The interaction of Apolipoprotein A5 gene promoter region T-1131C polymorphism (rs12286037) and lifestyle modification on plasma triglyceride levels in Japanese

  • BACKGROUND/OBJECTIVE: Apolipoprotein A5 gene promoter region T-1131C polymorphism (APOA5 T-1131C) is known to be associated with elevated plasma TG levels, although little is known of the influence of the interaction between APOA5 T-1131C and lifestyle modification on TG levels. To investigate this matter, we studied APOA5 T-1131C and plasma TG levels of subjects participating in a three-month lifestyle modification program. SUBJECTS/METHODS: A three-month lifestyle modification program was conducted with 297 participants (Age: $57{\pm}8years$) in Izumo City, Japan, from 2001-2007. Changes in energy balance (the difference between energy intake and energy expenditure) and BMI were used to evaluate the participants' responses to the lifestyle modification. RESULTS: Even after adjusting for confounding factors, plasma TG levels were significantly different at baseline among three genotype subgroups: TT, $126{\pm}68mg/dl$; TC, $134{\pm}74mg/dl$; and CC, $172{\pm}101mg/dl$. Lifestyle modification resulted in significant reductions in plasma TG levels in the TT, TC, and CC genotype subgroups: $-21.9{\pm}61.0mg/dl$, $-20.9{\pm}51.0mg/dl$, and $-42.6{\pm}78.5mg/dl$, respectively, with no significant differences between them. In a stepwise regression analysis, age, APOA5 T-1131C, body mass index (BMI), homeostasis model assessment-insulin resistance (HOMA-IR), and the 18:1/18:0 ratio showed independent association with plasma TG levels at baseline. In a general linear model analysis, APOA5 T-1131C C-allele carriers showed significantly greater TG reduction with decreased energy balance than wild type carriers after adjustment for age, gender, and baseline plasma TG levels. CONCLUSIONS: The genetic effects of APOA5 T-1131C independently affected plasma TG levels. However, lifestyle modification was effective in significantly reducing plasma TG levels despite the APOA5 T-1131C genotype background.

Enteric methane emissions, energy partitioning, and energetic efficiency of zebu beef cattle fed total mixed ration silage

  • Objective: The main objective of this study was to evaluate the effect of different feeding levels of a total mixed ration silage-based diet on feed intake, total tract digestion, enteric methane emissions, and energy partitioning in two beef cattle genotypes. Methods: Six mature bulls (three Thai natives, and three Thai natives - Charolais crossbreeds) were assigned in a replicated $3{\times}3$ Latin square design, with cattle breed genotype in separate squares, three periods of 21 days, and three energy feeding above maintenance levels (1.1, 1.5, and 2.0 MEm, where MEm is metabolizable energy requirement for maintenance). Bulls were placed in a metabolic cage equipped with a ventilated head box respiration system to evaluate digestibility, record respiration gases, and determine energy balance. Results: Increasing the feeding level had no significant effect on digestibility but drastically reduced the enteric methane emission rate (p<0.05). Increasing the feeding level also significantly increased the energy retention and utilization efficiency (p<0.01). The Thai native cattle had greater enteric methane emission rate, digestibility, and energy utilization efficiency than the Charolais crossbred cattle (p<0.05). The daily metabolizable energy requirement for maintenance in Thai native cattle ($388kJ/kg\;BW^{0.75}$, where $BW^{0.75}$ is metabolic body weight) was 15% less than that in Charolais crossbred cattle ($444kJ/kg\;BW^{0.75}$). Conclusion: Our results suggested that the greater feeding level in zebu beef cattle fed above maintenance levels resulted in improved energy retention and utilization efficiency because of the reduction in enteric methane energy loss. The results also indicated higher efficiency of metabolisable energy utilization for growth and a lower energy requirement for maintenance in Bos indicus than in Bos taurus.

  • The experiment was carried out to estimate the energy and protein requirements of Khaki Campbell $\times$ Thai Native laying ducks. To estimate the energy requirement, 5 experimental diets were formulated at levels of 2.70, 2.75, 2.80, 2.85 and 2.90 Mcal ME/kg. These diets were equal in ME/CP ratio (170/l). A total of 150 18-week old laying ducks were assigned to 5 energy level treatments, each comprising 3 replicates of 10 birds each, and they were fed the diets for 18 weeks. To estimate the protein requirement, 90 18-week old laying ducks were divided into 9 groups of 10 birds each, and they were assigned to 3 protein level treatments, each comprising 3 replicates. The levels of protein in the diets were 13.5%, 15.0% and 16.5%, and all diets were isocaloric (2.8 Mcal ME/kg). As a result, the 16.5% protein diet gave significantly better egg production than the 13.5% and 15.0% protein diets, however, no significant difference in egg production was found among the energy levels. Feed cost to produce 1k eggs was lower in the 2.70 Mcal and 16.5%protein diets than in the higher ME and lower protein diets.

