Improvement of Mechanical Properties of Mg alloys through Control of Grain Size and Texture (결정립크기와 집합조직제어를 통한 마그네슘 합금의 기계적 성질 개선)

  • The effects of lowering ECAP temperature during ECAP process and Post-ECAP annealing on microstructure, texture and mechanical properties of the AZ31 alloys have been investigated in the present study. The as-extruded materials were ECAP processed to 2 passes at 553K prior to subsequent pressing up to 6 passes at 523K or 493K. When this method of lowering ECAP temperature during ECAP was used, the rods could be successfully deformed up to 6 passes without any surface cracking. Grain refinement during ECAP process at 553K might have helped the material to endure further straining at lower deformation temperatures probably by increasing the strain accommodation effect by grain boundary sliding, causing stress relaxation. Texture modification during ECAP has a great influence on the strength of Mg alloys because HCP metals have limited number of slip systems. As slip is most prone to take place on basal planes in Mg at room temperature, the rotation of high fraction of basal planes to the directions favorable for slip as in ECAP decreases the yield stress appreciably. The strength of AZ31 Mg alloys increases with decrease of grain size if the texture is constant though ECAP deformation history is different. A standard positive strength dependence on the grain size for Mg alloys with the similar texture (Fig. 1) supports that the softening of ECAPed Mg alloys (a negative slope) typically observed despite the significant grain refinement is due to the texture modification where the rotation of basal planes occurs towards the orientation for easier slip. It could be predicted that if the original fiber texture is restored after ECAP treatment yielding marked grain refinement, yield stress as high as 500 MPa will be obtained at the grain size of ${\sim}1{\mu}m$. Differential speed rolling (DSR) with a high speed ratio between the upper and lower rolls was applied to alter the microstructure and texture of the AZ31 sheets. Significant grain refinement took place during the rolling owing to introduction of large shear deformation. Grain size as small as $1.4{\mu}m$ could be obtained at 423K after DSR. There was a good correlation between the (0002) pole intensity and tensile elongation. This result indicates that tensile ductility improvement in the asymmetrically rolled AZ31 Mg alloys is closely related to the weakening of basal texture during DSR. Further basal texture weakening occurred during annealing after DSR. According to Hall-Petch relation shown in Fig. 1, the strength of the asymmetrically rolled AZ31 is lower than that of the symmetrically rolled one when compared at the same grain size. This result was attributed to weakening of fiber texture during DSR. The DSRed AZ31, however, shows higher strength than the ECAPed AZ31 where texture has been completely replaced by a new texture associated with high Schmid factors.

Genetic Diversity, Pathogenicity, and Fungicide Response of Fusarium oxysporum f. sp. fragariae Isolated from Strawberry Plants in Korea (국내 딸기 시들음병균 Fusarium oxysporum f. sp. fragariae의 유전적 다양성, 병원성과 살균제 반응)

  • Fusarium wilt caused by Fusarium oxysporum f. sp. fragariae (Fof) is the most important diseases of a strawberry field in Korea. We surveyed phylogenetic analysis, pathogenicity test, and fungicide response about Fof isolates isolated from Korea. Twenty-seven isolates of F. oxysporum isolated from strawberry plants were conducted in this study. Specific amplification by Fof specific primer was confirmed in all 26 isolates except Fo080701 isolate. The nuclear ribosomal intergenic spacer region and the translation elongation factor EF-lα gene sequences of isolates revealed three main lineages. Most of all isolates were contained DNA lineage group 1, but 2 and 3 group was shown only one and three isolates, respectively. All isolates were shown in pathogenicity with cv. Seolhyang. The EC50 mean values of prochloraz ranged 0.02-0.1 ㎍/ml except for Fo080701 and effectively inhibited mycelial growth at low concentrations. The EC50 value of metconazole was also 0.04-0.22 ㎍/ml, showing a similar inhibitory effect to that of prochloraz. The EC50 value of pyraclostrobin was 0.23-168.01 ㎍/ml, which was different according to the strain. In the field trial, boscalid+fludioxonil, fluxapyroxad+pyraclostrobin, and prochloraz manganese were selected as the effective fungicides for controlling Fusarium wilt.

