Protective Effect of Nitric Oxide against Oxidative Stress under UV-B Radiation in Maize Leaves (UV-B 조사시 옥수수 잎의 산화적 스트레스에 대한 Nitric Oxide의 보호효과)

  • The effect of nitric oxide (NO) on antioxidant system and protective mechanism against oxidative stress under UV-B radiation was investigated in leaves of maize (Zea mays L.) seedlings during 3 days growth period. UV-B irradiation caused a decrease of leaf biomass including leaf length, width and weight during growth. Application of NO donor, sodium nitroprusside (SNP), significantly alleviated UV-B stress induced growth suppression. NO donor permitted the survival of more green leaf tissue preventing chlorophyll content reduction and of higher quantum yield for photosystem II than in non-treated controls under UV-B stress, suggesting that NO has protective effect on chloroplast membrane in maize leaves. Flavonoids and anthocyanin, UV-B absorbing compounds, were significantly accumulated in the maize leaves upon UV-B exposure. Moreover, the increase of these compounds was intensified in the NO treated seedlings. UV-B treatment resulted in lipid peroxidation and induced accumulation of hydrogen peroxide ($H_2O_2$) in maize leaves, while NO donor prevented UV-B induced increase in the contents of malondialdehyde (MDA) and $H_2O_2$. These results demonstrate that NO serves as antioxidant agent able to scavenge $H_2O_2$ to protect plant cells from oxidative damage. The activities of two antioxidant enzymes that scavenge reactive oxygen species, catalase (CAT) and ascorbate peroxidase (APX) in maize leaves in the presence of NO donor under UV-B stress were higher than those under UV-B stress alone. Application of 2-(4-carboxyphenyl)-4, 4, 5, 5-tetramethylimidazoline-1-oxyl-3- oxide (PTIO), a specific NO scavenger, to the maize leaves arrested NO donor mediated protective effect on leaf growth, photosynthetic pigment and free radical scavenging activity. However, PTIO had little effect on maize leaves under UV-B stress compared with that of UV-B stress alone. $N^{\omega}$-nitro-L-arginine (LNNA), an inhibitor of nitric oxide synthase (NOS), significantly increased $H_2O_2$ and MDA accumulation and decreased antioxidant enzyme activities in maize leaves under UV-B stress. This demonstrates that NOS inhibitor LNNA has opposite effects on oxidative resistance. From these results it is suggested that NO might act as a signal in activating active oxygen scavenging system that protects plants from oxidative stress induced by UV-B radiation and thus confer UV-B tolerance.

Herbicidal Property and Soil Behavior of a New Herbicide, Azimsulfuron (신제초제(新除草劑) Azimsulfuron의 제초활성(除草活性)과 토양중(土壤中) 행동(行動))

  • Azimsulfuron [1H-pyrazole-5-sulfonamide,N-(((4,6-dimethoxy-pyridine-2-yl-aminocarbonyl-4-(2-methyl-2H-tetrazole-5-yl)] is a new sulfonamide herbicide that selectively controls a wide range of weeds in lowland rice (Oryza sativa). It effectively controlled Cyperus serotinus, Eleocharis kuroguwai, Sagittaria pygmaea, S. trifolia, and Scirpus juncoides at 7.5 - 30 g ai/ha. In the tolerance test on grasses carried out in a nutrient solution containing 0.3 - 30 ppm of azimsulfuron, greater inhibition occurred in roots of both rice and barnyardgrass (Echinochloa crus galli) than in shoots. However, rice root was approximately 5-fold more tolerant than that of barnyardgrass. The downward movements as determined by 50% growth inhibition of S. juncoides were 4-cm in clay loam and 6.5-cm in sandy loam soil with 3-cm/day leaching for 3 days. When incubated at 20 and $30^{\circ}C$, the residual effect in clay loam soil lasted for 30 and 21 days, respectively. In a soil column applied at 15 g ai/ha of azimsulfuron followed by 3-cm/day leaching for 3 days, dry weights of S. trifolia emerging at 5, 10, and 15-cm depth were reduced to 87, 85, and 79% of the corresponding untreated control, respectively. Susceptibility of S. trifolia to azimsulfuron did not greatly vary with the emergence depth.

