Morphological Observation of Somatic Embryogenesis in Leaf Explant Cultures of Bupleurum falcatum L. (시호(Bupleurum falcatum L) 잎절편으로부터 형성된 체세포배 발생의 형태학적 관찰)

  • This study describes plant regeneration from leaf explant of Bupleurum falcatum through somatic embryogenesis, and the effect of 2,4-dichlorophenoxyacetic acid on somatic embryo abnormalities. The relationship between the cotyledon number of somatic embryo and its germinability is also described. Embryogenic calli were selected from calli formed on explants cultured on MS solid basal medium supplemented with 1 mg/L 2,4-D. Cotyledonary abnormalities were observed in somatic embryos which were developed from calli cultured on MS medium with 1 mg/L 2,4-D for 6-week and then subcultured on 2,4-D free MS medium for 3 weeks. The frequency of abnormalities was as follows: 7% of somatic embryos had one cotyledon, 65% of them had two cotyledons, 25% three cotyledons, 5% four cotyledons, 2% five cotyledons, and 3% trumpet-like cotyledons. The two cotyledon somatic embryos were germinated at a frequency of 80%. However the germination frequency of one cotyledon embryo and multicotyledonary embryo was lower than that of the two cotyledon somatic embryo. All of trumper-like somatic embryos did not germinate. Histological observations of multicotyledon embryo showed circular procambium in the root but pocambial strands in the cotyledonary node or upper hypocotyl. The number of the strands was equal to the cotyledon number.

Effects of NaOCl treatment on in vitro germination of seeds of a rare endemic plant, Oreorchis coreana Finet

  • Oreorchis coreana Finet is threatened globally by over-collection from its natural habitats for horticultural purposes. Its rarity in nature makes this plant one of the most endangered species in Korea. In this study, we investigated the effects of sodium hypochlorite (NaOCl) on orchid seed viability and seed germination. An in vitro bioassay swelling test using immature seeds was compared with a standard chemical procedure using triphenyl tetrazolium chloride (TTC) to test seed viability. In general, the bioassay was more appropriate for estimating embryo viability after a prolonged pre-treatment (more than 1 h) in 1% NaOCl, a surface sterilant often used to enhance germination of seeds of terrestrial plants. Therefore, an efficient method for investigating in vitro swelling of immature seeds is urgently needed. We established a method for determining the viability and swelling of O. coreana seeds via in vitro examination of immature seeds. Treatment of immature seeds with 1% NaOCl for 10 min greatly enhanced the extent of swelling of immature zygote embryos when compared to untreated seeds. These data obtained here appear to be comparable to viability and swelling that occurs in O. coreana seeds via asymbiotic germination.

Morphological and Morphophysiological Dormancy in Seeds of Two Genera (Heloniopsis and Thalictrum) Native to Korea (자생 관상용 Heloniopsis와 Thalictrum속 종자의 형태적 및 형태생리적 휴면)

  • This study was conducted to find basic information for the research of morphological dormancy in seeds of two genera (Heloniopsis and Thalictrum) native to Korea. Seeds of Heloniopsis koreana, Heloniopsis tubiflora, Thalictrum rochenbrunianum, Thalictrum uchiyamai, and Thalictrum coreanum were collected, and embryo morphology and seed germination were investigated. All seeds of five species had underdeveloped embryos which occupied about 10-20% of the full seed length at maturity. The seeds of H. koreana and H. tubiflora had rudimentary embryos. The seeds of T. rochenbrunianum, T. uchiyamai, and T. coreanum had intermediate type between rudimentary and linear embryo. The small embryos of all seeds of five species grew to a critical length before radicles emerge from the seeds. At 30 days after sowing, the seeds of H. koreana and H. tubiflora germinated to 22.7% and 40.7%, respectively. On the other hand, the seeds of T. rochenbrunianum, T. uchiyamai, and T. coreanum germinated to 12.5%, 3.3%, and 0%, respectively. If embryo growth and germination in seeds are completed in approximately 30 days, the seed are said to have morphological dormancy (MD). However, if the seeds require > 30 days for germination, they are considered to have morphophysiological dormancy (MPD). Thus, the seeds of H. koreana and H. tubiflora had about 78% and 60% MPD, respectively. On the other hand, the seeds of T. rochenbrunianum, T. uchiyamai, amd T. coreanum had about 87%, 96%, and 100% MPD, respectively. There was a different level of dormancy (MD and MPD) within the same seed population examined. Therefore, germination of the seeds was delayed due to the underdeveloped embryos. This result could contribute to determine morphological seed dormancy and germination mechanism in seeds of the Korean Peninsula.

