Effects of Growth Regulators on Somatic Embryogenesis from Ginseng Zygotic Embryos (인삼 접합자배로부터 체세포배의 발생에 미치는 생장조절제의 영향)

  • Intact mature zygotic embryos or their excised cotyledons of ginseng, were cultured on media containing various growth regulators such as auxin (2,4D, IAA) and cytokinin(BAP kinetin). In the culture of intact zygotic embryos, auxin inhibited germination but cytokinin did not Somatic embryogenesis occurred only from those of ungerminated embryos. In the culture of cotyledon segment, medium without growth regulators was the most appropriate to somatic embryogenesis. Somatic embryos were produced sporadically over the surfaces of zygotic embryos on medium containing auxin, while on medium without growth regulators, or media containing cytokinin, somatic embryos formed only on the proximal region of cotyledon. on medium containing 2,4-D, somatic embryos originated from multiple cells which comprised epidermal and subepidermal layers of cotyledon, which resulted in poly-somatic embryogenesis. When these somatic embryos were cultured on the same medium, the primary somatic embryos procured secondary embryos, which arose from epidermal or subepidermal single cells.

Callus induction and high-efficiency plant regeneration via somatic embryogenesis in Papaver nudicaule L., an ornamental medicinal plant

  • We describe culture conditions for a high-efficiency in vitro regeneration system of Papaver nudicaule through somatic embryogenesis and secondary somatic embryogenesis. The embryogenic callus induction rate was highest when petiole explants were cultured on Murashige and Skoog (MS) medium containing 1.0 mg $1^{-1}$ ${\alpha}$-naphthaleneacetic acid (NAA) and 0.1 mg $1^{-1}$ 6-benzyladenine (BA) (36.7%). When transferred to plant growth regulator (PGR)-free medium, 430 somatic embryos formed asynchronously from 90 mg of embryogenic callus in each 100-ml flask. Early-stage somatic embryos were transferred to MS medium containing 1.0 mg $1^{-1}$ BA and 1.0 mg $1^{-1}$ NAA to germinate at high frequency (97.6%). One-third-strength MS medium with 1.0% sucrose and 1.0 mg $1^{-1}$ $GA_3$ had the highest frequency of plantlet conversion from somatic embryos (91.2%). Over 90% of regenerated plantlets were successfully acclimated in the greenhouse. Secondary somatic embryos were frequently induced directly when the excised hypocotyls of the primary somatic embryos were cultured on MS medium without PGRs. Sucrose concentration significantly affected the induction of secondary embryos. The highest induction rate (89.5) and number of secondary somatic embryos per explant (9.3) were obtained by 1% sucrose. Most secondary embryos (87.2-94.3%) developed into the cotyledonary stage on induction medium. All cotyledonary secondary embryos were converted into plantlets both in liquid and on semisolid 1/3-strength MS medium with 1.0% sucrose.

  • The Cassava(Manihot esculenta Crantz) is a tropical root crop, originally from Amazonia, that provides the staple food of an estimated 800 million people worldwide. It belongs to the family Euphorbiaceae which also includes rubber (Hevea brasiliensis) and castor bean (Ricinus communis). Among tropical crops, rice, sugarcane, maize and cassava are the most important sources of calories for human consumption. Problems in the propagation of cassava are virus diseases and low rates of seed germination. So we tried to optimize protocols for mass production of somatic embryo amenable to large-scale vegetative propagation of Cassava. After in vitro eight-week culture of leaves of Cassava, the medium which contained the 2,4-D, BAP and IBA showed the highest callus induction rate, embryogenesis callus formation rate and somatic embryo formation in Cassava culture. In the medium with GA3 and myo-inositol, shoots were most vigorously regenerated from somatic embryos of Cassava. Our experiments confirmed that in vitro growth and multiplication of plantlets could depend on its reaction to the different medium composition, and this micropropagation techniques could be a useful system for healthy and vigorous plant production.