Changes in Microcystin Production in Microcystis aeruginosa Exposed to Different Concentrations of Filtered Water from Phytoplanktivorous and Omnivorous Fish (잡식성 및 플랑크톤 섭식어류의 간접노출 강도가 Microcystis aeruginosa의 microcystin 함량변화에 미치는 영향)

  • This study was to evaluate microcystin production by Microcystis aeruginosa in response to three different levels of indirect (0, 10, 50% of fish cultured media filtrate; control, FCMF1 and FCMF2) exposures to omnivorous and planktivorous fish (Carassius gibelio langsdorfi and Hypophthalmichthys molitrix, CCMF and HCMF, repectively). The cell biomass, intracellular microcystin (MC) and extracellular MC were measured everyday. The intracellular MC contents of all treatments were significantly increased than the controls (CCMF1, P=0.015; CCMF2, P<0.001; HCMF1, P<0.001; HCMF2, P<0.001). The intracellular MC contents of M. aeruginosa were significantly higher in CCMF2 than in CCMF1 (P=(0.023), Those of M, aeruginosa in HCMF2 were significantly higher than that in HCMF1 (P<0.001). The extracellular MC contents were not significantly different between control and CCMFs but those of M, aeruginosa in HCMF1 and HCMF2 were significantly higher than that in control (HCMF1, P=0.003; HCMF2, P<0.001). This study strongly supports that induced-defensive MC production (intra and extracellular MC) of potentially toxic cyanobacteria in response to kairomone concentration and this results can consider the biomanipulation of eutrophic waters as well as an information concerning strategies for recovering eutrophic waters.

Evaluation of Biological Activity on Hawthorn Tree (Crataegus pinnatifida) Extracts (산사나무 추출물의 생리활성 평가)

  • The wood and bark of Hawthorn tree (Crataegus pinnatifida Bunge) were immersed with 70% aqueous acetone for 3 days. After filtering, the wood extracts were fractionated with n-hexane, chloroform ($CHCl_3$), n-butanol (n-BuOH) and $H_2O$, and the bark extracts were fractionated with n-hexane, $CHCl_3$, ethylacetate (EtOAc) and $H_2O$. Then antioxidative and anti-inflammatory activities were evaluated on each fraction. Antioxidative activity indicated high activity in the n-butanol soluble fraction of wood and in the EtOAc soluble fraction of bark. Especially, the EtOAc soluble fraction of bark showed much higher antioxidative value compared to the controls, buthylated hydroxytoluene (BHT) and ${\alpha}$-tocopherol. In the anti-inflammatory activity, all of the tested fractions except the $H_2O$ soluble fraction of wood showed high inhibitory effect on nitric oxide (NO) production. Based on the above results, the extracts of hawthorn tree may be applied for one of the natural biomass sources that can be used as an antioxidant and an anti-inflammatory substance.

Effect of Antemortem and Postmortem Environmental Temperatures on Biochemical Metabolism and Tenderness in Chicken Muscels (도계처리 전후의 환경온도가 계육의 생화학적 대사 및 연도에 미치는 영향)

The effects of heat stress (38$^{\circ}C$), cold stress (4$^{\circ}C$) and extreme cold stress (-20$^{\circ}C$) before slaughter on the tenderness and postmortem glycolysis if the excised chicken breast muscle were studied Heat stress significantly (p 0.05) increased the toughness of breast muscle. Though statistically not significant, cold stress also adversely affected the tenderness. The heat-stressed birds showed higher zero hr glycogen higher zero hr pH and significantly (p 0.05) love. ultimate pH then the controls. The cold-stressed birds showed intermediate values in these parameters. Highly significant correlations. were observed between shear value and each of these three parameters. Glycolysis rate ana final moisture content were minor factors which affected the muscle tenderness to a limited extent. The slightly elevated lactate-dehydrogenase and creatine phosphokinase activities in serum and breast muscle of stressed birds failed to account for any variations in tenderness. 2. Chicken breast and thigh muscles were subjected to different environmental temperatures to determine if the phenomenon of cold shortening exists in chicken muscle. For both breast and thigh muscles, minimum shortening was observed in the 4-10$^{\circ}C$t range. Muscles held at 0$^{\circ}C$ showed a slightly higher extent of shortening than at 4$^{\circ}C$; where as muscles held at above 20$^{\circ}C$ showed a severe shortening effect. It was concluded that no apparent cold shortening was detected in chicken muscle except at 0$^{\circ}C$ and even at 0$^{\circ}C$ and even at 0$^{\circ}C$ the extent of shortening was of a small magnitude compared to bovine muscles. Since high temperature induces a much greater shortening, muscle temperature must be lowered to below 20$^{\circ}C$ as early as possible to prevent excessive muse]e shortening.