Ecophysiology of seed dormancy and germination in four Lonicera (Caprifoliaceae) species native to Korea

  • Background: To exploit the ornamental and medicinal purposes of Lonicera harae Makino, L. subsessilis Rehder, L. praeflorens Batalin, and L. insularis Nakai, native to Korea, it is necessary to understand their seed ecology for propagation. In this study, we investigated the seed dormancy type and germination characteristics of seeds of the four Korean native Lonicera species. Results: The seeds of the four Lonicera species imbibed water readily, suggesting that the species do not have physical dormancy. Furthermore, the seeds exhibited underdeveloped embryos with only about 15-25% of the length of the seeds at dispersal. The embryos grew to the critical length with approximately 50-80% of the length of the seeds' development before radicle protrusion. Further, 94.4% and 61.1% of freshly matured seeds of L. insularis and L. harae germinated within 4 weeks after sowing at 15 ℃ and 20 ℃, respectively. Contrarily, L. praeflorens and L. subsessilis seeds did not germinate within 4 weeks under all temperature treatments. At 15 ℃, L. praeflorens seeds started to germinate from 5 weeks and the final germination rate was 51.1% at 13 weeks. At 15 ℃, L. subsessilis seeds started to germinate from 5 weeks after sowing and the final germination rate was 85.6% at 17 weeks after sowing. Embryo growth and germination of L. praeflorens and L. subsessilis occurred at a relatively high temperature (≥ 15 ℃). Conclusions: Overall, L. insularis seeds have only morphological dormancy. The seeds of L. harae have approximately 60% and 40% of morphological dormancy and morphophysiological dormancy, respectively. Contrarily, L. praeflorens and L. subsessilis exhibited non-deep simple-type morphophysiological dormancy that requires relatively high temperature (≥ 15 ℃) for embryo growth and dormancy breaking. The optimum temperature for the germination of seeds of L. insularis, L. harae, L. praeflorens, and L. subsessilis was 15 ℃, 20 ℃, 15 ℃, and 20 ℃, respectively. There was interspecific variation in seed dormancy and germination patterns in the four Lonicera species. The difference in these characteristics within the four Lonicera species could be useful for understanding the seed ecophysiological mechanisms of Lonicera species.

  • This paper gives a description about the germination inhibition of American ginseng (Panax quinquefolium L.) seed coat. The existence of seed coat is one of the inhibitory factors which inhibit the embryo growth, particularly during the morphological after-ripening stage. The seed coat can obstruct the water absorption at the beginning of seed stratification, but it can not threaten seed germination. The inhibition of seed coat is not caused by the mechanical fetter neither. However, before splitting the seed coat, the inhibition of seed coat comes from both air-tight character and inhibitors, and after splitting the seed coat, the inhibition may come mainly from the inhibitors.

  • Palmitoyl CoA was found to inhibit corn embryo axis glucose-6-phosphate dehydrogenase and 6-phosphogluconate dehydrogenase, which were also inhibites by polyamines. However, reversal of inhibition of both enzymes by palmitoyl CoA was made by spermine. Activity of corn embryo axis protein kinase was found to increase steadily after germination. Activation and inhibition of protein kinase were made by MgCl$_2$and all polymines, respectively. Suc results suggest that fatty acid biosynthesis and lypolysis could be regulated to some extent by polyamines in corn embryo axis.

