혐기성 PCE 탈염소화 미생물 농화 배양 및 미생물 군집 해석

  • An anaerobic PCE(tetrachloroethylene) dechlorinating bacterial culture from a landfill soil was enriched and characterized. The enrichment culture could dechlorinate 60$\mu$mol/$m\ell$ of PCE during a month of incubation and cis-DCE(cis-dichloroethylene) was observed as a main product of PCE dechlorination. Microbial analysis of the dechlorinating enrichment culture by rising PCR-DGGE (Polymerase chain reaction-Denaturing gradient gel electrophoresis) method showed that at least three microorganisms were related to the anaerobic PCE dechlorination.

Stable Isotope Labeling of Proteins in Mammalian Cells

  • Stable isotope enrichment in proteins is necessary for high-resolution nuclear magnetic resonance (NMR) experiments. Although methods for 13C, 15N and 2H-enrichment in prokaryotic cells are well established, full processing and correct folding of complex protein systems require higher organisms as the expression host. In the present study, we review recent efforts to enrich stable isotopes in mammalian cells for protein NMR studies.

  • For ${\sim}240$ nearby stars their age and mass were determined and kinematic parameters determined for 362 stars, applying Woolley's three-dimensional potential. Metallicity and kinematic parameters of these stars were correlated with their age, suggesting the slow collapse ($t{\gtrsim}a$ few billion years) of the Galaxy and the initial rapid enrichment in metal abundance (${\Delta}Z{\approx}1/3Z_1$(present) for ${\sim}4{\times}10^8$ yrs). The late slow enrichment rate is given by $d(Z/Z_{\odot})/dt=5.9{\sim}7.0{\pm}3.4$ per Gyr.

Preconcentration of Cd by Continuous Hydroxide Precipitation-Dissolution in Atomic Emission Spectrometry

  • On-line preconcentration by direct precipitation with hydroxide has been developed and applied for the analysis of Cd in Inductively Coupled Plasma Atomic Emission Spectrometry. Cadmium is continuously precipitated with hydroxide and dissolved by nitric acid in on-line mode. Currently, the enrichment factor is more than 90 times for 20.0 mL of sample and could be further increased very easily. For a large sample throughput, 1.0 mL of sample loop is used and the enrichment factor is 4.5 with the sampling speed of 15/hr. The method has been applied to the analysis of NIST reference sample and has yielded good results with the certified value.

Development of an Enrichment Culture Growing at Low Temperature used for Ensiling Rice Straw

  • To speed up the conversion of rice straw into feeds in a low-temperature region, a start culture used for ensiling rice straw at low temperature was selected by continuous enrichment cultivation. During the selection, the microbial source for enrichment was rice straw and soil from two places in Northeast China. Lab-scale rice straw fermentation at $10^{\circ}C$ verified, compared with the commercial inoculant, that the selected start culture lowered the pH of the fermented rice straw more rapidly and produced more lactic acid. The results from denatured gradient gel eletrophoresis showed that the selected start culture could colonize into the rice straw fermentation system. To analyze the composition of the culture, a 16S rRNA gene clone library was constructed. Sequencing results showed that the culture mainly consisted of two bacterial species. One (A) belonged to Lactobacillus and another (B) belonged to Leuconostoc. To make clear the roles of composition microbes in the fermented system, quantitative PCR was used. For species A, the DNA mass increased continuously until sixteen days of the fermentation, which occupied 65%. For species B, the DNA mass amounted to 5.5% at six days of the fermentation, which was the maximum relative value during the fermentation. To the authors' best knowledge, this is the first report on ensiling rice straw with a selected starter at low temperature and investigation of the fermented characteristics.

자질별 관계 패턴의 다변화를 통한 온톨로지 확장 (Incremental Enrichment of Ontologies through Feature-based Pattern Variations)

  • 본 논문에서는 패턴의 다변화를 통하여 관계를 점진적으로 추출함으로써 온톨로지를 확장하는 모델을 제안한다. 패턴 다변화 과정에서 위키피디아로부터 추출한 관계 패턴 후보를 자질별로 다변화시킨다. 다변화된 패턴 후보로부터 말뭉치 빈도수에 따른 신뢰도를 이용하여 패턴을 선별한다. 선별된 패턴은 위키피디아로부터 관계를 추출하는 데 사용되며, 추출된 관계는 다시 관계 패턴 확장에 사용된다. 본 논문에서는 점진적 학습 과정에서의 패턴 다변화를 통하여 패턴 선택의 범위를 확장함으로써, 선택되는 패턴이 점진적으로 정제되는 모델을 제시한다. 이를 통하여, 관계의 확장성과 정확도를 향상시키고자 하였다. 단일 자질 패턴 모델에 대한 실험을 통하여, 어휘, 중심어, 상위어 정보는 신뢰도에, 품사, 구문 정보는 확장성에 유리하며, 구문 단위 유형별로 필요한 자질 유형이 다름을 관찰하였다. 이와 같은 특성에 기반하여 현재 연구 진행 중인복합 자질 패턴 모델을 제안한다.

