Non-Linear Behavior Analysis for Stratospheric Airship Envelope (성층권 비행선 기낭 막재료에 대한 비선형 거동 연구)

  • In this paper, geometrically non-linear finite element analyses were performed to study the mechanical behavior of the material system of the envelope of stratospheric airships. The microstructure of the load-bearing plain weave layer was identified and modeled. The Updated Lagrangian formulation was employed to consider the geometric non-linearity as well as the induced structural non-linearity for the fiber tows. The stress-strain behavior was predicted and the effective elastic modulus was calculated by numerical experiments. It was found the non-linear stress-strain curves were largely different from those by linear analysis. And non-linear elastic moduli were much higher than linear elastic moduli. The difference was more distinguishable when the tow waviness ratio was smaller.

  • The major glycoproteins of bovine gammaherpesvirus 4 (BoHV-4) are gB, gH, gM, gL, and gp180 with gB, gH, and gp180 being the most glycosylated. These glycoproteins participate in cell binding while some act as neutralization targets. Glycosylation of these envelope proteins may be involved in virion protection against neutralization by antibodies. In infected cattle, BoHV-4 induces an immune response characterized by low neutralizing antibody levels or an absence of such antibodies. Therefore, virus seroneutralization in vitro cannot always be easily demonstrated. The aim of this study was to evaluate the neutralizing capacity of 2 Argentine BoHV-4 strains and to associate those findings with the gene expression profiles of the major envelope glycoproteins. Expression of genes coding for the envelope glycoproteins occurred earlier in cells infected with isolate 10/154 than in cells infected with strain 07/435, demonstrating a distinct difference between the strains. Differences in serological response can be attributed to differences in the expression of antigenic proteins or to post-translational modifications that mask neutralizing epitopes. Strain 07/435 induced significantly high titers of neutralizing antibodies in several animal species in addition to bovines. The most relevant serological differences were observed in adult animals. This is the first comprehensive analysis of the expression kinetics of genes coding for BoHV-4 glycoproteins in 2 Argentine strains (genotypes 1 and 2). The results further elucidate the BoHV-4 life cycle and may also help determine the genetic variability of the strains circulating in Argentina.

  • The moment-curvature envelope describes the changes in the flexural capacity with deformation during a nonlinear analysis. Therefore, the moment-curvature analysis for reinforced concrete columns, indicating the available flexural strength and ductility, can be conducted providing the stress-strain relation for the concrete and steel are known. The moments and curvatures associated with increasing flexural deformations of the column may be computed for various column axial loads by incrementing the curvature and satisfying the requirements of strain compatibility and equilibrium of forces. Clearly it is important to have accurate information concerning the complete stress-strain curve of confined high-strength concrete in order to conduct reliable moment-curvature analysis to assess the ductility available from high-strength columns. However, it is not easy to explicitly characterize the mechanical behavior of confined high-strength concrete because of various parameter values, such as the confinement type of rectilinear ties, the compressive strength of concrete, the volumetric ratio and strength of rectangular ties, etc. So a stress-strain confinement model is developed which can simulate a complete inelastic moment-curvature relations of a high-strength reinforced concrete column

  • The neurotropic psudorabies virus(PRV) to replicate within neurons is very useful pathogen for neuronal tracing. I carried out this study to investigate the parametric analysis on the viral infection in the rat circadian control center with two genetically engineered strains out of PRV. The two strains are isogenic with the attenuated Bartha strain of PRV ; in one strain a lacZ reporter gene was inserted into the gC locus (PRV-BaBlu ; $4.75\times10^8pfu/ml$) and the other strain contained a PRV envelope glycoprotein gene(PRV-D ; $2.5\times10^8pfu/ml$) theat is absent in PRV-BaBlu. simultaneous or temporally separated sequential injection of$2{\mu}l$ of each strain into the vetreous body of eye produced a course of transsynaptic infection of retinohypothalamic circuitry. The results were as follows; 1. PRV-BaBlu and PRV-D infected the suprachiasmatic nucleus in hypothalamus and intergeniculate leaflet in lateral geniculate nucleus of thalamus. 2. The rate of PRV infection was dependent upon PRV strain. 3. Pre-infected neurons by PRV-D were interfered with the replication of PRV-BaBlu. 4. Dual injection of PRV-D and PRV-BaBlu showed more virulent than the parental strain.

Stress-Strain Response and Fracture of a Plain Concrete in Biaxial Loading (이축 하중을 받는 콘크리트의 응력-변형률 응답 및 파괴)

  • In this paper the biaxial failure criteria and stress-strain response for plain concrete are studied under uniaxial and biaxial stress(compression-compression, compression-tension, and tension-tension combined stress). The concrete specimens of a square plate type are used for uniaxial and biaxial loading. The experimental data indicate that the strength of concrete under biaxial compression, f2/fl=-l/-1, is 17 percent larger than under uniaxial compression and the poisson's ratio of concrete is 0.1745. On the base of the results, a biaxial failure envelope for plain concrete that the uniaxial strength is 398kgf/$cm^{2}$ are developed. The biaxial failure behaviors for three biaxial loading areas are also plotted respectively. In addition, the characteristics of stress-strain response under biaxial compression are compared and verified with the experimental and analytical results.

