내분비계 장애물질 (EDCs)은 생물체내에 유입되면 내분비계의 정상적 기능을 방해하는 특징이 있으며, 일반화학물질과는 다른 독성기전을 보이기 때문에 기존의 생태독성기법과 달리 EDCs만의 독성을 평가하기 위한 생태독성기법 및 독성종말점의 개발을 위한 연구가 필요하다. OECD, ASTM 및 USEPA와 같은 신뢰성 있는 국제기관에서는 EDCs의 생태독성기법들을 제안하고 있는 상황이다. EDCs가 유해성이 알려진 만큼 국내에서도 EDCs관련 정부지원 사업이 일부 EDCs에 대해 사업장에의 배출량, 환경매체에서 검출농도를 지속적으로 모니터링하고 있으며, EDCs관련 연구들이 보고되고 있다. 그러나 국내에는 EDCs를 대상으로 하는 생태독성기법이 부재한 상황이다. 본 연구에서는 국외 EDCs의 생태독성기법을 조사 및 분석하여 제안된 생물종과 독성종말점의 종류 및 시험조건 파악하고자 하였다. 그 결과, ISO에서는 아직 EDCs 생태독성기법을 따로 제안하고 있지 않은 상태이며, OECD, ASTM, USEPA에서만 수서생물종인 어류 양서류, 물벼룩 및 요각류, 토양생물종인 지렁이, 애지렁이, 톡토기, 진드기, 토양선충, 그리고 퇴적물서식종인 지렁이와 깔따구에 대한 EDCs 평가기법이 제시되어 있고, 생식, 호르몬, 성장, 비탈로제닌, 성비 및 발달 등과 관련된 독성종말점으로 제안되어 있었다. 결론적으로 EDCs 대상의 생태독성 평가방법은 매우 제한적이며, 생태독성 기법 개발 및 독성종말점을 개발하기 위한 연구가 필요한 것으로 판단된다.
Purpose: To investigate factors that affect behaviors of reducing exposure to endocrine disrupting chemicals (EDCs) in breastfeeding mothers. Methods: Subjects were 166 breastfeeding mothers who were recruited from January 25 to April 15, 2018. The questionnaire consisted of items associated with behaviors of reducing exposure to EDCs, attitude to behaviors of reducing exposure, subject norms of behavior of reducing exposure, and perceived behavior control. Data were analyzed by t-test, ANOVA, Pearson correlations, and stepwise multiple regression. Results: Factors influencing behaviors of reducing the exposure to EDCs were attitude toward behaviors of reducing exposure, perceived barrier, and period of lactation. The more positive the attitude toward behaviors of reducing exposure was, the lower the perceived barrier. The longer the feeding period, the higher the behaviors of reducing the exposure to EDCs. Conclusions: To enhance behaviors of reducing the exposure to EDCs, it is necessary to develop a positive educational program in breastfeeding department to improve attitude toward behaviors of reducing the exposure to EDCs, decrease perceived barrier, and increase the breastfeeding period.
내분비장애 물질은 분해가 어렵고, 생물체에 축적되기 때문에 먹이그물을 통하여 결국 인간에게 피해를 준다. 리그닌 분해효소 군을 가진 백색부후균들은 다양한 난분해성 물질의 분해능이 우수하며, 국내에서 분리한 아교버섯은 내분비장애 물질에 속하는 프탈레이트의 분해능이 우수하다. 내분비장애 물질분해와 관련된 laccase cDNA를 발현벡터로 재조합하고 이를 형질전환 방법에 의하여 아교버섯으로 도입하였으며 도입된 발현벡터는 형질전환체의 염색체에 안정하게 존재하였다. 형질전환체들 중 가장 효소활성이 좋은 균주를 대상으로 분석한 결과 laccase 활성이 증가되었을 뿐만 아니라 내분비장애 물질의 분해능도 향상되었고, 동시에 다양한 내분비장애물질에 의한 에스트로겐 활성도 야생형 균주에 비하여 빠르게 감소시켰다.
내분비장애물질은 분해가 매우 어려워 자연계에서 먹이그물을 통하여 사람에게 농축 전달된다. 이들은 정상적인 내분비계에 혼란을 일으키며, 특히 성호르몬의 작용에 많은 피해를 준다. 이를 효율적으로 분해하고 이들의 에스트로겐 활성을 제거하고자 백색부후균의 하나인 아교버섯(Phlebia tremellosa)을 활용하여 4가지 내분비계 장애물질의 분해에 대한 실험을 수행하였다. 아교버섯의 manganese peroxidase (MnP) 활성을 높이기 위하여 구름버섯의 MnP 유전자를 아교버섯에 도입하여 형질전환체를 확보하였으며 이들은 유전적으로 MnP 활성을 안정되게 나타냈다. 내분비 장애물질을 분해하는 조건에서 내분비장애 물질에 따라 30${\sim}$45%의 분해율을 보인 야생형에 비하여 이 형질전환체들 중 T5는 70${\sim}$88%의 분해율을 보였으며 에스트로겐 활성의 제거에도 약 2배 향상된 능력을 보였다.
Benzo(a)pyrene and bisphenol A have been classified as endocrine disrupting chemicals (EDCs), which have been of concern in toxicology and environmental sciences. Benzo(a)pyrene and bisphenol A were monitored by HPLC or GC-MS in Baekwoon and Ilwirl lakes of Kyonggi-Do province to investigate contamination levels of EDCs. During the period between June, 2000 and August, 2000, water samples were collected from four different sites of each lake once per month. Contamination levels of benzo(a)pyrene were 3.27~4.25 ppb in Ilwirl lake and 2.00~2.33 ppb in Baekwoon lake, respectively. Bisphenol A levels were detected with the range of 0.33~7.94 and 0.43~4.71 for Baekwoon lake and Ilwirl lake, respectively. pH levels were higher in Ilwirl lake than in Baekwoon lake, where the contamination was relatively lower. These data suggest that lakes in Kyonggi-Do province could be contaminated with EDCs and be subjected to the routine monitoring and water quality control.
