Optimal Protocol for Enumeration of Attached Bacteria on Glass Slides

  • In examining bacterial growth on glass surfaces immersed in sea water, we found serious differences between enumeration methods. Therefore, we compared various methods and found sonication and direct count methods were superior to other methods. Since the direct count method was not suitable for long-term investigation, we chose the sonication method and confirmed that sonication periods 8 times for 30 seconds was optimal for the detachment of bacteria from glass surfaces.

  • 스마트폰이 대중화되면서 안드로이드 애플리케이션의 보안문제가 대두되고 있다. PC환경의 소프트웨어는 개발단계에서부터 시큐어코딩을 통해 안전성을 확보하고 있으나 안드로이드 애플리케이션의 경우는 연구가 더 필요한 상황이다. 본 논문에서는 CWE(Common Weakness Enumeration)의 취약점 분류 체계와 CAPEC(Common Attack Pattern Enumeration and Classification)의 공격 패턴 분류 체계와 CERT(Computer Emergency Response Team)의 취약점 발생 예방을 위한 정책들을 통해 안드로이드 애플리케이션의 최신화된 취약점 목록을 도출하고 카테고리별로 분류하여 취약점을 효율적으로 분석하는 기법을 제안한다.

  • A novel approach for analysis of criticality with respect to path and to activity in networks with fuzzy activity durations is proposed. After recalling the Yager ranking method, the relative degree of criticality of activities and paths are defined. An efficient algorithm based on path enumeration to compute the relative degree of criticality of activities and paths in networks with fuzzy durations is proposed. Examples of former researches are employed to validate the proposed approach. The proposed algorithm has been tested on real world project networks and experimental results have shown that the algorithm can calculate the relative degree of criticality of activities and paths in a reasonable time.

Comparison of Bifidobacteria Selective Media for the Detection of Bifidobacteria in Korean Commercial Fermented Milk Products

  • This study was carried out to compare the efficacy and selectivity of TOS and BS media for enumeration of bifidobacteria in commercial fermented milk products. First, bifidobacteria was isolated from 20 fermented milk products, and all isolated bifidobacteria were identified by genomic technology as Bifidobacterium lactis. The two media significantly differed from each other with regard to the recovery of B. lactis, that is, the recovery of this organism was as much as 6 logs lower on BS medium than on TOS. When the concentration of BS solution (mixture of paromomycin sulfate, neomycin, sodium propionate, and lithium chloride) used in BS medium was reduced to 50% (BS50), a relatively high percentage recovery of bifidobacteria from pure cultures was achieved. Susceptibility tests to antibiotics and tests for selective agents for the isolated bifidobacteria and lactic acid bacteria were conducted. The BS solution inhibited some lactic acid bacteria and Bifidobacterium species, while mupirocin (MU) suppressed the growth of all tested lactic acid bacteria but not Bifidobacterium. As compared with BS50 medium, TOS with or without MU showed good bifidobacteria recovery and readily distinguishable colonies; in particular, TOS supplemented with MU had a high selectivity for bifidobacteria. In conclusion, all results suggested that TOS medium with or without MU was found to be suitable for selective enumeration of bifidobacteria from mixed cultures in fermented milk, and better in that capacity than BS medium.

Design of Composite Laminates Using Enumeration Method (나열법을 이용한 복합재 적층판 설계)

  • Fiber-reinforced composite laminates have high specific stiffness and strength and are expected to be useful for weight reduction in weight-sensitive industries, such as automotive and aerospace. However, designing composite laminates is often dependent on designer's experience and intuition because of difficulties in determining the number of plies and stacking sequence, which tends to lead to over-design. In this study, optimal design of composite laminates was performed to minimize weight, while withstanding the given load. Based on the enumeration method, all combinations of stacking sequence satisfying the design guideline for composite laminates were considered. Composite laminates were discretized into panels. Optimal number of plies and stacking sequence for each panel were determined considering local load on each panel and contiguity across adjacent panels. Failure index from Tsai-Wu criteria was optimized for strength and buckling analysis was performed for compressive load. Stacking angles of 0, ±45 and 90° were used.

Comparison of an Automated Technique TEMPO with Direct Plate Count Method for the Enumeration of Staphylococcus aureus (황색포도상구균의 TEMPO STA와 표준 평판 배지를 이용한 정량분석법 비교)

  • In this study the automated technique TEMPO STA method, a new automated enumeration based on MPN method, was compared to the direct plate count method by testing various ready-to-eat compound food products. Artificially inoculated food samples with Staphylococcus aureus - Bibimbap, Bibimnaengmyeon, Mulnaengmyeon, Kimmaki, Salad, Japchae, and Sushi- were tested. Statistical analysis of the results showed above 99% in agreement between the two enumeration methods. Also, we monitored 466 various ready-to-eat compound food products samples. The measure uncertainties of the two methods is also familiar, while TEMPO STA automated method is simpler, less time-consuming, more effective and more stable.

  • This study proposes a concept of Stepwise Transfer Coefficient(STC) which implies greater transfer cost with increasing the number of transfers. Thus, the public transport information system provides the choice sets of travel routes by the consideration of not only transportation time but also the optimum number of transfers. However, path choice problems that involve STC are found to include non additive cost, which requires additional route enumeration works. Discussions on route enumeration in actual transportation networks is very complicated, thereby warranting a theoretical examination of route search considering STC. From these points of view, this study results in a probability based transit trip assignment model including STC. This research also uses incoming link based entire route deletion method. The entire route deletion method proposed herein simplifies construction of an aggregation of possible routes by theoretically supporting the process of enumeration of the different routes from origin to destination. Conclusively, the STC reflected route based logit model is proposed as a public transportation transit trip assignment model.

Rapid Enumeration of Salmonella spp. in Contaminated Pork Meat Using Competitive PCR (Competitive PCR을 이용한 돼지고기 오염 살모넬라의 신속 계수)

  • In this study, the competitive polymerase chain reaction (cPCR) was used to develop a direct enumeration method of Salmonella spp. in pork meat. After comparing three DNA extraction methods, the modified guanidine thiocyanate-phenol-chloroform method was chosen for Salmonella DNA extraction in artificially inoculated pork meat. The previously reported 284-bp invA gene (Rahn et al. Mol. Cell. Probes 1992) was tested for specificity, and 57 Salmonella strains and 24 non-Salmonella strains were evaluated. All Salmonella strains tested were invA positive, and all non-Salmonella strains produced no false positive amplification products. The detection limit achieved was as low as 1,460 colony-forming units (cfu) per 0.1g of pork meat. For cPCR, the invA gene, which features a 82 bp-deletion, was cloned in the pGEM-4Z vector. A known amount of competitor DNA, which has the same primer binding sites, was co-amplified with Salmonella chromosomal DNA from the artificially inoculated pork meat. The cell-number determined by cPCR was approximately equal to the cfu from the most probable number (MPN) method. Finally, the whole procedure took only 5 hr.
