96 well plate and serum starved for overnight. The conditioned cells were exposed to a various concentration of $H_2O$$_2$ fur 24 hrs and loaded with neutral red (50$\mu\textrm{g}$/ml) for 4 hrs, washed with PBS for 2 min three times, and entrapped dye was dissolved out using acetic ethanol. Cytotoxicity was determined by reading the amount of dye in the medium using microplate reader. equipped with 575 nm filter. Relative amount of the dye entrapped within MB03 or HeLa were not significantly different when cells were exposed up to 0.4 mM $H_2O$$_2$. However, this sharply decreased down to 0.12% in HeLa cells when the cells were exposed to 0.8 mM $H_2O$$_2$, while it was approximately 54% in MB03 suggesting that this concentration of $H_2O$$_2$ is the defensive threshold for HeLa cells. The resistance to oxidative stimulation reversed, however, when cells were co-treated with BSO (L-buthionine- 〔S, R〕-sulfoximine) which chelates intracellular GSH. This result suggests that cellular GSH is the major defensive mechanism of human ES cells. Induction of enzymes involved in GSH metabolism and type of cell death is currently being studied.

  • 함정구조물은 함내 엔진 및 프로펠러 추진력의 환경진동에 노출되어 있다. 일반적으로 함상구조물은 함상진동규격인 MIL-STD-167-1A 에 따라 개발되고 있으며, 장기간 사용을 목적으로 하는 함상구조물의 진동에 대한 피로수명은 해석적 접근법과 진동실험을 통해 반드시 평가되야 한다. 본 논문에서는 함상구조재로 사용된 주조 알루미늄합금인 A356 의 피로강도를 14 S-N 법으로 평가하고, 구조물의 작용응력은 함상진동규격에 준하는 주파수응답해석을 통해 분석되었다. 최대등가응력의 주파수는 최대실험주파수에서 나타났으며, 함상장비의 진동피로수명은 누적손상법에 의해 평가되었다.

Isolation and Characterization of Cinnamoyl-CoA Reductase Gene from Panax ginseng C. A. Meyer

  • Cinnamoyl-CoA reductase (CCR, EC 1.2.1.44) catalyses the reduction of cinnamic acid CoA esters into their corresponding aldehydes, the first step of the phenylpropanoid pathway specially dedicated to monolignol biosynthesis. A cDNA clones encoding CCR have been isolated from Panax ginseng C.A. Meyer and its expression was investigated in response to abiotic stresses. The cDNA, designated PgCCR which is 865 nucleotides long and has an open reading frame of 590 bp with a deduced amino acid sequence of 176 residues. The PgCCR encoded protein possesses substantial homology with CCRs isolated and cloned from other sources; the highest identity (51.8%) was observed with CCR from Tomato (Lycopersicon esculentum). Under various stress conditions, expression patterns of the PgCCR were highly induced in adventitious and hairy roots by several abiotic stresses. These results indicated that PgCCR plays protective role against diverse environmental stresses.

저탄성 베이스플레이트 패드 적용에 따른 포장궤도 노반에서의 전달하중 저감에 관한 연구 (A Study on Transferred Load Reduction on Paved Track Roadbed with Low Elastic Base Plate Pad)

  • 포장궤도는 기존선의 유지보수 노력을 절감하기 위하여 개발되고 있다. 이 중 베이스플레이트 저탄성패드는 궤도 전체의 지지강성을 결정하는 구성품으로서 궤도의 응력-변위특성, 동적응답, 피로특성 등에 가장 큰 영향을 미친다. 따라서 저탄성패드의 적용은 궤도전체의 내구성과 안정성에 미치는 영향이 크다고 할 수 있다. 본 논문에서는 저탄성패드의 적용으로 인하여 획득되는 효과 중 노반에 전달되는 열차하중의 저감에 대한 연구를 수행하였다. 이를 위하여 패드스프링계수별 정적해석과 실물반복재하시험을 수행하고 그 결과를 비교 분석하여 저탄성패드의 하중 전달특성을 파악하였으며 전달하중 저감효과를 검토하였다.

  • 본 논문은 철근콘크리트와 프리스트레스트 콘크리트 보의 시간의존적 거동해석에 관한 내용으로 재령-보정 유효 탄생계수법에 토대를 두고 단면 내 힘의 평형관계와 변형의 Compatibility 조건을 사용한 반복해석을 통해 부재의 거동을 파악하였다. 콘크리트의 전 단면이 유효하고 부재가 선형 탄생체로 거동하며 강재의 효과는 단지 환산단면적 개념에 따라 해석시 강성만으로 보정되는 기존의 해석방법과는 달리 모든 재료가 고유의 성질을 유지할 수 있도록 함께 고려하며 균열의 영향도 동시에 고려함으로써 모든 하중조건에서 보다 정확한 결과를 얻을 수 있도록 하였다. 특히 예제해석을 통해 시간에 따른 각 재료의 응력 재분배, 장기처짐의 발생 등 시간의존적 거동에 영향을 미치는 여러 인자의 비교 검토와 균열의 영향분석을 하였으며 설계시 보다 정확하고 합리적으로 이들의 효과를 고려할 수 있도록 하였다.