  • High strength and fracture toughness of Al-Li-Cu alloy(2090 Al alloy) have been achieved by the improvement of melting and casting, extrusion and heat treatment techniques. To establish the sucessful process for semi-industrial scale ingot(20Kg) the following areas have been investigated: (1) Improvement of melting and casting techniques for ingot by introducing atmospheric modifications, vacuum and rotary degassing, and deslagging. (2) The effect of heat treatment on mechanical properties (3) Mechanical characterization by tensile test, fracture toughness test and fatigue crack propagation test. High mechanical properties were found to be intimately related with ingot soundness. Tensile strength of final products varied from 534MPa to 566MPa in peak aged condition while elongation/ductility ranged from 9.0% to 11.9%. From the fracture toughness test with using compact tensile specimen, plane strain fracture toughness($K_{Ic}$) appeared to be 39MPa${\surd}$m in peak aged condition and 23MPa${\surd}$ m in underaged condition. When load ratios of 0.1, 0.3 and 0.5 were given ${\Delta}K_{th}$ was 6.0MPa${\surd}$ m, 5.3MPa${\surd}$ m and 4.3MPa${\surd}$ m respectively.

Efficient Transformation Method of Soybean Using Meristematic Tissues of Germinating Seeds (발아종자의 분열조직을 이용한 효율적인 콩 형질전환 방법)

  • An efficient transformation method for soybean [Glycine max (L.) Merr.] using meristematic tissues of germinating seeds has been established. The embryonic axes were excised from germinating seeds of Korean soybean cultivar, Iksannamulkong and 0.5-2 cm long segment containing meristematic tissues were prepared by cutting hypocotyl region. The explants were inoculated with Agrobacterium tumefaciens strain LBA4404 harboring a binary vector with the bar gene as a selectable marker gene and a ${\beta}-glucuronidase$ (GUSINT) reporter gene, and then co-cultured for 7 days on co-cultivation medium (CCM). The meristematic tissues were cultured on shoot induction medium (SIMP6) supplemented with 0.4 mg/l $N_6-benzylaminopurine$ (BAP) and 0.1 mg/l indolebutyric acid (IBA) in the presence of 6 mg/l L-phosphinotricin (PPT) for 2 weeks and the surviving explants were transferred to shoot elongation medium (SEMP6). Transformation was confirmed by Southern blot analysis and the transformation efficiencies ranged from 1.48 to 2.07%. The new modified transformation method was successfully implemented for obtaining several transgenic lines with SMV-CP gene. It is expected that this method could efficiently be used for the transformation of recalcitrant soybean cultivars.

Effect of vacuum annealing and characterization of diecast ADC12 aluminum alloys (다이캐스팅 공정으로 제조한 ADC12 알루미늄 합금의 물성 향상 및 진공 열처리 효과)

  • We report structural, mechanical, and thermal properties of diecast ADC12 aluminum alloys characterized using synchrotron X-ray diffraction (XRD), scanning electron microscopy, energy dispersive X-ray (EDX) analysis, thermal conductivity (λ), Vickers hardness (Hv), and stress-strain measurements. We also studied the effect of post-annealing performed in a vacuum atmosphere on the mechanical properties of diecast ADC12 alloys. EDX and XRD results revealed that Al2Cu and AlCu3 grains are formed, well dispersed in Al base and highly crystalline. Ultimate tensile strength (UTS) of 307.9 ± 9.1 MPa and elongation of 2.98 ± 0.62 % were estimated. λ was 129.3 ± 0.27 W/m·K and Hv was approximately 130. Both values were significantly higher than the reported values. At annealing temperatures ranging from 25 to 200℃, UTS and Hv values remained constant, while as the annealing temperature increased to 500℃, these values gradually decreased. This is because stabilization of the microstructure improves toughness and ductility.

  • In this study, characteristics of effect on hydrogen gas was investigated to hydrogen embrittlement by disk and tensile tests. The developed and commercial alloy was fabricated to a plate material made from an alloy ingot. The prepared materials were processed in the form of a disk to measure rupture pressure by hydrogen and helium gas at a rate of 0.1 to 1,000 bar/min. In the hydrogen pre-charged tensile test, a specimen was hydrogenated using an anode charging method, and the yield strength, ultimate tensile strength, elongation, and reduction in area rate were carried by a strain rate test. Also, the microstructure was observed to the fracture surface of the tensile test specimen. As a result, the developed materials satisfied endurable hydrogen embrittlement, and the fractured surface showed a brittleness fracture surface with a depth of several ㎛, but dimple due to ductile fracture could be observed.