  • Polymer-Impregnated Concrete(PIC) can be considered composite material of concrete and polymer and has superior properties compared to conventional cement concrete, such as strength, stiffness, toughness, durability, water-proofing, chemical resistance. However, so far, the usage of PIC has been limited to repairing materials and non-structural applications, due to the lack of the design criteria and the analytical model to determine structural behavior. The objective of this study is to define the stress-strain response and strength characteristics of PIC in uniaxial and various biaxial compressive loading. On the bases of experimental results, general stress-strain relation, biaxial failure envelope and strength evaluation formular of PIC made with normal aggregate and methylmethacrylate(MMA) are proposed.

  • The vegetative nuclear divisions in hyphae and chromosome numbers were studied with the aid of Giemsa-HCl techniques from 10 strains of Fusarium oxysporum. The entire nuclear division process occurred within an intact nuclear envelope like other fungus. The results confirmed that 2 strains(F. oxysporum S Hongchun D2, F. oxysporum S Jinyang 4) were n=4; 3 strains(F. oxysporum f. sp. lini KFCC 32585, F. oxysporum f. sp. melongenae KFCC 34743 and F. oxysporum f. sp. raphani) n=5; 2 strains(F. oxysporum f. sp. vasinfectum, and F. oxysporum f. sp. mori KFCC 34742) n=6; 3 strains(F. oxysporum f. sp. cucumerium, F. oxysporum f. sp.niveum, and F. oxysporum f. sp. pisi) n=7.

Characterization of Bacillus thuringiensis Having Insecticidal Effects Against Larvae of Musca domestica

  • The entomopathogenic bacterium Bacillus thuringiensis is the most widely used biopesticide. Insecticidal proteins, coded by genes located in plasmids, form typical parasporal, crystalline inclusions during sporulation. We isolated a Bacillus thuringiensis strain having insecticidal activity against larvae of the house fly (M. domestica) from the soils at a pig farm in Korea, and named it Bacillus thuringiensis SM. The culture filtrate from Bacillus thuringiensis SM showed strong lethality (83.3%) against M. domestica larvae. The parasporal crystal is enclosed within the spores' outermost envelope, as determined by transmission electron microscopy, and exhibited a bipyramidal form. The crystal proteins of strain SM consisted of five proteins with molecular weights of approximately ~130, ~80, ~68, ~42, and ~27 kDa on a 10% SDS-PAGE (major band, a size characteristic of Cry protein). Examination of antibiotic resistance revealed that the strain SM showed multiple resistant. The strain SM had at least three different plasmids with sizes of 6.6, 9.3, and 54 kb. Polymerase chain reactions (PCRs) revealed the presence of cry1, cry4A2, and cry11A1 genes in the strain SM. The cry1 gene profile of the strain SM appeared in the three respective products of 487 bp [cry1A(c)], 414 bp [cry1D], and 238 bp [cry1A(b)]. However, the strain SM has not shown the cry4A2 md cry11A1 genes. In in vivo toxicity assays, the strain SM showed high toxicity on fly larvae (M. domestic) [with $LC_{50}$ of 4.2 mg/ml, $LC_{90}$ of 8.2 mg/ml].

Antigenic and Genetic Differences between the Prototype Nakayama-NIH Strain and Korean Strains of Japanese Encephalitis Virus (일본뇌염 바이러스 Nakayama-NIH주와 국내에서 분리된 일본 뇌염 바이러스주의 유전적 차이 및 항원성 차이의 조사)

  • The characterization of the 5 Korean isolates (K96P10, K94P05, K91P55, K87P39, and K82P01) of Japanese encephalitis virus (JEV) was compared with JE virus prototype Nakayama-NIH (NKY-NIH) using prM/M and envelope gene sequences of the JEV genome and phylogenetic analysis. The antigenic analysis of these viruses were done by the cross-hamagglutination inhibition (HI) test using polyclonal antibodies against Korean isolates and NKY-NIH. The sequence homology of the Korean isolates and NKY-NIH ranged between 87.4 % - 95.6 % at the nucleotide level and between 98.2 % - 97.2 % at the amino acid level over the E nucleotides compared. Alignment of E protein amino acid sequences revealed that residue positions E89, E129, E221, E244, E327, E366, E459, and E477 characterized the Korean strains. According to phylogenetic analysis bases on the E nucleotide, there are at least 2 genetic types of JEV existing in Korea and Korean strains were distinct from NKY-NIH. However, the cross HI test results of all the Korean isolates were serologically no different from NKY-NIH strain.