  • The experiment was conducted to study on the physiology of developing embryo and endosperm degradation during germination of Zea mays seeds treated with GA 20 and B-Nine 5000 ppm. Data obtained can be summerized as follows : 1. Swelling of seminal root increased the section of GA treatment more or less, on the other hand, the section of B-9 treatment inhibited slightly. 2. According to elapsing of seeding date, epithelial cell of Scutellum expansed in size, and the space of cell increased, that degree was enlarged as follows ; the section of GA, the section of Control, and the section of B-9. 3. According to the elapsing of seeding date, the formation of vascular organization embryo became clearer little by little, the lignification of vascular B-9 treatment section rather higher than the other section. 4. The degradation of Starch Grain is composed of near part of epithelial cell of Scutellum, the shape of degradation radiate from element of a disk shape, and the speed of degradation is the section of GA, the section of Control, the section of B-9 in turn.

Plant Regeneration from Embryogenic Suspension Cultures of Soybean (Glycine max L. Merrill)

  • In order to establish efficient plant regeneration from embryogenic suspension cultures of soybean, Glycine max L, we examined the effects of auxin type and concentration, cytokinin type and concentration, and amino acid type and concentration on the growth of embryogenic clumps from induced callus, and the effect of desiccation of mature somatic embryos obtained from these clumps on the frequency of somatic embryo germination. Embryogenic callus was induced from the edge of the cotyledons cultured on MS medium containing 6% sucrose, 40 mg/L 2,4-D, 0.2% gelrite and pH 5.7. The growth of embryogenic clumps was best in early staged, embryogenic callus that was placed in suspension culture of MS medium containing 5 mg/L 2,4-D and 0.5 mg/L asparagine. Single somatic embryos were isolated from the clumps and plated on the same medium for maturation. When the mature single somatic embryos were desiccated for 96 h, somatic embryo germination came up to approximately 90%. The plantlets germinated after embryos desiccation for 2 weeks were transfered to MS medium containing 3% sucrose,0.2% gelrite and pH 5.7.

The embryogenic competency and morphological changes during somatic embryogenesis in Iris pseudacorus

  • Embryogenic callus was obtained from bulb segments of Iris pseudacorus on Murashige and Skoog (MS) medium with 2,4-dichlorophenoxyacetic acid (2,4-D) alone or in combination with kinetin. When early globular somatic embryos were subcultured onto MS medium with $4.52{\mu}M$ 2,4-D, high frequency of somatic embryogenesis was obtained. Deprivation of 2,4-D was required for maturation. Mature somatic embryos had an elongated scutellum with a notch on the base of scutellum. Separation of embryos from embryo clusters was necessary to enhance the frequency of germination. Germination was stimulated by separation of embryos from embryo clusters and transfer onto fresh half-strength MS medium with 3% sucrose. After acclimation in artificial soil in greenhouse for 2 months, 96.4% of plantlets survived.

Effect of kinds and concentrations of osmoticum on somatic embryo induction and germination from suspended embryogenic cell in Larix kaempferi (낙엽송(Larix kaempferi) 현탁배양된 배발생세포로부터 체세포배 유도 및 발아를 위한 삼투압제 종류 및 농도 효과)

  • This study was conducted to examine suspended embryogenic cells growth with days of culture, effects of various kinds/concentrations of osmoticum for induction of somatic embryos (SEs), following somatic embryos germination or plantlet regeneration. The proliferation pattern of embryogenic cells in suspension culture is characterized by settled cells volume (SCV) increased with the duration of culture with marked the maxium of SCV (10.1 ml) in 18 days of culture, however the SCV of cells gradually decreased after that. In comparison of kinds/concentrations of osmoticum on somatic embryo induction, the highest induction number (352.3/g FW) of the SE was showed in 0.2 M sucrose, in addition, we also observed some effects with treatments of 0.2 M maltose (203.7) and 0.3 M maltose (193.7), respectively. However, no somatic embryos produced in treatments of 7.5% PEG plus 0.15 M sucrose or maltose. In comparison of germination efficiency of SEs which occurred from the treatments of various kinds/ concentrations of osmoticum, the highest induction frequency of cotyledon (25.2%) was obtained from SEs that produced 0.3 M maltose, however, the best occurrence rates of hypocotyl (39%), radicle (30.3%) and plantlet regeneration (3.5%) were observed from the 0.2 M sucrose treatment, respectively.