Differences in Chemical Composition of Soybean Seeds with Different Usages and Storage Durations (저장기간과 용도가 다른 대두종실의 화학성분 차이)

  • For the comparision of the differences in dry matter weight and the chemical composition of soybean seeds in relation to utilization styles and storage durations, the seed coats and cotyledons including embryo axis separated from the seeds of 3 recommended cultivars stored for 3 years and of 5 recommended cultivars and 2 local varieties with black seed coat and green cotyledons were observed in dry weight, germination percentage, protein, oil, soluble sugar, oilgosaccharide, and starch. In the seeds stored for 3 years, dry weight and germination rate were decreased distinctively. This decrease of seed dry matter resulted from the decrease of the cotyledon weight, and the ratio of seed coat to whole seed was increased. For the change of chemical compositions by the decrement of seed dry weight during the storage, protein, oil, and carbohydrate contents of the seed coat were decreased, and protein content of the cotyledon was markedly decreased in the seeds stored for 3 years. The ratio of seed coat to whole seed was higher in small seed than large seed cultivars. In the local varieties, protein and oil contents were 43.59% and 18.25% in the cotyledons, respectively, which were lower than those of recommended cultivars. But starch and sugar contents were 5.70% and 11.58%, respectively, which were higher than those of recommended cultivars. Sucrose, raffinose and stachyose contents were also higher in the cotyledon of the local varieties.

Dormancy of Somatic Embryos Derived from the Cotyledon of Korean Ginseng

  • Somatic embryos were induced directly from cotyledon explants of Korean ginseng (Panax ginseng C.A. Meyer) on Murashige and Skoog (MS) medium with 2,4-D, BAP, kinetin or lacking growth regulators. When somatic embryos formed on all media grew to cotyledonary stage, the further development of embryos was ceased and remained in white color. By gibberellic acid (over 1.0 mg/1 $GA_3$) treatment, all the somatic embryos turned rapidly to green and germinated within 3 weeks. Chilling treatment also induced the germination of somatic embryos. The effective temperature regime was $-2^{\circ}C$ for over 8 weeks but more higher temperature than $0^{\circ}C$ did not effective for germination of somatic embryos. Ultrastructural observation revealed that the cotyledon cells of somatic embryos without chilling or $GA_3$ treatment contained numerous lipid reserves, dense cytoplasm, proplastids and non-activated mitochondria with poorly differentiated internal structure, but the cotyledon cells of germinating somatic embryos after chilling or $GA_3$ treatment highly vacuolated and contained well-developed chloroplasts and active state of mitochondria enclosing numerous cristae. The above results indicate that in vitro developed somatic embryos of Panax ginseng may be dormant after mature similar to zygotic embryos.

  • Solomon's seal (Polygonatum odoratum) seedlings raised through its seeds can replace the rhizomes impelling higher cost for transplanting, This experiment was done to determine the seed characteristics and the germinating processes to give some information on bulk production of seedlings using the seeds. The external or internal morphology of seeds or seedlings grown in lab. or greenhouse was examined mainly with stereomicroscope. The external shape of Solomon's seal seed was hard seed-coat and orthotropous ovule with linear type embryo stretching to the center of seed. Germination proceeded through the several steps. The lower part of seed embryo having the primordia of bulbil and roots first grew before the bulbil and roots was developed from the primordia. The lower part of embryo was enlarged toward the endosperm of seed as soon as seed germinated. Then epicotyl was formed on the apex of bulbil. The epicotyl was elongated after at least 6-week chilling treatment for breaking its dormancy and the first leaf shape was affected by light intensity given during seedling emergence. The bulbil was the first organ of the rhizome used as tea or herb medicine.

Development of Zygotic Embryos and Seedlings is Affected by Radiation Spectral Compositions from Light Emitting Diode (LED) System in Chestnut (Castanea crenata S. et Z.)

  • Among the environmental conditions employed in micropropagation, light quality plays an important role in growth, specially morphogenesis and photosynthesis. The effect of radiation quality (350-740 nm) on the development and growth of zygotic embryos and in vitro plantlets of open-pollinated chestnut (Castanea crenata S. et Z.) were studied. Two types of explants were exposed for 4 weeks to cool white (W, as control), monochromatic red (R, peak emission 650 nm), monochromatic blue (B, peak emission 440 nm), red+blue (R+B, 1:1), or red+far-red (R+Fr, 1:1, far-red peak emission 720 nm) radiation from a light-emitting-diode (LED) system. While the zygotic embryos showed positive photoblastic behavior, their germination was inhibited by blue radiation. Hypocotyl elongation and root development were promoted by red radiation. The emergence of primary leaf and its expansion were faster under blue than under red radiation. In the plantlets, red and red+far-red radiation significantly increased the formation and growth of the root, whereas blue light reduced rooting. Therefore, radiation quality appears to influence some steps in the development of zygotic embryos and plantlets in the chestnut.