  • There are $10{\sim}30%$ polyphenol and $2{\sim}4%$ caffeine in green tea. Caffeine is a kind of alkaloid containing nitrogen which cause stimulation, impatience, headache, insomnia, low birth weight infant. Because of these negative effect, decaffeined beverage came out and decaffeined coffee already have a big market since 1970s. Having proving the physiologic functions of green tea, high consumption of coffee is shifting to green tea. Because of the carcinogenic effect of the organic solvents, decaffeine processing with supercritical carbon dioxide has industrialized and have an advantage in environment-friendly and minimized flavor loss. Decaffeined green tea using supercritical carbon dioxide is considered to be safe but there are not enough study, We investigated the chromosome aberration test with mammalian cell line, CHL. When the cells were treated with 5000, 2000, 1000 ${\mu}g/ml$ and compared with the negative controls, there were no significant (P>0.05) increased chromosome aberration. Same results was observed when adding S9 mixture or not. As a result, water extract of decaffeined green tea using supercritical carbon dioxide does not induce chromosome aberration.

Regulation of Chilling Tolerance in Rice Seedlings by Plant Hormones

  • Since the major important factors limiting plant growth and crop productivity are environmental stresses, of which low temperature is the most serious. It has been well known that many physiological processes are alterant in response to the environmental stress. With regard to the relationship between plant hormones and the regulation of chilling tolerance in rice seedlings, the major physiological roles of plant hormones: abscisic acid, ethylene and polyamines are evaluated and discussed in this paper. Rice seedlings were grown in culture solution to examine the effect of such plant hormones on physiological characters related to chilling tolerance and also to compare the different responses among tested cultivars. Intact seedlings about 14 day-old were chilled at conditions of 5$^{\circ}C$ and 80% relative humidity for various period. Cis-(+)-ABA content was measured by the indirect ELISA technique. Polyamine content and ethylene production in leaves were determined by means of HPLC and GC respectively. Chilling damage of seedlings was evaluated by electrolyte leakage, TTC viability assay or servival test. Our experiment results described here demonstrated the physiological functions of ABA, ethylene, and polyamines related to the regulation of chilling tolerance in rice seedlings. Levels of cis-(+)-ABA in leaves or xylem sap of rice seedlings increased rapidly in response to 5$^{\circ}C$ treatment. The tolerant cultivars had significant higher level of endogenous ABA than the sensitive ones. The ($\pm$)-ABA pretreatment for 48 h increased the chilling tolerance of the sensitive indica cultivar. One possible function of abscisic acid is the adjustment of plants to avoid chilling-induced water stress. Accumulation of proline and other compatible solutes is assumed to be another factor in the prevention of chilling injuies by abscisic acid. In addition, the expression of ABA-responsive gene is reported in some plants and may be involving in the acclimation to low temperature. Ethylene and its immediate precusor, 1-amincyclopropane-1-carboxylic acid(ACC) increased significantly after 5$^{\circ}C$ treatment. The activity of ACC synthase which converts S-adenosylmethionine (SAM) to ACC enhanced earlier than the increase of ethylene and ACC. Low temperature increased ACC synthase activity, whereas prolonged chilling treatment damaged the conversion of ACC to ethylene. It was shown that application of Ethphon was beneficial to recovering from chilling injury in rice seedlings. However, the physiological functions of chilling-induced ethylene are still unclear. Polyamines are thought to be a potential plant hormone and may be involving in the regulation of chilling response. Results indicated that chilling treatment induced a remarkable increase of polyamines, especially putrescine content in rice seedlings. The relative higher putrescine content was found in chilling-tolerant cultivar and the maximal level of enhanced putrescine in shoot of chilling cultivar(TNG. 67) was about 8 folds of controls at two days after chilling. The accumulation of polyamines may protect membrane structure or buffer ionic imbalance from chilling damage. Stress physiology is a rapidly expanding field. Plant growth regulators that improve tolerance to low temperature may affect stress protein production. The molecular or gene approaches will help us to elucidate the functions of plant hormones related to the regulation of chilling tolerance in plants in the near future.