Calculation on Surface Electronic State of $TiO_2$ Electrode (TiO2 전극 표면의 전자상태 계산)

  • The surface electronic state of rutile $TiO_2$, which is an oxide semiconductor and has a wide band gap of 3.1 $\sim$ 3.5 eV, was calculated by DV-$X_{\alpha}$ method, which is a sort of the first principle molecular orbital method and uses Hartre-Fock-Slater approximation. The $[Ti_{15}O_{56}]^{-52}$ cluster model was used for the calculation of bulk state and the $[OTi_{11}O_{34}]^{-24}$ model for the surface state calculation. After calculations, the energy level diagrams and the deformation electron density distribution map were compared in both models. As results, it was identified that the surface energy levels are found between the valence and conduction band of bulk $TiO_2$ on the surface area. The energy values of these surface-induced levels are lower than conduction band of bulk $TiO_2$ by 0.1 $\sim$ 1 eV. From this fact, it is expected that the surface energy levels act as donar levels in n-type semiconductor.

Effect of Nutritional Levels on the Growth and Meat Quality of Korean Native Black Pigs (사료의 영양수준에 따른 재래흑돼지의 산육능력 및 육질비교)

  • This study was conducted to evaluate the effects of nutritional levels on the growth and pork quality of Korean native black pigs (54 pigs from 35-75kg). Three nutritional levels (high, medium and low in ME and lysine) were tested. As nutritional levels increased, the average daily gain improved, and the days to market weight were reduced. There were no significant differences in dressing percentage, backfat thickness, loin eye area and % lean among the dietary treatments. However, barrows showed thicker (p<0.05) for backfat than gilt. The analysis of Korean native black pig carcass traits indicated that the fatty acid composition of loin-eye muscle, intra-muscular fat content, sarcomere length, and muscle ratio between red- and white-muscles did not show any statistically significant variation. However, regarding intra-muscular fat, H carcasses (high energy diet) contained higher levels of saturated fatty acids with lower levels of unsaturated fatty acids compared to the carcasses from the other two energy groups (M and L). In addition, the levels of n-3 tended to be lower as the energy/lysine level in the diet increased, while the ratio of n-6/n-3 tended to decrease as the energy/lysine level increased, even though the differences were not statistically significant. Even with no statistically significant differences, it was interesting to observe that the sarcomere length tended to increase as the energy level of the diet increased, and with muscle ratio analysis, the red muscle ratio from the L-group, and the white muscle ratio from the M-group were higher than the other energy groups.

Genetic Diversity among Korean Bermudagrass (Cynodon spp.) Ecotypes Characterized by Morphological, Cytological and Molecular Approaches

  • The genus Cynodon comprises ten species. The objective of this study was to evaluate the genetic diversity of Korean bermudagrasses at the morphological, cytological and molecular levels. Morphological parameters, the nuclear DNA content and ploidy levels were observed in 43 bermudagrass ecotypes. AFLP markers were evaluated to define the genetic diversity, and chromosome counts were made to confirm the inferred cytotypes. Nuclear DNA contents were in the ranges 1.42-1.56, 1.94-2.19, 2.54, and 2.77-2.85 pg/2C for the triploid, tetraploid, pentaploid, and hexaploid accessions, respectively. The inferred cytotypes were triploid (2n = 3x = 27), tetraploid (2n = 4x = 36), pentaploid (2n = 5x = 45), and hexaploid (2n = 6x = 54), but the majority of the collections were tetraploid (81%). Mitotic chromosome counts verified the corresponding ploidy levels. The fast growing fine-textured ecotypes had lower ploidy levels, while the pentaploids and hexaploids were coarse types. The genetic similarity ranged from 0.42 to 0.94 with an average of 0.64. UPGMA cluster analysis and principle coordinate analysis separated the ecotypes into 6 distinct groups. The genetic similarity suggests natural hybridization between the different cytotypes, which could be useful resources for future breeding and genetic studies.