도시가스용 바이오가스 메탄농축공정 최적화 (Optimization of Bio-Methane Gas Enrichment Process for City Gas Supply)

  • 국내의 늘어나는 천연가스의 수요를 충족시키면서 지구온난화에 대한 대안으로 바이오가스를 도시가스로 활용하는 방안이 자연스럽게 주목받고 있다. 도시가스사업법의 개정을 통해 바이오가스의 생산 및 공급의 제도적인 기반을 구축했지만, 바이오가스내의 불순물을 제거하고 이산화탄소를 분리하여 고순도의 메탄을 제조하는 국내 기술이 부족하여 실질적인 활용이 어려운 상황이다. 따라서 본 연구에서는 고순도의 바이오메탄 농축을 위하여 멤브레인(Membrane)법, 물흡수법(Water absorption), 화학흡수법(Chemical absorption), 흡착법 4가지의 공법을 적용하여 각 시스템별 운전시나리오를 작성하고 비교하여 최적공법을 도출하였으며, 시스템별 케이스 연구를 통해 최적의 시스템을 구축할 수 있었다. 이를 이용하여 순도 97%이상의 바이오메탄을 생산하여 도시가스로 이용하기에 충분하였으며, 회수율이 98%를 넘어 그간 경제성이 없어 버려지던 소량의 바이오 가스도 회수하여 활용 할 수 있게 되었다.

소 혈청 단백질 분획들의 기포분리 현상에 관한 연구 (Foam Separation of Bovine Serum Protein Fractions)

  • 기포분리 조작을 이용하여 소 혈청 단백질의 분리농축 현상을 전기영동 패턴에 의하여 분석하였다. 기포분리가 이루어지는 소 혈청 단백질 농도범위는 $20{\mu}g/ml$ $800{\mu}g/ml$이었으며 용액의 pH5부근에서 기포분리액의 부피는 최대가 되며 단백질 농축율은 최소치를 기록하였다. 기포분리 온도가 높아질수록 분리액의 부피는 감소하고 농축율은 증가하였으며, 가스유입속도가 25ml/min에서 100ml/min으로 증가 할 수록 기포분리액의 부피는 감소하고 농축율은 증가하였다. 염을 첨가할 때 이온강도 $1{\sim}3$부근에서 농축율은 최대치를 나타내었으며 이는 표면장력의 저하효과와 관계가 있었다. 일반적으로 소수성이 분자량이 작은 BSA, ${\alpha}_1$, 및 ${\alpha}_2-globulin$ 분획이 선택적으로 기포 분리 되는 경향을 나타내었으며 단백질 분획의 이동은 pH, 분리온도 가스유입속도, 첨가한 염의 이온강도 등에 따라 다르게 나타났다.

대청호에서 장기간 수질변동 및 인위적 Enclosure 영양염 투여실험에 따른 제한 영양염류 평가 (Long-term Water Quality Fluctuations in Daechung Reservoir and the Limiting Nutrient Evaluations Using In Situ Enclosure Nutrient Enrichment Bioassays (NEBs))

  • The objectives of this study were to elucidate spatio-temporal heterogeneity of water chemistry and develop empirical models using trophic variables in Daechung Reservoir during 2005-2010 along with in situ tests of nutrient enrichment bioassays (NEB). The relations of water quality parameters in regard to precipitation showed that seasonal and interannual fluctuations of biological oxygen demand (BOD), total nitrogen (TN) and pH were minor, whereas conductivity, suspended solids (SS), and total phosphorus (TP) were largely varied in response to the magnitude of rainfall. The CHL maxima occurred immediately after the spate of TP during the high flow, indicating that phytoplankton growth was directly controlled by phosphorus. Empirical linear models of CHL-TP indicated that the variation of CHL in premonsoon was accounted 60% ($R^2$ = 0.60, p < 0.05, n = 54) by TP. In the mean time, empirical models of annual CHL-TN showed that the variation of CHL was weakly accounted ($R^2$ = 0.16, p < 0.001) by TN and more strongly ($R^2$ = 0.44, p < 0.001) by TP. Thus, the variation of CHL was more explained by the variation of TP than TN. In situ tests of Nutrient Enrichment Bioassays (NEBs) showed that the growth of CHL was greater in the P-treatments (as $PO_4-P$) than the control and N-treatment (as $NO_3-P$). Overall, our results suggest that phosphorus was aprimary limiting nutrient controlling the seasonal phytoplankton growth, based on the in situ experiments of NEBs.

대구지역 부유분진 중 미량금속성분의 발생원 특성연구 (Source Characteristics of Particulate Trace Metals in Daegu Area)

  • This study was performed to understand the behavior and source characteristics of particulate trace metals in Daegu area. To do this, total of 84 samples had been collected from January to December 1999. TSP (total suspended particulate matter) and PM-10(particulate matter with aerodynamic diameters less 10${\mu}{\textrm}{m}$) were collected by filters on portable air sampler, and in TSP and PM-10 were analyzed by ICP(Inductively Coupled Plasma Spectrometer) after preliminary treatment. The results were follow as: first, annul means of TSP and PM-10 concentration were 123 and 69$\mu\textrm{g}$/㎤ respectively. The concentration of TSP adn PM-10 were highest in winter season compared to other seasons. Second, the concentration of Al, Fe, Mn were higher in TSP than in PM-10, indicating that these metals are generally associate with natural contributions. Third, a hierarchical clustering technique was used to group 9 metals. The results from the cluster analysis of TSP and PM-10 shows a similar clustering pattern : Fe, Al in a group and the rest of the metals such as Ni, Cr, As, Mn, Cd, Pb, Zn in the other group. One group of metal such as Fe, Al is associated with natural sources such as soil and dust. The other is closely related to urban anthropogenic sources such as fuel combustion, incineration, and refuse burning, Finally, using Al as a reference element, enrichment factors were used for identifying the major particulate contributors. The enrichment factors of Al. Fe<10 (standard value of enrichment factor) were considered to have a significant dust and soil source and termed nonenriched. Ni, Cr, As, Mn, Cd, Pb, Zn》10 is enriched and has a significant which is contributed by athropogenic sources.
