  • This study was carried out to investigate the elucidation of gene expression in embryo formation of the Kidney mutant, especially yolk proteins in unfertilized eggs and template activity of m-RNA extracted from them. The results were summarized as follow ; There was no recognized qualitative difference in yolk proteins of unferilized eggs between the Kidney mutant and normal. There was not any difference, between Kidney mutant and normal in the molecular species of m-RNA derived from maternal origin with the template activity in vitro.

  • The mechanisms controlling the intensity and duration of synaptic transmission are numerous. Once an action potential reaches a nerve terminal, the stored neurotransmitters are released in a quantum fashion into the synaptic cleft. At that point neurotransmitters can act on post-synaptic receptors to elicit an action on the post-synaptic cell or net at so-called auto-receptors that are located on the presynaptic side and which often regulate the further release of the neutotransmitter. Whereas the action of the neurotransmitter receptors is regulated by desensitization phenomenon, the major mechanism by which the intensity and duration of neurotransmitter action is presumably regulated by either its degradation or its removal from the synaptic cleft. In the central nervous system, specialized proteins located in fe plasma membrane of presynaptic terminals function to rapidly remove neurotransmitters from the synaptic cleft in a sodium chloride-dependent fashion. These proteins have been referred to as uptake sites or neurotransmitter transporters. Once taken up by the plasma membrane transporters, neurotransmitters are repackaged into secretory vesicles by distinct transporters which depend on a proton gradient.

  • 포자의 전자현미경사진과 mycelium의 광학현미경사진을 찍어 strain V-8 균주의 형태학적인 특성을 조사하였을 때 방선균 계열인 streptomyces로 확인되었다. 이 균주의 배양액은 노란색을 띠고 있고 이 노란색은 430 nm에서 특징적인 흡광을 보였다. 이 물질은 유기 용매로 추출되었다. 이 물질의 자외선 흡광스펙트럼의 특징을 기존에 보고된 화합물과 비교하여 이 물질이 actinomycin계열에 속함을 발견하였다. 이 물질의 FAB mass spectrum에서 각 분획들의 주성분들의 분자의 질량 + H 이온이 1290과 1292에서 각각 관찰되었다. 이 화합물들은 분자량이 1258인 actinomycin D와는 상이한 구조를 소유하고 있는 것으로 확인되었다. 각 분획 성분들의 proton 및 carbon HIR spectra를 얻어 기존에 알려진 화합물의 자료와 비교하였을 때 actinomycin계열에 속하는 신물질로 생각되었다. 지금까지 actinomycin 계열의 항생물질이 adenosine deaminase의 효소 활성을 저해한다는 사실은 보고된바 없다. 본 연구자는 분리한 2개의 화합물에 대하여 구조 연구를 계속하고 있다.

Association of Anti-apoptotic Mechanism Due to House Dust Mite in Neutrophils with Protein Synthesis and Bad

  • House dust mite is an essential allergen in the pathogenesis of allergic diseases. Abnormal regulation of neutrophil apoptosis is an important pathogenic process in allergic diseases. In the present study, we investigated the effects of house dust mites on spontaneous apoptosis of neutrophils and its associated mechanisms. Extract of Dermatophagoides pteronissinus (DP) inhibited neutrophil apoptosis in a time-dependent manner. Cycloheximide (CHX), an inhibitor of translation, increased apoptosis of DP-treated neutrophils as well as control cells. The pro-apoptotic effect of CHX was blocked by DP in neutrophils. In addition, DP increased the phosphorylation of Bad in a time-dependent manner, indicating that it exerted an inhibitory effect on the function of Bad. These results suggest that DP has anti-apoptotic effects of neutrophils and may regulate protein synthesis and activation of Bad. Moreover, these findings may shed light on elucidation of allergy pathogenesis due to house dust mites.

  • Light exerts two primary roles in plant growth and development. Plants acquire all biochemical energy required for growth and propagation solely from light energy via photosynthesis. In addition, light serves as a medium through which plants recognize environmental fluctuations, such as photoperiod and presence of neighboring animals and plants. Plants therefore constantly monitor the direction, intensity, duration, and wavelength of environmental light and integrate these light signals into the intrinsic regulatory programs to achieve an optimized growth in a given light condition. Although light regulates all aspects of plant growth and developmental aspects, the molecular mechanisms and signaling cascades involved have not been well established until recently. However, recent advances in genetic tools and plant transformation techniques greatly facilitated the elucidation of molecular events in plant photomorphogenesis. This mini-review summarizes the gist of recent findings in deetiolation and suppression of shade avoidance response as classic examples of the phytochrome-mediated photomorphogenesis.

  • A new ardimerin (1) was isolated from the herb of Ardisia japonica. Its structure was determined by the elucidation of spectroscopic data and a chemical transformation. Compound (1) showed potent radical scavenging effect on DPPH radical $(IC_{50},\;0.32\;{\m}M)$.

  • A new prenylated pyranoxanthone, inophyllin B (1), was isolated from the roots of Calophyllum inophyllum (Guttiferae). Together with this compound was also isolated the known pyranoxanthone brasilixanthone B (2) and two common triterpenes friedelin (3) and sitosterol (4). Structural elucidations of these compounds were achieved through $^1H,\;^{13}C$, DEPT, COSY, HSQC and HMBC experiments. The molecular mass was determined using MS techniques. The crude extract indicated weak toxicity to the larvae of Aedes aegypti. We report here the isolation, structural elucidation and bioassay data for Inophyllin B and brasilixanthone B.

  • Several flavonoid dimers were isolated and elucidated from the bark of Korean pine tree (Pinus densiflora). One of them was postulated to be a compound, epicatechin $(4{\rightarrow}7)$ 5,8,3',4'-tetrahydroxy-(2R,3R)-flavan-3-ol, whose structural determination was carried out by diverse NMR techniques.

  • The Anthocyanins in the Korean native Brassica juncea, which were favorably used directly for food or as condiments, were investigated. Two kinds of anthocyanin were separated from Korean native Brassica juncea by T.L chromatography and then Robinson test, spectrophotometry, T.L. chromatography, partial hydrolysis were tested for the structural elucidation of these two anthocyanins. It was known that two pigments were peonidin-3-glucoside and peonidin-3-galactoside.

  • An ${\alpha}-{\text\tiny{D}}-glucoside$ of sesterterpene, 11-epiterpestacin, was isolated from the culture of a filamentous fungus Bipolaris sorokiniana NSDR-011. The structure was elucidated by chemical studies and spectroscopic methods including NMR and ESI-MS. 11-ETG (1) named arbitrarily did not inhibit the root growth of Italian ryegrass seedlings even at the level of 200 ppm, while its aglycone 11-ET (2) completely inhibited root growth at level of 100 ppm.