Study on the optimization of additive manufacturing process parameters to fabricate high density STS316L alloy and its tensile properties (고밀도 STS316L 합금 적층 성형체의 제조공정 최적화 및 인장 특성 연구)

  • To optimize the process parameters of laser powder bed fusion process to fabricate the high density STS316L alloy, the effect of laser power, scanning speed and hatching distance on the relative density was studied. Tensile properties of additively manufactured STS316L alloy using optimized parameters was also evaluated according to the build direction. As a result of additive manufacturing process under the energy density of 55.6 J/mm3, 83.3 J/mm3 and 111.1 J/mm3, high density STS316L specimens was suitably fabricated when the energy density, power and scan speed were 83.3 J/mm3, 225 W and 1000 mm/s, respectively. The yield strength, ultimate tensile strength, and elongation of STS316L specimens in direction perpendicular to the build direction, show the most competitive values. Anisotropic shape of the pores and the lack of fusion defects probably caused strain localization which result in deterioration of tensile properties.

Stability of Human Centromeric Alphoid DNA Repeat during Propagation in Recombination-Deficient Yeast Strains (효모의 재조합 변이주를 이용한 인간 Centromeric Alphoid DNA Repeat의 안정성에 관한 연구)

  • The centromere is a highly differentiated structure of the chromosome that fulfills a multitude of essential mitotic and meiotic functions. Alphoid DNA (${\alpha}$-satellite) is the most abundant family of repeated DNA found at the centromere of all human chromosomes, and chromosomes of primates in general. The most important parts in the development of Human Artificial Chromosomes (HACs), are the isolation and maintenance of stability of centromeric region. For isolation of this region, we could use the targeting hook with alphoid DNA repeat and cloned by Transformation-Associated Recombination (TAR) cloning technique in yeast Saccharomyces cerevisiae. The method includes rolling-circle amplification (RCA) of repeats in vitro to 5 kb-length and elongation of the RCA products by homologous recombination in yeast. Four types of $35\;kb{\sim}50\;kb$ of centromeric DNA repeat arrays (2, 4, 5, 6 mer) are used to examine the stability of repeats in homologous recombination mutant strains (rad51, rad52, and rad54). Following the transformation into wild type, rad51 and rad54 mutant strains, there were frequent changes in inserted size. A rad52 mutant strain showed extremely low transformation frequency, but increased stability of centromeric DNA repeat arrays at least 3 times higher than other strains. Based on these results, the incidence of large mutations could be reduced using a rad52 mutant strain in maintenance of centromeric DNA repeat arrays. This genetic method may use more general application in the maintenance of tandem repeats in construction of HAC.

Plastic Behaviro of Two Phase Intermetallic Compounds Based on $Li_2$-type$(Ai, Cr)_3$/Ti ($Li_2$$(Ai, Cr)_3$/Ti기 2상 금속간화합물의 소성거동)

  • Plastic behavior of two-phase intermetallic compounds based on $LI_{2}$-type $(Al, Cr)_3$ Ti was investigated using compression test at R.T. and 77K. $LI_{2}$ single phase alloys and two-phase alloys consisting of mainly $LI_{2}$ phase and a few or 20% second phases were selected from AI-Ti-Cr phase diagram. In general, compared with Llz single phase, two-phase alloys consisting of 20% second phase showed relatively high yield strength and poor ductility. Among the alloys, however, AI-21Ti-23Cr alloy consisting of 20% $Cr_{2}Al$ phase showed available ductility as well as high yield strength. Plastic behavior of $LI_{2}$ single phase alloys and two-phase alloys consisting of a few% $Cr_{2}Al$ was also investigated. Homogenization of arc melted ingots substantially reduced the amount of second phases but introduced extensive pore. When Cr content increased in $Ll_{2}$ single phase alloys after the homogenization, the volume fraction of pore in the alloys decreased, and no residual pore was observed in two-phase alloys consisting of a few% $Cr_{2}Al$ phase. Environmental effect on the ductility of the alloys was investigated using compression test at different strain rates($1.2 \times 10^{-4}/s$ and $1.2 \times 10^{-2}/s$). Environmental embrittlement was least significant in A1-25Ti-10Cr alloy consisting of LIZ single phase among the alloys tested in this study. However, based on the combined estimation of the pore formation, environmental embrittlement and ingot cast structure, AI-21Ti-23Cr alloy consisting of 20% $Cr_{2}Al$ as the second phase is expected to show the best tensile elongation behavior.