Food Ingestion Standards for Nuclear Emergency Exposure Situations

  • Background: This study presents food ingestion standards for radioactivity that can be applied in nuclear emergency exposure situations, and discusses the validity of the current domestic standards. Materials and Methods: This study derived food ingestion standards for radiocesium and radioiodine using domestic food intake rates and intervention levels, which serve as a basis for determining the necessity of public protective actions, and then compared them with the existing guidelines. Operational intervention levels were also derived using domestic food intake rates, and were compared with those of the International Atomic Energy Agency. Results and Discussion: The derived activity concentrations for food ingestion standards of radiocesium for infants were higher than those in the Act on Physical Protection and Radiological Emergency (APPRE) for all food categories, while for adults, the derived activity concentrations for drinking water and milk appeared to be slightly lower. The derived activity concentrations for vegetables, fruits, and grains were greater than those in the guidelines of the APPRE, while the derived activity concentrations for meat and seafood were similar to those in the APPRE. The derived activity concentrations for radioiodine were greater than both domestic and global standards. The calculated operational intervention levels (OILs) based on domestic food intake rates were greater than the IAEA's default OIL6 values for most radionuclides, except for a few ${\alpha}$-radionuclides. Conclusion: The current domestic guidelines turned out to be conservative overall, compared to the present results that were calculated using domestic food intake rates. It is recommended that the domestic guidelines should be revised and complemented transparently through an in-depth review by stakeholders on a solid scientific basis.

Effects of Dietary Energy Levels on the Physiological Parameters and Reproductive Performance of Gestating Gilts

  • This experiment was conducted to investigate the effects of dietary energy levels on the physiological parameters and reproductive performance of gestating first parity sows. A total of 52 F1 gilts ($Yorkshire{\times}Landrace$) were allocated to 4 dietary treatments using a completely randomized design. Each treatment contained diets with 3,100, 3,200, 3,300, or 3,400 kcal of metabolizable energy (ME)/kg, and the daily energy intake of the gestating gilts in each treatment were 6,200, 6,400, 6,600, and 6,800 kcal of ME, respectively. During gestation, the body weight (p = 0.04) and weight gain (p = 0.01) of gilts linearly increased with increasing dietary energy levels. Backfat thickness was not affected at d110 of gestation by dietary treatments, but increased linearly (p = 0.05) from breeding to d 110 of gestation. There were no significant differences on the litter size or litter birth weight. During lactation, the voluntary feed intake of sows tended to decrease when the dietary energy levels increased (p = 0.08). No difference was observed in backfat thickness of the sows within treatments; increasing energy levels linearly decreased the body weight of sows (p<0.05) at d 21 of lactation and body weight gain during lactation (p<0.01). No significant differences were observed in the chemical compositions of colostrum and milk. Therefore, these results indicated that high-energy diets influenced the bodyweight and backfat thickness of sows during gestation and lactation. NRC (2012) suggested that the energy requirement of the gestation gilt should be between 6,678 and 7,932 kcal of ME/d. Similarly, our results suggested that 3,100 kcal of ME/kg is not enough to maintain the reproductive performance for gilts during gestation with 2 kg feed daily. Gilts in the treatment 3,400 kcal of ME/kg have a higher weaning number of piglets, but bodyweight and backfat loss were higher than other treatments during lactation. But bodyweight and backfat loss were higher than other treatments during lactation. Consequently, an adequate energy requirement of gestating gilts is 6,400 kcal of ME/d.

  • Dietary protein is the most expensive component in flounder feed because of the high protein requirement of this species. The non-protein energy sources may also influence the protein utilization of fish. The inclusion of adequate levels of non-protein energy sources in diets can minimize use of protein as an energy source. Protein sparing effect by fat and carbohydrate has been studied in other fish.   • The Institute of Medicine of the National Academies Food and Nutrition Board in conjunction with Health Canada recently released the sixth in a series of reports on dietary reference values for the intake of energy and macronutrients. This paper discusses the Dietary Reference Intakes (DRIs) for energy and physical activity. Several new approaches were used with the DRIs for the macronutrients. Those discussed in this paper include: $.$Estimated Energy Requirements (EER) at four levels of energy expenditure based on doubly labeled water measurements of total energy expenditure (TEE). $.$Recommendations for levels of physical activity to decrease the risk of chronic disease.