Modeling of cyclic joint shear deformation contributions in RC beam-column connections to overall frame behavior

  • In seismic analysis of moment-resisting frames, beam-column connections are often modeled with rigid joint zones. However, it has been demonstrated that, in ductile reinforced concrete (RC) moment-resisting frames designed based on current codes (to say nothing of older non-ductile frames), the joint zones are in fact not rigid, but rather undergo significant shear deformations that contribute greatly to global drift. Therefore, the "rigid joint" assumption may result in misinterpretation of the global performance characteristics of frames and could consequently lead to miscalculation of strength and ductility demands on constituent frame members. The primary objective of this paper is to propose a rational method for estimating the hysteretic joint shear behavior of RC connections and for incorporating this behavior into frame analysis. The authors tested four RC edge beam-column-slab connection subassemblies subjected to earthquake-type lateral loading; hysteretic joint shear behavior is investigated based on these tests and other laboratory tests reported in the literature. An analytical scheme employing the modified compression field theory (MCFT) is developed to approximate joint shear stress vs. joint shear strain response. A connection model capable of explicitly considering hysteretic joint shear behavior is then formulated for nonlinear structural analysis. In the model, a joint is represented by rigid elements located along the joint edges and nonlinear rotational springs embedded in one of the four hinges linking adjacent rigid elements. The connection model is able to well represent the experimental hysteretic joint shear behavior and overall load-displacement response of connection subassemblies.

Upregulation of Heme Oxygenase-1 as an Adaptive Mechanism against Acrolein in RAW 264.7 Macrophages

  • Acrolein, a known toxin in cigarette smoke, is the most abundant electrophilic $\alpha$, $\beta$-unsaturated aldehyde to which humans are exposed in a variety of environmental pollutants, and is also product of lipid peroxidation. Increased unsaturated aldehyde levels and reduced antioxidant status plays a major role in the pathogenesis of various diseases such as diabetes, Alzheimer's and atherosclerosis. The findings reported here show that low concentrations of acrolein induce heme oxygenase-1 (HO-1) expression in RAW 264.7 macrophages. HO-1 induction by acrolein and signal pathways was measured using reverse transcription-polymerase chain reaction, Western blot and immunofluorescence staining analyses. Inhibition of extracellular signal-regulated kinase activity significantly attenuated the induction of HO-1 protein by acrolein, while suppression of Jun N-terminal kinase and p38 activity did not affect induction of HO-1 expression. Moreover, rottlerin, an inhibitor of protein kinase $\delta$, suppressed the upregulation of HO-1 protein production, possibly involving the interaction of NF-E2-related factor 2 (Nrf2), which has a key role as a HO-1 transcription factor. Acrolein elevated the nuclear translocation of Nrf2 in nuclear extraction. The results suggest that RAW 264.7 may protect against acrolein-mediated cellular damage via the upregulation of HO-1, which is an adaptive response to oxidative stress.

Comparative proteomic analysis of plant responses to sound waves in Arabidopsis

  • Environmental factors greatly influence the growth, development, and even genetic characteristics of plants. The mechanisms by which sound influences plant growth, however, remain obscure. Previously, our group reported that several genes were differentially regulated by specific frequenciesof sound treatmentusing a sound-treated subtractive library. In this study, we used a proteomic approach to investigate plant responses to sound waves in Arabidopsis. The plants were exposed to 250-Hz or 500-Hz sound waves, and total proteins were extracted from leaves 8 h and 24 h after treatment. Proteins extracted from leaves were subjected to 2-DE analysis. Thirty-eight spots were found to be differentially regulated in response to sound waves and were identified using MALDI-TOF MS and MALDI-TOF/TOF MS. The functions of the identified proteins were classified into photosynthesis, stress and defense, nitrogen metabolism, and carbohydrate metabolism. To the best of our knowledge, this is the first report on the analysis of protein changes in response to sound waves in Arabidopsis leaves. These findings provide a better understanding of the molecular basis of responses to sound waves in Arabidopsis.

Anti-aging Effect and Gene Expression Profiling of Aged Rats Treated with G. bimaculatus Extract

  • Extract from Gryllus bimaculatus crickets inhibits oxidation at the DNA level, with reduced production of 8-hydroxy-2'-deoxyguanosine (8-OHdG). Microarray analyses were performed with a rat 28K cDNA clone set array to identify the gene expression profiles of aged (10 months old) Wistar Kyoto rats treated for one month with 100 mg/kg G. bimaculatus ethanol extract to assess the effects. The extract produced a meaningful anti-edema effect, evident by the inhibition of creatinine phosphokinase activity. The weights of abdominal and ovarian adipose tissues were reduced and the proportion of unsaturated fatty acids in adipose tissues was increased in an extract dose-dependent manner. Compared with untreated control rats, rats treated with the extract displayed the upregulation of 1053 genes including Fas (tumor necrosis factor receptor superfamily, member 6), Amigo3 (adhesion molecule with an immunoglobulin-like domain), Reticulon 4, 3-hydroxy-3-methylglutaryl-coenzyme (Hmgcr; a reductase), related anti-fatigue (enzyme metabolism), and Rtn antioxidant, and the downregulation of 73 genes including Ugt2b (UDP glycosyltransferase 2 family), Early growth response 1, and Glycoprotein m6a. Data suggest that G. bimaculatus extract may have value in lessening the effects of aging, resulting in a differential gene expression pattern indicative of a marked stress response and lower expression of metabolic and biosynthetic genes.