Effects of Temperature, Photon Irradiance, and Photoperiod on the Growth of Embryos of Sargassum horneri in Laboratory Culture (괭생이모자반(Sargassum horneri) 유배의 생장에 미치는 온도, 광량, 광주기의 영향)

  • The effects of temperature, photon irradiance, and photoperiod on the growth of Sargassum horneri embryos were examined for female plants collected at Chokpo in Tongyeoung, Gyeongnam Prefecture, Korea on 27 December 2011. Mature plant receptacles were detached, and fertilized eggs were cultured in the laboratory at temperatures of $5-25^{\circ}C$ with photon irradiances of $10-80{\mu}mol\;m^{-2}s^{-1}$ under 14L:10D and 10L:14D photoperiods. Germination and embryo growth were rapid at $20-25^{\circ}C$ and $40{\mu}mol\;m^{-2}s^{-1}$ under 14L:10D. The number of rhizoids in the germinated young thalli was high at high photon irradiances ($40-80{\mu}mol\;m^{-2}s^{-1}$) and $15-20^{\circ}C$. At $20^{\circ}C$ and 14L:10D, vigorous growth of main and lateral branches was observed; however, at $5^{\circ}C$ under both photoperiods, plant growth decreased markedly. In the present study, S. horneri grew well at high temperatures ($20-25^{\circ}C$) and high photon irradiances ($40-80{\mu}mol\;m^{-2}s^{-1}$) with a 14L:10D photoperiod.

Induction of somatic embryogenesis from immature zygotic embryos and young apical leaves in cork oak (Quercus suber L.)

  • The present work aims to study the induction of somatic embryogenesis in cork oak (Quercus suber L.) from immature zygotic embryos and young apical leaves obtained from 2-month-old seedlings through acorn germination on sterilized peat. The immature zygotic embryos were grown for 1 month on the mineral solution of MS in the presence of 4.52 µM 2,4-D and 30 g/L sucrose. They were then transferred to the same mineral solution with no added growth regulators. In the third subculture, yellow somatic embryos, characterized by two voluminous cotyledons, were differentiated from the radicle of the immature zygotic embryos. The induction of somatic embryogenesis in young leaves required a series of transfers on different culture media containing 30 g/L sucrose and 100 mg/L myo-inositol. Secondary or recurrent somatic embryogenesis occurred within the immature somatic embryo radicles after 1 month of culture on growth regulator-free medium containing WPM macronutrients, MS micronutrients, and vitamins.

Effect of GA3 Concentrations on the Vernalization Period in the Immature Embryo Culture of Barley (Hordeum vulgare) (미숙배 배양시 GA$_3$농도가 보리의 춘화처리 효과에 미치는 영향)

  • Immature embryos were tested for investigating the effect of cold treatment duration and GA$_3$ concentration on the vernalization response in barley. Immature embryos were cultured on B$_{5}$ medium with GA$_3$ for 5 days, and were transplanted under 20/15$^{\circ}C$ temperature after cold treatment. Germination rate and shoot length were increased more in 20-day-old embryos than those in 13-day-old ones but germination day was decreased. Addition of 1 or 10 ppm of GA$_3$ to B5 medium was effective on the growth of immature embryos. The higher GA$_3$ concentration was, the shorter time from embryo culture to flag leaf emergence and heading was. The earlist flag leaf emergence and heading were showed on treatment of 1 or 10 ppm of GA$_3$, but GA$_3$ did not completely replace vernalization. The days to flag leaf emergence of immature embryo-cultured barley with cold treatment for two and three weeks was shortened by 3 and 18 days at 1 ppm GA$_3$, and 16 and 20 days at 10 ppm GA$_3$, respectively, when compared to 0 ppm GA$_3$ culture with cold treatment for three weeks. It was at 10 ppm GA$_3$ with 3 weeks cold treatment that showed the lowest culm length, spike length and number of grain per spike. GA$_3$ concentration was not correlated significantly with each character in 1 week cold treatment, but was significant with each character in 2 weeks. In 3 weeks cold treatment, it was except for days to heading. Correlation between cold treatment duration and culm length was negative in 0 ppm GA$_3$. In 1 or 10 ppm of GA$_3$, all characters had highly negative correlation with cold treatment duration.n.