Development of a PCR Assay for Detection of the Protozoan Parasite Perkinsus (PCR 기법을 이용한 바지락포자충 Perkinsus 진단 기술개발)

  • Detection of protozoan parasites Perkinsus sp. and P. atlanticus was developed in this study using a specific polymerase chain reaction (PCR) to diagnose the presence of those organisms that causes extensive mortalities of marine shellfishes. The PCR was conducted together with fluid thioglycollate medium (FTM) method and 2 M NaOH lysis method. For the test, Manila clams, Ruditapes philippinarum, were collected from four coastal locations in Korea including Wando Island, Gimnyeong, Sungsan and Sogwipo in Jeju. In addition, trophozites of Perkinsus sp. cultivated in vitro and the granular ark clam, Tegillarca granosa, taken from Gangjin on the south coast of Korea, were used as positive and negative controls, respectively. Expected DNA bands were detected in the samples from Wando Island, Sungsan and the in vitro cultured Perkinsus sp. when the probes specific for the genus Perkinsus and P. atlanticus were used. The samples were also positively diagnosed by the FTM and 2 M NaOH methods. In contrast, the Manila clams from Gimnyeong and Sogwipo, and the granular arks clams from Gangjin showed no detectable signs of infection with the PCR, the FTM method and the 2 M NaOH lysis method. On the other hand, being amplified by p. atlanticus specific primer, it is suggested that the protozoan parasite Perkinsus sp. found in the Korean Manila clam is P. atlanticus. Finally the PCR- based assay developed in the present study can be used in detection of Perkinsus infection and discrimination of Peykinsus species in quarantine stations or laboratories due to the high sensitivity and specificity as well as its rapid detection.

Toxicity Evaluation of Burkholderia pyrrocinia CAB08106-4 in Cyprinus carpio and Daphnia magna (Burkholderia pyrrocinia CAB08106-4 원제가 잉어 및 물벼룩에 미치는 영향 연구)

  • Bukholderia pyrrocinia CAB08106-4 has an anti-fungal effect on Garlic White Rot caused by Sclereotium cepivorum and Sclereotium sp. It is environmentally friendly microbial product that prevents and controls a variety of phytopathogens including Garlic White Rot caused by Sclereotium cepivorum and Sclereotium sp. The aim of this study was to assess the environmental toxicity using Cyprinus carpio and Daphnia magna. Bukholderia pyrrocinia CAB08106-4 ($1.0{\times}10^9cfu/mL$) was adminatrated to Cyprinus carpio and Daphnia magna according to the toxicity test guideline for peciticide. $LC_{50}$ of Bukholderia pyrrocinia CAB08106-4 is over $6.67{\times}10^4cfu/mL$ in Cyprinus carpio and Daphnia magna and no adverse effect was observed. Based on these results, we concluded that Bukholderia pyrrocinia CAB08106-4 has no toxiciy for Cyprinus carpio and Daphnia magna.

Risk Factors for Cervical Cancer in Rural Areas of Wuhan China: a Matched Case-control Study

  • Cervical cancer is a serious public health problem in developing countries. We investigated possible risk factors for cervical cancer in rural areas of Wuhan China using a matched case-control study with 33 women diagnosed with cervical cancer and 132 healthy women selected from the same area as matched controls. A questionnaire, which included questions about general demography conditions, environmental and genetic factors, the first sexual intercourse, first marriage age, age at first pregnancy, pregnancy first child's age, female personal health history, social psychological factors, dietary habits, smoking and alcohol status and other living habits was presented to all participants. At the same time, HPV infection of every participant was examined in laboratory testing. Results showed HPV infection (P<0.000, OR=23.4) and pregnancy first child's age (P<0.000, OR=13.1) to be risk factors for cervical cancer. Menopause (P=0.003, OR=0.073) was a protective factor against cervical cancer. However, there was no indication of associations of environmental (drinking water, insecticide, disinfectant) genetic (cancer family history), or life-style factors (smoking status, alcohol status, physical training, sleep quality), including dietary habits (intake of fruit and vegetable, meat, fried food, bean products and pickled food) or social psychological factors with cervical cancer. The results suggest that the risk of cervical cancer in Chinese rural women may be associated with HPV infection, menopause and the pregnancy first child's age.