Purpose: This study attempted to determine the factors related to behaviors of reducing exposure to endocrine disrupting chemicals (BRE to EDCs) in pregnancy, based on the PRECEDE model. Methods: A cross-sectional survey was conducted with participants (N=239) who met the medical records and eligibility criteria from the Women's Hospital and Public Health Center. Data were collected using a specially-designed questionnaire based on the PRECEDE model and included BRE to EDCs predisposing factors, reinforcing factors, and enabling factors. The data were analyzed using the chi-square test, independent t-test, one way ANOVA, Pearson's correlation coefficients and a hierarchical multiple regression analysis. Results: In Model I it was found that participation in prenatal education influenced the BRE to EDCs significantly; and a regression showed that the determinant variables accounted for 3.4%. In Model II, the predisposing factors of perceived barriers of BRE to EDCs and environmental self-efficacy were added. It was shown that they significantly influenced BRE to EDCs in the order named, and a regression revealed that increases in the determinant variables accounted for 22.5%. In Model III, to which enabling factors were added, the information acquisition experience of BRE to EDCs interacted significantly with BRE to EDCs and a regression showed an increase in the determinant variables accounting for 25.3%. Conclusion: The results of this study, the content of endocrine disruptors must be included in the prenatal care education program for pregnant women, and it should be composed of contents that can reduce the perceived obstacle to BRE to EDCs, enhance the environmental self-efficacy, and provide the information in regard to reducing exposure to EDCs.
An efficient method for the simultaneous determination of eleven phenolic endocrine-disrupting chemicals (EDCs) present in wastewater influent samples was described. The 11 phenolic EDCs including alkylphenols, chlorophenols, and bisphenol A were determined by gas chromatography/mass spectrometry-selected ion monitoring (GC/MS-SIM) following two work-up methods for comparison; isobutoxycarbonyl (isoBOC) derivatization and tert-butyldimethylsilyl (TBDMS) derivatization. The wastewater influent samples containing the 11 EDCs were adjusted to pH 2 with $H_2SO_4$ and then cleaned up with n-hexane. Next, they were subjected to solid-phase extraction (SPE) with XAD-4 resin and subsequently converted to isoBOC or TBDMS derivatives for sensitivity analysis with gas chromatography/mass spectrometry-selected ion monitoring (GC/MSSIM). Following isoBOC derivatization and TBDMS derivatization, the recoveries were 86.6-105.2% and 97.6-142.7%, the limits of quantitation (LOQ) for the 11 phenolic EDCs for SIM was 0.001-0.050 ng/mL and 0.003-0.050 ng/mL, and the SIM responses were linear with the correlation coefficient varying by 0.9717-0.9995 and 0.9842-0.9980, respectively. When these methods were applied to five selected wastewater influent samples, for isoBOC derivatization and TBDMS derivatization the ranges of concentration detected were 0.2-99.6 ng/mL and 0.4-147.4 ng/mL, respectively.

A method for determination 11 endocrine disrupting chemicals of phenols in various samples was deloped. The alkylphenols, chlorophenols and bisphenol A were determined by gas chromatography/mass spectrometry-selected ion monitoring (GC/MS-SIM) followed by two work-up methods for comparison; isobutoxycarbonyl (isoBOC) derivatization method and tert-butyldimethylsilyl (TBDMS) derivatization method. In this study, three of the representative EDCs, $17{\beta}$-estradiol, bisphenol A, and styrene, were employed to find their mode of toxic actions in E. coli. To accomplish this, four different stress response genes, recA, katG, fabA, and grpE genes, were used as a representative for DNA, oxidative, membrane, or protein damage, respectively. The expression levels of these four genes were quantified using a real-time RT-PCR after challenge with three different EDCs individually. Bisphenol A and styrene caused high-level expression of recA and katG genes, respectively, whereas $17{\beta}$-estradiol made no significant changes in expression of any of those genes. These results lead to the classification of the mode of toxic actions of EDCs on E. coli.
Purpose: This study aimed to enhance health management and prevention strategies for precocious puberty by comparing the knowledge, motivation, and behaviors related to reducing exposure to endocrine disrupting chemicals (EDCs) between mothers of children with precocious puberty and those of typically developing children. Methods: Participants were 66 mothers of children with precocious puberty from a hospital and 71 mothers of typically developing children from an elementary school. The research questionnaire assessed knowledge of EDCs, motivation, self-efficacy, and behaviors towards reduced exposure to EDCs, in addition to general characteristics of children with precocious puberty. Data analysis utilized descriptive statistics, chi-square tests, t-tests, ANOVA, and Pearson's correlation. Results: There were statistically significant differences in self-efficacy (p=.015) and behavior (p=.001) regarding reduced exposure to EDCs between the two groups. Behaviors toward reduced exposure to EDCs were positively associated with knowledge of EDCs, motivation, and self-efficacy among mothers of children with precocious puberty. Additionally, these behaviors correlated positively with knowledge of EDCs, motivation, self-efficacy, and the child's birth weight and waist circumference among mothers of typically developing children. Conclusion: The results of this study revealed that mothers of children with precocious puberty exhibited less proactive behavior in reducing exposure to EDCs compared to mothers of typical children. Therefore, it is important to foster interest in the impact of environmental factors, beyond characteristic factors, on behaviors aimed at reducing exposure to EDCs.
